Aim & Scope
The Food Science and Biotechnology (Food Sci. Biotechnol.; FSB) was launched in 1992 as the Food Biotechnology and changed to the present name in 1998. It is an international peer-reviewed journal published monthly by the Korean Society of Food Science and Technology (KoSFoST). The FSB journal covers; Food chemistry/food component analysis Food microbiology and biotechnology Food processing and engineering Food hygiene and toxicology Biological activity and nutrition in foods Sensory and consumer science s Consumer perception and sensory evaluation on processed foods are accepted only when they are relevant to the laboratory research work. As a general rule, manuscripts dealing with analysis and efficacy of extracts from natural resources prior to the processing or without any related food processing may not be considered within the scope of the journal. The FSB journal does not deal with only local interest and a lack of significant scientific merit. The main scope of our journal is seeking for human health and wellness through constructive works and new findings in food science and biotechnology field.

Bacillus thuringiensis was isolated as a pathogen of the sotto disease of silkmoth larvae about a hundred years ago. Since then, this bacterium has attracted attentions of not only insect pathologists but also many other scientists who are interested in its strong and specific insecticidal activity. This has led to the recent worldwide development of B. thuringiensis-based microbial insecticides and insect-resistant transgenic plants, as well as a landmark discovery of par asp orin, a cancer cell-specific cytotoxin produced by B. thuringiensis. In this review, we describe examination of interaction between inclusion proteins of B. thuringiensis and brush border membrane of insects using a surface plasmon resonance-based biosensor, identification and characterization of parasporin-4, the latest parasporin produced by the B. thuringiensis A1470 strain, and an effective method for preparing the parasporin-4 from inclusion bodies expressed in the recombinant Escherichia coli cells.

Considerable progress has been made in functionalization of the soy isoflavones through enzymatic modification of daidzin, genistin, and glycitin. After hydrolysis of
$\beta$ -glucosides into their corresponding aglycones, these compounds were structurally modified via biotransformations such as regioselective hydroxylation, enantioselective reduction, regioselective methylation, and polymerization. These reactions often resulted in an increase of the biological activities (e.g., anti oxidative activity, antiproliferative activity) and/or improvement of the physico-chemcial properties (e.g., water solubility, bioavailability). This review briefly summarizes on-going research activities on the biofunctionalization of the soy isoflavones. -
The optimal conditions for the production of branched dextrin from com starch (CSBD) using branching enzyme (BE) were established by investigating the degree of retrogradation of the gelatinized starch. The physicochemical properties of CSBD prepared using the established process were evaluated. It was found that physicochemical properties of com starch were greatly modified by BE treatment. CSBD had a higher dextrose-equivalent value and water solubility than the corresponding control. On the other hand, the viscosities in gelatinized solution and amylose contents of CSBD were lower than those of the control. A high-performance size-exclusion chromatography/multiangle laser light scattering/retractive index (HPSEC/MALLS/RI) system showed that the average molecular weight of CSBD was lower than that of the control. The pasting viscosities of CSBD were stable during the entire temperature cycle. In general, the BE treatment resulted in the retrogradation during storage being lower for CSBD than for the control.

A nutritional supplement was developed aiming at correcting the most common nutrient and caloric deficiencies encountered in elderly people (
${\geq}60$ years old). The protein source was a mixture of whey protein isolates (WPI) and bovine collagen hydrolysate (BCH) with high nutritional and functional qualities making up 12% of the formulation. The carbohydrate fraction was composed of sucrose, inulin (soluble fiber), and fructo-oligosaccharide (prebiotic). The most commonly deficient essential minerals and vitamins were also included. Acceptance of the product was good according to both an elderly panel and a laboratory panel composing of both sexes and various ages. The stability of the formulations was evaluated and the estimated shelf life at room temperature (ca.$27^{\circ}C$ ) was approximately 4 months. -

