한국 한방화장품 발달 배경 및 연구 현황 (The Background and Current Research Applied to Development of Korean Cosmetics Based on Traditional Asian Medicine)

  • Traditional Asian medicine has an extensive evidence base built upon thousands of years of experience within Asia, of curing various diseases. Only recently, within the past two centuries, have modern medical scientists developed interest in traditional Asian medicine. Asian Medicine seems to be regarded only as an adjunctive medicine and viewed as alargely un-proven alternative medicine to complement western medicine, used in some cases to establish a new paradigm of "integrative medicine". This article reviews how Korean herbal cosmetics emerged by applying traditional Asian medicine to the science of cosmetics. The characteristics of Korean herbal cosmetics are examined through examples of history, concepts and traditions. With the advancements in biotechnology, studies are now being conducted on the dermatological effects and processing methods of herbal ingredients, including ginseng. The authors explain the current research on the identification on the active ingredients of herbs, extraction methods, and bio-processing of ingredients to improve the biological efficacies of herbs on the skin. A summary of studies focused on modern reinterpretations of ageing theories, such as 'Seven year aging cycle', are provided. In conclusion, the development of Korean cosmetics products are based on the accumulated knowledge of thousands of years of experience including; 1) practical heritage of traditional Asian medicines such as Donguibogam; 2) excellent medicinal plants, such as ginseng, which are native to Korea; and 3) innovative attempts to modernize materials, processes, and principles.

노간주나무(Juniperus rigida Sieb.)의 주름개선 효과 (Anti-wrinkling Effects of Juniperus rigida Sied)

  • 사람의 피부는 자외선, 오염된 공기, 화학제품 등 환경에 끊임없이 노출된다. 특히 자외선은 피부에 다양한 형태로 영향을 주어 주름, 잔주름, 피부거침, 피부건조와 같은 현상을 발생시켜 피부노화를 유발시킨다. 본 연구에서는 전 세계적으로 향나무과의 일종인 다년생 수목인 노간주나무(Juniperus rigida Sieb.) 추출물의 주름개선 효과에 대하여 검증하였다. 주름개선 효과 측정으로 elastase 저해활성을 측정한 결과 노간주나무 열수 추출물 $1,000{\mu}g/ml$에서 10% 이하의 낮은 저해활성을 나타낸 반면에 노간주나무 에탄올 추출물의 $1,000{\mu}g/ml$에서 68.5%의 저해활성을 나타내었으며, collagenase 저해활성을 측정한 결과 $1,000{\mu}g/ml$에서 열수 및 에탄올 추출물은 44.9%, 97.2%로 에탄올 추출물의 경우 ascorbic acid (99.6%)와 유사한 효과로 높은 저해활성을 나타내었다. 또한 섬유아 세포를 이용한 collagen 생합성량을 측정한 결과 노간주나무 에탄올 추출물 $50{\mu}g/ml$에서 151.52%의 생합성 촉진 효과를 나타내어 노간주나무 에탄올 추출물의 주름개선 효과가 우수하다는 것을 확인할 수 있었다. UVA에 의해 발현이 증가되는 MMP-1에 노간주나무 에탄올 추출물이 미치는 영향을 알아본 결과 노간주나무 에탄올 추출물은 $1,00{\mu}g/ml$에서 67.1%의 MMP-1 발현 저해효과를 나타내었다. 노간주나무 에탄올 추출물의 MMP-1의 단백질 및 mRNA의 발현은 $50{\mu}g/ml$의 농도에서 각각 35%, 39%의 저해율을 나타냄에 따라 노간주나무 에탄올 추출물은 collagen을 분해하는 효소인 MMP-1의 발현을 억제하고 광노화에 의한 주름생성억제 활성을 보이는 것을 확인하였다.

β-glucosidase 활성 균주 발효에 의한 황기 Isoflavonoid 성분변화 및 생리활성 (The Isoflavonoid Constituents and Biological Active of Astragalus Radix by Fermentation of β-glucosidase Strains)

