After cell removal from fermentation broth the supernatant containing poly (${\gamma}-glutamic acid$) (${\gamma}-PGA$) was concentrated by ultrafiltration in order to reduce the amount of organic solvents such as methanol, ethanol, propanol required for the recover of ${\gamma}-PGA$ with precipitation. The concentration and volume reduction of cell-free solution by ultrafiltration could reduce 3 times the amount of ethanol for the recover of ${\gamma}-PGA$ from cell-free fermentation broth.

The poly(3-hydroxybutyrate) (PHB) synthase of Ralstonia. eutropha, which was produced by a recombinant strain E. coli and purified in one-step with a methyl-HIC column to a purity of more than 90%, was used to polymerize 3-hydroxypropionyl-CoA (3HPCoA) and to copolymerize 3HPCoA with 3-hydroxybutyryl-CoA (3HBCoA) in vitro. A $K_m$ of $189\;{\mu}M$ and a $k_{cat}$ of $10\;sec^{-1}$ were determined for the activity of the enzyme in the polymerization reaction of 3HPCoA based on the assumption that the dimer form of PHB synthase was the active form. Free coenzyme A was found to be a very effective competitive inhibitor for the polymerization of 3HPCoA with a $K_i$ of $85\;{\mu}M$. The maximum degree of conversion of 3HPCoA to polymer was less than 40 %. In the simultaneous copolymerization reactions of these two monomers, both the turnover number for the copolymerization reaction and the maximum degree of conversion of 3HPCoA and 3HBCoA to copolymers increased with an increase in the amount of 3HBCoA in the monomer mixture. However, the maximum conversion of 3HPCoA to a copolymer was less than 35 % regardless of the ratio of 3HPCoA to 3HBCoA. Block copolymers were obtained by the sequential copolymerization of the two monomers and these copolymers had a much narrower molecular weight distribution than those obtained by the simultaneous copolymerization of the same molar ratio of 3HPCoA and 3HBCoA.

Structually organized mono- and multilayers were developed on gold for the catalytic and affinity biosensing using hyper-branched dendrimers. For the catalytic biosensing interface, a new approach to construct a multilayered enzyme film on the electrode surface was developed. The film was prepared by layer-by-layer depositions of dendrimers and periodate-oxidized glucose oxidase. The voltammograms obtained from the GOx/dendrimer multilayered electrodes revealed that bioelectrocatalytic response is directly correlated to the number of deposited bilayers. From the analysis of voltammetric and ellipsometric signals, the coverage of active enzyme per layer during the layering steps was estimated, demonstrating the spatially-ordered multilayer formation. As an extension of the study, dendrimers having various degrees of ferrocenyl modification were prepared and used. The resulting electrodes were electrochemically characterized, and the density of ferrocenyl groups, active enzyme coverage, and sensitivity were estimated. For the affinity-sensing surrface, a biosensor system based on avidin-biotin interaction was developed. As the building block of affinity monolayer, G4 dendrimer having partial ferrocenyl-tethered surface groups was prepared and used. And the biotinylated and electroactive dendritic monolayer was used for the affinity-sensing surface interacting with avidin. Electrochemical characterization of the resulting biosensor was conducted using free enzyme in electrolyte in terms of degree of surface coverage with avidin and subsequent surface shielding.

Thermophilic bacterium, Bacillus stearothermophilus NUB3621, was engineered to produce ethanol from glucose by introducing cloned thermostable pyruvate decarboxylase and alcohol dehydrogenase genes. A novel promoter sequence was screened and used for the enhancement of these two enzymes. Successful redirection of metabolic flux into ethanol was obtained. In addition, gene expression profiling using Bacillus subtilis DNA microarray was analyzed to overcome the intrinsic low glucose utilization of B.stearothermophilus. Many known and unknown genes were identified to be up or down regulated under glucose-containing media.

본 연구에서 현미의 발아에 키토산을 활용함으로써 곰팡이 발생을 억제하고 기능성 생리활성 물질인 GABA의 생성을 증진시킬 수 있음을 확인하였다. 100 ppm 키토산액에서 72시간 발아는 현미파우더 그램당 1,035 nmole의 GABA를 보유하게 되어, 발아되지 않은 현미의 136 nmole, 물침종 발아 현미의 771 nmole, 100 ppm 젖산침종 발아 현미의 728 nmole GABA에 비하여 높은 것으로 조사었다. 발아에 의한 GABA 생성증진과 더불어 알라닌의 생성증진 및 글루탐산, 아스파틱산, 세린의 함량 감소가 뚜렸하였다. 키토산처리에 의한 GABA의 생성 증진은 GABA 생성에 관여하는 글루탐산 탈탄산효소의 활성증진이 기여한 것으로 조사되었다. 키토산액에서의 침종 발아는 물침종 발아나 젖산침종 발아에 비하여 곰팡의 발생빈도를 현저하게 낮추었다. 이들결과를 종합할 때 발아현미 제조시 키토산액을 사용하면 기능성 물질인 GABA 함량이 증진되고 곰팡이등의 잡균의 오염을 줄일 수 있어 양질의 발아현미를 얻을 수 있을 것으로 기대된다.

Chlorella vulgaris를 이용하여 기본배지에서 초기 주입농도 $0.7{\times}10^4\;cells/mL$ 상태에서 300시간 배양 후 $1.21{\times}10^5\;cells/mL$의 생체수를 얻었으며, 초기 cell 농도가 0.30g/L인 경우가 성장률이 $0.162g/L\;{\cdot}\;day$로 우수하였다. Glucose 초기농도가 2.00g/L인 경우 초기 cell 농도 0.025g/L에서 140시간 후 0.874g/L이 되었으며 비성장속도는 $0.243h^{-1}$을 나타내었고, 72시간내에 가스를 268mL를 생성하였으며 이후로는 수소 생성이 종료으나 순수 소소를 19.87mol% 함유하고 있었다. Chlorella vulgaris의 조성을 원소분석기로 분석한 결과 $C_{1.000}\;H_{1.774}\;N_{0.125}\;O_{0.557}$가 되었으며 이산화탄소 전환율은 $0.616cells/\;g\;CO_2$가 됨을 알 수 있었다.

A novel, continuous bioreactor system combining a bubble column (absorption section) and a two-phase bioreactor (degradation section) has been designed to treat a gas stream containing benzene. The bubble column contained hexadecane as an absorbent for benzene, and was systemically chosen considering physical, biological, environmental, operational and economic factors. This solvent has infinite solubility for benzene and very low volatility. After absorbing benzene in the bubble column, the hexadecane served as the organic phase of the two-phase partitioning bioreactor, transferring benzene into the aqueous phase where it was degraded by Alcaligenes xylosoxidans Y234. The hexadecane was then continuously recirculated back to the absorber section for the removal of additional benzene. All mass transfer and biodegradation characteristics in this system were investigated prior to operation of the integrated unit, and these included: the mass transfer rate of benzene in the absorption column, the mass transfer rate of benzene from the organic phase into the aqueous phase in the two-phase bioreactor, the stripping rate of benzene out of the two-phase bioreactor, etc. All of these parameters were incorporated into model equations, which were used to investigate the effects of operating conditions on the performance of the system. Several experiments were conducted to show the feasibility of this system. This process is believed to be very practical for the treatment of high concentrations of gaseous pollutants.

Various analytical methods such as electron microscopy, quinone analysis, and 16S rDNA sequencing studies were used to investigate the microbial communities and to identify the microorganisms responsible for enhanced biological phosphorus removal (EBPR) in an anaerobic/aerobic sequencing batch reactor (SBR) fed with acetate. Electron photomicrographs showed that oval-shaped microorganisms of about $0.7\;{\sim}\;1\;{\mu}m$ in diameter dominated the microbial sludge. These microorganisms contained polyphosphate granules and glycogen inclusions, which suggests that they are a kind of phosphorus accumulating organism. Quinone and 16S rRNA sequence analyses showed that the members of Proteobacteria beta subclass were the most abundant species, which were affiliated with the Rhodocyclus-likes group. Phylogenetic analysis revealed that the two dominating clones of the beta subclass were most distantly related to Propionivibrio dicarboxylicus DSM 5885 and Rhodocyclus tenuis DSM 109 with about 95% and 96% sequence similarity, respectively. Therefore, it was concluded that the oval-shaped organisms related to the Rhodocyclus-likes group are likely to be responsible for biological phosphorus removal in SBR operation supplied with acetate.

Biosensors consist principally of two basic components, receptor and transducer. The transducer part of the sensor serves the signal transfer from the output domain of the recognition system. Optical method of transducer have many benefits, based on phenomena like emission and absorption. This paper will review the methode of optical transduction in brief and out some new development.

미세 가공 기술을 이용하여 제작된 IDA 전극을 활용하여 임피던스 측정방식의 cell chip으로의 응용에 대해 고찰하였다. IDA 전극은 기존의 반도체 공정으로서 손쉽게 제작할 수 있고, 대량 제작시 전극의 재현성 확보가 용이하고 소형화 할 수 있으며 낮은 단가로 제작될 수 있는 장점이 있다. IDA 전극을 채용한 cell chip을 B16-F1 melanoma 세포 배양에 적용한 결과, 세포성장과 임피던스 변화량이 상관성을 보였고, 세포의 성장을 저해하는 약물의 투과시 cell chip의 임피던스 변화 역시 기존의 방법과 유사한 결과를 보여 주었다.

Biological science is being revolutionized by the availability of much sequence information from many genome project With the advanced technology at hand, main trend in biological research is rapidly changing from a structural DNA analysis to understanding cellular function of the DNA sequences. Combined with mechanics, computer, bioinformatics and other advanced technologies, DNA chip technology provides numerous applications because of its robustness, accuracy, and automation. DNA chip is expected to become an indispensable tool in fields of biology, biotechnology, drug discovery, and other application areas. DNA chip can be used for mutation and polymorphism detection, gene expression monitoring and phenotypic analysis as well. If DNA chip is used for the development of pharmaceutical products, it can considerably reduce the cost and time for the entire process of drug discovery and development, and can also contribute in developing personal drugs.

Bio-electronics has been considered as one of the most appropriate candidates to overcome the frequently encountered problems in the development of future electronic devices. It has some advantages such as ultra fast electron transfer rate and high-energy efficiency compared with the silicon-based electronic devices. In silicon-based electronics, there are some of limitations of manufacturing process and physical problems. Bio-electronics can overcome the limitation and problem of silicon-based electronics. Bio-electronics has possible application areas as biosensor, biochip, bio-transistor and bio-computer. In the future, bio-electronics can substitute the silicon-based electronics.

A bacterial strain producing a large amount of EPS was isolated from marine sediment sample collected from the Cheju Island, Republic of Korea. In the present study, the isolation and identification of this isolate, which is named Hahella chejuensis gen. nov., sp. nov., the effects of nutrients on the production of EPS, and some properties of this EPS are reported.

Numerous novel marine natural products have been discovered and isolated from varied marine organisms by the diligent bio-prospectors over the past decades. An assessment of the current status of commercial development of these natural compounds indicates only minimal commercialization due to the lack of sustainable supply. To bridge the gaps between discovery and commercialization of these tantalizing bioactive compounds, marine bioprocess engineering is the key for its success. The problems, challenges and opportunities for marine bioprocess engineers are examined for the timely transformation of the discovery into commercial reality. Marine bioprocess engineers will find it the most rewarding practice of their expertise in diving into the ocean.

크로마토그래피 칼럼 검증전용으로 미니 칼럼을 이용한 자동화된 크로마토그래피 시스템(MiniValChrom)을 개발하였다. 이 시스템은 생산용 칼럼을 scale-down한 미니칼럼을 자동적으로 반복사용 하면서 실험실 내에서 ex-situ로 크로마토그래피 공정의 검증을 수행할 수 있게 설계되었다.

배큘로바이러스와 actinomycin D에 의해 유도되는 곤충세포 apoptosis의 누에체액에 의한 저해 효과에 대해 연구하였다. 감염 전 또는 감염 중, 배양 배지에 누에체액을 첨가함으로써 배큘로바이러스에 의해 감염된 숙주 세포의 생존율을 높은 수준으로 유지시켜 주었다. 누에체액은 또한, RNA 합성 저해제인 actinomycin D에 의해 유도된 apoptosis 역시 저해시켰다. 이러한 효과들은 TUNEL assay와 flow cytometry로 확인하였다. 추가적으로, 누에체액은 감염 후 세포 내 유전자의 발현에는 영향을 미치지 않으나 바이러스 유전자의 발현을 증진시킴을 알 수 있었다.

Angelica gigas Nakai 뿌리배양을 통한 decursinol angelate생산을 증대시키기 위하여 여러 가지 elicitor들을 처리하여 생산 증진을 도모하였다. 가장 효과적인 elicitor로는 hydrogen peroxide와 $CM-\;{\beta}\;-chitin$이었으며, 이때의 g당 생산되는 decursinol angelate 양은 각각 대조구에 비해 2.12, 1.62배 증대된 값이다.

유가식배양을 통한 고농도배양에서 계면활성제와 산소전달물질의 첨가는 세포의 고밀도와 배지의 고점도로 인해 발생하는 nutrient의 물질전달제한과 산소부족현상을 효과적으로 해결하였다. Pluronic 10R-5는 Pluronic F-68과 같은 triblock copolymer 구조이며 세포생장의 저해를 나타내지 않았다. 그러나 diblock copolymer구조이며 소수성 부분이 hydrocarbon block인 Plurafac A-38은 세포의 생장을 저해함을 관찰할 수 있었다. 따라서 장기간의 유가식배양이나 perfusion 배양을 수행할 경우 세포의 생존도를 증대시켜 유용물질의 생산성을 높일 수 있으며 여러 형태의 bioreactor에서 고농도 배양을 가능케 하여 유용물질 생산성을 높일수 있을 것이라고 기대된다.

본 연구에서는 천연피혁제조공장에서 발생하는 대표적인 악취성분인 황화수소, 암모니아 및 benzene을 활성슬러지에서 획득한 혼합균주와 황산화균인 Thiobacillus sp.IW가 동시에 접종된 생물막반응기를 이용하여 효과적으로 제거하고자 하였다. Benzene의 경우 단일 및 복합성분의 악취에 대해 90%의 제거효율을 유지하였고, 암모니아는 99%의 아주 우수한 제거효율을 나타내었으나, 황화수소의 경우 처리효율은 85%정도로 다른 성분에 비해 낮았다.

B. Stearothermophilus 단백질 분해 효소는 2가 금속 이온인 $Ca^{2+}$에 의하여 내열성이 향상되며 $75^{\circ}C$에서 최대 역가를 나타내었다. 2mM $Ca^{2+}$를 첨가하여 B. Stearothermophilus를 이용한 호열 ${\cdot}$ 호기성 소화공정 운전할 경우 각각 최대량 대비 51%와 27%의 DOC 및 세포외단백질의 분해가 이루어지는 것을 확인하였다.

본 연구는 용존산소의 양에 따른 MnP의 생산양상을 체계적으로 연구하였으며, 과량의 산소 공급 시 MnP 생산이 저해될 수 있음을 산소공급에 따른 과량의 $H_2O_2$생산 측면에서 설명하고 있다. 또한 보다 높은 MnP 생산을 위한 산소공급방법을 제시하고자 하였다.