The$CHCl_3$ , EtOAc, and n-BuOH fractions showed a marked inhibition of 5% fatal bovine serum (FBS)-induced cell proliferation. The$IC_{50}$ values of the chloroform fractions from leaf, stem, and root as well as the n-BuOH and EtOAc fraction from root on cell proliferation were$1.2{\pm}0.4$ ,$17.2{\pm}6.4$ ,$81.8{\pm}33.2$ ,$40.8{\pm}8.0$ , and$237.1{\pm}85.6\;{\mu}g/mL$ , respectively. On the other hand, the EtOAc fractions, and the$CHCl_3$ fraction significantly inhibited collagen-, arachidonic acid-, U46619-, and thrombin-induced platelet aggregations. The$IC_{50}$ values of EtOAc fraction from leaf, and the$CHCl_3$ and EtOAc fraction from stem were$214.1{\pm}12.2$ ,$134.3{\pm}2.5$ , and$42.6{\pm}7.0\;{\mu}g/mL$ with collagen,$312.4{\pm}7.5$ ,$158.9{\pm}1.7$ , and$82.2{\pm}2.7\;{\mu}g/mL$ with arachidonic acid,$31.1{\pm}2.4$ ,$48.7{\pm}0.3$ , and$29.7{\pm}1.1\;{\mu}g/mL$ with U46619, and$36.7{\pm}2.4$ ,$69.1{\pm}11.3$ , and$34.2{\pm}0.1\;{\mu}g/mL$ with thrombin, respectively. Taken together, these data provide new evidence that fractions from Allium victorialis var. platyphyllum (AVP) are able to inhibit vascular smooth muscle cell (VSMC) proliferation and platelet aggregation, which may be a novel resource for the development of anti-atherothrombotic agents. -
Changes in antioxidant activity of Rubus coreanus fruit of 3 clones (S13, S114, and S16), which were selected from different sites, were studied at different ripening stages. Antioxidant activities (tree radical scavenging activity and reducing power) were determined and their relationships to total phenolic contents and ascorbic acid were analyzed. The highest tree radical scavenging activities of 3 clones (S13, S14, and S16) were 79.39, 75.80, and 81.16% at
$125\;{\mu}g/mL$ , respectively. In general, the antioxidant activity and the related parameters, including total phenolic content and vitamin C content decreased during fruit ripening. Total phenolic contents of the R. coreanus fruits (S13, S14, and S16) were correlated with tree radical scavenging activity ($R^2=0.8114$ , 0.9186, and 0.9714). These results improve knowledge of the effect of ripening on the antioxidant activity and related compounds contents that could help to establish the optimum R. coreanus fruit harvest data for various usages. -
Previous studies have suggested a possible correlation between cell surface hydrophobicity (CSH) and stress tolerance in Bifidobacterium. In this study, the relationship was examined between CSH and environmental stress tolerance in Lactobacillus spp. By measuring the adhesion to hexadecane, 2 Lactobacillus fermentum strains- KLB 261 and KLB 231 were found to have high and low CSH, respectively. To measure their tolerance to various stresses, cells were subjected to salt (2 M NaCl), acid (pH 2),
$H_2O_2$ (0.01 %, v/v), ethanol (20%, v/v), heat ($60^{\circ}C$ ), and cold ($-20^{\circ}C$ ). Compared with KLB 231, the hydrophobic KLB 261 was found to be much more resistant to the various stresses examined. After being subjected to different stresses for a period of time, KLB 261 and KLB 231 showed 50 and 0% survivability in 2 M NaCl, 108.2 and 0.6% in 0.01 %(v/v)$H_2O_2$ , 40.2%(v/v), and 3.7% at$60^{\circ}C$ incubation, 4 and 0.6% at$-20^{\circ}C$ , 12.9 and 0.1 % in pH 2, 33.8 and 0.2% in 20%(v/v) ethanol, respectively. Autoaggregation test and morphological observation were also conducted in an attempt to explain these differences. These results suggested that high CSH could strengthen the stress tolerance of lactobacilli. -

This study was conducted to provide a vitamin$B_{12}$ database for the representative Korean food items and to assess the dietary intake assessment of vitamin$B_{12}$ for Koreans. The vitamin$B_{12}$ content of 106 foods had been determined by high performance liquid chromatography (HPLC) using column switching method. Rich sources of vitamin$B_{12}$ were meats, milk, and egg ($0.3-3.4\;{\mu}g/100g$ ). Vegetables and fruits contained vitamin$B_{12}$ below limit of detection (LOD). The major food sources for vitamin$B_{12}$ intake were milk (72.0%), meats (22.3%), egg (3.6%), and fishes & shellfishes (2.1%). Mean vitamin$B_{12}$ intake of Koreans was$3.16\;{\mu}g$ /person/day. The proportion of population with intake below estimate average requirement (EAR) and above recommended intake (RI) of vitamin$B_{12}$ was 60.7 and 36.5%, respectively. The vitamin$B_{12}$ intake level of young children with 1-2 years which was 834.6% of RI while the intake level of the older adults 50 years and older was only 70.0% of RI. Also, there were regional differences between urban and rural area. The population with intake below RI was larger than that with intake above RI in Korea. -
Water-soluble fraction (WSF) from edible mushroom hinmogi (Tremella fuciformis) were obtained by water extraction, and polysaccharides in the WSF were separated by ethanol precipitation. The inhibitory effects of the polysaccharides on 3T3-L1 adipocyte differentiation were evaluated by the reduction of peroxisome proliferators-activated receptor
${\gamma}$ ($PPAR{\gamma}$ ) translation, triglyceride accumulation, Oil Red-O staining, and expression levels of$PPAR{\gamma}$ , CCAAT/enhancer binding protein a (C/$EBP{\alpha}$ ), and leptin. The$PPAR{\gamma}$ translation in 3T3-L1 cells was inhibited by the treatment with polysaccharide precipitated by 80% ethanol (P80) which showed highest inhibitory activity among polysaccharides tested. In addition, treatment of P80 to 3T3-L1 cells significantly inhibited the triglyceride accumulation, Oil Red-O staining, and mRNA expression of$PPAR{\gamma}$ , C/$EBP{\alpha}$ , and leptin in a dose-dependent manner. Based upon these results, P80 from edible mushroom hinmogi shows the inhibitory activity on the differentiation of 3T3-L1 adipocytes. Therefore, it might be employed as a potential anti-obesity material. -
Ethanol extracts from the edible mushroom hinmogi (Tremella fuciformis, TF) were used in the investigation of effects on
$\alpha$ -glucosidase in vitro and on glucose-uptake in 3T3-L1 mature adipocytes. Addition of the extract significantly inhibited$\alpha$ -glucosidase from small intestine of porcine and of rat (about 42 and 35% of control, respectively), and stimulated glucose uptake (about 100% of control), of which activity was higher than that of maitake (Grifola frondosa) X-fraction, a well known anti-diabetic substance. When the ethanol extracts were further partitioned successively by organic solvents and purified by silica gel chromatography, the non-polar layer (F-7) from hexane layer showed highest stimulatory activity of glucose-uptake among layers tested. The major components of the F-7 were 1-monooleoylglycerol and 1-monopalmitoylglycerol. Our report is the first description of TF with stimulatory activity of glucose-uptake. These results suggest that TF extracts may constitute a new source of glucose transport activator and could be employed as a potential anti-diabetic material for treatment and preventing diabetes. -

