Water temperature is a key environmental factor controlling the epizootics of viral diseases in fish. High water temperature is associated with the rapid spread of rock bream iridovirus (RBIV) disease and with high mortality of RBIV infected fish. Although protection of fish against iridoviral disease by active immunization has been reported, little information is available concerning whether fish survived from an epizootic of iridoviral disease can naturally acquire resistance against the viral disease. In the present study, we have demonstrated that juvenile rock bream, which survived from a natural epizootic of RBIV, acquired resistance against recurrence or reinfection of RBIV, and this resistance was established during the subsequent low water temperature period. Furthermore, the possible involvement of the adaptive humoral immune response in the resistance of the juvenile rock bream was suggested by in vivo neutralization experiment.

수산생물질병관리법이 제정 시행 (2009)된 이후 매년 우리나라 양식산 흰다리새우에 대한 국내 미발생 법정전염병을 모니터링하고 있다. 최근 외래질병의 유입가능성이 증가함에 따라 질병 감시를 강화하고자 조사대상 법정전염병을 2종 (YHD, IMN)에서 5종 (YHD, IMN, IHHN, TS, WTD)으로 확대하고 조사지역과 양식장 개소를 매년 상향 조절하여 실시하였다. 2013년에는 지정 법정전염병 5종에 대해 흰다리새우 종묘생산장 및 양식장 29개소에서 총 2,650마리의 샘플을 조사한 결과, 검사한 모든 시료에서 바이러스가 검출되지 않았다. 새우 양식장에서의 향후 새로운 질병 유입에 대한 위험성을 낮추고 국가적 질병관리 및 차단 방역을 위해서는 지속적인 질병 예찰 및 질병 감시가 필수적일 것이다.

Biosensors consist of biochemical recognition agents like antibodies immobilized on the surfaces of transducers that change the recognition into a measurable electronic signal. Here we report a piezoelectric immunosensor made to detect Vibrio vulnificus. A 9MHz AT-cut piezoelectric wafer attached with two gold electrodes of 5mm diameter was used as the transducer of the QCM biosensor with a reproducibility of ${\pm}0.1Hz$ in frequency response. We have tried different approaches to immobilize antibody on the sensor chip. Concerning the orientation of antibody for the best antigen binding capacity, the antibody was immobilized by specific binding to protein G or by cross-linking through hydrazine. In addition, protein G was cross-linked on glutaraldehyde activated immine layer (PEI) or EDC/NHS activated sulfide monolayer (MPA). PEI was found to be more effective to immobilize protein G following glutaraldehyde activation than MPA. However, hydrazine chip showed a better capability to immobilize more IgG than protein G chip and a higher sensitivity. The sensor system was able to detect V. vulnificus in dose dependent manner and was able to detect bacterial cells within 5 minutes by monitoring frequency shifts in real time. The detection limit can be improved by preincubation to enrich the bacterial cell number.

