Bioavailability of Fermented Korean Red Ginseng

  • In an effort to improve ginsenoside bioavailability, the ginsenosides of fermented red ginseng were examined with respect to bioavailability and physiological activity. The results showed that the fermented red ginseng (FRG) had a high level of ginsenoside metabolites. The total ginsenoside contents in non-fermented red ginseng (NFRG) and FRG were 35715.2 ${\mu}g$/mL and 34822.9 ${\mu}g$/mL, respectively. However, RFG had a higher content (14914.3 ${\mu}g$/mL) of ginsenoside metabolites (Rg3, Rg5, Rk1, CK, Rh1, F2, and Rg2) compared to NFRG (5697.9 ${\mu}g$/mL). The skin permeability of RFG was higher than that of NFRG using Franz diffusion cells. Particularly, after 5 hr, the skin permeability of RFG was significantly (p<0.05) higher than that of NFRG. Using everted instestinal sacs of rats, RFG showed a high transport level (10.3 mg of polyphenols/g sac) compared to NFRG (6.67 of mg of polyphenols/g sac) after 1 hr. After oral administration of NFRG and FRG to rats, serum concentrations were determined by HPLC. Peak concentrations of Rk1, Rh1, Rc, and Rg5 were approximately 1.64, 2.35, 1.13, and 1.25-fold higher, respectively, for FRG than for NFRG. Furthermore, Rk1, Rh1, and Rg5 increased more rapidly in the blood by the oral administration of FRG versus NFRG. FRG had dramatically improved bioavailability compared to NFRG as indicated by skin permeation, intestinal permeability, and ginsenoside levels in the blood. The significantly greater bioavailability of FRG may have been due to the transformation of its ginsenosides by fermentation to more easily absorbable forms (ginsenoside metabolites).

발효홍삼이 제2형 당뇨병 환자의 혈당 및 인슐린저항성에 미치는 영향 (Effects of Fermented Red Ginseng Supplementation on Blood Glucose and Insulin Resistance in Type 2 Diabetic Patients)

  • 본 연구는 홍삼추출물을 Bifidobacterium longum H-1으로 발효시켜 인삼사포닌을 체내에서 흡수가 잘 되는 형태로 변화시킨 발효홍삼을 제2형 당뇨병환자에게 아침과 저녁 식전에 한 캡슐씩 하루 780 mg을 섭취하였다. 위약군은 동량의 cellulose를 실험군과 동일하게 제조한 후 같은 방법으로 섭취하도록 하였다. 연구대상자는 약물이나 식사로만 치료하는 총 38명의 제2형 당뇨병 환자로 발효홍삼군 20명(남자 12, 여자 8), 위약군 18명(남자 11, 여자 7)으로 구성되었으며, 두 군의 일반적 특성 및 생활습관은 유의적인 차이가 없었다. 연구결과 발효홍삼을 섭취한 군에서의 공복혈당은 $136.29{\pm}16.45$ mg/dL에서 $127.71{\pm}17.74$ mg/dL로 유의하게(p<0.05) 감소하였으며, 당화혈색소 역시 $7.36{\pm}1.02%$에서 $6.96{\pm}0.93%$로 감소하였다. 특히 당화혈색소가 8% 이상인 고당화혈색소군에서 유의적으로(p<0.05) 감소하는 것으로 나타났다. 인슐린 저항성은 대조군에서는 유의적 변화가 없는 반면, 발효홍삼군에서는 섭취 전 $3.11{\pm}1.13$ mmol/L에서 섭취 후 $2.23{\pm}0.71$ mmol/L로 유의한(p<0.01) 감소를 나타내었다. 혈청지질 변화는 없었으며, LDL-콜레스테롤도 발효홍삼군에서 유의한 변화가 없었다. 간 기능 지표인 AST, ALT, ${\gamma}$-GTP와 신장기능 지표인 BUN, creatinine에서 유의적인 차이를 나타내지 않았으며, 정상범위에 속하여 발효홍삼의 안전성을 확인하였다. 이상의 연구 결과 한국인 제2형 당뇨병 환자에 있어 발효홍삼은 혈당 및 당화혈색소 감소와 인슐린 저항성 개선에 효과적인 것으로 나타났으며, 인체 안전성 지표인 간 및 신장 기능도 정상으로 확인되었으므로 당뇨병 환자의 혈당 개선을 통해 합병증지연 또는 예방에 있어 효과를 기대할 수 있을 것으로 사료된다.

인삼 첨가 비율 및 첨가 시기에 따른 인삼 약주 품질 특성 (Determination of Ginseng Yakju Quality using Different Percentages and Application Dates of Ginseng)

  • We investigated the quality characteristics of fermented ginseng Yakju produced using different percentage contents and application dates of ginseng. Although the pH of fermented ginseng Yakju was not significantly different among the various treatment groups, the total acidity was changed depending on the percentage of ginseng content. the application date of ginseng did not influence the total acid content. The amount of volatile acids was higher at a 0.2~0.3% content of white or red ginseng than in other treatment groups. The ginsenoside contents of white and red ginseng used for making ginseng Yakju were 3,555.0 and 4,447.4 ppm, respectively. In ginseng Yakju, total ginsenoside contents were detected at very small amounts of <10 ppm. A total of 15 volatile components were detected in ginseng Yakju. Higher levels of methanol were produced at early application dates. In sensory evaluations, the preference for aroma was higher at white ginseng levels of 0.5% and 1.0% than for other treatment groups, and the taste was judged to be best at a white ginseng content of 0.3%. In the red ginseng treatment group, a red ginseng content of 0.3% was most preferred for both aroma and taste. The taste preference for white ginseng was at 4 days after application, while the taste of the red ginseng treated group was good at the beginning of fermentation.