The antioxidative activity of various enzymatic extracts from leaves of Perilla frutescens var. japonica was evaluated by measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. For this study, the leaves were enzymatically hydrolyzed by 8 carbohydrases (Dextrozyme, AMG, Promozyme, Maltogenase, Termamyl, Viscozyme, Celluclast, and BAN) and 9 proteases [Flavourzyme, Neutrase, Protamex, Alcalase, PP-trypsin (trypsin from porcine pancreas), papain, pepsin,$\alpha$ -chymotrypsin, and BP-trypsin (trypsin from bovine pancreas)]. The DPPH radical scavenging activities of Promozyme and Alcalase extracts were the highest, and the$IC_{50}$ values were 77.25 and$109.66\;{\mu}g/mL$ , respectively. All enzymatic extracts of the leaves scavenged hydroxyl radical, and the$IC_{50}$ values of Celluclast and pepsin extracts which were the highest activity were 243.34 and$241.86\;{\mu}g/mL$ , respectively. The BAN and$\alpha$ -chymotrypsin extracts showed the highest scavenging activities, and the$IC_{50}$ values were 21.13 and$33.23\;{\mu}g/mL$ , respectively. The pepsin extracts from the leaves showed protective effect on$H_2O_2$ -induced DNA damage. In addition, the pepsin extracts decreased cell death in PC-12 cells against$H_2O_2$ -induced oxidative damage. The findings of the present study suggest that enzymatic extracts of the leaves possess antioxidative activity. -
Protective effects of cheonggukjang fermented with 20% red ginseng (RC) were observed in streptozotocin (STZ)-induced diabetic rats by measuring levels of blood glucose, serum lipid profiles, and hepatic reactive oxygen species generating and scavenging enzymc activities. RC diet was prepared by mixing with AIN-76 diet at the final concentration 2%, and it was fed to STZ-induced diabetic rats for 3 weeks. The RC dict was significantly improved body weight, feed efficiency ratio, levels of serum glucose, and serum and hepatic lipids in diabetes. The significantly elevated O type activity of xanthine oxidase in diabetes was also greatly decreased by the RC diet. The treatment of RC showed the improved hepatic glutathione s-transferase activities in the diabetic animals. The present study indicates that cheonggukjang fermented with red ginseng could ameliorate STZ-induccd diabetic symptoms such as aggravated blood glucose levels, serum lipid profiles, and even the conditions of oxidative stress.

In this study, the antioxidant activities of red pepper seed kimchi extracts were examined. The extracts were evaluated by various antioxidant assays that included determinations of total phenolic and flavonoid contents, DPPH radical scavenging, superoxide anion radical scavenging, nitric oxide scavenging, superoxide dismutase (SOD) activity, metal chelating activity, and reducing power. All the kimchi extracts showed strong antioxidant activities by the tested methods. The 7% red pepper seed kimchi that was fermented for 6 days possessed the highest activity according to the performed tests. Yet, the highest scavenging activity was exhibited against nitrite, by extracts from kimchi for 0 day of fermentation rather than 6 days. All the activities for the kimchi fermented for 0 day with the addition of 7% red pepper seed increased markedly with an increase in concentration. With the exception of metal chelating and SOD activities, for the antioxidant properties assayed, the red pepper seed kimchi extracts were found to be highly effective.

