Effects of Lycium chinense Miller Fruit and its Constituent Betaine on Immunomodulation in Balb/c Mice

  • BACKGROUND: The aim of this study is to investigate the effects of Lycium chinense Miller fruit and its representative constituent betaine on reduction of immobility time and blood parameters in balb/c mice. METHODS AND RESULTS: We investigated the immobility time and the changes in aspects of blood biochemical parameters by the administration of L. chinense Miller fruit and its representative constituent betaine, after the forced swimming test. The immobility time was significantly reduced about 41.3% and 53.6%, respectively, in the animal of L. chinense Miller fruit and its representative constituent betaine-administrated group for 7 days, in comparison with that of the control group. The level of blood urea nitrogen was significantly decreased in L. chinense Miller fruit and its representative constituent betaine-treated group compared with the control group (P < 0.05), respectively. In addition, the interlukin-2 levels of mice in L. chinense Miller fruit and betaine treated group was increased compared with the control group. CONCLUSION: These results indicate that L. chinense Miller fruit and betaine might be helpful in the immune function improvement, enhance physical stamina, and fatigue recovery.

  • According to the research of the cultivation result on the spot farmers for 5 years in order to investigate its cultivation conditions and problems of Lycium chinetrse Mill, the development of the variety with early mature, pest tolerance, and high yielding is urgent and it is call for the development of rooted cutting cultivation by testing of bed soil, growth regulator and cutting times. The development of cultivation method with planting year, planting density, supplementary period and amounts of supplementary of Lycium chinense Mill planted in growing field are required and for the disease and the effective control of disease and insect pest resistance is needed by auxotrophic fertilizing and organic agricultural method, met by agricultural chemicals.

A Convenient HPLC/ELSD Method for the Quantitative Analysis of Betaine in Lycium chinense

  • In order to facilitate the quality control of betaine from the fruits of Lycium chinense, we have developed a rapid and simple method for quantitative determination. Determination was achieved on a Discovery C18 column with an isocratic solvent system of 0.32% perfluoropentanoic acid aqueous-acetonitrile at a flow-rate of 0.5 mL/min and detected an ELSD. The method was reproducible with intra- and inter-day variations of less than 6% (R.S.D). The recoveries were in the range of 90.01~100.05%. The method turned out to be fast and simple, furthermore, to have a good selectivity and sensitivity for the quantity determination of betaine in the fruits of L.chinense.

  • Further investigation of Lycium chinense fruits gave a mixture of (6'-O-palmitoyl)- and (6'-O­stearoyl)-$\beta$-sitosterol-3-O-$\beta$-D-glucopyranoside (1) and two glycolipids, 1-O-(9Z, 12Z, 15Z-octa­decatrienoyl)-2-O-(9Z, 12Z, 15Z-octadecatrienoyl)-3-O-$\beta$-D-galactopyranosyl glycerol (2) and 1-O-(9Z, 12Z-octadecadienoyl)-2-O-(9Z, 12Z, 15Z-octadecatrienoyl)-3-O-$\beta$-D-galactopyranosyl glycerol (3). These compounds were newly isolated as constituents of L. chinense.

구기자 추출물과 생과즙의 간독성 보호효과 (Hepatoprotective Effects of Lycium chinense Mill Fruit Extracts and Fresh Fruit Juice)