복분자와 홍삼 발효 추출물의 복합투여가 고지방 고콜레스테롤 식이를 섭취한 흰쥐의 지질대사 및 비만에 미치는 영향 (Co-treatment with Fermented Black Raspberry and Red Ginseng Extracts Improves Lipid Metabolism and Obesity in Rats Fed with a High-fat and High-cholesterol Diet)

  • 이 연구는 고지방 고콜레스테롤 식이를 12주 동안 랫트에게 공급하여 복분자와 홍삼, 복분자와 홍삼 발효 추출물의 지질대사 및 비만 개선효과를 조사하였다. 정상식이, 고지방 고콜레스테롤 식이, 양성대조군, 유산균발효군, 복분자와 홍삼, 복분자와 홍삼 발효 추출물 투여군의 체중증가, 음수, 식이 섭취량은 그룹간의 유의적인 차이를 보이지 않았다. 하지만 혈중 HDL-콜레스테롤은 고지방 고콜레스테롤 식이와 비교 했을 때 복분자와 홍삼 발효 추출물에서 유의적인 증가를 보였으며, LDL-콜레스테롤, 중성지방은 유의적인 감소율을 보였다. 또한, 간 조직에서 복분자와 홍삼 발효추출물 처치에 의해 HMG-CoA reductase, LDL receptor 및 SREBP-2 mRNA의 발현 증가와 지방생성 억제를 확인하였다. 그리고 복분자와 홍삼 발효 추출물 투여군은 비만인자인 혈중 leptin과 FAS의 농도를 유의적으로 감소시켰을 뿐만 아니라 대변에서 체내 콜레스테롤 배출을 증가시켰다. 이러한 결과로 미루어 보아 복분자와 홍삼 발효 추출물은 혈중 지질 대사 및 비만의 예방에 기여할 것으로 판단된다.

당귀.지황.홍삼 첨가에 따른 발효 청국장의 기능성 변화 연구 (Changes in the Functionality of Cheonggukjang During Fermentation Supplemented with Angelica gigas, Rehmanniae Radix, and Red ginseng)

  • 일반 청국장과 한약재를 첨가한 CKJ-DD, CKJ-RG, CKJDD+RG, CKJ-RED의 기호도 및 기능성을 측정하였다. 색도분석 결과, 5가지 청국장 모두 브라운 계열이었으며, a값 (redness), b값(yellowness)은 5가지 청국장에서 모두 유의적인 차이를 나타냈다. 유리아미노산 분석 결과, 구수한 맛을 나타내는 Aspartic acid, Glutamic acid의 함량이 CKJ보다 한약재를 첨가한 청국장(CKJ-DD, CKJ-RG, CKJ-DD+RG, CKJ-RED)이 더 많은 함량을 나타났으며, 단맛을 나타내는 alanine, glycine, lysine 또한 CKJ에 비해 한약재를 첨가한 청국장(CKJ-DD, CKJ-RG, CKJ-DD+RG, CKJ-RED)이 3~4.5배 더 많은 함량을 나타냈다. 반면, 한약재의 고유한 향에 의해서 쓴맛을 나타내는 leucine은 CKJ에 비해 CKJ-DD, CKJ-RED이 2배의 함량을 더 나타냈다. 한편 혈당강하 조절기전과 관련된 항당뇨 활성 결과에서는 농도 의존적으로 혈당상승억제효과가 증가하는 경향을 나타내지만, CKJ과 한약재를 첨가한 청국장(CKJ-DD, CKJ-RG, CKJ-DD+RG, CKJRED CKJRED) 간의 큰 유의적인 차이는 나타나지 않았다. 또한 5종의 청국장의 기능성을 알아보기 위해, 총 피놀릭성분의 함량 분석 결과, CKJ에 비해 한약재를 첨가한 청국장(CKJDD, CKJ-RG, CKJ-DD+RG, CKJ-RED)의 총 피놀릭 함량이 유의적으로 증가하는 경향을 나타냈다. 항산화 활성(Peroxyl radical 소거능)을 측정한 결과, $10\;{\mu}g/mL$ 농도에서 대조군인 CKJ과 비교하여 CKJ-DD은 비슷한 활성을 보였으나 CKJ-RG는 1.5배, CKJ-DD+RG은 3배 높은 항산화 활성을 나타냈다. 이상의 결과로 다양한 한방청국장 제품의 표준화 및 제품화를 위한 다양한 지표들을 확인 할 수 있었다.