Novel amylase inhibitors were synthesized via transglycosylation by Thermotoga maritima glucosidase (TMG). TMG hydrolyzes acarbose, acarviosine-glucose, and maltooligosaccharide by releasing$^{14}C$ -labeled glucose from the reducing end of each molecule. When TMG was incubated with acarviosine-glucose (the donor) and glucose (the acceptor), two major transfer products, compounds 1 and 2, were formed via transglycosylation. The structures of the transfer products were determined using thin-layer chromatography (TLC), high-performance ion chromatography (HPIC), and$^{13}C$ nuclear magnetic resonance (NMR) spectroscopy. The results indicate that acarviosine was transferred to glucose at either C-6, to give a$\alpha-(1{\rightarrow}6$ ) glycosidic linkage, or at C-3, to produce an$\alpha-(1{\rightarrow}3$ ) glycosidic linkage. The transfer products showed a mixed-type inhibition against porcine pancreatic$\alpha$ -amylase; therefore, they may be useful not only as inhibitors but also as acarbose transition-state analogs to study the mechanism of amylase inhibition. -
The flower buds of Tussilago farfara (TF) have been traditionally used in oriental medicine for the treatment of bronchitis and asthma. In our study, the primary objective was to determine the mechanisms that are inherent to TF-induced cytotoxicity and apoptosis, using the methanolic extract of TF (TFM) in HT-29 human colon cancer cells. We found that TFM-induced induced cytotoxicity in HT-29 cells in a dose-dependent manner. This effect was verified via an MTT reduction assay, an lactate dehydrogenase (LDH) release assay, and a colony formation assay. Interestingly, we also detected apoptotic bodies on Hoechst staining, and attempted to determine whether TFM-induced apoptosis involved the caspase pathway using a caspase-3/7 activity assay. Overall, the results indicate that TFM contain chemotherapeutic agents and potential candidates use for against human colon cancer cells.

This study was conducted to evaluate in vitro antioxidative and antimicrobial properties of ethanolic extracts from Allium victoriallis L. var. platyphyllum (AVP) with 6 different parts. The antioxidative activities of these samples were determined using the 4 separate methods that involved reducing power, DPPH, hydrogen radical scavenging, and lipid peroxidation with use of a
$\beta$ -carotene/linoleic acid system. The leaf part ethanolic extracts (1,000 ppm) showed the strongest inhibitory potential for reducing power, DPPH, and hydroxyl radicals to 99.8, 49.4, and 52.8%, respectively. Inhibition values of linoleic oxidation were calculated as 58.0, 39.5, and 38.0% for seed, flower, and leaf ethanolic extracts (1,000 ppm), respectively, from AVP. In addition, the ethanolic extracts of the root part showed the most effective antimicrobial activity. The inhibition zones of the root ethanolic extracts ($200\;{\mu}g/disc$ ) of AVP against Bacillus cereus and Staphylococcus aureus were 17 and 14 mm, respectively. In a micro-dilution assay, B. cereus, S. aureus, and Vibrio parahaemolyticus exhibited sensitivity to root part ethanolic extracts with an minimum inhibition concentration (MIC) value of 20, 28, and 18 mg/mL, respectively. Therefore, the AVP extracts may be suitable as antimicrobial and antioxidative agents in the food industry. -

Mutant rice cv. Goami2 (G2) and Baegjinjoo (BJJ) derived from a high-quality japonica rice cv. Ilpumbyeo (IP) were extruded under different feed moisture (20 and 30%) and barrel temperature (90, 110, and$130^{\circ}C$ ). Increasing feed moisture at fixed barrel temperature increased extrudate density (ED) in IP and BJJ. Whereas, G2 showed a varied ED depending on extrusion conditions; increasing barrel temperature decreased the ED of G2 extrudate with low feed moisture, but increased with high moisture. Results indicated a positive barrcl temperature effect on volume expansion in IP and G2, but a negative effect on 811, probably due to shrinkage of expanded products containing low-amylose contents. A significant increase of water absorption was found in G2 and BJJ extruded flour, while an increase of water solubility in those from IP. Non-digestible carbohydrates measured by total dietary fiber (TDF) indicated that extrusion increased slightly TDF in IP and BJJ extrudates, but decreased in G2 products, which might be variety-dependent. -
The effect of ultra-high pressure homogenization on the emulsifying properties of whey protein was investigated in a model emulsion made with whey protein isolate and soya oil under various pH. The emulsifying properties, the average diameter of the oil droplets (
$d_{vs}$ ), and the protein load, were measured for each emulsion produced at different homogenization pressures (50 to 200 MPa) and pH values (4.6 to 8.0). According to the results of variance analysis and response surface, the pH had more influence on oil droplet size and protein load than homogenization pressure. The model equations, which were obtained by response surface analysis, show that pH and homogenization pressure had the major effect on oil droplet size and protein load. Higher homogenization pressure decreased the average droplet size and the protein load. Homogenization at high pressure, as opposed to low pressure, causes no overprocessing, but the effect was pH-dependent. The average diameter of the oil droplets increased slightly by decreasing the pH from 8.0 to 6.5 and then increased dramatically toward the isoelectric point of whey protein (i.e., at pH 4.6). Moreover associated droplets were found at acidic pH and their size was increased at high temperature. -
Water vapor adsorption kinetics of 3 different types of chitosan-based films, i.e., control chitosan, chitosan/montmorillionite (Na-MMT), and chitosan/silver-zeolite (Ag-Ion) nanocomposite films, were investigated at temperature range of
$10-40^{\circ}C$ . In all the films, water vapor is initially adsorbed rapidly and then it comes slowly to reach equilibrium condition. Reasonably good straight lines were obtained with plotting of 1/($m-m_0$ ) vs. l/t. It was found that water vapor adsorption kinetics of chitosan-based films was accurately described by a simple empirical model and the rate constant of the model followed temperature dependence according to Arrhenius equation. Arrhenius kinetic parameters ($E_a$ and$k_o$ ) for water vapor adsorption by chitosan-based films showed a kinetic compensation effect between the parameters with the isokinetic temperature of 315.52 K. -
The aim of the present study was to assess the anti-obesity effects of fermented kochujang supplement in C57BL/6J mice. Thirty mice were divided into 3 groups; normal diet control group (ND), high fat diet control group (HD), and high fat diet plus kochujang supplemented group (HDK). Results were as follows: 1. Fennented kochujang supplement in high fat diet decreased body weight and epidydimal and back fat weight compared to non-supplement in HD group. 2. Lipid content and blood glucose level were lower in HDK group than HD group. 3. Fermented kochujang supplement increased mRNA level of lipolytic genes such as acyl-CoA synthetase (ACS), carnitine palmitoyltransferase-1 (CPT-1), and uncoupling proteins-1 (UCP-1) expression, whereas decreased mRNA level of adipogenic genes such as acetyl CoA carboxylase (ACC) expression. These findings suggest that fermented kochujang supplement in high fat diet normalized body weight, epididymal and back fat weight, lipid content, and blood glucose levels through controlling lipid metabolism and provides basic information on the control of obesity.

