Peroxynitrite (ONOO$^{-}$) is one of cytotoxic species produced by the reaction between superoxide(ㆍ $O_2$$^{-}$) and nitric oxide (NO). The aim of this study was to characterize ONOO$^{-}$ scavenging constituents from herbs. Methanolic extract derived from one hundred fifty nine herbs were screened out for their ONOO$^{-}$ scavenging activities. It was investigated that about 33 herbs was excellent scavengers of ONOO$^{-}$. The extracts exhibited dose-dependent ONOO$^{-}$ scavenging activities.
The radical scavenging activities of 9 medicinal plants on peroxynitrite ($ONOO^-$) and hydroxyl (${\cdot}OH$) radical were investigated using in vitro system. The water extracts of 9 medicinal plants showed the protective effect against $ONOO^-$ and ${\cdot}OH$ radical. In particular, Akebia quinata, Aster scaber, Cudrania tricuspidata, Diospyros kaki, Eriobotrya japonica, Lycium chinense, Parthenocissus tricuspidata and Polygonum aviculare exhibited $ONOO^-$-scavenging activity by about 50% at the concentration of $10{\mu}g/ml$. Although those $ONOO^-$-scavenging activities were lower than that of penicillamine (94.08${\pm}$3.04%) as a positive control, Eriobotrya japonica (89.87${\pm}$4.57%) was the most potent scavenger of $ONOO^-$ at the concentration of $10{\mu}g/ml$. Also, Diospyros kaki and Urtica angustifolia showed the strong${\cdot}$OH-scavenging activity than thiourea, positive control, at the concentration of lmg/ml. Our results indicate that 9medicinal plants may act as free radical scavengers and reduce damages caused by oxidative stress associated with $ONOO^-$ and${\cdot}$OH radical.

Korean red ginseng (KRG) has been used worldwide as a traditional medicine for the treatment of various diseases, including cancer. In this study, we determined the effect of KRG on the responses of HaCaT cells to peroxynitrite ($ONOO^-$). In this study, we determined the effect of KRG on the responses of HaCaT cells to peroxynitrite ($ONOO^-$). Cells treated with $ONOO^-$ (2 mM) prior to incubation with control medium for 12 hours displayed reduced viability, as determined using the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay (viability about 48% of that of non-treated control cells). When KRG was added to the post-incubation medium, the negative effects of $ONOO^-$ on cell viability were significantly reduced. Reverse transcription-polymerase chain reaction analysis indicated that KRG alone did not significantly alter p53 or "growth arrest and DNA damage" (GADD)45 mRNA levels. However, the addition of KRG to the post-incubation medium significantly and dose-dependently reduced levels of p53 and GADD45 mRNA in $ONOO^-$-treated cells. Western blot analyses revealed that incubation with KRG decreased p53 and GADD45 protein levels in $ONOO^-$-treated cells, relative to those in cells incubated with control medium. Collectively, these results suggest that Korean red ginseng extract protects cells against $ONOO^-$-induced genotoxicity by increasing cell viability through modulating the expression of p53 signaling intermediates.
Crude extracts of Limonium tetragonum and their solvent-partitioned fractions were evaluated for their potential to scavenge authentic $ONOO^-$, and $ONOO^-$ derived from 3-morpholinosydnonimine (SIN-1). Four flavonol glycosides (1-4) were isolated by activity-guided separation. Their chemical structures were elucidated by extensive 2 D NMR experiments and by comparison with published spectral data. These compounds were also estimated for their peroxynitrite scavenging effects. The scavenging ratios of compounds 1-4 on authentic $ONOO^-$ were 56, 37, 56, and 54%, respectively, at a concentration of 1 ${\mu}M$. On the other hand, the inbihition ratios of compounds 1-4 against $ONOO^-$ generation from SIN-1 were 59, 39, 44, and 54% at the same concentration, respectively.

