Microbiome of Haemaphysalis longicornis Tick in Korea

  • Ticks can transmit pathogenic bacteria, protozoa, and viruses to humans and animals. In this study, we investigated the microbiomes of Haemaphysalis longicornis according to sex and life stages. The Shannon index was significantly higher for nymphs than adult ticks. Principal coordinates analysis showed that the microbiome composition of female adult and male adult ticks were different. Notably, Coxiella-like bacterium (AB001519), known as a tick symbiont, was found in all nymphs and female adult ticks, but only one out of 4 male adult ticks had Coxiella-like bacterium (AB001519). In addition, Rickettsia rickettsii, Coxiella burnetii, and Anaplasma bovis were detected in this study.

  • Staphylococcus epidermidis is well-known not only as an innocuous normal flora species commonly isolated from human skin, but also as an important bacterial species to keep skin healthy, because this species can protect the human skin from pathogenic microorganisms. However, S. epidermidis turns into a potential pathogen in damaged skin, because these bacteria can easily form a biofilm on the wound area and provide antimicrobial resistance to other microorganisms embedded in the biofilm. Thus, it is important to kill S. epidermidis in the early stage of wound treatment and block the formation of biofilms in advance. In the present study, hydrogel wound dressings were fabricated using polyvinyl alcohol/polyethylene glycol containing silver citrate nanorods, which have been proven to have strong antimicrobial activity, especially against S. epidermidis, and their wound-healing efficacy was investigated in vivo using a rat experiment.

도축장 출하차량에서 Lawsonia intracellularis 분포율 조사 (Distribution of Lawsonia intracellularis in livestock transport car of slaughterhouse, Korea)

  • Lawsonia intracellularis is the pathogenic agent of porcine proliferative enteritis (PPE). The bacterial pathogen infects the intestinal crypt cells which causes hyperplasia of the infected cells and leads to the process of intestinal pathogenesis. PPE includes some clinical maninfestations, including acute hemorrhagic diarrhea with sudden death in growing pigs and porcine intestinal adenomatosis, to a chronic diarrhea with reduced productivity of the infected pigs. The purpose of the present studies were carried out to determine L. intracellularis in livestock transport car of slaughterhouse. Distribution of L. intracellularis in livestock transport car were conducted using real-time polymerase chain reaction (real-time PCR) testing method, total 300 samples. Of 300 samples, 119 (39.7%) were detected as positive to L. intracellularis in livestock transport car. In seasonal analysis, 42 (28.0%) out of 150 samples in spring and summer season. 77 (51.3%) out of 150 sample in autumn and winter season. In regional analysis, 53 (88.3%) out of 60 cars and the detection ratio showed that regional variation in livestock transport car.

Volatile Metabolic Markers for Monitoring Pectobacterium carotovorum subsp. carotovorum Using Headspace Solid-Phase Microextraction Coupled with Gas Chromatography-Mass Spectrometry

  • Identifying the extracellular metabolites of microorganisms in fresh vegetables is industrially useful for assessing the quality of processed foods. Pectobacterium carotovorum subsp. carotovorum (PCC) is a plant pathogenic bacterium that causes soft rot disease in cabbages. This microbial species in plant tissues can emit specific volatile molecules with odors that are characteristic of the host cell tissues and PCC species. In this study, we used headspace solid-phase microextraction followed by gas chromatography coupled with mass spectrometry (HS-SPME-GC-MS) to identify volatile compounds (VCs) in PCC-inoculated cabbage at different storage temperatures. HS-SPME-GC-MS allowed for recognition of extracellular metabolites in PCC-infected cabbages by identifying specific volatile metabolic markers. We identified 4-ethyl-5-methylthiazole and 3-butenyl isothiocyanate as markers of fresh cabbages, whereas 2,3-butanediol and ethyl acetate were identified as markers of soft rot in PCC-infected cabbages. These analytical results demonstrate a suitable approach for establishing non-destructive plant pathogen-diagnosis techniques as alternatives to standard methods, within the framework of developing rapid and efficient analytical techniques for monitoring plant-borne bacterial pathogens. Moreover, our techniques could have promising applications in managing the freshness and quality control of cabbages.