In this study, antioxidant activity of methanol extract of Glycyrrhiza uralensis Fisch (GUE) against$H_2O_2$ -induced DNA damage in human leucocytcs and cell death in PC12 cells was determined. The effect of GUE on$H_2O_2$ -induced DNA damage in human leucocytcs was evaluated by the comet assay, where GUE ($1-50\;{\mu}g/mL$ ) was a dose dependent inhibitor of DNA damage induced by$H_2O_2$ . The protective effect of GUE against$H_2O_2$ -induced damage on PC12 cells was investigated by MTT reduction assay and lactate dehydrogenase release assay. A marked reduction in cell survival induced by$H_2O_2$ was significantly prevented by$1-50\;{\mu}g/mL$ of GUE. The enzyme activity of caspase-3 was elevated in$H_2O_2$ -treated PC12 cells, while preincubation with GUE for 30 min inhibited$H_2O_2$ -induced caspase-3 activation in a dose-dependent manner. In conclusion, GUE ameliorates$H_2O_2$ -induced DNA damage in human leucocytes and has neuroprotective effect by preventing cell death in PC12 cell, suggesting that GU may be a potential candidate for novel therapeutic agents for neuronal diseases associated with oxidative stress. -
The green tea market is rapidly growing and identifying the driving factors of consumers' liking for the green tea is important in the tea industries. The objectives of this study were to investigate the effects of manufacturing conditions of the green tea on its sensory characteristics, to elucidate its relationship with the consumer liking. A descriptive analysis and consumer acceptability test were conducted for various green tea samples. The samples differed with regard to the source of the tea, the amino acid content, and the processing methods including the roasting temperature. Partial least square regressions (PLS-R) were performed to establish the relationship between the descriptive data and the consumer acceptability data. The PLS-R results showed that the majority of the consumers liked a green tea which has a stronger 'sweet taste' and roasting-related flavors such as 'roasted barley' and 'burnt leaf'. Such sensory characteristics were produced when a sample made of tea leaves mixed with the tea stem was roasted at a high temperature (
$250^{\circ}C$ ) in this study. -
One of the major allergenic proteins in common buckwheat (Fagopyrum elculentum) was found to be a BW10KD. In this work, allergenic BW10KD genomic DNAs from the native common buckwheat 'Pyeongchang' and Tartarian buckwheat 'Clfa47' were cloned by polymerase chain reaction (PCR), and their nucleotide sequences were determined. In addition, a novel PCR assay targeting the allergenic BW10KD gene was developed to detect and differentiate both buckwheat species in food. The nucleotide sequences of the BW10KD genomic DNA from 'Pyeongchang' and 'Clfa47' were 94% identical. Base differences in the nucleotide sequences of the BW10KD genes are probably useful as a molecular marker for species-specific identification. The 'Pyeongchang'-specific primer set 154PF/400PR and the 'Clfa47'-specific primer set 154DF/253DR generated 247 and 100 bp fragments in singleplex PCR, respectively. A duplex PCR assay with 2 species-specific primer sets simultaneously differentiated the 'Pyeongchang' and 'Clfa47' in a single reaction. The PCR assay also successfully allowed for the rapid detection of buckwheat ingredients in foods.

Anti-lipogenic effects of tannic acid on 3T3-L1 adipocytes as well as on rats fed high fat diet (HFD) were investigated. Tannic acid stimulated lipolysis through suppression of the leptin secretion and an increase of glycerol release in a dose dependent manner in 3T3-L1 adipocytes. For animal study, the rats were fed either HFD or HFD supplemented with 1%(w/w) tannic acid (HFDT) for 12 weeks, respectively. Body weight gain, liver weight, and visceral fat mass in rats fed HFDT were significantly decreased compared to those of rats fed HFD. The lipid profiles of HFDT group were significantly decreased compared with HFD group in the serum and liver, whereas fecal total cholesterol excretion was increased in HFDT group. These results suggest that anti-lipogenic effect of tannic acid in 3T3-L1 adipocytes and in rats fed HFD may be due to the stimulation of lipolysis and the reduction of lipid levels.