Biodegradability of the $poly({\beta}-hydroxybutyrate-co-3 -hydroxyvalerate)$ [PHB/V] containing 0, 10 and 15mol% hydroxyvalerate [HV] was studied. Bioderadability of PHB/V was evaluated at $30^{\circ}C$ for 58 days and $55^{\circ}C$ for 33 days by monitoring the time-dependent changes in weight loss(erosion) of aerobic conditions in a temperature-controlled microcosms containing the earthworm cast($30^{\circ}C$) and compost ($55^{\circ}C$). It was found that PHB/V biodegradability occurred with increasing HV monomer concentration from 0 mol% to 15 mol%.

Toluene-degrading bacterium, Microbacterium esteraromaticum CS3-1 was isolated from the biofilter for the removal of BTEX. Microbacterium esteraromaticum CS3-1 was shown to utilize toluene as a primary carbon and energy source. Effect of mixed BTEX gases on toluene degradation rate by M. esteraromaticum CS3-1 was investigated in this study. Toluene degradation rate was 2.26(only toluene), 2.06(toluene+benzene), 2.57(toluene+ethylbenzene), and 4.74(toluene+xylene) mmole $toluene\;{\cdot}\;g-DCW^{-1}\;{\cdot}\;h^{-1}$. Toluene degradation rate was 2.26(only toluene), 1.23(toluene+benzene+ethylbenzene), 1.52 (toluene+ethylbenzene+xylene), and 1.76(toluene+benzene+ethylbenzene+xylene) mmole $toluene\;{\cdot}\;g-DCW^{-1}\;{\cdot}\;h^{-1}$. The presence of BTEX compounds over three mixtures had a negative effect on toluene degradation rate. Toluene degradation rates were enhanced by the presence of ethylbenzene or xylene, whereas the presence of benzene had a negative effect on toluene degradation rate in comparison with toluene degradation rate when only toluene is existent.

Recombinant E. coli JM109(pZH3-5/pMT) harboring manganese transport gene(mntA) and metal sequestering protein, metallothionein(MT), was cultivated to accumulate cadmium in aqueous phase. Bioaccumulation followed Michaelis-Menten type kinetics. Equilibrium isotherm showed Langmuir type isotherm. IPTG induction cell showed fast $Cd^{2+}$ uptake and had higher uptake rate than wild type and no-induced cell. The optimum pH and temperature for $Cd^{2+}$ uptake was 7 and $37^{\circ}C$, respectively. Manganese (0.01M) inhibited the $Cd^{2+}$ accumulation. However, $Cu^{2+}$, $Zn^{2+}$ and $Pb^{2+}$ did not affect the $Cd^{2+}$ bioaccumulation.

본 연구는 5가지의 발광성 미생물을 이용하여 유해 방사선으로 알려져 있는 ${\gamma}-rays$가 여러가지 cellular stresses 중, 특히 유전자 손상과 생물막 손상을 유발하였는데, 이들의 손상 정도가 총 방사선량과 상관관계가 있음을 발생하는 bioluminescence 로써 확인하였다. 뿐만 아니라, 선량률의 변화를 통하여 방사선으로 인한 유전자 손상 및 일반적인 독성 효과가 큰 영향을 받는 것을 확인하였는데, 선량률 증가에 따라 이들 손상정도가 증가하는 것으로 보아 선량률이 genetic 및 radioprotecion에 심각한 영향을 미치는 것을 확인하였다.

B. macerans 유래의 CGTase를 yeast surface display기술을 이용하여 S. cerevisiae의 표면에 발현된 것을 halo-test와 immunofluorescence microscopy와 flow cytometry를 통하여 확인하였다. 재조합 효모는 효소의 cyclization작용을 저해하고 CD의 분해작용을 촉진하는 glucose와 maltose를 제거하는 발효공정과 표면 발현된 CGTase의 cyclization 공정을 동시에 수행할 수 있어 CD의 생산, 분리공정을 효율적으로 개선하였다.

The color of textile-wastewater hindered spectrometric measurements of $H_2O_2$ and enzyme activity during enzymatic decoloring. By using ABTS, we developed a new method for measuring peroxidase activity and $H_2O_2$ concentration. The ratio of enzyme and $H_2O_2$ was optimized as 1:150 by investigating the effects of $H_2O_2$ on enzymatic decoloring. Pulse feeding of $H_2O_2$, upon depletion, significantly increased the decoloring of Congo Red.

The capacity of micelle formation between astaxanthin and various surfactants was compared. Tween 20 was identified the most suitable surfactant in terms of astaxanthin extraction capacity. The ethylene oxide group of Tween 20 was identified as the most significant factor to increase the HLB value that determined the extraction capacity. The effect of micelle formation condition, such as molar ratio of astaxanthin and Tween 20, pH and ionic strength was also investigated. pH and ionic strength showed no significant effects. Antioxidant activity of astaxanthin was twice of ${\alpha}-tocopherol$ and 4 times of ${\beta} -carotene$. Crude astaxanthin extract from the yeast cell seemed to be less degraded than pure astaxanthin by air and light exposure, probably because of the presence of other carotenoids and lipids. Under the optimal conditions, the molar ratio of micelle formed was found to be 1 : 12 for astaxanthin : Tween 20.

A gene coding $endo-{\beta}$,-1, 4 glucanase from Actinomyces sp. KNG40 and phytase from Hansenula polymorpha were cloned into Esherichia coli JM101 by using E. coli/Lactobacillus shuttle vector pNZ3004 and pNZ123. The plasmid p3PS(1-4) and p123(1-4) have slpA promoter and slpA signal sequence. So, I constructed expression vectors, p3PS(1-4)Endo, phy and p123(1-4)Endo, phy. These constructed vector was transformed in target host Lactobacillus gasseri and reutri. These transformed host expressed endoglucanase and phytase as extracellular fraction. In the enzyme activity of the same vector, host L, gasseri was higher activity than L. reuteri. This indicates that L. gasseri recongnize promoter and signal sequence very well.

스크린프린팅 된 탄소전극과 항체가 결합된 리포좀을 이용한 일회용 전기화학발광 면역센서를 구성하였다. 분석물질농도에 비례하여 분석물질-면역리포좀 복합체는 모세관현상에 의해 membrane strip에서 전극 상으로 전달되었고, 이 리포좀은 계면 활성제에 의해 파괴되고 신호발생에 이용되는 ruthenium이 방출되었다. 이와 같은 면역센서의 성능은 의료현장검사 혹은 자가진단에 적합할 것으로 예측된다.

본 연구를 통해 환경 유해성을 평가하기 위한 동물세포를 개발했고 이를 이용한 모니터링 방법 연구와 다양한 독성 물질에 대한 반응성을 확인했다. 개발된 독성 모니터링 동물세포(KFC-A10)는 각 독성 물질에 따라 빛의 발현 양이 증가하는 성향을 가지므로 이번 실험에서 사용된 MMC, BPA, ${\gamma}-ray$에 농도 의존적으로 빛의 양이 증가함을 관찰할 수 있었다. 특히 BPA의 경우는 환경호르몬으로 알려진 바 그 estrogenic 효과를 관찰할 수 있었다.

토양오염의 독성을 탐지하기 위해 재조합 발광 박테리아의 고정화를 이용하여 바이오 센서를 제작하였으며 이를 이용하여 대표적 토양오염물질인 PAHs의 독성을 측정할 수 있었다. 또한, 이 바이오 센서를 이용하여 토양오염처리 전후의 처리 효율을 신속히 탐지할 수 있음을 확인하였다.

목질계 바이오매스의 아세톤상 가용매분해는 목재의 구성성분인 셀룰로오스, 헤미셀룰로오스, 리그닌중에 가장 전환율이 낮은 리그닌의 전환율을 증가시킴으로 전체적인 목재의 전환율의 증가에 기여하였다 그러나 $300^{\circ}C$ 이상에서는 리그닌의 전환율이 감소하였다. 아세톤의 첨가에 관계없이 셀룰로오스와 헤미셀룰로오스의 전환율의 변화는 온도에 의존하기 때문에 주어진 열분해 온도에 따라 생성물을 선택적으로 얻을 수 있었다. 자일란의 경우 $250^{\circ}C$ 이하에서 전환되고 셀룰로오스의 경우 $250^{\circ}C$이하에서는 거의 전환되지 않지만 $250^{\circ}C$와 $300^{\circ}C$ 사이에서 급격한 전환율의 증가가 나타났다. 생성물은 주로 방향족류와 페놀류이며, 특히 리그닌의 아세톤 상 가용매 분해의 경우 $300^{\circ}C$에서의 메톡시 페놀류가 가장 많이 얻어졌다. $400^{\circ}C$이상의 반응에서는 메톡시기가 없는 methyl-, dimethyl-, trimethyl-, phenol 류가 생성되는 것으로 나타났다. 이것은 메톡시기를 제거시킬 만한 에너지가 $350^{\circ}C$ 이상에서 공급되는 것으로 사료된다.

Sophorolipid, a glycolipid type biosurfactant produced from Candida Bombicola, inhibited the growth of bacteria and fungi. Between two types of sophorolipid, acid and lactone form, the latter had more strong antimicrobial activity. Culturing parameters including oil substrate, initial glucose concentration, aeration, culture mode, affected the ratio of two forms. The fatty acid moety varied depending on the oil having different fatty acid chains. Minimum inhibitory concentration of lactonic sophorolipid toward Propionibacterium acne was below 0.5ppm. This result indicated the potentials of sophorolipid as antimicrobial agent in various area including cosmetics.

본 연구에서는 균주의 배양과 동시에 생산물을 회수할 수 있는 동시추출공정을 건조질량의 $15{\sim}75%$의 탄화수소를 생산한다고 알려진 B. braunii 배양에 적용하고자 한다. 일반적인 two-phase 동시추출공정의 적용시 B. braunii의 경우 생산된 탄화수소가 균주 외벽의 matrix에 강하게 부착되어 있기 때문에, two phase 추출공정 적용시 bubble column내에서 단지 폭기에 의한 교반만으로는 충분한 탄화수소의 회수율을 얻을 수가 없었다. 본 연구에서는 배양액과 유기용매층의 접촉기회를 증대시킨 two-stage 동시추출 공정을 개발하여 기존의 two-phase 동시추출 공정보다 2배 이상 높은 56.2%의 탄화수소 회수율을 얻을 수가 있었다.

Bacillus thuringiensis (bt)는 생물학적 살충제 시장에서 가장 널리 사용되는 미생물 살충제이다. Bt의 고농도로 세포배양을 하면 생물반응기에서 원하는 생산물의 농도를 이룰 수 있을 뿐만 아니라 생산물의 생성속도도 이룰 수 있다. 이런 목적 하에서 우리는 세라믹 막 모듈을 반응기 내부에 장착한 새로운 형태의 생물반응기를 개발하였다. 이 새로운 형태의 생물반응기에서 포도당 제한 상태하의 운전을 통해서 bt의 세포와 포자 수율을 현저하게 증가시킬 수 있었다. 최대로 $1.2\;{\times}\;10^{10}\;CFU/ml$의 포자 수율을 달성할 수 있었다. 세라믹 막 모듈 생물반응기에서 포도당 제한 상태 하에서 bt의 성장특성을 연구하였다. 포도당 제한 상태에서는 세포의 성장은 선형 (linear) 성장특성을 보였는데 이것은 간단한 비구조적 수학적 모델의 결과와 잘 일치하였다.

회분식 발효를 실시하여 균주의 최적 발효조건을 구하여 세포 재순환식 연속 발효 공정에 적용한 결과, CSL의 최적 함량은 2.5%로 나타났으며, 수율과 생산성은 각각 0.80g total acid/g glucose, 0.26g total acid/L/h이였다. 연속 발효 결과, 최대 생산물 생산성은 희석률 0.2/h에서 3.32g organic acid/L/h을 얻었다. 최적 추출시스템은 30%w/w TOA/MIBK로 나타났으며, Membrane Contactor을 이용한 추출에서는 수용액상의 200ml/min, 추출상은 100ml/min의 유량을 갖는 Counter-current가 최적 조건임을 확인했고, 또한 수용액상과 추출상의 접촉 부피비는 2:1이 여러 가지 요인을 고려했을 때 최적 조건임을 확인했다. 실제 발효를 통해 생성된 유기산의 추출은 1:1의 부피비로 counter-current로 접촉시켰을 때 Acetic acid 가 56% Propionic acid가 72.41%의 추출력을 얻을 수 있었다.

CO로부터 수소를 생산할 수 있는 미생물인 R. palustris P4를 반응기에 적용하여 연속적으로 수소를 생산하는 연구를 진행하였다. 고농도 배양을 위해 R. palustris P4를 호기적 조건에서 영양요구성 성장을 시켰고 그 결과 13 g/L정도의 높은 균체 농도를 얻을 수 있었다. CO는 물에 잘 녹지 않는 기상의 기질이므로 반응기내에서 기체의 충분한 체류시간을 제공할 수 있는 반응기인 TBR을 수소생산 단계에 적용하였다. 기체 체류시간이 50분으로 운전되는 조건에서 CO의 분압이 0.4 atm일 때 최대 CO 소모 속도가 16 mmol/L/hr였다. 또한 광합성 미생물 R. rubrum을 적용하여 TBR에서 수소생산을 시도한 Gaddy group의 수소생산속도 3-4 mmol/L/hr에 4-5배 높은 수소생산 속도이다(3).

Agrobacterium sp.에 의해 생산되는 커들란은 2차 부산물이기 때문에 이의 생산을 증대시키기 위해서는 고농도 세포배양이 필요하다. 본 연구에서는 고농도 세포배양을 실현하기 위하여 탄소원 과 질소원을 제어 공급하는 방법을 이용하였다. 초기 탄소원의 농도가 20g/L일 때 24시간 내에 모두 소모되었으며 생산된 세포의 농도는 6g/L였다. 고농도의 탄소원을 이용하여 배양조내의 설탕의 농도가 10g/L 이상이 되도록 제어하는 방법을 사용하였다.

Antimicrobial activity of sophorolipid produced by Candida bombicola ATCC 22214 was investigated, Minimum inhibitory concentration of sophorolipid to Propionibacterium acne and Bacillus subtilis were 0.5 ppm and 4 ppm, respectively. However no antimicrobial activity to E. coli was detected. Indicating the antimicrobial activity of sophorolipid only toward gram positive bacteria. Antifungal activity of sophorolipid was also detected in the hyphae growth assay for Botrytis cinerea. Malate dehydrogenase, a cytoplasmic enzyme, of B. subtilis was remarkably increased with the sophorolipid treatment, indicating the leakage of membrane by sophorolipid. This result shows the potentials of sophorolipid as mild, non-toxic antimicrobial agent.

We researched effects of growth on initial inoculum size, liquid volume, and medium feeding rate etc. Cell suspension inoculated at low cell concentrations showed a typical growth reduction, whereas root cultures showed an improvement in growth. In this paper, Hairy roots showed high growth rate at 0.4 % inoculum size and 100 mL liquid volume in 250 mL flask cultures.