This study was to investigate the inhibitory effects of Mori Fructus on the generation of peroxynitrite ($ONOO^-$), nitric oxide (NO) and superoxide anion radical (${\cdot}O_2{^-}$) in the endothelial cells of rat vessels. The aim of this study was to investigate the $ONOO^-$, NO, ${\cdot}O_2{^-}$ scavenging and anti-inflammatory activitives of Mori Fructus. For this study, the fluorescent probes, namely dihydrorhodamine 123 (DHR 123), 4,5-diaminofluorescein (DAF-2) and 2',7'-dichlorodihydrofluorescein diacetate (DCFDA) were used. Western blotting was performed using anti-NF-${\kappa}B$ (p50, p65), anti-COX-2, anti-iNOS antibodies, respectively. Mori Fructus prevented lipopolysaccharide (LPS)-induced cell death in YPEN cells. Mori Fructus inhibited the generation of $ONOO^-$, NO and ${\cdot}O_2{^-}$ in the LPS-treated cells. Mori Fructus inhibited the expression of COX-2 and iNOS genes by means of decreasing the NF-${\kappa}B$ activation. These results suggest Mori Fructus is effective on inhibiting the generation of $ONOO^-$, NO and ${\cdot}O_2{^-}$, and that therefore it might have a potential role as a treatment for the inflammatory process and inflammation-related diseases.
A peroxynitrite is formed when superoxide and nitric oxide exist at near eqimolar ratio in biological systems. Although not a free radical by chemical nature, peroxynitrite is a powerful oxidant having a wide array of tissue damaging effects ranging from lipid oxidation and inactivation of enzymes and ion channels through protein oxidation and nitration to inhibition of mitochondrial respiration. During our search for new antioxidizing components from natural resources, twenty salt marsh plants were screened for their ONOO and DPPH radical scavenging activities. Among them, methanol extract of Rosa rugosa, lxeris tamagawaensis, Erigeron annus, Tetragonia tetragonoides, Imperata cylindrica, and Suaeda japonica inhibited more than 85% of peroxynitrite produced by 3-morpholinsydnonimine (SIN-1) at a concentration of 5 $\mu\textrm{g}$/$m\ell$. In addition, Rosa rugosa, Artemisia capillaris, Erigeron annus and Ixeris tamagawaensis showed significant scavenging effect against DPPH (1,1-diphenyl-2-picrylhydrazyl radical).
To find the anti-Alzheimer's activity of Cacalia hastata var. orientalis (Compositae), caffeoylquinic acids were analyzed in this plant by HPLC and the inhibitory activities of the extract on cholinesterases including acetylcholinesterase (AChE) and butyrylcholinesterse (BChE) and peroxynitrite ($ONOO^-$) were assayed. Sum of caffeoylquinic acids in the 80% MeOH extract was quantitatively higher in leaves (35.8% of the extract) than in stems (3.7%). The compound, 3,5-dicaffeoylquinic acid, was contained in the highest amount in the leaf (44.32 mg/g dry weight). The $IC_{50}s$ of the 80% MeOH extract were shown to be $67.45{\mu}g/ml$ for AChE, and $8.59{\mu}g/ml$ (for $ONOO^-$), respectively, suggesting that the leaves would have the anti-Alzheimer's activity due to the high content of caffeoylquinic acids.
Peroxynitrite formation in vivo is implicated in numerous human diseases and there is considerable interest in the use of antioxidants and natural products for their treatment. The three components (1-3) isolated from Sargassum thunbergii as well as the organic solvent-soluble fractions and the aqueous layer of S. thunbergii were evaluated for their potential to scavenge authentic ONOO$\^$-/ and ONOO$\^$-/ derived from 3-morpholinosydnonimine (SIN-1). The antioxidant activity of the individual fractions was in the order of 85% aqueous (aq.) MeOH>n$\^$-/ BuOH>n-hexane>H$_2$O. The three known compounds, sargahydroquinoic acid (1), sargaquinoic acid (2) and sargachromenol (3) showed peroxynitrite-scavenging activities comparable to those of L-ascorbic acid and penicillamine. These results showed a possible antioxidant activity in major constituents of S. thunbergii.

This study examined the antioxidative activity of delphinidin, a kind of anthocyanidin from eggplant. Cellular protective potential from oxidative damage by nitric oxide (NO), superoxide anion ($O_2^-$), and peroxynitrite ($ONOO^-$) using epithelial cell line LLC-PK1 cell as well as in vitro radical scavenging effects were investigated. Delphinidin showed strong in vitro radical scavenging effects against NO, $O_2^-$, and hydroxyl radical (${\cdot}OH$) in dose-dependent manners. In addition, delphinidin increased cell viability in LLC-PK1 cells in a concentration-dependent manner when viability was reduced by $ONOO^-$-induced oxidative damage. To elucidate the protective mechanisms of delphinidin from $ONOO^-$, sodium nitroprusside (SNP), and pyrogallol were also employed to generate NO and $O_2^-$, respectively. The treatment of delphinidin recovered reductions in cell viability caused by SNP and pyrogallol, indicating that delphinidin can attenuate oxidative stress induced by NO and $O_2^-$. The present study suggests that delphinidin is a promising anti oxidative agent.