Cellular protection against carcinogens could be achieved by the induction of phase 2 detoxifying and antioxidant enzymes such as glutathione S-transferase (GST), NAD(P)H:quinone oxidoreductase 1 (NQO1) and heme oxygenase 1 (HO1). Nuclear transcription factor E2-related factor 2 (Nrf2) binds to antioxidant response element (ARE) in the promoter region of these genes and the resulting transactivation occurs. In the present study the effect of gujeolcho (Chrysanthemum zawadskii) roots on the Nrf2-ARE pathway were investigated. C. zawadskii root extract was fractionated with a series of organic solvents and their ability to induce Nrf2-ARE pathway was examined. We separated the most potent dichloromethane (DCM) fraction into 12 sub-fractions and found several sub-fractions with strong effects on the Nrf2-ARE pathway. Fraction 4 strongly induced the ARE-reporter gene activity as well as Nrf2 expression. Sitosterol was isolated as a major compound in fraction 4 although its activity was not as potent as its mother fraction. These results indicate that C. zawadskii roots might be used as a potential natural chemopreventive source. -
Contents of phytochemicals in citron (Citrus junos Sieb. ex Tanaka) seeds and their effects on antioxidant activities were investigated. Methanol extract of defatted citron seeds contained the highest amounts of total polyphenols followed by 70% ethanol and water extracts. Neohesperidin was the most predominant citrus flavanones in these extracts. The highest amounts of limonoids were found in methanol extract, and this extract was the most efficient in scavenging both DPPH and ABTS radicals. All 3 extracts also exhibited good antioxidant activities against attack of linoleate free radicals on
$\beta$ -carotene. When methanol extract was sequentially fractionated into ether, ethyl acetate, n-butanol, and water fractions, butanol fraction contained the highest amounts of polyphenols otherwise most of limonoids were concentrated in ethyl acetate fraction. A positive relationship between radical scavenging activities and total polyphenol contents in fractions was observed while antioxidant activity on$\beta$ -carotene seemed more related with contents of limonoids and other hydrophobic polyphenols. -
Extraction characteristics of garlic and functional properties of corresponding extract were monitored by response surface methodology (RSM). Maximum extraction yield of 26.41% was obtained at microwave power of 146.29 W, ethanol concentration of 63.31 %, and extraction time of 5.88 min. At microwave power, ethanol concentration, and extraction time of 114.84 W, 58.83%, and 1.42 min, respectively, maximum electron-donating ability (EDA) was 72.86%. Maximum nitrite-scavenging ability was 94.62% at microwave power, ethanol concentration, and extraction time of 81.83 W, 2.65%, and 3.83 min, respectively. Superoxide dismutase (SOD) showed maximum pseudo-activity of 49.12% at microwave power of 34.23 W, ethanol concentration of 33.11 %, and extraction time of 4.40 min. Based on superimposition of 4-dimensional RSM with respect to extraction yield, electron-donating ability, nitrite-scavenging ability, and pseudo-activity of SOD, optimum ranges of extraction conditions were microwave power of 0-100 W, ethanol concentration of 40-70%, and extraction time of 2-8 min.

Alzheimer's disease (AD) is the most common neurodegenerative disorder and is responsible for more than 50% of all dementia cases. There is significant interest in finding new sources of compounds that inhibit acetylcholinesterase (AChE) to be used in the treatment of AD, since only a few AChE inhibitors, such as galanthamine, physostigmine, and tacrine, are available for clinical use. In the present study, ICR mice were treated with a 1 mg/kg scopolamine, which caused impaired cognitive ability. The steady consumption of a water extract of Chrysanthemum indicum Linne flowers for 3 months significantly prevented the scopolamine induced deficit of the spatial cognitive capability of mice. It also improved long-term memory in mice with amnesia induced by scopolamine, as assessed by the Morris water maze and passive avoidance tests. In addition, water extract consumption significantly decreased AChE activity in mouse brain, leading to inhibition of acetylcholine hydrolysis.

The objective of this study was to examine the effects of crude protease from Bacillus polyfermenticus SCD and marination time on quality of pork and beef jerky. Neither pork nor beef jerky showed a significant difference in pH among all treatments, and each protease was found to have a greater effect on the color of beef jerky. The hardness was significantly lower in all jerky treated with each protease, however the textural properties of jerky were not significantly different with regard to marination times. Water content was not affected by protease addition or marination times, however the water activity was lower in jerky treated with protease. The rehydration capacity of pork jerky was higher in jerky treated with protease, whereas that of beef jerky was higher in jerky dried after tumbled and held for 24 hr. Sensory characteristics were higher in jerky treated with protease, not affected by holding time after marinated. -
Effects of replacing pork backfat with a combination (ICM) of isolated soy protein (ISP), carrageenan, and maltodextrin, or with ICM +olive oil, on the quality characteristics of sausages were investigated. Both treatments had lower fat content (p<0.05), but higher protein and moisture contents than the control (p<0.05). The fat content of low-fat sausage containing the ICM was increased on day 30 compared to day 1 and 15 (p<0.05), and that of ICM+olive oil was increased after day 15. The water holding capacity of ICM was lower than the control through day 30 (p<0.05). The ICM+olive oil had a lower cooking loss than ICM on day 1 and 15 (p<0.05). On day 1, the ICM had lower lightness and higher redness values than the control (p<0.05), and the ICM+olive oil had a higher yellowness value than the control and ICM (p<0.05). Both treatments presented higher hardness, cohesiveness, gumminess, and chewiness values than the control (p<0.05). The lipid oxidation values of both treatments were lower than the control on day 15 and 30 (p<0.05), and those were affected by the addition of olive oil. The ICM was rated higher for sensory color and overall acceptability than the ICM+olive oil (p<0.05).