  • Background: In this study, the radix of Astragalus membranaceus Bunge extract fermented by Saccharomyces cerevisiae, Weissella cibaria, and Pediococcus pentosaceus to increase the levels of isoflavonoid aglycone contents. Methods and Results: In order to change the in isoflavonoids, we fermented the radix of A. membranaceus extracts with microorganisms that have β-glucosidase activity. Besed on the β-glucosidase activity, we selected three strains, Weissella cibaria, Pediococcus pentosaceus, and Saccharomyces cerevisiae. HPLC analysis revealed that the levels of isoflavonoid aglycones were increased in all fermentation cases, and the extracts fermented by S. cerevisiae showed the highest levels of isoflavonoid aglycones. We evaluated the antioxidant activity, anti-wrinkle effects and whitening effects of the S. cerevisiae-fermented extracts using the DPPH assay, tyrosinase inhibition activity assay, and collagenase inhibition activity assay. We confirmed higher activity in S. cerevisiae-fermented extracts than in control, with the half maximal inhibitory concentration (IC50) value of 565.1 ± 59.1 ㎍/㎖ in DPPH radical scavenging activity, tyrosinase inhibition rate of 78.4 ± 0.9%, and collagenase inhibition rate of 83.8 ± 1.1%. Conclusions: We selected three stains of microorganisms showing high β-glucosidase activity, W. cibaria, P. pentosaceus and S. cerevisiae. Isoflavonoid glycones in the radix of A. membranaceus were converted to isoflavonoid aglycones by fermentation. In addition, the fermented radix of A. membranaceus exhibited antioxidant activity, anti-wrinkle effect, whitening effect and radical scavenging activity.

선학초 추출물의 미백활성 (Whitening Activities of the Agrimonia pilosa L. Extracts)

  • 선학초 추출물을 기능성 화장품 소재로 활용하기 위하여 미백효과를 검증하였다. 세포 내 tyrosinase 저해활성 측정결과 선학초 에탄올 추출물 $500{\mu}g/mL$에서 42%의 저해활성을 나타내었다. 이는 선학초 추출물이 세포 내 tyrosinase 발현을 억제시킴으로서 멜라닌 합성 또한 저해 하는 것임을 확인 할 수 있었다. 선학초 추출물의 단백질 발현과 mRNA 발현 억제효과를 검토한 결과 선학초 열수 및 에탄올 추출물을 처리한 B16F10군에서는 tyrosinase protein의 발현이 처리하지 않은 군보다 감소함을 확인할 수 있었다. 결과적으로 선학초 추출물의 미백효능을 확인할 수 있었으며, 식품 및 화장품의 기능성 소재로 이용이 가능할 것으로 판단된다.

산수유 헥산 추출물의 항균효과 및 세포독성 (Screening of Cytotoxicity and Antimicrobial Effects of Hexane Extracts from Cornis fructus)

  • Cornis fructus was extracted by successive extraction and then fractionated with hexane extract to get active fractions. This study was performed to determine the cytotoxic effect of hexane extract from Cornis fructus on NIH 3T3 fibroblasts and cancer cell lines using MTT assay. Hexane extract showed cytotoxic effect against A549, B16 melanoma and MDA-MB-231. Futher fractionation with hexane extract was performed to obtain effective fraction, fraction 3 showed the cytotoxic effect against A549 and MDA-MB-231. In antimicrobial test of each fraction of hexane extract, fraction 5 showed antimicrobial activities against P. putida and P. aeruginosa.

  • The effects of the methanol subfraction from Pueraria Radix on hydrogen peroxide-induced lipid peroxidation and cadmium-induced cytotoxicity were investigated in NIH 3T3 fibroblast cells. After the methanol subfraction treatment, the content of MDA induced by 600 ${\mu}g$ $H_2O_2$ significantly decreased in proportion to the subfraction concentrations as well as 50 ${\mu}M$ $CdCl_2$-induced cytotoxicity. Especially, 200 ${\mu}g/mL$ concentration of methanol subfraction was strongly shown inhibition of lipid peroxidation and detoxification of cadmium. These results suggest that the methanol subfraction from Pueraria Radix retains a potential antioxidant and protective effect against cadmium.

  • "Eating cosmetic" market comes to prominence worldwide. Inner Beauty has established itself as the top trend nowadays and is leading the health functional food market. Another noticeable trend other than beauty-from-within is that products using "plant-based" natural botanical raw material are increasing. Based on these trends above, we have developed a plant extract named Agatri® for skin health using Agastache rugosa. By ingesting Agatri®, it is expected that the formation of skin wrinkles is suppressed, and skin elasticity is increased by suppressing the decrease in collagen content caused by ultraviolet rays. At the same time, it is possible to prevent skin aging by improving skin hydration and trans-epidermal water loss (TEWL). The studies show that it significantly increased production of hyaluronic acid and collagen, and decreased production of matrix metalloproteinases (MMPs). And Agatri® has been approved as an "Individually recognized functional ingredient for skin health by the Ministry of Food and Drug Safety, Korea in 2020. As a result, Agatri® is a good solution for those who are in need of skin protection from exposure to UV and aging.

