Separation and purification of elements from alkaline and carbonate nuclear waste solutions

  • This article provides a survey of wet (aqueous) methods for recovery, separation, and purification of uranium from fission products in carbonate solutions during the reprocessing of spent nuclear fuel and methods for removal of radionuclides from alkaline radioactive waste. The main methods such as selective direct precipitation, ion exchange, and solvent extraction are considered. These methods were compared and evaluated for reprocessing of spent nuclear fuel in carbonate media according to novel alternative non-acidic methods and for treatment processes of alkaline radioactive waste.

Separation and Purification of Lipase Inhibitory Peptide from Fermented Milk by Lactobacillus plantarum Q180

  • In this study, we separated and purified lipase inhibitory peptide from fermented milk by Lactobacillus plantarum Q180 with the aim of developing a new functional anti-lipase activity yogurt product. L. plantarum 180 was inoculated into 10% reconstituted skimmed milk and incubated at 37℃ until the pH of the culture reached pH 4.4. The lipase activity was measured using porcine pancreatic lipase. The lipase inhibitory peptides were gradually isolated by ultrafiltration, reversed phase column chromatography (RPC), reversed phase high-performance liquid chromatography (RP-HPLC), and gel permeation high-performance liquid chromatography (GP-HPLC) from the fermented milk by L. plantarum Q180. An ODS-AQ column was used for the RPC, a Vydac C18 column for the RP-HPLC, and a Superdex Peptide HR column for the GP-HPLC. The peptide was composed of Asp, Thr, Ile, Ser, Ala, and Gln, and the anti-lipase activity (IC50) was 2,817 ㎍/mL.

  • A novel prepurification method was developed aiming at increasing yield and purity, also reducing solvent usage for purification of paclitaxel. The use of a micelle and precipitation in the prepurification process allows for rapid separation of paclitaxel from interfering compounds and dramatically reduces solvent usage compared to alternative methodologies. The prepurification process serves to minimize the size and complexity of the HPLC operations for paclitaxel purification. The process is readily scalable to a pilot plant and eventually to a production environment where multikilogram quantities of material are expected to be produced. As much as possible, the process has been optimized to minimize solvent usage, complexity, and operating costs.

  • Gas separation membranes have been developed for decades in various areas to replace the conventional processes. Membrane processes for gas separation have many advantages of energy saving, compact size, and easy scale-up. Nowadays, gas separation processes is widely spreaded in nitrogen generating system, hydrogen generating system, membrane dryer, on board inert gas generating system, natural gas purification, biogas purification and fuel cells. Carbon dioxide separation process using membrane would be a strong candidate of carbon dioxide capturing process. In order to broaden the scope of application of gas separation membranes, development of new materials which can overcome the borderline of Robeson's plot should be necessary, so that many researchers and companies are trying to develop the new materials like polymers containing cardo and spiro group and PIMs (polymers for intrinsic microporosity).

  • A combination separation system is composed of three parts, simple microfiltration unit for the pretreatment of real waste IPA, pervaporation unit with plate and frame type module(the effective membrane area 9,040$cm^2$), and simple ultrafiltration unit as a refiner. Utrafiltration module with hollow fiber membrane(MWCO 10,000) used to purify waste aqueous IPA solution. In addition, the flux of $CMPA-K^+$ composite membrane for waste aqueous IPA solution was very steady-state with long experiment time(30 days). And the standard deviation($\sigma$) was 0.152 and then the coefficient of variation($CV\%$)was 10.82 The IPA concentration on the membrane performance using pervaporation module system could be increased from $89.85wt(\%)$ to more than $99.90wt\%$ in about 8hr at operation temperature of $70^{\circ}C$ using the pervaporation module system. Therefore, a combination separation process system of simple filtration and pervaporation was very effective for the purpose of the IPA purification and reuse front industrial electronic components cleaning process.