Synthesis and Evaluation of Antimicrobial Activity of 1, 3, 4-Thiadiazole Analogues for Potential Scaffold

  • Objectives: Pathogenic microbes are causal agents for various types of severe and even lethal infectious diseases. Despite of development in medication, bacterial and fungal infections still persist to be a vital problem in health care. Bacteria and several fungal species have shown resistance to antibiotics used in treatment to current medications. Therefore, it is a considerable field of interest in the design and development of novel compounds with antimicrobial activity. Methods: The compounds bearing a heterocyclic ring play an imperative role among other organic compounds with pharmacological activity used as drugs in human for control and cure of various infections. Thiadiazoles containing nitrogen-sulfur atom as part of their cyclic structure which shown wide-ranging application as structural units of biologically active molecules and are very useful intermediates in Medicinal Chemistry. Results: The effectiveness of the thiadiazole nucleus was established by the drugs currently used for the treatment of various infections. 1,3,4-Thiadiazoles and some of their derivatives are widely studied because of their broad spectrum of pharmacological activities. Conclusion: In the present work, a series of 1,3,4-Thiadiazole derivatives were synthesized by cyclization of a group of various benzaldehyde with thiosemicarbazide in the presence of various reagent like FeCl3, HCHO by losing a molecule of water. These derivatives were found to possess prominent antimicrobial activity.

Biocontrol Activity of Aspergillus terreus ANU-301 against Two Distinct Plant Diseases, Tomato Fusarium Wilt and Potato Soft Rot

  • To screen antagonistic fungi against plant pathogens, dual culture assay (DCA) and culture filtrate assay (CFA) were performed with unknown soil-born fungi. Among the different fungi isolated and screened from the soil, fungal isolate ANU-301 successfully inhibited growth of different plant pathogenic fungi, Colletotrichum acutatum, Alternaria alternata, and Fusarium oxysporum, in DCA and CFA. Morphological characteristics and rDNA internal transcribed spacer sequence analysis identified ANU-301 as Aspergillus terreus. Inoculation of tomato plants with Fusarium oxysporum f. sp. lycopersici (FOL) induced severe wilting symptom; however, co-inoculation with ANU-301 significantly enhanced resistance of tomato plants against FOL. In addition, culture filtrate (CF) of ANU-301 not only showed bacterial growth inhibition activity against Dickeya chrysanthemi (Dc), but also demonstrated protective effect in potato tuber against soft rot disease. Gas chromatography-tandem mass spectrometry analysis of CF of ANU-301 identified 2,4-bis(1-methyl-1-phenylethyl)-phenol (MPP) as the most abundant compound. MPP inhibited growth of Dc, but not of FOL, in a dose-dependent manner, and protected potato tuber from the soft rot disease induced by Dc. In conclusion, Aspergillus terreus ANU-301 could be used and further tested as a potential biological control agent.

Genome Wide Analysis of the Potato Soft Rot Pathogen Pectobacterium carotovorum Strain ICMP 5702 to Predict Novel Insights into Its Genetic Features

  • Pectobacterium carotovorum subsp. carotovorum (Pcc) is a gram-negative, broad host range bacterial pathogen which causes soft rot disease in potatoes as well as other vegetables worldwide. While Pectobacterium infection relies on the production of major cell wall degrading enzymes, other virulence factors and the mechanism of genetic adaptation of this pathogen is not yet clear. In the present study, we have performed an in-depth genome-wide characterization of Pcc strain ICMP5702 isolated from potato and compared it with other pathogenic bacteria from the Pectobacterium genus to identify key virulent determinants. The draft genome of Pcc ICMP5702 contains 4,774,457 bp with a G + C content of 51.90% and 4,520 open reading frames. Genome annotation revealed prominent genes encoding key virulence factors such as plant cell wall degrading enzymes, flagella-based motility, phage proteins, cell membrane structures, and secretion systems. Whereas, a majority of determinants were conserved among the Pectobacterium strains, few notable genes encoding AvrE-family type III secretion system effectors, pectate lyase and metalloprotease in addition to the CRISPR-Cas based adaptive immune system were uniquely represented. Overall, the information generated through this study will contribute to decipher the mechanism of infection and adaptive immunity in Pcc.