The synergistic antimicrobial effect of Achyranthes japonica Nakai (AJN) and Bifidobacterium extracellular factors against Clostridium difficile were measured using a turbidity method. Each broth supernatant of Bifidobacterium infantis (
$68.8{\pm}0.02%$ ) and Bifidobacterium adolescentis ($33.2{\pm}0.2%$ ) obtained by adding ethyl acetate soluble fractionate from A. japonica Nakai ethanolic extracts (AJNEA, 100 ppm, no inhibition) showed high synergistic antimicrobial activity against C. difficile. In addition, the antimicrobial activity in a laboratory medium and yogurt products against C. difficile were evaluated. In yogurt prepared with a starter 5 (Lactobacillus acidophilus: Streptococcus thermophilus: B. adolescentis =1 : 1 : 1) and a starter 4 (L. acidophilus: S. thermophilus: B. infantis=1 : 1 : 1) and 0.5% AJNEA powder, high antimicrobial effects were recorded that measured 79.0 and 65.2%, respectively. The results indicated the potential of AJN extract for use as an antimicrobial agent. In addition, the efficiency of the antimicrobial activity of the extracts was further improved in combination with lactic acid bacteria, which suggests that they have the potential to be used as a highly effective antibiotic-tolerant microorganism prevention system. Such a strategy can be used for alternative drugs or functional food additives for treatment of antibiotic-associated diarrhea. -

Determining the elution of water-soluble substances from herbal teas is an important factor in their efficient use in terms of taste, perfume, and content of health-related components. The antioxidant activity and content of catechins in commercial Chrysanthemum indicum (gamguk) teas were determined for optimum elution conditions. The water extract of gamguk teas did not differ significantly in yield compared to methanol extracts and showed stronger antioxidant activity. Catechin contents in gamguk teas were 8-18% of the extracts when individual peaks in high-performance liquid chromatography analysis were compared to standard catechin peaks. Gamguk teas exhibited faster release of antioxidants, and the antioxidant activity was positively correlated with the thermal treatments. Gukhwacha (GC) was the best tea for rapid release (30 sec) of antioxidants with the$50^{\circ}C$ treatment, whereas antioxidants in other teas were relatively slower released. -
Effects of extracts obtained from the flowers of Elsholtzia splendens on the serum lipid profile and hepatotoxicity in mice were investigated. Female ICR mice were given E. splendens ethanolic extract (ESEs) orally at a dose of 10 or 50 mg/kg BW for 50 days. Significant dose-dependent decreases in triglyceride and low-density lipoprotein (LDL)-cholesterol of serum were observed. In addition, ESEs prolonged the lag-time of LDL oxidation in vitro. In the serum of ICE mice given ESEs orally at 10 and 50 mg/kg BW, the serum levels of aspartate aminotransferase (AST) and lactic dehydrogenase (LDH) increased significantly, while total protein, albumin, creatinine, alanine aminotransferase (ALT), and total bilirubin did not change. Therefore, ESEs may be beneficial to human health, although it has some hepatotoxicity.

Rheological properties of sweet potato starch (SPS)-glucose composites (5%, w/w) at different concentrations (0, 10, 20, and 30%, w/w) of glucose were investigated in steady and dynamic shear. The steady shear rheological properties of SPS-glucose composites were determined from rheological parameters for power law and Casson flow models. At
$25^{\circ}C$ all the samples showed a pronounced shear-thinning behaviors (n=0.29-0.37) with high Casson yield stress. In general, the presence of glucose resulted in the decrease in consistence index (K), apparent viscosity (${\eta}_{a,100}$ ), and yield stress (${\sigma}_{oc}$ ). Storage (G') and loss (G") moduli increased with an increase in frequency ($\omega$ ), while complex viscosity (${\eta}*$ ) decreased. Dynamic moduli (G', G", and${\eta}*$ ) of the SPS-glucose composites at higher glucose concentrations (20 and 30%) were higher than those of the control (0% glucose) and also increased with increasing glucose concentration from 10 to 30%. The effect of glucose on steady and dynamic shear rheological properties of the SPS pastes appears to greatly depend on glucose concentration in the range of 10-30%. -