Purification of a Mosquitocidal Toxic Protein from B. thuringiensis strain H9B by Immuno-Affinity Chromatography (Immuno-Affinity Chromatography에 의한 B. thuringiensis H9B 균주의 모기살충성 내독소 단백질의 정제)

  • For purification of a 70kDa toxic protein of mosquitocidal delta-endotoxin from B. thuringiensis strain H9B, immuno-affinity chromatography was performed. After separation of 70kDa toxic proteins from the delta-endotoxin of the strain H9B on SDS-PAGE, the 70kDa toxic protein was subcutaneously injected into rabbit for making a polyclonal antibody. A anti-70kDa toxic protein was purified by a column chromatography packed with protein A-sepharose 4B gels. The 70kDa toxic protein from delta-endotoxin of the strain H9B was also purified by an immuno-affinity chromatography packed with CNBr-activated sepharose 4B gels conjugated anti-70kDa toxic protein after elution with 1/10M citric acid-1/5M Na$_{2}$HPO$_{4}$ buffer(pH3.2) containing 0.5M NaCl. The 70kDa toxic protein was purified through only one step-separation system, was demonstrated by SDS-PAGE and immunoblot.

Identification and Characterization of Homoharringtonine from Cephalotaxus koreana

  • A novel purification method was developed for producing homoharringtonine from Cephalotaxus koreana, to guarantee high purity and yield. Our simple, efficient procedure for isolating and purifying homoharringtonine from C. koreanabiomass consisted of solvent extraction, synthetic adsorbent treatment, low-pressure chromatography, followed by high performance liquid chromatography (HPLC). The use of active clay treatment and silica gel low-pressure chromatography in the pre-purification process allowed for the rapid, efficient separation of homoharringtonine from interfering compounds and dramatically increased the yield and purity of crude homoharringtonine for high-performance liquid chromatography (HPLC) purification steps compared with alternative processes. Homoharringtonine could be obtained simply with high yield and purity from biomass using this purification method, while minimizing solvent use and the scale and complexity of HPLC operations for homoharringtonine purification. Purified homoharringtonine was identified and characterized.

Synthesis of molecularly imprinted polymer (MIP) by radiation-induced polymerization and separation of ferulic acid from rice oil using MIP-packed column

  • A molecularly imprinted polymer (MIP) was synthesized by radiation-induced polymerization (RIP), where the ferulic acid was used as a template molecule, 4-vinylpyridine as a monomer and ethylene glycoldimethacrylate (EGDMA) as a cross-linking monomer. The MIP was packed in a glass column using a slurry method for use in medium pressure liquid chromatography (MPLC). The MPLC column was tested for separation and purification of ferulic acid from the rice oil. When repeated three times, the MPLC separation/purification yielded the ferulic acid with the purity higher than ~99%. The chemiluminescence of the luminal (5-amino-2,3-dihydro-1,4-phtalazinedione) measured on a potato disc slide (5.0 mm thick) was enhanced in the presence of ferulic acid, while, without the ferulic acid, the chemiluminescence of luminol on the potato slice disc was not observed, which suggests the ferulic acid obtained from the rice oil can be useful for immunoassay.

Investigation on purification of α-Fe2O3 from zinc smelting iron slag by superconducting HGMS technology

  • Comprehensive utilization of zinc smelting iron slag not only solves environmental problems but also creates huge economic benefits. This study was conducted on the enrichment and recovery of ${\alpha}-Fe_2O_3$ from zinc smelting iron slag by superconducting HGMS technology. Several variables such as slurry flow velocity, slag concentration, magnetic field intensity and the amount of dispersing agent were tested in magnetic separation. In the experiments, obtained optimal magnetic separation parameters were 1.60 T of magnetic flux intensity, 600 mL/min of slurry flow velocity of, 15 g/L of slag concentration of, 0.10 g/L of dispersing agent. Under this condition, the content of ${\alpha}-Fe_2O_3$ was increased from 86.22% to 94.39% that can approach the Chinese national standard requirements (A level) of iron oxide red. It was concluded that using superconducting HGMS technology was an effective method for the purification of ${\alpha}-Fe_2O_3$ from zinc smelting iron slag.