Inhibition of Quorum Sensing Regulated Virulence Factors and Biofilm Formation by Eucalyptus globulus against Multidrug-Resistant Pseudomonas aeruginosa

  • Objectives: The quorum-sensing-inhibitory and anti-biofilm activities of the methanol extract of E. globulus leaves were determined against clinically isolated multidrug-resistant Pseudomonas aeruginosa. Methods: The preliminary anti-quorum-sensing (AQS) activity of eucalyptus was investigated against a biosensor strain Chromobacterium violaceum ATCC 12472 (CV12472) by using the agar well diffusion method. The effect of sub-minimum inhibitory concentrations (sub-MICs) of the methanol extract of eucalyptus on different quorum-sensing-regulated virulence factors, such as swarming motility, pyocyanin pigment, exopolysaccharide (EPS), and biofilm formation, against clinical isolates (CIs 2, 3, and 4) and reference PA01 of Pseudomonas aeruginosa were determined using the swarm diameter (mm)-measurement method, chloroform extraction method, phenol (5%)-sulphuric acid (concentrated) method, and the microtiter plate assay respectively, and the inhibition (%) in formation were calculated. Results: The preliminary AQS activity (violacein pigment inhibition) of eucalyptus was confirmed against Chromobacterium violaceum ATCC 12472 (CV12472). The eucalyptus extract also showed concentration-dependent inhibition (%) of swarming motility, pyocyanin pigment, EPS, and biofilm formation in different CIs and PA01 of P. aeruginosa. Conclusion: Our results revealed the effectiveness of the E. globulus extract for the regulation of quorum-sensing-dependent virulence factors and biofilm formation at a reduced dose (sub-MICs) and suggest that E. globulus may be a therapeutic agent for curing and controlling bacterial infection and thereby reducing the possibility of resistance development in pathogenic strains.

온라인에서 유통 중인 국내산 및 수입산 오징어(Todarodes pacificus) 젓갈의 미생물학적 오염도 조사 (Microbiological Contamination in Domestic and Imported Squid Todarodes pacificus Jeotgal Distributed at On-line Marketplaces: An Investigation)

  • Squid jeotgal is a very popular traditional, salted, and fermented seafood in Korea. Due to the development of a distribution structure, a lot of imported squid jeotgal has recently entered Korea. In this study, we analyzed and compared the microbial contamination levels of domestic and imported squid jeotgal. The total viable bacterial count in domestic and imported jeotgal was 5.20 and 5.38 log CFU/g, respectively. The highest contamination level was 6.94 log CFU/g in one of the imported squid jeotgal samples. Coliforms were not detected in domestic jeotgal but were detected at 50% with 2.21 log CFU/g in imported jeotgal. Escherichia coli and Staphylococcus aureus were not detected (ND: <1 log CFU/g) in both domestic and imported jeotgal. All samples of jeotgal were negative for Bacillus cereus and Clostridium perfringens. The microbial contamination levels determined in the current study may be utilized as primary data to execute microbial risk assessments on domestic and (or) imported jeotgal.

Antimicrobial Evaluation and Characterization of Copper Nanoparticles Synthesized by the Simple Chemical Method

  • Copper nanoparticles (CuNPs) are considered of great importance due to their high catalytic and antimicrobial activities. This study focuses on the preparation and characterization of CuNPs, and on their antibacterial/antifungal activities. A copper salt (copper sulfate pentahydrate) as precursor, starch as stabilizing agent, and ascorbic acid as reducing agent were used to fabricate CuNPs. The resulting product was characterized via different techniques such as X-ray diffractrometry (XRD), Fourier Transform Infrared (FTIR) spectroscopy, and Scanning electron microscopy (SEM) to confirm its characteristic properties. Employing the Scherrer formula, the mean crystallite sizes of copper (Cu) and cuprous oxide (Cu2O) nanocrystals were found to be 29.21 and 25.33 nm, respectively, as measured from the main X-ray diffraction peaks. The functional groups present in the resulting CuNPs were confirmed by FTIR. In addition, the engineered CuNPs showed antibacterial and antifungal activity against tested pathogenic bacterial and fungal strains.