  • 국내에서 선별 육종된 불로 구기자의 처리방법에 따른 간기능 보호효과를 측정하기 위하여 사염화탄소$(CCl_4)$ 투여로 급성 간독성을 유발시킨 흰쥐를 대상으로 연구하였다. 건조구기자 추출물(DFE), 볶은 구기자 추출물 (RFE), 그리고 생과즙(FFE)을 경구 투여한 결과 군간의 체중증가량은 차이를 나타내지 않았다. 한편, 체중 100 g 당 간 무게는 처리조건에 관계없이 구기자 추출물 및 생과즙 투여군이 유의적으로 낮은 간 무게를 나타내었다(p<0.01). 이것은 구기자 추출물과 생과즙 섭취가 $CCl_4$ 투여에 의한 지방간 형성 또는 간 독성에 의한 간 무게증가를 다소 완화하여준 효과를 나타내는 것으로 판단된다. 사염화탄소$(CCl_4)$ 투여에 의하여 흰쥐의 혈청 GPT 및 GOT 활성과 LDH 활성은 유의적으로 증가하였으나 볶은 구기자 추출물 투여군(RFEC)과 생과즙 투여군(FFJC) 모두 GPT, GOT 활성이 유의적으로 감소되었다. 혈중 총 콜레스테롤 함량은 구기자 추출물과 생과즙 투여군 모두 대조군(CON)과 비교하여 유의적인 증가를 나타내지 않았고 중성 지방함량은 대조군에 비해 유의적인 증가를 나타내었다(p<0.05). 간 조직의 MDA 함량과 GST, cytochrome P-450 활성 결과로 보아 구기자 추출물과 생과즙을 섭취 시 사염화탄소에 의한 독성을 완화시키는 것으로 나타났다. 따라서 이러한 모든 결과를 종합해 볼 때, 구기자 추출물과 생과즙은 $CCl_4$ 투여로 인한 지질 과산화, 지방변성, 간 세포 괴사 등을 억제 시키는 간 독성 보호 효과가 있는 것으로 생각된다.

흰쥐에 있어서 구기자 알콜 추출물이 Oxygen Free Radical 및 Alcohol 대사효소 활성에 미치는 영향 (Effect of the Ethanol Extract of Lycium chinense on the Oxygen Free Radical and Alcohol Metabolizing Enzyme Activities in Rats)

  • To investigate an effect of the ethanol extract of Lycium chinense(EELC) on the activities of enzymes scavenging oxygen free radicals or detoxicating alcohol. The ground Lycium chinense was extracted with 30% edible ethanol and then diluted with 6% ethanol to contain 2% EELC(w/v). Three different groups of male Sprague-Dawley rats had taken a drink EELC, ethanol(ETH) or water(control), respectively for 2 months. At the end of experimental period, the animals were sacrificed and obtained the following findings. The EELC-treated animals showed the highest activity of hepatic glucose-6-phosphatase among three groups. The activities of xanthine oxidase and cytochrome p-450 from EELC treatment group were lower than those from ETH-treated group. However, the activity of superoxide dismutase was higher in the EELC-treated group than the ETH-treated(p<0.005). Furthermore, hepatic alcohol or aldehyde dehydrogenase activity, and glutathione content and glutathione peroxidase were significantly higher in EELC-treated animals than in ETH-treated those. The activity of glutathione S-transferase in liver was appeared the orderly higher value in EELC, ETH and control-treated group. As the result, EELC may affect the reduction of oxygen free radical production and help the detoxication of ethanol.

젖산균 발효 구기자(Lycium chinense ) 추출물의 항염증 및 항노화 효능 연구 (Anti-inflammation and Anti-aging Effect of Lactic Acid Bacteria Fermented Lycium chinense Extracts)

  • 본 연구에서는 발효 과정을 거친 구기자(Lycium chinense) 추출물을 이용하여 항염증 및 항노화 관련 기능성 화장품 소재로서의 활용가치를 검증하고자 nitric oxide (NO) 및 항노화 관련 인자들의 억제 효능을 분석하였다. 물과 70% 에탄올을 용매로 구기자를 추출, 발효, 농축, 동결건조하여 시료를 제조한 다음, MTT assay, NO inhibition activity assay, western blot assay, UPLC 분석을 진행하였다. MTT assay를 통해 세포독성을 측정한 다음, 500 ㎍/mL의 농도를 세포실험에 적용하여 진행하였다. NO 억제 효능의 분석 결과, 발효 구기자 열수 추출물(FLW), 발효 구기자 70% 에탄올 추출물(FLE) 각각 47.96%, 56.71%의 우수한 NO 억제 효능을 나타내었다. Western blot을 통해 주름 관련 단백질인 MMP-1, TRPV-1의 발현양상을 측정한 결과, FLW, FLE 모두 MMP-1, TRPV-1에 대한 발현 억제 효능을 나타내었다. 따라서 발효 구기자 추출물은 항염증 및 항노화 관련 기능성 화장품 소재로서의 높은 활용 가치가 있을 것으로 판단된다.