형질전환된 인삼 모상근의 여러 생물반응기에서의 대량배양 가능성을 알아보기 위하여 1/2 MS 배지를 기본배지(호르몬 무첨가, 3% sucrose)로 하여 실험을 수행하였다. 그 결과 플라스크 배양에 비하여 $2{\sim}3.5$배 높은 성장률을 보였으며, 그 성장이 왕성함을 알 수 있었다. 배양 후반기에 모상근의 뭉치 형성으로 물질전달의 어려움으로 인하여 성장에 장해를 미치는 것으로 사려된다.

혐기성 박테리아균인 Zymomonas mobilis을 이용한 에탄올 발효에서 사용한 균주가 pH변화에 대하여 강한 내성을 지니고 있으므로 pH 변화에 대한 생산성의 변화는 나타나지 않았다. 다만 주입 기질의 농도가 증가함에 따라 생성되는 알코올의 농도도 증가함을 알 수 있으나 생산성을 고려하여 glucose의 농도가 10%일 때 가장 경제성이 있음을 알 수 있었다. 연속발효에서 glucose의 농도를 10%로 하고 CSL을 1.2%로 하여 실험한 결과 Maximum Productivity는 14.06g EtOH/L/hr을 나타냈으며 glucose의 전환율은 0.47g EtOH / g glucose을 나타냈다. 알코올의 분리 농축을 위한 투과증발 실험에서는 공급액의 유속이 482ml/min이고 공급액의 농도가 8%, 온도가 $70^{\circ}C$에서 투과물의 Flux는 $0.04kg/m^2/hr$ 이었으며 Selesctivity는 7.5로 나타났다. 5{\sim}8%$의 희석된 에탄올 수용액을 투과증발실험을 한 결과 7배 이상 농축되었다.

대량발효 후 미세여과막과 MWCO 100K, 50K의 한외여과막을 이용한 막분리시스템을 적용하여 MTGase를 분리한 결과 pore size 1.6, $0.7\;{\mu}m$의 cellulose fiber 재질의 예비여과막과 $0.45\;{\mu}m$ cellulose acetate재질의 미세여과막을 이용하여 얻어진 분리액을 MWCO 100K와 50K를이용하여 농축시킨 결과 enzyme의 농도가 1.29 units/ml, 효소 비활성도는 약 0.2 units/mg protein 으로 나타났으며 초기 배양액에 비해 3.7배의 농축효과를 보였다.

Helicobacter pylori(H. pylori) is the causative agent of chronic gastritis and the single most important factor in peptic ulcer disease, however, the pathogenetic mechanisms underlying H, pylori infection are not well understood. Futhermore, there is a strong association between H. pylori infection and gastric cancer. Various diagnostic methods for detecting H. pylori infection are available. These can be divided into invasive methods, requiring endoscopy, and non-invasive tests, mainly 13C-urea breath tests and serologic detection of antibodies. Rapid urease test is the most recommendable endoscopic test for the diagnosis of H. pylori infection, presently. CLO test kit is the represent of rapid urease test kits. The principles of CLO test kit is that hydrolysis of urea by urease Is detected by a dye indicators showing a color change. Our device is used same principle but we improved the reaction time is more faster and positive color change is more distinctive from the color of the negative specimen. So, this kit is more reliable because it response faster and accuracy.

홍자두 쥬스($20^{\circ}\;brix$)에 Saccharomyces cerevisiae를 5%접종하여 $25^{\circ}C$에서 5일간 발효시켰을 때 에탄올이 가장 많이 생성(약11%)되었고 홍자두 발효주와 증류액, 후모사와 홍자두 각각의 침출주 가운데 홍자두 발효주의 기호도가 제일 높았고 ACE 저해활성, 혈전용해활성 및 항산화활성 등이 가장 우수하였다.

Marine fungus, Thraustochytrium aureum ATCC 34034, was incubated in artificial sea water media in order to produce docosahexaenoic acid(DHA). Cultures were performed at $24^{\circ}C$ in light for 13 days with orbital shaker at 100rpm. Maximum cell productivity of 1.34g/L and DHA yield of 41.4 mg/L were obtained by using this method, which is almost the twice level of DHA yield obtained for the strains reported previously. This strain did produce much more DHA after sporangium disruption.

수소 생산량은 기질로부터 생산될 수 있는 수소의 분자수와 균주간의 특성에 의해서 좌우된다. 광합성 세균에 의한 수소 가스 생산은 유기물질 및 물을 전자 공여체로 하여 광합성에 의해 생산되는 것으로 알려져 있다. 이론적으로 수소생산량은 glucose 한 분자로부터 12분자의 수소가스가 생산된다.

본 연구에서는 fed-batch를 통하여 Agaricus blazei의 균체증식과 다당체 생산을 증가시키고자 공급배지와 배지공급속도에 변화를 주어 균체증식과 다당체 생성에 미치는 영향을 비교 ${\cdot}$ 검토하였다. 유가식 배양에서의 최종균체량 및 다당체 생성량은 회분식 배양에 비해 훨씬 높았으며, 초기에 균체증식을 위해 공급배지로 yeast extract를 먼저 공급하고 4일째부터 다당체 생성을 위해서 glucose를 공급할 경우 균체량은 18.2 g/L, 다당체 생성량은 10.4 g/L를 생산하여 배양결과 최대 생산량을 나타내었다.

회분배양에서 교반속도와 통기량에 대한 영향을 살펴본 결과 400 rpm과 1.0 vvm의 조건에서 균체량은 25.1 g/L, 다당체는 2.5 g/L로 가장 높게 생성되었다. 회분배양을 기초로 균체량과 다당체 생성을 높이기 위해 glucose를 대수기에 공급한 결과 배양 6일째 균체량은 29,2 g/L, 다당체 생성은 3.3 g/L로 회분배양보다 우수한 결과를 보였다.

The production of polyhydroxybutrate(PHB) by Alcaligenes eutrophus NCIB 11599 was studied in a synthetic medium from whey as a sole carbon source. Especially pH-effect was treated and compared in this study. At the end of fermentation (A) unadjusted to pH, the dry cell weight, PHB concentration, and PHB conversion rate were 10.3g/L, 3.1g/L, and 30%, respectively. At the end of fermentation (B) adjusted to $pH(7.0{\sim}7.5)$, the dry cell weight, PHB concentration, and PHB conversion rate were 12.5g/L, 4.8g/L, and 41%, respectively. PHB conversion rate was about 10% higher on the fermentation (B) than on the fermentation (A).

Effect of light intensity on ALA production was, investigated. The culture condition and medium optimization were also examined for the biosynthesis of ALA using Rhodobacter sphaeroides, non-sulfur bacteria, and investigated for enhancement of the production of ALA. In the dark condition, extracellular ALA formation and cell growth were not observed. Optimum light intensity for cell growth and ALA production were 4 kLux and 5 kLux, respectively.

Succinic acid was produced by Enterococcus faecalis RKY1 cells immobilized in hollow fiber bioreactor as an alternatively immobilized culture in bioconversion of fumaric acid to succinic acid. The feed was pumped through the shell side. As the flow rate of the feed was increased, the steady state was obtained more quickly. The steady state was reached after 24 hr cultivation in 0.25 ml/min, 12 hr in 0.5 ml/min, and 9 hr in 1.0 ml/min, respectively. The effect of medium pH on succinate production was also investigated. By changing the medium pH of 8.0, the succinic acid produced was increased about 16% than that of pH 7.0.

Fed-batch culture was designed to increase cell concentration and astaxanthin content by mutant AJ-6 of Phaffia rhodozyma. Fed-batch culture was performed in the continuous feeding with manual adjustment of flow rate to control glucose concentration. When the final glucose concentration was 100 g/L, the cell and astaxanthin were 38.3 g/L, 34.8 mg/L, respectively. Addition of ethanol(10 g/L), when glucose was depleted, the cell and astaxanthin concentration were 37.2 g/L and 45.6, respectively, 5 g/L of acetic acid supplied, 40.6 g/L, 43.9 mg/L were obtained. Ethanol and acetic acid enhanced the astaxanthin content act as precursor of carotenoid synthesis.

A halophilic bacterium was isolated from domestic marine. The bacterium was gram negative and motile. Transmission electron micrograph after cultivating for 6-, 20-, 72-, and 144 hrs showed that it was bacilli and contained intracellular granules, which were pleomorphic, larger in density by the time and considered to be PHB since they were positive on the sudan black B staining. The presence of sodium chloride was critical, because the isolated marine bacterium could not multiply and even produce any immunostimulant in the deficiency of sodium chloride. The strain produced an immunostimulant, which was investigated for the biological characteristics. The optimal conditions for the production of the immunostimulant were 1 % dextrose and 1 % yeast extract in artificial sea water for carbon and nitrogen sources, respectively. The initial pH and growth temperature for the production were 8.0 and $30^{\circ}C$ under the presence of oxigen, respectively.

The effects of initial sugar concentration and the various nitrogen sources on the fermentation characteristics of Thraustochytrium aureum were investigated. The sugar conversion ratios decrease with increase in the initial sugar concentration regardless of the kinds of nitrogen sources. The specific growth rate, the biomass yield, and the DHA yield also decreased but approached constant values higher than the concentration of 30g/L. The sugar conversion ratios and the specific growth rate showed the highest values in case of using the yeast extract as nitrogen source. It was difficult to conclude that the variation of nitrogen source had some effects on the biomass yield. However, the initial sugar concentration had a rather effects on the biomass yield for the sugar concentration of 40 g/L showed 0.27. The nitrogen source that showed the greatest yield of DHA was sodium glutamate.

The optimal temperature and pH for both mycelial growth and exe-biopolymer production by Cordyceps millitaris in shake flask culture were found to be $20^{\circ}C$ and 6.0, respectively. Sucrose (4%) and corn steep powder (1%) were the most suitable carbon and nitrogen source for mycelial growth and exo-biopolymer production. The maximum specific growth rate $(0.142h^{-1})$ was achieved when sucrose was used as the sole carbon source. Exo-biopolymer production was increased with the increase in C/N molar ratio concentration, probably due to the facilitated carbon uptake. Under the optimal culture conditions, the maximum mycelial growth exe-biopolymer concentration were reached to around 13.3 g dry cell weigh/l and 3.33 g/l, respectively.

Monascus purpureus ATCC 16365에 의한 5-liter 배양기에서 pH 조절을 통하여 세포외로 이차대사물인 수용성 적색소의 생산 증가 현상 와 황색소 보다 적색소로의 대사과정을 촉진시켜 적색소의 생산성을 향상시킬수 있는지의 가능성에 대하여 실험을 수행하였다. 황색소와 적색소의 분석은 UV-Vis spectrophotometer를 이용하여 각각 385nm 와 495nm에서 측정하였다. 실험결과 곰팡이의 최대균체량과 황${\cdot}$적색소를 포함한 총 색소 생산양은 pH 8보다 pH 4.0-5.5인 산성조건에서 2배 및 4배정도 증가하였다. 적색소 및 황색소의 전구물질인 홍색소는 pH 4.0에서 많이 생성되며 또한 홍색소에서 적색소로의 대사과정도 산성 조건 하에서 촉진된다고 보고되었다. 본 실험에서도 같은 경향을 보였으며 또한, 산성 조건 하에서 특히 수용성 적색소가 곰팡이 세포 외로 유출되는 양이 증가하는 경향을 나타내었다.

The effects of DO and pH on the mass production of pullulan with high-molecular weight from A. pullulans ATCC 42023 were evaluated. The maximum pullulan production yield (51%) was obtained at pH non control (initial pH 6.5) and DO control (above 50%) condition. The pullulan degrading enzyme was activated when the pH of broth reached lower than 5.0 and portion of low molecular weight pullulan was increased. The formation of a black pigment was observed at the initial stationary phase, 40hr of fermentation. Therefore, the fermentation should be carried out in pH non control and DO control condition and harvested before reaching stationary phase around 40h for the production of high molecular weight pullulan.

A marine bacterium, Pseudomonas aeruginosa BYK-2 KCTC 18012P was immobilized in modified polyvinyl alcohol for the continuous production of rhamnolipids. The stability of rhamnolipids production, the mechanical strength of beads and the scanning electron microscope of immobilized cell were determined in a repeated batch culture. The rhamnolipids production was maintained $80{\sim}90%$ stability of initial production, and the mechanical strength also was stable during the repeated batch culture more than 14 cycles. In the case of SEM studies, the internal distribution pattern of the cell entrapped in modified PVA beads was observed. On the basis of optimal conditions, the continuous culture was investigated in 1.8L air lift bioreactor. The result suggested 0.1g/h rhamnolipids was obtained from 1%(v/v) fish oil continuously in conditions of 1.2L working volume, 0.5vvm and 20ml/h flow rate.

본 연구에서 개발하고자 하는 김치가공후 부산물인 배추즙을 이용하여 최적의 배양 조건으로 배지를 개발 하는것인데, 배추즙은 비교적 다른 이물질이 포함되어 있지 않은 장점을 가지고 있으며, 효과적인 방법을 모색함으로써 유용한 미생물을 생산할 수 있는 값싼 배지를 개발할 수 있다. 길항력이 있는 Bacillus lentimorbus G-74를 배추즙 배지로 이용하여 배양조건을 최적화한 결과 50%에서 가장 높은 균체 성장을 보였다.

Extracellular polysaccharide was produced by Pseudomonas elodea NK-2000 under nitrogen limitation and aerobic condition, The effect of agiatation speed on cell growth and production of exopolymer was investigated. The agitation speed of 7.5 L fermentor ranged from 200 to 500 rpm. Production of exopolymer increased with higher agitation speed. Maximal cell growth and production of exopolymer from 2% glucose were 3.35 g/l and 3.80 g/l, respectively when agitation speed was 400 rpm.

An internal membrane bioreactor system was employed for continuous succinic ac id production from glucose in order to prove its performance and practicality. Succinic acid-producing Anaerobiospirillum succiniciproducens required more $CO_2$ for the proper growth and succinic acid production in cell recycled continuous culture than in batch culture. The maximum productivity obtained in cell recycled continuous culture was about 3.3 g/L-h which was ca. 3.3 times higher than that obtained in batch culture.

Effect of volcanic ash on cell growth of Aspergillus sp. and production of exopolymers by Agrobacterium sp. and Aureobasidium pullualns was investigated. The volcanic ash contained various mineral salts such as $SiO_2$, $Al_2O_3$, CaO, $K_2O$. Maximal cell growth of Aspergillus sp. was obtained when 0.3% volcanic ash was added to medium. Cell growth of Aspergillus sp. increased with higher concentration of volcanic ash in medium. Amount of cell growth with 0.3 % volcanic ash was 6.7 times higher than that without volcanic ash. Volcanic ash also stimulated production of exopolymer as well as cell growth. Production of curdlan with 0.1% volcanic ash was 12.40 g/l whereas that without volcanic ash was 9.15 g/l. Production of pullulan with volcanic ash was also higher than that without volcanic ash.