섬쑥부쟁이(Aster glehni Fr. Schm.) 분획물의 미백 및 주름 개선 효과 (The Effect of Aster glehni Fr. Schm. Extracts on Whitening and Anti-Wrinkle)

  • 천연의 식물과 추출물은 비용의 효율성과 풍부한 자원을 이점으로 식품, 약, 의약 분야와 마찬가지로 화장품 분야에서도 이용될 수 있다. 현재까지 섬쑥부쟁이는 항산화 활성을 제외하고는 그 효능에 대해 알려진 바가 없다. 따라서 본 연구에서는 울릉도의 대표적인 특산 식물인 섬쑥부쟁이 추출물을 이용하여 화장품 분야에서의 활용방안에 대한 연구를 조사한바, 효소적 in vitro 검색법을 측정하여 이들 중 가장 우수한 EtOAc 추출물을 대상으로 이러한 저해능이 멜라닌 생성에 관련된 단백질 발현과도 연관성이 있는지를 확인하기 위해 B16F10 melanoma cell line 내 단백질 수준을 확인하였다. Tyrosinase, TRP-1의 항체를 이용한 western blot으로 관련 단백질의 발현량을 조사한 결과 섬쑥부쟁이 에틸아세테이트 분획물 100 ug/ml을 처리한 군에서는 tyrosinase protein을 30.5%를 억제하였고, TRP-1은 41.5% 억제하였다. 한편, 섬쑥부쟁이 분획물의 생육 저해환 측정에서는 Staphylococcus epidermidis, Propionibacterium acnes, Escherichia coli에 대하여 헥산 분획물 2 mg/disc에서 각각 21.0 mm, 12.0 mm, 14.0 mm의 저해환을 나타내어 섬쑥부쟁이 헥산 분획물만이 유일하게 높은 항균활성을 나타났다. 이들 실험 결과를 미루어보아, 섬쑥부쟁이의 화장품 분야에서의 활용적 가치가 충분하다고 사료되나, 그 활성 성분에 대한 명확한 규명을 위한 추가적인 연구를 제시하는 바이다.

초임계 유체로 추출된 산삼 부정 배양근 오일의 효모균 처리에 의한 비사포닌계 지방산 함량 변화 및 Collagenase 및 Elastase 저해 활성 증대 (Changes in Non-saponin Fatty Acid Content and Increases in Inhibitory Activities of Collagenase and Elastase by Treatment with Saccharomyces cerevisiae of the Supercritical Fluid Extracted Oil of the Adventitious Roots Culture of Wild Mountain Ginseng)

  • Background: To obtain useful cosmetic resources, this study aimed to determine the non-saponin fatty acid and inhibitory activities of collagenase and elastase by treatment of Saccharomyces cerevisiae in supercritical fluid extracted oil of the adventitious root culture of wild mountain ginseng. Methods and Results: We performed supercritical fluid extraction at various conditions such as pressure, temperature, time, and use of co-solvents, unlike the n-hexane extraction for the adventitious roots culture of wild mountain ginseng. The non-saponin-fatty acid obtained from the oil of the adventitious roots culture was incresed by treatment with S. cerevisiae. The supercritical fluid extraction was conducted using gas chromatography. Non-saponin-fatty acid content, in the oil of adventitious roots culture of wild mountain ginseng treated with S. cerevisiae for 2 days were three times higher than that in the control. In addition, the oil of the adventitious roots culture treated with S. cerevisiae was investigated for the anti-wrinkle effect by using collagenase and elastase. The oil of adventitious roots culture treated with S. cerevisiae exhibited higher collagenase and elastase inhibitory activities than those in the control. Conclusions: Supercritical fluid extracted oil of the adventitious roots culture of wild mountain ginseng treated with S. cerevisiae was found to have decreased ratio of saturated fatty acids and incresed ratio and content of unsaturated fatty acids increased. Furthermore, it showed anti-wrinkle effects in vitro.

Antioxidative Activity and Irritation Response of Lespedeza bicolor

  • In the present study, we evaluated the free radical scavenging and xanthine oxidase inhibitory activities exhibited by extracts obtained from the dried stems (and leaves) of Lespedeza bicolor We also assessed its potential irritation activities with regard to cosmetic use. When the DPPH radical scavenging activities of L. bicolor were assessed at six different concentrations (0, 50, 100, 250, 500 and 1000 ${\mu}g/ml$), the concentration of L. bicolor required to inhibit DPPH radical formation by $50\%$ was found to be $164.90{\mu}g/ml$. The effects of L. bicolor on the inhibition of xanthine oxidase were determined at seven different concentrations. The $50\%$ effective concentration was found to be $282.75{\mu}g/ml$. In the skin irritation test, all animals survived for the duration of the study, and all exhibited normal gains in body weight. The control sites exhibited no response to the control procedures. No edema, erythema, or eschar formation was observed in any of the tested rabbits. In the ocular irritation study, all of the rabbit eyes remained normal. In summary, L. bicolor extracts were considered to be non-irritating to the skin and eye.