  • 두 품종 (청양재래, 명안)의 구기자나무 (Lycium chinense Mill.) 유식물체를 자엽하배축의 자엽 일부분만 남기고 절단 후, 절단면이 1.0 mg/L zeatin이 첨가된 MS배지에 접하도록 하여 암소에서 3주 배양한 후 명소로 옮겨 배양하였을 때 자엽하배축 절단면으로부터 부정아가 고빈도 (80% 이상)로 유도되었다. 그러나 자엽하배축절편을 암소에서 전 배양하지 않고, 명소에서만 배양 하였을 경우 부정아는 유도되지 않았다. 자엽하배축 절단면에서 유도된 부정아를 분리하여 1.0 mg/L IAA가 함유된 발근배지에서 4주간 배양하여 뿌리를 유도하였다. 잎과 뿌리가 잘 발달한 소식물체는 화분에 옮겨 키웠을 때 정상적인 형태의 재분화 식물체로 성장하였다.

구기자 세척기 개발을 위한 구기자의 세척특성 (The Washing Characteristics of Lycium chinense Miller with Different Washing Methods)

  • This study was conducted to define the optimal Lycium chinense miller washing method for developing the Lycium chinense cleaner and we analyzed the Lycium chinense miller washing characteristics for removing pesticides and microorganism according to washing methods; habitual washing method, air bubble washing method and nozzle spray washing method. The results were summarized as follows; 1. In case of measuring physical properties according to the varieties, maximum yield strength of Hokwang was 2.562 kgf, minimum yield strength of Hokwang was 0.269 kgf and average yield strength was about 1 kgf. 2. In case of measuring change of bacteria according to washing methods, the number of bacteria of non-washing method was more than the number of bacteria of habitual washing method or mechanical washing method and the number of nozzle spray washing method was least. 3. Ahjoksiseuteurobin of 0.218 ppm was detected in the untreated sample, 0.051 ppm was detected in the habitual washing method, 0.047 ppm was detected in the air bubble washing method and 0.034 ppm was detected in nozzle spray washing method. Every amount detected were less than 2 ppm that is reference value and the detected amount was least in case of nozzle spay cleaning method. Cypermethrin of 0.772 ppm was detected in the non-cleaned sample, 0.089ppm was detected in habitual washing method, 0.26 ppm was detected in the air bubble washing method and 0.292 ppm was detected in the nozzle spray washing method. Every detected amount of Cypermethrin were less than 5 ppm that is reference value and the detected amount was least in case of habitual washing method.

양돈용 사료 첨가제 개발을 위하여 구기자 부산물로부터 메탄올수용액을 이용한 총 폴리페놀 추출조건 최적화 (Optimization of Aqueous Methanol Extraction Condition of Total Polyphenol from Spent $Lycium$ $chinense$ Miller to Develop Feed Additives for Pig)

  • This study was conducted to develop a functional feed additive for pig with spent $Lycium$ $chinense$ Mill fruit. We investigated the optimum conditions for the extraction of polyphenol from spent $Lycium$ $chinense$ Mill using methanol. Methanol concentration as a solvent for extraction, extraction time and the volume of solvent per a gram of solid (ground spent Lyceum chinense Mill) were selected as parameters. Three levels of parameters were configured according to Box Behnken experiment design, a fractional factorial design, and total 15 trials were employed. Total polyphenol concentration from each trial was used as response from experiment system and effects of parameters on total polyphenol extraction efficiency were determined using response surface model. As a result, all terms in analysis of variance, regression ($p$ = 0.001), linear ($p$ = 0.002), square ($p$ = 0.017) and interaction ($p$ = 0.047) was significant and adjusted determination coefficient ($R^2$) was 94.7%. Total polyphenol extraction efficiency was elevated along increased methanol content and decreased solvent to solid ratio. However extraction time did not affect the efficiency. This study provides a primary information for the optimum extraction conditions to maximize total polyphenol recovery from spent Lycium chinens Mill fruit and this result could be applied to re-use of argo-industrial by-products and to develop of functional feed additives in organic farming.