In order to investigate the characteristics for RNA accumulation, continuous cultures of S. cerevisiae MTY62 under the carbon-limitation or carbon-phosphate-limitation were performed. In the carbon-limited chemostat, the highest RNA content of 144 mg-RNA/g-dry cell was observed at the dilution rate of $0.30\;hr^{-1}$. In the carbon and phosphate-limited chemostat, the culture condition with the dilution rate of $0.25\;hr^{-1}$ gave the maximal RNA content of 209 mg-RNA/g-dry cell. The accumulation rate of RNA $(mg-RNA/L\;{\cdot}\;hr)$ was decreased at higher dilution rate in the carbon and phosphate-limited chemostat.

We isolated cellulose-producing bacteria from persimmon vinegar(Korea traditional fermentation food). Some of these strains were selected for cellulose production in agitation culture. On the other hand, it was also found that strains suitable for static culture production were not necessarily suitable for agitation culture. Therefore we estimated the cellulose production of these isolates in static culture. To determine nutritional requirement for the production of bacterial cellulose, several nutrients as carbon source, nitrogen and mineral salt were tested.

Production of pullulan by Aureobasidium pullulans HP-2001 with agro-industrial byproducts was investigated. Agro-industrial byproducts from the rice processing industry for the traditional Korean food (AIB-A), apple juice production (AIB-B), and soybean sauce production (AIB-C) were used for carbon and nitrogen source for production of pullulan. Major components of AIB-A were glucose, maltose, maltotriose, and dextran. AIB-A and B were found to be good substitute to glucose as carbon source. Productivity of pullulan with AIB-A and B as carbon source was similar to that glucose. Molecular weight of pullulan produced with AIB-A and B was higher than that with glucose. Major components of AIB-B and C were carbohydrate, protein, fat and ash. AIB-C was also a good substitute to yeast extract as nitrogen source. Some of physiological conditions were examined for the large scale production of pullulan.

Xylitol의 생산성을 높이기 위해 two-stage fed-batch를 수행하였다. Glucose가 고갈되어 pH가 5.7에서 올라가면 glucose를 공급하는 방법에서 최종세포의 OD 185.0과 최종 ethanol 농도 1.0 g/L를 얻었다. 산소전달에 대한 xylitol의 생성 영향에서는 통기량 1 vvm에서 500 rpm의 교반속도일 때 xylitol 수율 55.2%와 생산성 $2.19\;g-xylitol/L\;{\cdot}\;h$를 얻었다.

The anaerobic bacterium P. productus was known to produce acetate from CO, $CO_2$ and $H_2$. In this research the production of acetate at high concentration from carbon monoxide was studied. In this research the effect of $CO_2$, $H_2$, and CODH co-factor on cell growth and acetate production was studied. Also the toxicity of acetate on cell growth was examined. Acetate formation in lab scale fermenter was tested and specific acetate production rate of $0.43\;g/L \;{\cdot}\;hr\;{\cdot}\;O.D.$ was obtained and the concentration of the acetate obtained in a fermenter higher than 40g/L.

토양속의 인산 가용화를 위한 Pantoea agglomerans 배양의 최적조건과 균주 고정화를 위한 안정성을 조사한 결과 $30^{\circ}C$, pH 7.0, 공기유량 2vvm에서 각각의 흡광도 값이 51시간에서1.756과 1,713, 50시간대에 1.819로써 가장 균의 성장이 활발한 것으로 나타났으며, 1% $CaCl_2$ 용액에서 172시간대에 흡광도값 2.361로 가장 안정하였다.

Pantoea agglomerans의 인산 가용화 정도를 조사하기 위하여 합성배지인 HY와 G broth에서 실험을 수행한 결과 pH와 glucose의 감소 정도에 따른 인산의 가용화 정도는 역상 관계임을 알 수 있었다. aeration조건에서 240시간대에 pH 3.48, glucose양 61245ppm일 때 인산의 가용화 정도가 1675ppm으로 가장 많이 나타났으며, 이때 균은 모두 사멸하였다. 따라서 균이 glucose인 탄소원을 기질로 이용하면서 생성되는 유기산에 의해 시간이 경과함에 따라 pH와 glucose양은 감소하게 되고, 유기산에 의해 불용성 인산인 Hydroxyapatite가 가용성 인산으로 바뀌게된다.

Lactobacillus cristatus KLB 46 isolated from Korean woman was grown on supplemented whey medium and medium compositions were optimized for maximum viable cell count. Among the nitrogen sources tested, beef extract yielded the highest viable cell number. When corn steep liquor was applied as an additional nitrogen source, the viable cell number was highest $(3.11{\times}10^9\;CFU/ml)$ in the medium containing 50g/ l corn steep liquor and 10g/ l beef extract. The highest viable cell $count(5.00{\times}10^9\;CFU/ml)$ was obtained from the supplemented whey medium that contains beef extract(10g/ l ), corn steep liquor(50g/ l ), tween 80(0.1%, v/v) and trace amounts of sodium acetate(5g/ l ), dipotassium phosphate(2g/ l ), magnesium sulfate(0.1g/ l ), and manganese sulfate (0.05g/ l ).

메탄올 자화효모인 Pichis pastoris는 메탄올에 의해 유도되는 강력한 AOX1 프로모터의 존재로 인하여 외래 단백질의 생산을 위한 가장 좋은 숙주중의 하나이다. methanol fed batch phase(MFP)동안에 메탄올의 공급은 그 메탄올이 단백질의 발현을 유도하며 또한 숙주에게 에너지원으로 쓰이기 때문에 매우 중요하다. 과량의 메탄올은 세포의 성장을 저해하며, 반면에 불충분한 메탄올의 공급은 세포를 느리게 자라게 하고 생산성도 떨어뜨린다. 본 연구는 새로운 혈소판 응집 억제제인 saxatilin의 생산성, 혹은 수율을 최대화 하기 위해서 메탄올의 공급속도와 세포의 비성장속도를 조절하였다.

불완전 곰팡이의 일종인 Aureobasidium pullulans ATCC 42023 균주를 이용한 pullulan의 최적조건 및 pH 조절에 따른 pullulan의 생성 그리고 균체형태와 고분자 pullulan 생산에 미치는 영향을 조사 하였다. 삼각플라스크 내에서 최대의 pullulan 생산은 초기 pH 6.5일때 11.98 g/l의 pullulan을 생산하였고, 회분식 발효에서는 산소 공급속도 0.5 vvm일때 15.16 g/l의 최대의 pullulan생산량을 나타내었다. pH조절에 따른 실험에서는 pH 4.5일때 13.31 g/l의 pullulan 생산량을 보였고, 이때 균체의 형태는 효모형과 균사형태의 성장을 하였다. 그러나 분자량 생성면에서는 pH 6.5일때 100만 이상의 고분자가 가장 많이 생산되었다.

Heteropolysaccharide-7 (PS-7) was produced by Beijerinckia indica HS-2001 under aerobic condition. Production of PS-7 was investigated under various ratios of glucose as carbon source to ammonium nitrate as nitrogen source. Maximal production of PS-7 was 7.13 g/l when concentrations of glucose and ammonium nitrate were 10 g/l and 0.3 g/l, respectively. But its conversion rate from glucose was as low as 7 %. The highest conversion rate of PS-7 was 46% when those of glucose and ammonium nitrate were 1.0 g/l and 0.3 g/l, respectively.

의료용 단백질인 hGM-CSF유전자가 형질전환된 담배세포를 배양하여 배양 7일째 hGM-CSF가 배지중에서 최대 $162\;{\mu}g/L$의 농도로 축적되었다. 하지만 배양을 계속할수록 생산된 단백질의 양이 다시 감소하는 것으로 나타났다. 이것은 배양말기로 갈수록 배지의 삼투압이 낮아지고 세포가 분해되는 등의 이유로 hGM-CSF가 안정화되지않는 배양환경이 조성됨을 알 수있다.

동결건조로 레트로바이러스를 저장 시 활성유지에 가장 좋은 첨가물로 trehalose , trehalose&PVP 를 첨가해 주었고 DSC 를 통해 열적분석을 하여 온도에 따른 레트로바이러스의 열적변화를 살펴 본 결과 첨가물을 넣어 준 경우 더욱 높은 온도에서 열적 변화가 있었다. 이것을 바탕으로 저장 온도에 따른 레트로바이러스의 활성유지를 조사하기 위해 다양한 온도에서 장기간 보관 한 결과 trehalose 를 첨가한 경우 $4^{\circ}C$ 의 온도 까지 애서도 8 주동안 60% 이상의 높은 활성을 유지하였고 trehalose&PVP 의 경우에도 비슷한 활성을 보였으나 첨가물을 넣지 않은 경우에는 모든 온도에서 40%미만의 낮은 활성도를 보였다.

$TGF-{\beta}_1$을 insect cell-baculovirus system을 이용하여 생산하였으며, 이 경우 낮은 세포밀도와 암모니아 같은 노폐물에 의한 생산성 저해 문제를 해결하기 위해 유가식 배양과 흡착제에 의한 암모니아 제거를 동시에 수행함으로써 $TGF-{\beta}_1$의 생산성을 향상시킬 수 있었다.

Bioreactor을 좀더 효율적으로 운전하기 위하여 여러 가지 gas 성분의 조성을 on-line으로 자동제어 및 분석할 수 있는 시스템을 연구하였으며, 이 시스템을 이용하여 각 gas 성분이 식물세포에 미치는 영향을 여러 관점에서 비교 ${\cdot}$ 분석하였다. 실험결과, 산소는 exponential phase을 증가시켰으며, 이산화탄소와 ethylene은 lag phase을 줄여주는 결과를 보였다. 이산화탄소는 2차대사물질인 aloin의 생산량을 증대시켰으며, 다당의 경우를 보면 현저한 생산량의 차이(약 1.8배)를 나타내었다. Ethylene을 주입한 샘플에서는 glucose 소비속도가 매우 빨랐으며, aloin의 생산량을 높여주는 결과를 보였다.

In this study, we have developed an ultrasonic separation system for plant cells and its operating conditions in terms of voltage, flow rate and concentration were examined. For plant cell, the operation of ultrasonic separator highly depended on concentration of cells. Holding capacity highly depended on flow rate in chamber. Optimum voltage was 30V in high density culture

In hybridoma cell culture, $NH_4\;^+$ is the most important toxic byproduct so far identified. It has been postulated that $NH_4\;^+$, which is similar to $K^+$ in size, is taken up non-specifically by the cells through a potassium transport system, and that the addition of $K^+$ to the culture medium may have a detoxifying effect of $NH_4\;^+$. Thus, in this study the effects of varying $Na^+/K^+$ ratios by adding potassium were investigated for hybridoma physiology and CHO cell morphology respectively. The possible use of potassium addition for the adaptation of CHO cells to suspension culture is discussed.

Panax ginseng hairy roots, transformed by Agrobacterium rhizogenes KTCT 2744, was incubated flask, and investigated its optimal growth conditions. Transformed P. ginseng hairy roots showed active branching pattern and fast growth at free-hormone medium. Optimal conditions of hairy roots showed maximum growth at $23^{\circ}C$, pH 5.8, 1/2 MS medium, 3% sucrose. Effects of combined sugar showed that it had a better growth at only sucrose without glucose or fructose than with glucose or fructose. Also the case of sucrose with glucose had a better growth than with fructose.

Cartilage injuries are frequent nowadays. The previous surgical treatment of cartilage defect was limited. Another approach in the treatment of cartilage injuries is the use of reconstitute cartilage consisting of chondrocytes cultured in suitable biodegradable scaffolds. Current studies have demonstrated the compatibility of chondrocytes with different biomaterials and the chondrogenesis in various types of porous scaffolds. The cell ingrowth into the porous scaffolds is modulated by initial cell loading efficiency. Therefore, well-interconnected pore structure and even pore distribution of the scaffolds are essential for efficient cell seeding. According to our previous work, well-interconnected macroporous scaffolds can be prepared by gas-foaming/salt-leaching method using ammonium bicarbonate salt as porogen additives. In this work, primary chondrocytes were cultured in PLGA 65/35 scaffolds fabricated by using our method. Cells seeded in the scaffolds showed well distribution by agitated seeding method. Histochemical staining of proteoglycans present in the scaffolds was used to visualize the chondrocyte ingrowth in the scaffolds. At 3 weeks, the population of chondrocytes was increased for the most part of the scaffolds, and extra cellular matrix (ECM) secretion was increased as culture periods progressed.

본 연구에서는 재조합단백질 생산을 위한 식물세포배양용 세포배양기 운전에서 배지교환식 배양과 기체조성에 따른 영향을 관찰하였다. 배지교환식 배양에서는 6일째에 배지교환을 시작하는 것이 세포의 생장을 저해하지 않았으며 교환용 배지의 조성을 120 g/L sucrose와 2배 농축된 염농도를 하였을 때 가장 높은 세포농도인 20.0 gDCW/L를 나타내었으며 GUS 생산량은 60 g/L sucrose와 기본배지와 같은 농도의 염농도로 배지를 교환했을 때 12.9 U/mL로 다른 경우에 비하여 $1.5^{\sim}4.6$ 배 증가된 결과를 보였다. 그리고 $CO_2$의 농도를 5%로 높인 공기로 통기했을 때는 세포의 초기생장을 증진시켰으며 0.2 vvm의 통기량에서 5% $CO_2$로 통기시킨 경우는 일반 공기를 0.1 vvm으로 통기시킨 경우보다도 77% 높은 GUS의 생산량을 나타내었다.

생체 적합성, 생분해성, 항균성 등의 특징을 갖는 키토산 지지체는 type I -p collagen과 bFGF 또는 fibronectin을 함께 코팅함으로써 세포적합성을 향상시켜 섬유아세포의 증식과 ECM의 분비를 증가시킬 수 있으며, 인공피부를 위한 적합한 지지체로 사용될 수 있다고 사료된다.

토양으로부터 2종류 그리고 해양으로부터 1종류의 원유 분해 미생물을 분리하였고, 이들을 strain A132, strain F722 그리고 strain OM1으로 각각 명명하였다. 이들 미생물은 Acinetobacter sp., Pseudomonas aeruginosa, Acinetobacter calcoaceticus로 각각 동정되었다. Strain A132. F722의 최적 배양 및 분해온도는 $35^{\circ}C$ 이고, 최적 생장 pH는 각각 8과 9에서 나타났다. 원유를 유일한 탄소원으로 하여 배양을 하였을 때 원유농도 2.0%(w/v)에서 생장이 높았다. 한편, 해양에서 분리된 strain OM1은 pH 7, 원유농도 3.0%(w/v)에서 생장이 높았다. 원유 분해능 조사에서는 Eleuthera (OMAN) 원유를 2.0%(w/v) 기질로 하였을 때, strain A132가 $25^{\circ}C$에서 $5.49g/\;l\;{\cdot}\;day$의 분해능을 나타내었다. 반면, L-Zakum (AFRICA) 원유에서는 strain F722가 $35^{\circ}C$에서 $1.19g/\;l\;{\cdot}\;day$의 분해능을 보였다. 등유$(nC_9-nC_{20})$와 경유$(nC_9-nC_{28})$에 대하여 분해특성을 조사한 결과 strain OM1은 배양 7일 후 95, 75%를 각각 분해하였다. Strain F722는 배양10일 후 80%의 분해율을 나타내었다.

