The experimental evidences of steamed and freeze-dried mature silkworm powder as the calorie restriction mimetics

  • Steamed and freeze-dried mature silkworm powder (SMSP) is a natural food containing a large amount of various functional materials and has various health promoting effects. SMSP is known to increase the life expectancy and healthspan, simultaneously. The accomplishment of extension of healthspan should be possible to achieve by activating various signaling pathways delaying aging in various tissues, not by regulating only a few signaling pathways. Consistent with this notion, SMSP increased the resistant to Parkinson disease by enhancing olfaction and mitochondrial activity in neurons of animal models. In addition, SMSP could enhance the gastrointestinal functions. The animals consumed SMSP showed enhanced alcohol metabolisms, reduced cholesterols in bloods, increased resistance to carcinogens causing liver cancers, and protective effects in alcohol induced stomach ulcers. Furthermore, SMSP was also effective in appearance. The SMSP consumed animals showed reduced skin pigmentations and more hair growth compared with control animals. Taken together, the functional enhancement effects of SMSPs in various tissues and organs, which have been discovered to date, are combined to extend healthspan. Therefore, SMSP can be regarded as calorie restriction mimetics. Further studies in the health promoting effects of SMSP will contribute to identifying new applicable diseases, resulted in increased sales of SMSP and incomes of sericulture farmers.

  • Mature silkworms (MS) of Bombyx mori have enlarged silk glands, which are known to be enriched with various functional materials. A special steaming method has been developed for processing MS with enlarged silk glands edible, by which the MS have become a potential health supplement for improving the quality of life. Steamed and lyophilized MS powder (SMSP) has been strongly believed to have various unidentified beneficial effects for humans. Therefore, the purpose of this mini-review is to summarize the various health promoting functionalities of SMSP, which have been published in the paper so far. Firstly, SMSP increased healthspan as well as lifespan, locomotor ability, and resistance to Parkinson's disease by enhancing olfaction and mitochondrial activity in neurons. Secondly, SMSP increased resistance to carcinogens causing liver cancers, improved both acute and chronic liver function with alcohol metabolism, and prevented alcohol induced stomach ulcers. Thirdly, SMSP reduced UVB-induced skin pigmentations and melanogenesis. Taken together, SMSP can be regarded as a promising natural health product to increase incomes for sericultural farm households in Korea as well as global human health and welfare.

  • Helianthus tuberosus Linne (Compositae) has been widely used as a folk remedy to treat various ailments including fever, bleeding, fracture and contusion. This study was designed to elucidate the lifespan extending activities MeOH extract of the tubers of Helianthus tuberosus Linne (MHT) using Caenorhabditis elegans (C. elegans) model system. In the current study, we found that the lifespan of worms was significantly extended by MHT supplement, dose-dependently. MHT also provided robust protection against various stress environments such as osmotic, thermal and oxidative condition. In addition, elevated antioxidant enzyme activities by MHT resulted in attenuation of intracellular reactive oxygen spices (ROS) levels, suggesting antioxidant capacity of MHT might be associated with longevity properties. Herein, we showed that altered food intake and growth of worms were also involved in the MHT activity. Furthermore, MHT increased body movement in aged worms, indicating possible role for MHT in healthspan.

  • Online application for survival analysis (OASIS) and its update, OASIS 2, have been widely used for survival analysis in biological and medical sciences. Here, we provide a portable version of OASIS, an all-in-one offline suite, to facilitate secure survival analysis without uploading the data to online servers. OASIS portable provides a virtualized and isolated instance of the OASIS 2 webserver, operating on the users' personal computers, and enables user-friendly survival analysis without internet connection and security issues.

  • Humans have been breeding the mulberry silkworm for the long period of time to obtain silk fabric and nutrient-rich pupae. Currently, silkworm larvae, pupae, and silk-Fibroin hydrolysates are registered as food raw materials, while silkworm feces and Bombyx batryticatus are registered as Korean traditional medicines. Among sericulture products, individually recognized health functional food ingredients include silk-protein acid-hydrolysates for immunity enhancement, Fibroin-hydrolysates for memory improvement, and freeze-dried 5th instar and 3rd-day-silkworm powder for lowering-blood sugar. Recently, HongJam produced by steaming and freeze-drying mature silkworms were reported to have various health-promoting effects such as preventing the onset of Alzheimer's disease and Parkinson's disease, enhancing gastro-intestinal functions, improving skin-whitening and hair growth, and extending healthspan. By consuming silkworm products with various health-promoting effects, it is possible to increase the healthspan of human beings, thereby reducing personal and national medical expenses, resulting in increasing the individual's happiness.