Edible red algae paper coated with green tea ext Tact was prepared and determined the microbial growth and quality change of kimbab wrapped with the paper during storage. The paper coated with green tea extract had the antimicrobial activity against Listeria monocytogenes and Escherichia coli. After 12 hr of storage of kimbab, packaging with the paper coated with green tea extract decreased populations of total aerobic bacteria by 1 log cycle. Kimbab wrapped with the red algae paper coated with green tea extract had 5.5 mg malondialdehyde (MDA)/kg after 12 hr, in contrast to the control of 7.4 mg MDA/kg. Kimbab with red algae paper with green tea extract was better than the control in terms of sensory qualities. These results clearly indicate that kimbab can be packaged with red algae paper coated with green tea extract, resulting in extending its shelf life. -

The anti-obesity effect of chokong, Rhynchosia nolubilis seeds pickled in vinegars for 2 weeks at$4^{\circ}C$ , was investigated. During the differentiation of 3T3-L1 adipocytes, the addition of ethanolic extracts of chokongs lowered the cellular triglyceride content by 8.1-9.0%, and glucose content by 12.2-27.6%, depending on the kinds of vinegar used. The activity of glycerol-3-phosphate dehydrogenase also decreased up to 56.0-59.3% by supplying those extracts. In addition, vinegars were superior to acetic acid, citric acid, and hydrochloric acid solutions, and distilled water in anti-obesity of the pickled seeds. -
A high performance liquid chromatography (HPLC) method has been successfully developed to identify and quantify major ginsenosides in Korean ginseng seeds and roots. Using gradient elution of acetonitrile and water without buffer, the 6 major ginsenosides (
$Rb_1,\;Rb_2$ , Rc, Rd, Re, and$Rg_1$ ) were identified. Compared with ginseng roots, the amount of ginsenoside Re and Rd in ginseng seeds were significantly higher than those in ginseng roots (p<0.05). In ginseng seeds, the content of protopanaxtriol (PPT) was higher than that of protopanaxdiol (PPD) and the ratio of PPT and PPD was approximately 2.2 : 1. However, the content of PPT was lower than that of PPD in ginseng roots. It should be mentioned that both content of PPT and PPD in ginseng seeds were much higher than those in ginseng roots. -
The aim of this study was to investigate whether Monascus-fermented soybean extracts (MFSE) containing natural estrogen-like compounds such as isoflavones and mevinolins has potential effects on human osteoblast-like SaOS2 cells using 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) and alkaline phophatase (ALP) assaies. MFSE exerted biphasic dose-dependent effect; stimulating osteoblastic activity at low concentrations and inhibiting SaOS2 cells viability at high concentrations. At
$10^{-8}-10^{-4}\;mg/mL$ , MFSE is not only non-cytotoxic but also induced comparatively high ALP activity on SaOS2 cells. ALP activity (%) significantly increased (220.1%, p<0.05) when SaOS2 cells were treated with MFSE at a concentration of$10^{-5}\;mg/mL$ , whereas slowly increased (185.6%, p<0.05) in unfermented soybean extracts (UFSE) at$10^{-3}\;mg/mL$ . The potentially greater ALP activity of MFSE compared to the UFSE might partially be caused by its mevinolin, which was derived from the soybean during Monascus-fermentation. Our findings indicate that supplementation of MFSE may accelerate the speed of intracellular ALP synthesis by the bone cells when provided at optimal dosages. -
A novel lactobacilli replicon from plasmids of Lactobacillus reuteri KCTC 3678 was isolated. Eight L. reuteri strains from Korean Collection for Type Cultures (KCTC) and Korea Food Research Institute (KFRI) were screened for cryptic plasmids and most strains harbored 1 or 2 plasm ids. Particularly, L. reuteri KCTC 3678 contained 6 plasm ids which all were used for screening of lactobacilli replicon. EcoRI digests of the plasmid DNA prep from L. reuteri KCTC 3678 were ligated with pUC19 and the recombinant DNAs were serially named from pLR1 to pLR7. A cat (chloramphenicol acetyltransferase;
$Cm^r$ ) gene originated from pC194 was introduced into pLR1-7, resulting in pLR1cat-pLR7cat, respectively. The recombinant plasmids were introduced into L. reuteri KCTC 3679, and only transformants harboring pLR5cat were obtained, indicating that the insert in pLR5 functioned as a lactobacilli replicon. -

Meju, a major ingredient of doenjang which is a popular Korean traditional fermented soyfood, was manufactured by fermenting steamed soybeans in natural environment in which steamed soy was exposed to airborne microorganism, in particular, fungi. Total isoflavone content was reduced from 1,849 mg/kg of cooked soy to 816 mg/kg of meju at the$90^{th}$ day of fermentation. Total glycosides and aglycones of isoflavones in meju were 1,827 and 22 mg/kg at 0 day and changed into 487 and 329 mg/kg at the 90th day of fermentation, respectively. Meanwhile, the ratio of glycosides to aglycones of isoflavones was not changed during aging of doenjang but remained relatively constant with 592 and 644 mg/kg aglycones at the 0 and$160^{th}$ day, respectively. When cooked soy was fermented with Aspergillus oryzae and Aspergillus flavus, isoflavone profiles were significantly different from each other while A. oryzae caused more extensive metabolism of isoflavones than A. flavus.