인산가용화균 Aspergillus sp. PS-104의 포자를 6개월 이상 장기보존함과 동시에 균의 생활력 유지를 위하여 다양한 농도의 유기물을 첨가하여 $25^{\circ}C$에서 보관한 후 2주, 4주, 2개월, 4개월, 6개월 간격으로 시료를 채취하여 PDA 평판배지에서의 균수를 측정하였다. 그 결과에 의하면 PEG 200의 경우 6개월 저장시 0.1%의 첨가구에서 대조구에 비해 10배 이상 균의 생활력이 증가하였다. Glycerol의 경우 $0.01{\sim}5.0%$의 첨가구에서 $1.5{\sim}2$배 균의 생활력이 증가하였다.

예건 공정을 거친 양파를 구입하여 2평 정도의 콘테이너 창고에 넣어 실온에서 보관을 하면서 3ppm의 농도로 오존을 처리한 결과 무처리구에 비해 무게 감소율이 현저하게 낮았으며 부패 발생율도 무처리구는 30일 경과 시 10% 정도인 것에 비해 오존 처리 시 2%로 아주 낮았다. 또한 보관 중에 발생되는 양파의 냄새도 완전히 억제하였다.

An in vitro liquid culture media for the cultivation of entomopathogenic nematode Steinernema carpocapsae was developed. Supplementation of whole milk powder with basal liquid culture media showed a remarkable increase in productivity compared to that without whole milk powder and the maximum nematode concentration reached about $1.5{\times}10^5/mL$ within 20 days. Five to twenty gram per liter of liver extract addition revealed highest pathogenicity against 3rd instar of Galleria mellonella which was above about 90% mortality after 48 hr.

Disinfection of pathogenic protozoa Giardia has been done by using $TiO_2$ prepared by hydrothermal process. $TiO_2$ suspended in a photoreactor or immobilized on the optical-fiber surface immersed in a photoreactor has been applied with the ultraviolet light. It has been shown that disinfection effect with $TiO_2/UV$ system 2 times as that with only the UV light and disinfection capability of Giardia increased with an increased $TiO_2$ concentration up to 0.1g/L in a $suspended-TiO_2$ reactor.

대두단백을 응고시키는 균체외 효소를 생산하는 박테리아를 포함하는 미생물을 토양으로부터 분리하였다. 이 미생물효소를 이용하여 대두 펩타이드를 생산하는 최적분해조건을 검토하였고, pH 6.1, $65^{\circ}C$에서 1시간 효소처리를 하여 11S globulin를 가수분해, 대두 펩타이드를 얻었다. 효소반응으로 얻은 응고물은 치밀한 구조를 지녔다. 효소반응 가수분해물인 펩타이드 Y와 W는 등전침전으로 얻은 가수분해물에 비해 그 용해성이 뛰어났다.

This study was carried out to investigate the effects of hydraulic backwash load and porous ceramic media on the biological nitrogen removal efficiencies of a biological aerated filter. An upflow anoxic-oxic biological aerated filter(AO-BAF) with porous ceramic media can remove nitrogen by nitrification and denitrification in single unit. After the AO-BAF backwash, nitrogen removal efficiency was lowest and gradually increased to the steady state. Nitrification efficiency, however, showed the opposite result. It is likely that the biofilms are exposed to aerobic condition as the excess biofilms were sloughed off by backwashing

Characteristics of $CO_2$ fixation by Chlorella sp. HA-1 were studied in a semi-continuous mode in a series operation. A high $CO_2$ fixation rate for a long time was obtained, when the method of semi-continuous mode was employed, in which the controlling parameter was the dilution ratio. A constant $CO_2$ fixation rate was maintained even when the dilution ratio was increased with the increment of 0.1 at the initial value of 0.5. The method of series operation was used to improve the efficiency of $CO_2$ fixation. The total amount of $CO_2$ fixation was proportion to the number of reactor in series operation.

Biofilm airlift 반응기를 이용한 선택적 질산화는 air가 2.5 L/min, $NH_4\;^+-N$ load가 $2.5\;kg-N/m^3/d$에서 약 90% 이상의 질소 제거율과 75-90%의 아질산성 질소의 축적이 일어났다. nitrite oxidizer가 낮은 DO의 영향으로 activity가 저해를 받아서 nitrite를 산화시키지 못하여 축적을 일으킨 것으로 보인다. 반응기 내부의 미생물의 양이 가장 많은 기간에 질소 제거율이 저하하고 nitrite ratio가 약간 감소됨을 보였다. 이는 많은 미생물의 양으로 용존산소가 감소되었기 때문이다. 미생물의 양에 따른 질소 부하의 증거로 F/M 비를 맞추어 그에 따른 질산화를 관찰하는 연구를 할 수 있을 것 같다.

본 연구에서는 활성슬러지법으로 처리하는 제약폐수를 대상으로 공정내 미생물의 산소소모량을 측정하고 미생물의 증식정도를 알아보기 위해 동력학 계수를 측정하였다. 미생물 접종량에 따른 산소소모율은 $43{\sim}64\;mgO_2/hr$이었고, 기질 농도 변화에 따른 산소소모율은 $4{\sim}42\;mgO_2/L/hr$로 나타났다. 또한 최대비성장율$(\;{\mu}\;_{max})$, 수율계수, 그리고 반속도상수(Ks)는 각각 $2.7day^{-1}$, 0.655, 36.11 mg/L로 측정되었다.

The characterization of mixed microorganisms that have activities of protein- and lipid-degradation were studied to reduce marine pollution by fish wastes and to utilize them again as feeds or fertilizers. There was no mutual inhibition among the microorganisms. The pH and temperature for the mixed culture were found to be 7 and $32^{\circ}C$, respectively. The fish waste were started to degrade after one day, and were almost degraded after 4 days without any odor.

4종의 각 개별균주와 6가지 2균주조합별 실험을 실시한 결과 개별균주의 경우 평균 15%로 낮은 COD제거율을 보였다. 2균주조합의 경우는 NAR3A와 NBY3A가 50.3%로 가장 우수하게 나타났으며 평균 34.9%의 제거율을 보였다. 이러한 제거율은 단일균주에 비해 약 20%향상된 수치로서 단일균주 보다는 2균주조합에 의한 것이 보다 효율적인 것으로 나타났다. 반월의 염색단지 내 폐수처리장의 폐수를 이용하여 실험한 결과 거의 비슷하게 낮은 색도제거율을 나타냈다. 이는 본 연구에 이용된 균주의 경우 단일염료의 색도를 제거하는 실험에 의해 선별되어 다양한 성상의 염료가 함유되어 있는 실제폐수의 염료를 분해하기 힘들었기 때문이다. COD제거의 경우는 비교적 우수한 효율을 나타냈다.

오존의 강력한 산화력은 돈사 내 공기중에 부유하는 세균에 대하여 큰 살균효과를 나타냈는데, 이는 $0.05{\sim}0.07ppm$의 작업자와 돼지에게 피해를 최소화 할 수 있는 오존 농도 범위여서 그 효과가 크다 하겠다. 본 실험에서 오존은 발생장치 내에서의 원료공기(산소)의 체류시간에 따라 각기 다른 발생농도를 나타냈으며, 풍량이 14.7m^3/min$ 일 때 가장 높은 오존 농도를 보임을 알 수 있었다. 또한 오존 발생기를 개방형에서 폐쇄형으로 변형함으로써 같은 조건에서 더 높은 오존발생농도(폐쇄형: 0.13ppm, 개방형: 0.08ppm)를 얻을 수 있었다.

For an integrated nitrification-denitrification process, nitrite formation in the aerobic stage leads to big savings. Dissolved oxygen concentration, temperature, pH and free ammonia concentration have been meet for nitrite accumulation. Also their effects over the ammonia oxidizers and nitrite oxidizers have been studied. Dissolved oxygen limitation and free ammonia inhibition led to slow nitrification and nitrite build up. In this study batch kinetics of ammonium and nitrite oxidations were performed with free ammonia accumulated nitrifiers. From the results it is likely the nitrite oxidizers are inhibited by oxygen limitation rather than free ammonia.

Degradation of aromatic compound by Pseudomonas mendocina BCNU 154 has been investigated. The microorganism utilizes xylene, toluene, nitrobenzene, ethylbenzene and cumene. This strain is tolerant to some heavy metals, such as Mn, Cu, Si, and Mo, and resistant to some antibiotics, such as vancomycin, chloramphenicol and ampicillin. The metabolic pathway of toluene in Pseudomonas mendocina BCNU 154 is also elucidated.

The eventual goal of this study is to elucidate roles of plant terpenoids (e.g., cymene, limonene and others) as natural substrates in the cometabolic biodegradation of PCBs and to develop an effective PCB bioremediation technology. The aim of this study was to examine how plant terpenoids, as natural substrates or inducers would affect the biodegradation of PCB congeners. Various PCB degraders that could grow on biphenyl and several terpenoids were tested for their PCB degradation capabilities. The PCB congener degradation activities were first monitored through resting cell assay technique that could detect degradation products of the substrate. The congener removal was also confirmed by concommitant GC analysis. The PCB degraders, Pseudononas sp. P166 and Caynebacterium sp. T104 were found to grow on both biphenyl and terpenoids ((S)-(-) limonene, p-cymene and ${\alpha}-terpinene$) whereas Arthrobacter B1B could not grow on the terpenoids as a sole carbon source. The strain B1B grown on biphenyl showed a good degradation activity for 4,4'-dichlorobiphenyl (DCBp) while strains P166 and T104 gave about 25% of B1B activity. Induction of degradation by cymene, limonene and terpine was hardly detected by the resting cell assay technique. This appeared to be due to relatively lower induction effect of these terpenoids compared with biphenyl. However, a subsequent GC analysis showed that the congener could be removed up to 30% by the resting cells of T104 grown on the terpenoids. This indicates that terpenoids, widely distributed in nature, could be utilized as both growth and/or inducer substrate for PCB biodegradation.

The relative affinity of seaweed alginate, polyguluronate and polymannuronate for cations was investigated. The cations used in this study were $Ca^{2+}$, $Cd^{2+}$, $Co^{2+}$, $Cu^{2+}$, $Fe^{3+}$, $Hg^{2+}$, $Mg^{2+}$, $Mn^{2+}$, $Pb^{2+}$, $Rb^{1+}$, $Sr^{2+}$ and $Zn^{2+}$. The ability of cations to precipitated polymers was determined as the relative affinity of seaweed alginate, polyguluronate and polymannuronate for cations. The relative affinity of polymers for cations in order are as follow: Seaweed alginate: $Fe^{3+}$,$Cu^{2+}$,$Cd^{2+}>Pb^{2+}>Co^{2+}$,$Zn^{2+}>Ca^{2+}>Sr^{2+}$,$Rb^{1+}>Mn^{2+}>Mg^{2+}$,$Hg^{2+}$ Ployguluronate:$Fe^{3+}$,$Cu^{2+}$,$Cd^{2+}>Ca^{2+}$,$Co^{2+}$,$Pb^{2+}>Sr^{2+}$,$Rb^{1+}$,$Zn^{2+}>Hg^{2+}$,$Mn^{2+}>Mg^{2+}$ Polymannuronate:$Fe^{3+}$,$Cd^{2+}$,$Cu^{2+}>Ca^{2+}$,$Pb^{2+}>Zn^{2+}$,$Rb^{1+}$$Sr^{2+}$,$Hg^{2+}>Co^{2+}>Mn^{2+}>Mg^{2+}$

미생물을 이용한 새로운 독성 측정 방법으로서, 본 연구에서는 자성 박테리아 Magnetospirillum sp. AMB-1 세포들의 유영 속력 측정을 통한 독성 측정의 가능성과, 발광 박테리아를 이용한 기존의 $Microtox^{circledR}$ 시스템에 견줄 수 있는 새로운 독성 측정 방법이라는 사실을 확인하였다. 또한, AMB-1 세포들의 주기성 관찰을 통해 또 다른 독성 측정 방법의 가능성을 확인하였다.

Enzymatic hydrolysis of cellulose was studied with emphasis on the effect of cellulase loading and pulp sludge concentration on glucose yield. Enzyme loading appeared to have a significant effect on glucose yield. Chemical pretreatment had no effect on enzymatic hydrolysis of pulp sludge. High glucose yield was obtained from enzymatic hydrolysis, especially at sludge concentrations lower than twenty percent. The optimum concentrations of crude cellulase and ${\beta}-glucosidase$ were 5 U/mL and 8 U/mL, respectively, considering the amount of enzymes used and glucose produced.

In situ hybridization with fluorescent oligonucleotides(FISH) was used to detect and localize microorganisms in the granules of lab-scale upflow anaerobic sludge blanket(UASB) reactors. An UASB reactor was seeded with mesophilically-grown($35^{\circ}\;C$) granular sludge, and thermophilically($55^{\circ}\;C$) operated by feeding with a synthetic wastewater. Sections of the granules were hybridized with 16S rRNA-targeted oligonucleotide probes for Eubacteria, Archaeabacteria, and specific phylogenetic groups of methanogens. FISH clearly showed the layed structure of thermophilic granules, which was consisted of outer bacterial cells and inner archaeal cells. Methanoseata-, Methanosarcina-like cells were also found to be localized inside the granules. These results demonstrated FISH was useful in studying the spatial organizations of methanogens and in situ morphologies and metabolic functions in thermophilic granular sludges.

A parallel reactor system which is consisted of two trickle bed reactors (TBR) was developed for the biodegradation of trichloroethylene (TCE) in waste gas stream. The reactor were packed with porous ceramic materials and Burkholderia cepacia G4 was inoculated to form biofilms. Each reactor was operated alternatively in TCE degradation or reactivation mode, and the effect of switching time on TBR performance was investigated. The MO (monooxygenase) activity during the TCE transformation decreased below 10 % within 24 hr, but could be recovered to the initial high level within 10 hr after supplying the reactivation medium supplemented with phenol as a carbon source. This shows that the parallel TBR system has a great potential for the long-term stable treatment of TCE.

암모니아와 황화수소를 동시 제거하기 위한 biofilter가 연구되었다. 암모니아 제거의 경우 공탑속도 30초에서 유입 암모니아 농도 80 ppmv까지 100 %의 제거율을 달성하였다. 암모니아와 황화수소의 동시 제거의 경우 공탑속도 30초에서 유입 황화수소 농도 40 ppmv와 암모니아 농도 80 ppmv가 제거율 100 %로 처리되었고 이 경우 pH의 변화가 암모니아의 산화에 매우 큰 영향을 미침을 알 수 있었다.

Phenol and other phenolic compounds are common constituents of aqueous effluents from processes such as polymeric resin production, oil refining and cokeing plants. Phenol is a both toxic and lethal of fish at relatively low concentrations e.g. 5-25 mg/L and imparts objectionable tastes to drinking water at far lower concentration. Therefore, the treatment of phenol effluent is important. Among the various techniques of phenol wastewater treatment, microbial teratment is a popular process. The breakdown of phenols by microorganisms has recived considerable attention, because of its biochemical interest and its industrial importance in effluent treatment. This research was performed to investigate the dynamics of microbial community, biofilm growth and the comparison of phenol removal efficiency by RBC (Rotating Biological Contactor) using Rhodococcus sp. EL-GT The experiment was carried out at rotating speed of 10ppm and hydraulic retention time of 7 hours. As time passed, phenol removal efficiency was gained highly. The RBC using Rhodococcus sp. EL-GT completely degraded 15 mM.