세균성 어류질병의 치료 및 예방을 위하여 어류와 인체에 안전한 수산용 probiotic의 개발이 요구되고 있다. 본 연구에서는 남해안 일대에서 양식되고 있는 참굴 (Crassostrea gigas), 바지락 (Ruditapes philippinarum), 피조개 (Scapharca broughtonii), 새조개 (Fulvia mutica)의 가식 부위로부터 17균주의 candidate probiotic bacteria (CPB)를 분리하였다. 나아가 다양한 연쇄상구균 (Lactococcus garvieae, L. piscium, Streptococcus sp., S. iniae, and S. parauberis)에 대하여 강한 생장 억제력을 보이는 균주(CPB-St)를 선별하여 어류의 연쇄구균증 관리를 위한 probiotic 균주로서의 개발 가능성을 알아보았다. CPB-St 균주를 double layer test를 통하여 다양한 연쇄상구균들에 대한 생장 억제 정도를 알아보았으며, 혼합 배양에서의 Streptococcus sp.에 대한 생장 억제 능력을 확인하였고, CPB-St의 어류에 대한 안전성 및 장내 생존 유무를 평가하였다. CPB-St는 대부분의 연쇄상구균에 대하여 18~26 mm의 생장 억제대를 형성함으로써 높은 생장 억제 능력을 보였다. CPB-St와의 혼합 배양에서 Streptococcus sp.는 6시간째부터 생장 억제 현상이 관찰되기 시작하여 12시간 전후로 8~55배 정도의 감소를 나타내었다. CPB-St의 어류에 대한 병원성 유 무를 알아본 결과, 2주일 동안 CPB-St로 인한 어류 폐사는 관찰되지 않았다. CPB-St의 1회 강제 경구 투여 후, 위와 장에서 CPB-St의 생존을 확인한 결과 투여 24시간 후에는 CPB-St가 모두 배출되는 것으로 나타났다. CPB-St를 probiotic bacteria로 개발하기 위해서는 어류의 장내 정착 가능성과 먹이를 통한 효과의 정확한 검증이 추후 이루어져야 할 것이다.

본 연구에서는 수온변화에 따른 비소 (As) 노출의 영향을 틸라피아 Oreochromis niloticus의 간과 아가미에서 항산화 방어기작 (antioxidant defense system)을 통해 알아보고자 한다. 틸라피아를 수온이 각각, 20, 25 및 $30^{\circ}C$ 일때, 비소 농도 0, 200 및 $400{\mu}g/L$에서 10일간 노출시킨 후, glutathione (GSH) levels, glutathione reductase (GR), glutathione peroxidase (GPx) and glutathione S-treansferase (GST) 효소 활성을 측정하였다. 비소 노출 이후, 틸라피아의 간과 아가미에서 이들 항산화 효소는 수온 변화에 따라 유의하게 변화하였다. 특히, 다른 온도구간에 비하여 수온이 $30^{\circ}C$ 일 때, 비소에 노출된 틸라피아의 간에서 이들 효소의 변동폭은 가장 유의하게 증가하였다. 즉, 본 연구는 틸라피아의 간과 아가미에서 GSH 및 항산화 효소인 GR, GPx 및 GST에 미치는 비소의 영향은 수온 상승이 동반되었을 때, 어류의 산화 스트레스에 대한 방어 기작의 감소를 촉진시켰음을 보여준다.

넙치에 적합한 표지방법을 조사하기 위하여 표지 후 누적폐사율, 혈액생화학적 성상과 조직병리학적 조사를 하였다. 본 연구에서는 아가미뚜껑 절단, 배지느러미 50%와 100% 절단 방법을 사용하였다. 누적폐사율은 아가미뚜껑 절단구가 가장 높았다. 배지느러미 100% 절단구와 아가미뚜껑 절단구는 절단 후 13일 째 조직병리학적 변화를 나타내었다: 간에서 화농성 염증반응, 체신에서 섬유성 염증반응이 관찰되었다. 본 연구결과 50% 배지느러미 절단구가 가장 효과적인 표지방법인 것으로 판단된다.

본 연구에서는 2013년 4월 제주 소재 육상수조에서 무지개송어 해수순치 과정중에 대량폐사가 발생하여 질병검사를 실시하였다. 병어의 대부분은 체표에 궤양이 형성되어 있었고, 비장 비대, 유문수 부위 및 복강 근육의 출혈 및 위속에 액성물질이 가득 차 있었다. 병원체 검사결과, 기생충, 진균, 바이러스는 검출되지 않았지만 검사 개체 5마리에서 간, 비장 및 신장조직에서 동일한 형태의 세균 colony가 시료 당 200개 이상 분리되었다. 분리세균의 16S rDNA 염기서열 분석 결과, Vibrio anguillarum과 100% (875/875 bp) 일치하였다. 이상의 결과, 무지개송어 해수 순치과정 중에 발생된 질병은 Vibrio anguillarum 감염증으로 사료되었다.