Lifespan-extending Effects of Allium hookeri Leaves in Caenorhabditis elegans (삼채 잎 추출물의 예쁜꼬마선충에 대한 수명연장 효과)

  • To evaluate the longevity property of Allium hookeri (Liliaceae) leaves, this study was performed. Ethanol extract of A. hookeri was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions, and lifespan-extending effects of the fractions were checked using Caenorhabditis elegans model system. The most active ethyl acetate-soluble fraction elevated heat stress tolerance, and increased expression of stress resistance protein. Further studies were performed to investigate several aging-related factors such as reproduction, food intake, growth and movement of C. elegans. The results revealed that there were no significant changes in aging-related factors including reproduction and food intake, however, ethyl acetate-soluble fraction treatment led to up-regulation of locomotory ability and growth of aged worms, suggesting ethyl acetate-soluble fraction affected healthspan as well as lifespan of nematode.

Genistein from Vigna angularis Extends Lifespan in Caenorhabditis elegans

  • The seed of Vigna angularis has long been cultivated as a food or a folk medicine in East Asia. Genistein (4',5,7-trihydroxyisoflavone), a dietary phytoestrogen present in this plant, has been known to possess various biological properties. In this study, we investigated the possible lifespan-extending effects of genistein using Caenorhabditis elegans model system. We found that the lifespan of nematode was significantly prolonged in the presence of genistein under normal culture condition. In addition, genistein elevated the survival rate of nematode against stressful environment including heat and oxidative conditions. Further studies demonstrated that genistein-mediated increased stress tolerance of nematode could be attributed to enhanced expressions of stress resistance proteins such as superoxide dismutase (SOD-3) and heat shock protein (HSP-16.2). Moreover, we failed to find genistein-induced significant change in aging-related factors including reproduction, food intake, and growth, indicating genistein exerts longevity activity independent of affecting these factors. Genistein treatment also led to an up-regulation of locomotory ability of aged nematode, suggesting genistein affects healthspan as well as lifespan of nematode. Our results represent that genistein has beneficial effects on the lifespan of C. elegans under both of normal and stress condition via elevating expressions of stress resistance proteins.

Lindera obtusiloba Extends Lifespan of Caenorhabditis elegans

  • Lindera obtusiloba has been widely used as a traditional medicine for the treatment of lots of diseases, including abdominal pain, bruise, and hepatocirrhosis. Here in this study, we elucidated the lifespan-extending effect of methanolic extract of Lindera obtusiloba (MLO) using Caenorhabditis elegans model system. We found that MLO has potent lifespan extension activities under normal culture condition. Then, we determined the protective effects of MLO on the stress conditions such as osmotic, thermal and oxidative stress. To reveal possible mechanism of MLO-mediated lifespan, we further investigated the effect of MLO on the antioxidant enzyme activities and intracellular ROS levels. Our results demonstrated that superoxide dismutase and catalase activities were significantly up-regulated by MLO treatment, resulted in reduced intracellular ROS levels. In this work, we also tested whether MLO-mediated longevity activity was associated with aging-related factors such as food intake and growth. Our data revealed that both of pharyngeal pumping rate and body length were significantly shifted by MLO treatment, indicating these factors were involved in MLO's lifespan-extension effects. Although MLO induces reduction in food intake, the body movement of MLO-fed aged worms was not decreased, compared to untreated control worms, indicating MLO might extend lifespan without affecting healthspan.

Lifespan Extending and Stress Resistant Properties of Vitexin from Vigna angularis in Caenorhabditis elegans

  • Several theories emphasize that aging is closely related to oxidative stress and disease. The formation of excess ROS can lead to DNA damage and the acceleration of aging. Vigna angularis is one of the important medicinal plants in Korea. We isolated vitexin from V. angularis and elucidated the lifespan-extending effect of vitexin using the Caenorhabditis elegans model system. Vitexin showed potent lifespan extensive activity and it elevated the survival rates of nematodes against the stressful environments including heat and oxidative conditions. In addition, our results showed that vitexin was able to elevate antioxidant enzyme activities of worms and reduce intracellular ROS accumulation in a dose-dependent manner. These studies demonstrated that the increased stress tolerance of vitexin-mediated nematode could be attributed to increased expressions of stress resistance proteins such as superoxide dismutase (SOD-3) and heat shock protein (HSP-16.2). In this work, we also studied whether vitexin-mediated longevity activity was associated with aging-related factors such as progeny, food intake, growth and movement. The data revealed that these factors were not affected by vitexin treatment except movement. Vitexin treatment improved the body movement of aged nematode, suggesting vitexin affects healthspan as well as lifespan of nematode. These results suggest that vitexin might be a probable candidate which could extend the human lifespan.

Longevity Property of Ethyl acetate Fraction of Dried Zingiber officinale in Caenorhabditis elegans (건강 Ethyl acetate 분획의 선충에 대한 수명연장 효과)

  • This study was performed to evaluate the lifespan extending effects of the dried ginger (Zingiber officnale Roscoe, Zingiberaceae).