An electroenzymatic system to oxidize veratryl alcohol of on electrodes with in-situ generated hydrogen peroxide was studied. We investigated hydrogen peroxide generation, current efficiency, and veratryl alcohol oxidation in the electrode system at various conditions. The reaction rates of veratryl alcohol oxidation were compared in an electrochemical, an electroenzymatic, and an usual biochemical systems to prove the concept of electroenzymatic oxidation.

식물의 토양전염병을 유발하는 Pythium sp. 및 Rhizoctonia sp.에 대한 길항력과 생장속도를 고려한 RPI (relative performance indices) 기법을 이용하여 100 여개 후보균 중 #19, #42 등 길항균을 선정하였다. 후보균들은 일반적으로 Pythium sp.보다 Rhizoctonia sp.에 대한 길항력이 우수하였고 Bacillus sp.의 배양 상등액은 항진균활성과 blood agar에서 용혈현상을 보여 상등액으로부터 항균물질인 lipopeptide 계열물질을 분리하였다.

고분자 담채 PE에 대한 미생물의 초기부착 현상과 그 형태를 광학현미경, SEM, 형광현미경을 이용하여 관찰하였는데 광학현미경 관찰에 의한 관찰에서 미생물의 초기부착은 거친 표면의 홈에서 시작한다는 것을 알 수 있었고 SEM 사진에 의해서는 미생물 형태를 정확히 파악할 수 없고 특히 메탄 형성 미생물의 존재를 규명하기 어려워 형광현미경을 이용하였다. 형광현미경에 의한 혐기성 부유슬러지와 담체에 부착된 미생물의 관찰 결과 메탄생성균을 확인할 수 있었고 초기 부착에 메탄생성균이 관여함을 알 수 있었다.

A simple and effective method has been developed for the immobilization of the cell on polyurethane foam. Two types of commercially available polyurethane foam and Hydro-filt were tested. The ultimate purpose of the process is to produce low-cost materials for hydrogen sulfide removal which are being increasingly used for industrial application. Effect of several parameters were studied on the cell loading. These parameters were type, size, and amount of polyurethane foam. MC-70 was the best immobilization material of three type of carriers, and optical particle size was $5{\sim}8mm$ and amount of polyurethane foam was 8g/L.

본연구에서는 Rhodococcus rhodochrous IGTS8 균주를 사용하여 고농도의 유상에서 탈황효율을 높이기 위한 오일 함유비, pH, 영양물질의 영향을 조사하였다. 오일 함유비에 의한 영향은 유상이 30%이하일 경우 그리 크지 않았으며, pH 조절에 의해 50%, 영양물질의 강화에 의해 32%의 탈황효율이 증가했다. 강화배지에서 pH를 조절하며 배양한 결과, 기존의 배양에 비해 136% 탈황효율이 증가했다.

수계로부터 anthraquinone계 염료인 Remazol brilliant blue R를 분해 ${\cdot}$ 자화하는 균주를 분리하여 Strain RBB로 명명하고 성장특성을 조사하였다. 최적 배양온도와 pH는 $35^{\circ}C$, 9.0으로 나타났다. 진탕배양의 경우가 정치배양의 경우보다 미생물 성장속도가 현저히 상승하였다.

본 연구의 석탄에서 분리한 미생물은 유기황인 dibenzotiophene을 2-hydroxybiphenyl로 탈황하며 yeast extract의 첨가로 그 속도가 증가할 수 있음을 관찰할 수 있었다. Resting cell을 이용한 탈황의 경우 화학 계면활성제의 첨가에 의해 그 속도와 정도가 증가함을 관찰할 수 있었다.

Possibility of biological nitrogen treatment was tested in wastewaters with low C/N ratio. Chlorella kessleri was inoculated at $10^6\;cell/mL$ of initial density in two different artificial wastewaters: one that contained glucose for organic carbon source and the other without carbon source. Nitrate could be successfully reduced below 10 mg $NO_3/mL$ from initial nitrate concentration of 560 mg $NO_3/mL$ in 10 days even in the wastewater without carbon source, This 98% removal of nitrate without extra organic carbon source lights up the future of biological wastewater treatment, where the insufficient ability of nitrogen removal is a major problem.

내염성이며 세포외 protease를 강력하게 생산하는 효모를 재래식 메주효모들과 자연계에서 분리된 내염성 효모들 가운데서 선별하여 Hansenula sp. S-9으로 동정하였다. S-9균주는 $30^{\circ}C$, pH 7.5, 0.5M NaCl을 함유한 배지에서 잘 생육하였고 1.0% beef extract와 1% glucose 및 0.5M NaCl를 함유한 BD배지에서 $30^{\circ}C$로 72시간 배양하였을 때 가장 많은 효소가 생산되었다.

키토산으로부터 키토산 올리고당을 생산하기 위한 유용 효소원을 개발하기 위해서 토양에서 4종의 미생물을 분리하였으며, 가장 높은 활성의 chitosanase를 생산하는 Bacillus sp. HSB-21 균주를 선정하였다. 분리균주로부터 생산된 chitosanase는 endo-type의 효소로 분자량이 약 21,000이었다. 최적 pH와 온도는 각각 5.5, $50^{\circ}C$이었으며, pH $3{\sim}8$ 범위와 $40^{\circ}C$까지 비교적 안정한 효소로 나타났다. 본 효소는 키토산을 분해하여 단당을 형성하지 않았으며 주로 $2{\sim}8$당을 포함하는 키토산 올리고당을 만들었다. 그러므로 본 효소는 고중합도 키토산 올리고당의 생산을 위한 응용 가능성이 큰 것으로 판단된다.

DFA IV를 생산하는 levan fructotransferase를 ${\kappa}\;-carrageenan$을 이용하여 고정화한 결과 카라기난의 농도가 2%일 때 가장 높은 활성을 나타내었다. 이 때에 고정화 담체에 포괄되어지는 효소의 활성도는 0.81 units으로서 soluble enzyme 7.7 units에 비해 상대적으로 낮은 활성을 보여주었다. 고정화 및 soluble enzyme의 최고 활성온도와 최적 pH는 동일하게 $55^{\circ}C$, pH6.0 이었다. 가교제를 처리할 경우에 적당한 농도는 0.5%로 생각되어진다. $37^{\circ}C$에서 시간에 따른 고정화 및 soluble enzyme의 DFA IV 생산량을 HPLC로 분석한 결과 60시간 이후의 전환률이 각각 32%, 61%로 나타났다.

Hydrolase는 생물학적 공정에서 가장 널리 사용되는 효소군 중의 하나이다. 본 연구에서는 hydrolase들 중에 xylanase, lysozyme, glucoamylase, barnase가 대상 hydrolase로 선택하여 최적 pH를 예측하는 방법론을 제시하였다. Tanford-Kirkwood(TK) model에 기반을 두고 선택된 효소들의 catalytic residue들의 전하 변화가 optimum pH에 중요한 인자로서 그 catalytic residue들의 전하 차이가 최대일 때의 pH가 optimum pH로 나타났다. 이렇게 예측된 optimum pH 결과들은 실험적으로 결정된 optimum pH와 서로 잘 일치하고 있음을 보여준다.

pNAN15b와 E. coli BL21(${\lambda}DE3$)를 써서 클로닝한 E. coli 유래의 NALase는 높은 수준으로 발현이 되었다. Escherichia coli, Haemophilus influenzae, Clostridium perfringens 세 가지 균주 유래의 NALase를 비교해본 결과 E. coli 유래의 NALase가 발현이 잘되어 전체 활성도는 높았지만 활성저해를 받는 경향이나 pH 11에서의 안정성을 보면 H. influenzae 유래의 NALase가 Neu5Ac 합성에 더 유리한 특성을 갖고 있어서 H. influenzae 유래의 NALase를 E. coli에서 충분히 발현할 수 있다면 H. influenzae 유래의 NALase가 더 효과적이라고 할 수 있다.

(R,S)-3-amino-n-butanoic acid에 대한 광학분할 능력이 우수한 균주 Alcaligenes denitrificans Y2k-2를 토양으로부터 분리하였다. 광학분할 반응에 관계하는 효소는 트랜스아미나제로 추정되며, 아민 수용체에 대한 특이성에서 크기가 작을수록 반응성이 높아지는 경향이 있다. pyruvate에 의한 기질저해 현상이 나타나는데, 효율적인 생산 체계를 갖추기 위해서 이 문제를 해결할 필요가 있으며 현재 효소의 정확한 특성을 파악하기 위한 실험을 진행 중이다.

Many different oligosaccharides were produced by transferring the fructose residue of sucrose to maltose, cellobiose, lactose and sucrose (self-transfer), where their yields of fructosylated acceptor products accounted for $26{\sim}30%$ (w/w). The maximum conversion yield (30%) was obtained in fructosyl cellobioside formation with 500 g sucrose/l (substrate) and 200 g cellobiose/l (acceptor). These four acceptors gave various products having DP (degree of polymerization) $2{\sim}7$ by successive transfer reactions.

The production of inulooligosaccharides from inulin by a dual endoinulinase system of Pseudomonas sp and Xanthomonas sp. was investigated the optimum conditions for a dual endoinulinase reaction were as follows : pH,5.8; temperature, $50^{\circ}C$; substrate concentration, 50 g/l; enzyme ratio, 3:1 as Xanthomonas endoinulinase to Pseudomonas endoinulinase. Under optimum conditions, the maximum yield of oligosaccharides was 90.5% in total sugar basis by dual endoinulinase system

The indole tolerance level of Pseudomonas savastanoi BCNU 106 was as high as 300 mg/ml when toluene or p-xylene was added to the medium to 20% by volume. Pseudomonas savastanoi BCNU 106 grown in a two-phase system containing the various concentrations of indole and solvents produced indigo and indirubin. The optimal condition in the production of indigo and indirubin was also studied.

Chiral epoxides are useful chiral synthons in organic synthesis and various biological methods have been investigated for the production of chiral epoxides. In this work, enantioselective resolution of racemic styrene oxide was investigated using an isolated Aspergillus niger sp. for the production of optically pure (S) -styrene oxide. The enantioselectivity and initial hydrolysis rates of racemic substrate were highly dependent on the pH, temperature, and the volume ratio of cosolvent. The experimental sets of pH, temperature, and the volume ratio of cosolvent were designed using central composite experimental design, and the reaction conditions were optimized using response surface analysis. The optimal conditions of pH, temperature, and the volume ration of cosolvent were determined to be 7.78, $28.32^{\circ}C$, and 2.4 %(v/v), respectively, and optically pure (S)-styrene oxide (> 99% ee) could be obtained with the 35 % yield by microbial enantioselective hydrolysis reaction.

In this study the enzymatic synthesis of ester-linked conjugates of amino acid and monosaccharide in pyridine was tested by the catalysis of Optimase M-440, an alkaline serine pretense. Optimase M-440 showed the higher activity in the reaction of monosaccharides which have one or more primary -OH groups. And also Optimase M-440 showed high regioselectivity; The transesterification of primary -OH group selectively occurred.

Enzyme production of calcium stearate was attempted by lipase. Lipase having the highed hydrolyzing activity at $60^{\circ}C$, a melting point of substrate (hydrogenated beef tallow),was selected. 60% of hydrogenated beef tallow was converted into calcium stearate in 48hr. This result showed the possible replacement of conventional waste-producing process into enzymetic reaction.

Preparation for the D-phenylalanine using chemo-enzymatic reaction was investigated. D,L-5-benzylhydantoin was synthesized chemically from L-phenylalanine and converted to N-carbamoyl-D-phenylalanine by the immobilized hydantoinase. The pH and temperature affected the solubility and racemization rate of benzylhydantoin. The optimal temperature and pH of the process were $50^{\circ}C$, 8.5, respectively and the conversion yield hadn't much difference with the hydantoinase content in 10hrs. This produced N-carbamoyl-D-phenylalanine was transformed chemically into D-phenylalanine.

To separate chitosanoligosaccharides easily by size exclusion, an coencapsulating technology of substrate and enzyme was developed. Chitosan and chitosanase were enveloped in this membrane and the product released to medium by size exclusion. The lower limit of the alginate concentration and the agitation speed were 0.5% and 40 rpm, respectively. Membrane thickness and capsules diameter were $10{\mu}m$ and approx. 3.0mm, 1.5mm, respectively. The molecular weight difference by concentration and cps of alginate were of little significance. And also, the molecular weight of distribution according to enzyme concentration was low concentration of enzyme produced high molecular weight of oligosaccharides. At 1.5mm size of capsule, product diffusion rate to outer part was higher than other capsules. The molecular weight distribution of the released oligosaccharides ranged from 1000 to 6000 Da.

The purpose of present study was to examine the efficacy of PAB (para-amino benzamidine) affinity column chromatography, pasteurization ($60^{\circ}C$ heat treatment for 10 h), and lyophilization steps, employed in the manufacture of urokinase from human urine, in the removal and/or inactivation of urine-born viruses. Bovine herpes virus (BHV) and Murine encephalomyocarditis virus (EMCV) were selected for this study. Samples from the relevant stages of the production process were spiked with the viruses and the amount of virus in each fraction was quantified by 50% tissue culture infectious dose ($TCID_{50}$). BHV and EMCV were effectively partitioned from urokinase during PAB chromatography with the log reduction factors of 6.71 and 5.27, respectively. Pasteurization was a robust and effective step in inactivating BHV and EMCV, of which titers were reduced from initial titers of $8.65\;log_{10}\;TCID_{50}$ and $7.81\;log_{10}\;TCID_{50}$, respectively, to undetectable levels within 1 hour of treatment. The log reduction factors achieved during lyophilization were 2.06 for BHV and 4.54 for EMCV. These results indicate that the production process for urokinase has sufficient virus reducing capacity to achieve a high margin of virus safety.

For the production of $B^{30}-homoserine$ human insulin precursor, four types of fusion peptides LacZ, MBP, GST, and His-tagged sequence were studied in this work. Recombinant E. coli JM 103 and E. coli JM 109 containing fusion peptides were cultivated at $37^{\circ}C$ for 1hr, and gene expression was occurred when 0.5mM of isopropyl-D-thiogalactoside(IPTG) was added to the culture broth, and followed by longer than 4hr fermentation respectively. DEAE-Sphacel and gel filtration chromatography, amylose and glutathione-Sepharose 4B affinity chromatography, and nickel-affinity chromatography system were employed as purification of $B^{30}-homoserine$ human insulin precursor. Recovery yields of His-tagged, LacZ, GST, and MBP fused $B^{30}-homoserine$ human insulin precursor resulted in 47%, 20%, 20%, and 18%, respectively.

Bacteriocin을 생산하는 유산균을 김치에서 분리하였고 Lactococcus latis subsp. lactis로 확인 되었으며 bacteriocin의 최대생산조건은 $30^{\circ}C$, 6시간으로 다양한 유산균 및 Pseudomonas synsantha, Acetobacter aceti의 생육을 저해하였고 여러 peptidases에 의해 불활성화되었다. 또한 다양한 범위의 pH와 열처리에 안정하였고 SDS-PAGE결과 약 8 Kda으로 확인되었다.

The Pseudomonas aeruginosa BYK-2(KCTC 18012p) produced three kinds of glycolipids on olive oil as a substrate and purified two types of major glycolipids(Rf=0.48, BS-1; Rf=0.65, BS-2) using silica gel chromatography, TLC, HPLC, etc. From the analysis of the chemical structure, the glycolipid of BS-1 was estimated as rhamnolipid($2-O-{\alpha}-L-rhamnopyranosyl- {\alpha}-L-rhamnopyranosyl-{\beta}-hydroxyldecanoyl-{\beta}-hydroxydecanoic$ acid; M.W. 650) and BS-2 was detected as rhamnolipid methyl ester($2-O-{\alpha}-L-rhamnopyranosyl-{\alpha}-L-rhamnopyranosyl-{\beta}-hydroxyldecanoyl-{\beta}-hydroxydecanoic$ acid methyl ester; M.W. 664) by FT-IR, FAB Mass spectrometry, $^1H-NMR$, $^{13}C$ FT-NMR, DEPT, 2D-NMR (TOCSY, RELAY, NOESY, HSQC, HMBC). In particular, It was found that a marine bacterium Pseudomonas aeruginosa BYK-2(KCTC 18012P) remarkably produced rhamnolipid and rhamnolipid methyl ester simultaneously.

지방산과 당의 transesterification에 의하여 제조되는 당에스테르를 분리정제하는 방법으로 크로마토그라피방법을 조사하였다. 당에스테르제조에 사용되고 있는 각종 지방산(oleic, stearic, palmitic acid)과 당에스테르의 크로마토그라피 용출특성을 이동상의 유속과 온도, 그리고 주입량을 변화시켜 관찰하였다. 본 실험에서 사용한 column은 Silica 계통의$C_{18}$ column($3.9{\times}300mm$) 이었으며 본 실험에서 조사한 지방산과 당에스테르의 혼합물은 모두 높은 해상도로 분리되는 것을 관찰하였다.

Lincomycin의 추출을 위한 최적용매 및 최적 추출조건을 조사하였고 회분식 다단 추출을 시도하는 한편, 연속식 3단 향류추출장치를 개발하여 lincomycin의 추출효율을 조사하였다. 연속식 3단 향류추출장치를 운전한 결과 회분식 추출과 마찬가지로 단수가 증가함에 따라 lincomycin의 추출효율이 증가함이 확인되었고, 3단을 거친 수용액상에서는 lincomycin이 거의 발견되지 않음이 확인되었다.

본 연구에서는, 최근 관심의 대상이 되어 다방면에서 그 사용의 예가 늘고 있는 CD와 alfalfa 식물세포 현탁액으로부터 발현되는 휘발성 향기성분을 비롯한 대사물질의 포접형성에 대해 관찰하고, 식물 세포배양계에서의 CD의 역할에 대해 확인해보고자 하였다. 실험결과 CD는 alfalfa로부터 발현되는 향기성분과 inclusion complex을 이룬다는 것을 확인하였고, 또한 CD가 alfalfa 식물 세포로부터 대사물질을 더 많이 이끌어 내는 elicitor로서 작용할 수도 있음을 알 수 있었다.

Quinone reductase was purified to electrophoretic homogeneity from bovine liver by using ammonium sulfate fractionation, ion-exchange chromatography, and gel filtration chromatography. The enzyme utilized either NADH or NADPH as the electron donor. The optimum pH of the enzyme was pH 8.5, and the activity of the enzyme was greatly inhibited by $Cu^{2+}$ and $Hg^{2+}$ ions, dicumarol and cibacron blue 3GA. The enzyme catalyzed the reduction of several quinones and other artificial electron acceptors. Furthermore, the enzyme catalyzed NAD(P)H-dependent reduction of azobenzene or 4-nitroso-N,N-dimethylaniline. The apparent $K_m$ for 1,4-benzoquinone, azobenzene, and 4-nitroso-N,N-dimethylaniline was 1.64mM, 0.524mM and 0.225mM, respectively. The reduction of azobenzene or 4-nitroso-N,N-dimethylaniline by quinone reductase was strongly inhibited by dicumarol or cibacron blue 3GA, potent inhibitors of quinone reductase.

본 연구는 항산화작용, 항암작용, estrogen 유사작용, 항골다공증작용 등 다양한 생리적기능을 가진 isoflavones에 대한 추출조건을 추출용매의 농도, 추출온도, 추출pH, 추출시간으로 나누어 단계적으로 isoflavones을 추출하는 실험을 실시하였고, 보다 기능이 잘 알려진 4종 isoflavones 추출에 대한 결과를 분석하여 최적추출 조건을 선정하였다. 그 결과 75% ethanol, $80^{\circ}C$, pH4, 3시간의 추출 조건에서 $4,024{\mu}g/ml$의 가장 높은 총 isoflavones 추출량을 나타내었고, 대두 추출물 중의 단백질 제거를 위해 이용된 염화칼슘의 농도가 증가할수록 보다 높은 함량의 isoflavones을 얻을 수 있었다.

본 연구에서는 pH 변화에 따른 poly(2-ethyl-2-oxazoline)[PEOx]과 poly(acrylic acid)[PAA]의 수소결합 특성을 고찰하였다. PEOx와 PAA는 산성 조건(pH 4.3이하)하에서 PAA의 수소와 PEOx의 산소간에 수소결합으로 안정한 복합체를 형성하여 침전하였다. PEOx와 PAA 혼합용액을 수성이상계에 적용하기 위한 최적 반응비는 질량비 1:1.5이었고, 이 고분자 혼합용액은 수소결합을 제거한 상태에서 dextran용액과 수성이상계를 형성하였다.

본 연구에서는 재조합 E. coli 시스템에 의한 재조합 cystein proteinase의 대량 생산을 위하여 4L 회분 배양 및 최적 지수 생장기에서의 IPTG- induction을 연속적으로 실시하였다. 발현된 rPwCP1 양은 진탕 배양 시스템에서의 결과와 비교해 볼때 현저하게 향상되었음을 알 수 있었으며, 더불어 metal affinity chromatography 방법으로 처리하여 전기영동을 수행한 결과, 목적 재조합 단백질만을 정제 및 농축시킬 수 있음이 확인되었다.

토양에서 분리한 endo-inulinase 생산 균주인 Xanthomonas oryzae #5로 부터 11.5kb의 endo-inulinase 유전자를 포함하는 재조합 plasmid를 함유한 형질 전환주를 분리하였다. 11.5kb의 단편으로부터 8.6kb, 4.1kb의 단편을 포함한 pDI 2, pDI4 재조합 plasmid를 제작하여 활성을 확인한 결과 endo-inuliase 활성을 나타내었으며, 재조합 plasmid pDI 2를 이용하여 DNA sequence를 한 결과 endo-inulinase 유전자는 1,333개의 아미노산으로 구성된 ORF를 가지고 있었다. 또한 B. circulans MCI-2554의 CFTase와 아미노산 배열에 있어은 약 72%의 높은 homology를 나타냈었으며, 다른 fructan hydrolases, inulinase, levanase와의 아미노산 비교로부터 ${\beta}-fructouranosidase$ motif를 포함한 6개의 유사부위를 확인하였다.

UK-58,852의 생합성에 관여하는 유전자를 분리하기 위해 Actinomadura roseorufa의 genomic DNA와 E. coli-Streptomyces shuttle cosmid vector인 pOJ446이 genomic library를 만들었다. Genomic library는 dehydratase PCR product와 eryA 유전자를 probe로 하여 sugar 생합성 유전자와 polyketide typel 유전자가 집단으로 존재하는 cosmid pHD54를 분리하였고, 이를 제한 효소인 BamHI, SmaI와 Sonicater를 이용해서 subcloning 하였다. 이들의 염기서열을 부분 분석한 결과, polyketide 생합성에 관여하는 ketoacyl synthase, methylmalonyl acyltransferase, ketoreductase, enolreductase 그리고 PKS loading domain 등 polyketide synthase type I 임을 보여주고 있고, BLAST 분석된 결과를 보면 polyketide synthase 유전자는 rifamycin 생합성 유전자와 유사성이 높다. 그리고 sugar 생합성에 관여하는 유전자로는 oxidoreductase, dTDP-D-glucose 4,6 dehydratase, dTDP-D-glucose synthase 그리고 dTDP-4-keto-6-deoxy-D-glycose 3,5-epimerase으로 구성된 gene cluster를 확인하였다. 그리고 염기서열 분석된 유전자중 dTDP-D-glucose synthase를 발현하여 유전자의 기능을 확인하였다.

Recombinant Escherichia coli NZN111 ($F-{\Delta}pfl::Cam$ ldhA::Kan) harboring pTrcML, a plasmid containing the E. coli malic enzyme gene, produced considerable amount of malic acid along with the desired product succinic acid. This seemed to be due to the unmatched redox states between glucose and succinic acid. Therefore, a more reduced carbon substrate sorbitol was examined for the possibility of matching the potential during succinic acid production. When NZN111 (pTrcML) was cultured in LB medium containing 20 g/L sorbitol under $CO_2$ atmosphere, 10 g/L of succinic acid was produced. The apparent yield of succinic acid was 1.1 g succinic acid per g sorbitol, which is 85% of the maximum theoretical yield.

Antimicrobial cationic peptides have attracted increasing research and clinical interest as a natural antibiotics due to their broad spectrum of antimicrobial activites and the rapid development of multidrug-resistant pathogenic microorganisms. In this study, first, we synthesized artificial fusion partner and cationic peptide genes (lactoferricin, magainin, protegrin-1, and indolicidin). Second, we constructed recombinant expression vectors and then transformed Pichia pastoris. Finally, expressed cationic peptides were purified and tested for their antimicrobial activites. Antimicrobial activity has been tested upon the appearance of clearing zone on the plate with the lawn of gram negative E.coli XL- I blue and garm positive Staphylococcus aureus. Protegrin-1 and Indolicidin have apparant activity of cationic peotides. This fusion technique may lead to a general and suitable tool for production of pure antimicrobial cationic peptides in Pichia pastoris.

E.coli ATCC 33456은 6가 크롬 환원능이 있다고 보고되어진 미생물보다 비교적 높은 활성을 지닌다. E.coli ATCC 33456에서 6가 크롬 환원효소를 클로닝하기 위해서 colony hybridization과 southern hybridization을 통하여 2개의 형질 전환 균주를 획득할 수 있었다. 두 균주 모두 E.coli ATCC 33456이 가진 6가 크롬 환원능보다 약 2.5-4배정도 높은 활성을 보였으며, 두 균주의 조효소액을 가지고 immuno-blotting을 실시한 결과, E.coli ATCC 33456에서 분리 정제된 6가 크롬 환원효소의 항체가 약 42Kda에서 반응하는 것을 확인할 수 있었으며, 이 결과는 분리 정제된 6가 크롬 환원효소와 매우 유사한 유전자 산물이 두 균주에서 나온다는 것을 보여주었다.

고온성 박테리아인 Hydrogenobacter thermophilus가 생산하는 cytochrome c-552 유전자에 대한 primer를 제작하여 PCR을 통해 증폭한 후 E. coli에 cloning하였다. 또한 point mutagenesis를 통해 fusion protein을 만들기 위한 기초자료를 마련하였다. 앞으로 cytochrome c-552가 E. coli에서 mutant type으로 발현되기 위한 발현벡터의 개발과 이를 정제하는 기술을 개발하는 것이 필요할 것이다.

The super-producing recombinant H.polymorpha mutant is obtained by double membrane screening technique combined with optimum mutation method. The characterization of mutant is carried out to find the change of mutant in m-RNA level, cell wall leakage, protease level and methanol utilization metabolic flux. The change of these properties of mutant was figured out.

Ferritin, an iron-storage protein, is found in bacteria and some animal tissues such as liver, spleen, and bone marrow. It is more effective and causes less side reactions than the traditional ferrous sulfate, and thus has been used primarily to treat iron deficiency in pregnancy anaemia. Currently, the ferritin extracted from the bovine and equine spleens are sold as a commercial product. Its market is estimated as several hundreds of million US dollars. However, because of the recent warnings against the viral diseases of animal origins such as mad cow disease, a safer ferritin is sought after. Our research team has successfully developed a production process for recombinant human ferritin. Its expression titer from yeast is high enough to be economically viable, and its particle formation characteristics are as effective as well.

We studied the viability and function of islet with monomethoxy polyethylene glycol (mPEG) grafted onto its membrane. Islets were isolated from rat and were repeatedly reacted with activated mPEG (mw 5000) in order to increase grafting density. The density of grafted PEG on the islet membrane was confirmed by Fluorescein-PEG-NHS. An assessment of islet viability using AO / PI staining method showed that multiple PEGylation did not reduce islet viability. The function of PEG grafted islets was evaluated by measuring released insulin from islets. Insulin secreted from the PEGylated islets for 1 h did not show any significant difference compared to control (non-PEGylated) islets. In addition, PEGylated islets responded in the same pattern as control islets in the perifusion test.

대장균에 대한 항체를 수정결정상의 금전극에 고정화하여 압전류적 항체센서 시스템을 제작하고 그 특성을 평가하였다. 그 결과, 정상상태의 센서반응을 얻는데 소요되는 시간이 20분 이내로 단축되었으며 항원 ${\cdot}$ 항체결합에 의한 진동수변화도 대단히 안정적으로 나타났다.

We constructed a biospecific affinity surface using hyper-branched dendrimers on gold for biospecific recognition, and characterized the resulting surfaces by using confocal fluorescence microscopy. The dendrimer monolayer was firstly constructed on the mercaptoundecanoic acid SAM/Au with pentafluorophenyl ester activation and further functionalized with sulfo-NHS-biotin, an activated ester of biotin. To confirm the formation of biospecific affinity surface, FITC(fluorescein isothiocyanate)-labeled avidin was loaded onto the biotinylated dendrimer monolayer, and fluorescence images of the bound avidins were investigated with a confocal microscope. The constructed biospecific affinity surface showed a much more dense and uniform fluorescence compared to those from poly-L-lysine- and cystamine SAM-based affinity surfaces. For the dependency on the concentration of added FITC-labeled avidin on the affinity surface, derived fluorescence could be detectable from as low as $1{\mu}g/ml$, and intensified up to $50{\mu}g/ml$. Further reaction of FITC-labeled avidin layer with TMR(tetramethylrhodamine)-biocytins resulted in the efficient FRET(fluorescence resonance energy transfer) phenomenon. As an extension of the study, we attempted a patterning of the affinity surfaces on gold by microcontact printing. Fluorescence of the patterned surface demonstrated that FITC-labeled avidin molecules were specifically bound to the biotinylated patches.

Membrane strip 면역 크로마토그래피 방법을 이용하여 정량분석을 수행하기 위해 스크린프린팅 기술에 의해 제작된 membrane 전극 상에 항체를 고정화하였고 신호발생물질로써 전도성고분자인 polyaniline이 결합된 colloidal gold를 사용하였다. 이때 사용이 적합한 전도성 고분자는 수용액에 대해 용해도가 높아야 하고 또한 면역반응 최적조건인 중성 pH에서 전기전도도를 유지할 수 있어야 한다. 이를 위해 기존의 polyaniline을 $LiPF_6$로 doping 하거나 새로운 수용성 고분자인 LEB-SPAN을 사용하여 전기전도도 측정용 면역 gold를 합성하였다.

유전자센서 기술은 biomedical analysis를 위해 일반적으로 고체 상에 고정화된 DNA 분자를 이용한다. 이 센서의 검출능력은 주로 capture probe의 서열뿐만 아니라 oligonucleotide의 고체 상에 고정화 방법에 달려있다. 본 연구에서는 glass 표면에 DNA 분자를 고정화시키는 두 가지 다른 방법을 비교하였고 유전자센서의 구성에 대해 검토하였다.

DNA probe assay comprising a microwell as' solid matrix for the immobilization of streptavidin (SA) and an oligonucleotide with covalently bound fluorecein as detection probe was developed. The insolubilized SA captured the biotinylated DNA product of polymerase chain reaction (PCR), and the product was denatured under a basic condition. The remaining single-stranded DNA on the solid surface was hybridized with the probe for signal generation that was performed based on enzyme-linked immuno -reactions.

Six monoclonal antibodies to human cardiac troponin I (hcTnI) were produced to eventually develop an immunosensor for acute myocardial infarction (AMI). For the characterization of these antibodies, a set of 11 different peptides covering selected ranges of the complete amino acid sequence of hcTnI was prepared and used for epitope mapping. Such analysis allowed to select an appropriate pair of antibodies that can form a sandwich type of immune complexes and was consequently used for an immunoassay.

본 실험을 통하여 각 채널에서의 독성 농도에 따른 다양한 발광 패턴을 확보 할 수 있었으며 이를 바탕으로 본 시스템의 실제 현장 적용 시 나타나는 발광 패턴과 비교하여 독성을 분석하고 정성, 정량화 하는데 지표로 사용될 수 있다.

재조합 단백질 생산에서 문제가 되고 있는 번역 후 과정인 glycosylation 을 in vitro 상에서 수행하였다. 원핵생물 시스템에서 재조합 단백질을 생산하고, 이후 효소를 이용하여 올리고당을 붙여 원래의 당단백질과 유사한 단백질을 생산하는 것이 산업적으로 경쟁력을 가질 수 있으므로 이를 위하여 glucose oxidase와 fetuin을 모델 당단백질로, 가수분해 효소인 peptide-N-glycosidase F 의 역반응 활성을 이용하여 glycosylation 을 시도하였다. 역가수분해로의 평형 이동을 위하여 그 기질인 올리고당과 암모니아를 과량 첨가하고, 반응 온도를 높였다. Glucose oxidase의 경우에는 denaturation 했을 때 완전한 deglycosylation 이 일어났지만, fetuin의 경우에는 그렇지 못했다. Glucose oxidase 의 glycosylation 은 수용액상에서는 불가능 했지만 acetone 을 media로 사용하여 $50^{\circ}C$에서 4 시간동안 반응시켰을 때 SDS-PAGE 분석 결과 reglycosylation이 일어나 단백질 밴드가 위로 올라감을 관찰할 수 있었다.

The effects of agitation rate and agitation time on the synthesis of sphingosome were studied. As increase in the agitation rates 4,000, 6,000, 8,000, 10,000 and 12,000 rpm the viscosity of sphingosome were decreased. The most sufficient agitation rate was 8,000rpm for which micell viscosity and stability. The effect of agitation time on the sphingosome viscosity and stability was investigated by changing the agitation times 2, 4, 6, 8 and 10 minute at 8,000rpm. 4 minute was the most sufficient for the viscosity and stability. The sphingolipid activity of cutaneous wound healing in impaired mice was examined. As a result, we could prove that phytosphingosine-HCl medically worked on wound healing well. For the phytosphingosine-HCl, it was found that the experimentally determined medical action more effective than that of tetra-acetylphytosphingosine.

양파의 저온저장중 부패방지를 위한 천연항균제 개발의 일환으로 저장양파로부터 부패세균을 분리한 후 이 균에 대한 식물추출물의 항균활성을 조사하였다. 양파의 저온저장중 과육으로부터 분리한 곰팡이를 MEA와 PDA에서 배양하면서 분생자병과 분생자의 형태적 특성을 조사한 결과 플라스크형의 phialide, 분생자병의 branching type이 simple type이며, conidial head가 columnar shape 가지고 있는 것으로 확인되어 Penicillium sp. ON-211으로 동정하였다. 또한 자생 식물은 채집하여 각종 유기용매로 추출한 후 항균활성을 조사하였다. 정향나무, 탱자나무의 잎은 chloroform 분액에서 높은 활성을 나타내었고, 측백나무 열매는 물분획에서 높은 활성을 나타내었으며, 연필향나무의 잎은 ethyl acetate 분획에서 비교적 높은 항균 활성이 나타났다.

목질계 바이오매스의 구성성분 중 건조중량으로 약 $18{\sim}33%$를 차지하고있는 리그닌은, 기본적 단위물질이 가솔린의 성분물질과 화학적으로 유사한 구조를 형성하고 있기 때문에, 분해하여 저분자물질로 전환시킨다면 연료 또는 연료첨가제로 사용될 수 있다. 본 연구에서는 실험실용 관형반응기를 사용하여 반응온도 $250{\sim}450^{\circ}C$, 반응시간 $20{\sim}80$분의 범위에서 용매상 열분해 ${\cdot}$ 액화반응을 수행하였으며 리그닌의 열분해 ${\cdot}$ 액화반응특성을 조사하기 위하여 전환율을 측정하였는데 아세톤을 열분해 용매로 사용한 경우 가장 높은 전환율은 $350^{\circ}C$, 50분의 55.5%로 측정되었으며, 타르의 발생량은 $250^{\circ}C$의 경우 $260{\sim}350mg/g\;{\cdot}\;lignin$으로 가장 높게 나타났다. 타르성분을 제거한 후 전환율을 측정한 경우 가장 높은 전환율은 $300^{\circ}C$, 30분의 76.88%로 측정되어 열분해시 일차적으로 생성되는 타르의 분해도에 따라 전환율 값이 좌우되며 생성물의 조성과 생성량이 좌우됨을 확인할 수 있었다.

Two groups of bacteria, harmful and useful bacteria, coexist in our body. Harmful bacteria and free radicals which cause many diseases and agings could be controlled by not synthetic materials but natural products. 50 kinds of natural products were searched for this purpose, some of them expressed bacterial controlling ability. Clostridium difficle was controlled by Paeonia Japonica and Lycii Fructus, etc., Clostridium perfringens was inhibited by Scutellaria Baicalensis and Astragali Radix, etc., and same way, Listeria monocytogenes; Paeonia Japonica and Scutellaria Baicalensis, E. coli; Scutellaria Baicalensis and Schizandrae Fructus, Staphylococcus aureus; Paeonla Japonica and Scutellaria Baicalensis, Streptococcus mutans; Coptis Rhizoma, respectively. These natural products will play an important role in bio-Industry.

부상공정을 이용한 수용액으로부터 색을 제거하기 위한 실험을 수행한 결과 다음과 같은 결론을 얻었다. 1. NaLS를 첨가하였을 때 색 제거율은 투여량과 색 농도의 몰비가 0.6에서 96%, 1.0에서 99%로 나타났으며, NaOl을 첨가하였을 때는 0.6에서 70%, 1.0에서 90%로나타나 NaOl 보다 NaLS를 주입한 경우 색 제거효율이 더 우수함을 알 수 있었다. 2. 3종의 아민류 계면활성제를 첨가하여 Direct Orange 31의 색을 제거한 실험에서는 투여량과 색 농도의 몰비가 0.6미만에서는 pH에 의존하였으나 0.9이상인 경우에는 pH에 의존하지 않았으며, dodecylamine과 색 농도의 몰비가 $0.9{\sim}l.0$, 즉 임계투여량인 12 mg/L에서 색 제거 효율이 가장 우수하였다.

Anaerobic digester coupled with hollow fiber membrane unit. Treatment of starch waste with anaerobic digester-membrane system was studied. $0.17\;m^2$ area of hollow fiber membrane unit of known pore size was immersed into laboratory-scale anaerobic digestion system. The gas production was about $0.74\;m^3/kg$ COD treated. The COD removal efficient was about 80-95% depending on the hydraulic retention time. Crossflow ultrafiltration as Post treatment to anaerobic filter. The study conducted with different membrane pore size indicated that membrane with 1,000,000 molecular weight cut-off size gave a higher COD removal efficiency in the range of 83-87% while giving a study flux of $120-130\;L/m^2\;{\cdot}\;h$. A study was conducted to see the long term clogging effect of membrane also.

알려진 dTDP-D-glucose 4,6-dehydratase의 amino acid 서열로 부터 primer를 제작하여 내열성 균주인 Thermus caldophilus GK24에서 colony hybridization 과정을 거쳐 dTDP-D-glucose 4,6-dehydratase를 포함하는 cosmid DNA를 얻었다. 유전자 분석을 위해 cosmid DNA를 subclone 하여 작은 크기로 분리하였다. 분리된 cosmid를 pSMTC-1 으로 명명하고 pSMTC-1를 BamHI으로 반응시켜 BamHI 단편 모두를 pGEM 7(+)를 이용하여 subclone 하였다. 각각의 이름은 크기에 따라 pKCB10(1.2kb-BamHI), pKCB20(1.6kb-BamHI), pKCB30(2.Ikb-BamHI), pKCB40(2.5kb-BamHI), pKCB50(2.5kb-BamHI), pKCB60(2.7kb-BamHI), pKCB70(3.4kb-BamHI), pKCB80(4.4kb-BamHI), pKCB90(7.0kb-BomHI) 으로 명명하였다. 각각의 subclone된 유전자를 분석하기 위해 Erase-a-base 방법을 이용하여 template를 준비하였고 이를 자동 염기서열 분석기를 이용하여 염기서열을 분석하였다. 염기서열분석 결과 pKCB80(4.2kb)에 dTDP-D-glucose synthase(orfA) 유전자를 비롯하여 dTDP-D-glucose 4,6-dehydratase(orfB), orfC (dTDP-4-keto-L-rhamnose reductase) 그리고 orfD(dTDP-4-keto-6-deoxy-D-glucose 3,5-epimerase)와 유사한 유전자들이 있음이 확인 되었고 dTDP-L-rhamnose의 생합성 과정을 예상할 수 있었다.

포도당 결합성 효소의 결합 부위에 관여하는 아미노산중 하나인 asparagine을 이용하여 리포솜 표면에 결합시켜 포도당 민감성 리포솜-아미노산 결합체를 제조하였다. Asparagine이 결합된 리포솜이 그렇지 않은 리포솜이나 증류수에 비해 포도당 결합정도가 컸다. DPPC에 대한 콜레스테롤 함량이 증가할수록 리포솜의 안정성도 증가하였고, zeta sizer로 입도를 측정한 결과 그 분포를 관찰할 수 있었다.

5-fluorouracil loaded chitosan-cellulose microspheres was prepared by W/O/W multiple emulsions solvent evaporation technique which is appropriate to oral drug delivery. The influences of process parameters on the physical characteristics of microspheres and on in vitro drug release were investigated.

폐신문지는 MSW의 많은 부분을 차지하고 있다. 폐신문지는 바이오매스의 성분인 셀룰로오스, 헤미셀룰로오스, 리그닌을 함유하고 있고, 특히 리그닌은 열분해하였을 때 연료 및 연료첨가제를 얻을 수 있었다. 따라서 폐신문지를 열분해 하였을 때도 이와 같은 현상을 얻을 수 있었다. 폐신문지의 열분해 공정에서 최적조건은 $300{\sim}400^{\circ}C$, $40{\sim}50$분으로 확인할 수 있었다. 반응온도는 촉매를 써서 낮출 수가 있을 것이다. 열분해 생성물질은 방향족화합물이 많았는데, 이는 리그닌에서 기인하였다고 볼 수 있을 것이다.

Cerebrosides were isolated from soybean, deunjang and chungkukjang and their constituents were investigated. The principal fatty acid of soybean cerebroside was 2-hydroxy-palmitic acid but chungkukjang cerebroside consisted palmitic acid, elaidic acid(trans 18:1), oleic acid and stearic acid. The only sugar in cerebrosides was glucose. TLC analysis of cerebroside hydrolysate elucidated soybean and chungkukjang had same sphingold bases, mainly dihydroxy bases, trans-4, trans-sphingadienine. But, alkali stable glycolipid fractions of chungkukjang and deunjang had $2{\sim}3$ different primary amines.

강원도 동해안에서 다량 어획되어 가공되는 오징어 및 명태 내장으로부터 추출되는 내장유 용도 확대 및 고부가가치화를 위한 미세 캡슐화를 제조를 위한 조건을 시험하였다. 미세 캡슐 소재로 wall material은 gum arabic과 유화제로 gelatin이 적절하고, gum arabic과 gelatin을 소재로하여 배합비를 달리하였을 때, 전체적인 경향을 유화제가 wall material 보다 많은 것이 양호한 미세 캡슐이 된다는 것을 확인하였다. 상기한 소재로 제조된 미세 캡슐의 캡슐화율은 25-45.5% 정도의 범위로 전체적으로 미세캡슐화율은 낮았다. 향후 CMC, maltodextrin 그리고 키토산 등을 소재로 미세 캡슐화 조건을 좀 더 검색할 필요가 있을 것으로 판단되었다.

시판중인 분말 와사비를 물로 추출하여 제조된 시료를 가지고 Candida bambicola ATCC 11858, Vibrio vulnificus M06-2410, Staphylococcus aureas ATCC 25923, Psudomonas aeruginosa ATCC 27853의 미생물에 미치는 증식 저해효과에 대하여 연구하였다.

Propolis는 꿀벌이 다양한 식물로부터 모아온 수지상의 물질이며 꿀벌의 분비물과 수목의 삼출액 등의 다양한 형태를 함유하고 있다. Propolis는 고대로부터 항생작용과 상처치유에 효과적인 혼합물로 알려져 있다. 또한 에탄올 추출 Propolis는 항박테리아, 항미생물, 항곰팡이, 마취성, 항염증, 통증저하, 면역상승과 세포증식억제작용 특성과 같은 약물학적 활성을 나타낸다고 알려져 있다. 이 연구에서는 국내산 Crude Propolis로부터 얻어진 EEP의 첨가가 미생물의 성장에 미치는 영향을 조사하였다. Paper Disc Diffusion Method와 균주의 성장에 따른 저해 정도를 확인하기 위한 20mg EEP/100mL MH배지에서도 균주의 성장을 저해함을 확인할 수 있었다. 70% EEP와 95% EEP에서 모두 저해를 확인할 수 있었고95% EEP에서 저해활성이 더 좋았다.

A combination of Poisson-Boltzman (PB) equation and quantum mechanics methodologies has been used for the calculation of the solvation energies and $pK_a$ values of biomolecules such as amino acids and nucleotide bases.