The bioavailability of red ginseng extract fermented by Phellinus linteus

  • For the improvement of ginsenoside bioavailability, the ginsenosides of fermented red ginseng by Phellinus linteus (FRG) were examined with respect to bioavailability and physiological activity. The polyphenol content of FRG ($19.14{\pm}0.50$ mg/g) was significantly higher (p<0.05) compared with that of non-fermented red ginseng (NFRG, $11.31{\pm}1.15$ mg/g). The antioxidant activities in FRG, such as 2,2'-diphenyl-1-picrylhydrazyl, 2,2-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid, and ferric reducing antioxidant power, were significantly higher (p<0.05) than those in NFRG. The HPLC analysis results showed that the FRG had a high level of ginsenoside metabolites. The total ginsenoside contents in NFRG and FRG were $41.65{\pm}1.53$ mg/g and $50.12{\pm}1.43$ mg/g, respectively. However, FRG had a significantly higher content ($33.90{\pm}0.97$ mg/g) of ginsenoside metabolites (Rg3, Rg5, Rk1, compound K, Rh1, F2, and Rg2) compared with NFRG ($14.75{\pm}0.46$ mg/g). The skin permeability of FRG was higher than that of NFRG using Franz diffusion cell models. In particular, after 3 h, the skin permeability of FRG was significantly higher (p<0.05) than that of NFRG. Using a rat everted intestinal sac model, FRG showed a high transport level compared with NFRG after 1 h. FRG had dramatically improved bioavailability compared with NFRG as indicated by skin permeation and intestinal permeability. The significantly greater bioavailability of FRG may have been due to the transformation of its ginsenosides by fermentation to more easily absorbable forms (ginsenoside metabolites).

Inhibitory Effect of Fermented Red Ginseng against Passive Cutaneous Anaphylaxis Reaction and Scratching behaviors in Mice

  • To evaluate the antiatopic effect of Korea red ginseng (RG, steamed root of Panax ginseng CA Meyer, Family Araliaceae) fermented by Bifidobacterium longum H-1 (FRG), its inhibitory effect on passive cutaneous anaphylaxis (PCA) reaction and itching in mice was measured. FRG and its ingredient saponin fraction (FSF) potently inhibited PCA reaction and scratching behaviors. FRG at a dose of 200 mg/kg and FSF at a dose of 50 mg/kg significantly inhibited the scratching frequency by 45% and 47%, respectively. FRG and FSF also inhibited the degranulation and protein expression of tumor-necrosis $factor-{\alpha}$ and interleukin-4 of RBL-2H3 cells induced by IgE-complex. However, polysaccharide fraction of FRG (FPF) weakly inhibited it, compared with FSF. The inhibitory effect of FRG against PCA reaction and scratching behaviors more potently inhibited than that of RG. Based on these findings, FRG can improve allergic skin disorders atopic dermatitis by the regulation of $TNF-{\alpha}$, and IL-4 produced by mast cells and basophils and its degranulation.

Diet of Red Ginseng-Cheonggukjang Improves Streptozotocin-induced Diabetes Symptoms and Oxidative Stress

  • Protective effects of cheonggukjang fermented with 20% red ginseng (RC) were observed in streptozotocin (STZ)-induced diabetic rats by measuring levels of blood glucose, serum lipid profiles, and hepatic reactive oxygen species generating and scavenging enzymc activities. RC diet was prepared by mixing with AIN-76 diet at the final concentration 2%, and it was fed to STZ-induced diabetic rats for 3 weeks. The RC dict was significantly improved body weight, feed efficiency ratio, levels of serum glucose, and serum and hepatic lipids in diabetes. The significantly elevated O type activity of xanthine oxidase in diabetes was also greatly decreased by the RC diet. The treatment of RC showed the improved hepatic glutathione s-transferase activities in the diabetic animals. The present study indicates that cheonggukjang fermented with red ginseng could ameliorate STZ-induccd diabetic symptoms such as aggravated blood glucose levels, serum lipid profiles, and even the conditions of oxidative stress.

붉은덕다리버섯 균사체로 발효한 홍삼 배양액의 cell migration 및 항염 효능에 관한 연구 (Cell migration and Anti-inflammatory Effect of Red Ginseng Extracts Fermented with Laetiporus Sulphureus)

  • 홍삼(Red Ginseng; RG)은 인삼보다 더 높은 생체 흡수율과 다양한 약리효과를 갖는 특이한 진세노사이드(Rg2, Rg3)를 함유하고 있다. 따라서 오랫동안 많은 사람들의 건강을 위해 이용되어 왔다. 또한 발효는 유효한 생리활성을 갖는 저분자의 물질들을 생성하기 때문에 많은 연구자들이 생물학적 활성에 대해 오랫동안 연구해오고 있다. 본 연구에서는 홍삼을 붉은덕다리버섯 균사체로 7일 동안 발효하였다. HPLC 분석 결과 진세노사이드 Rg1, Re 및 Rb2가 각각 0.24, 0.25, 0.16 mg/g에서 0.12, 0.1, 0.03 mg/g으로 함량 감소를 확인하였고, 홍삼 붉은덕다리 균사체배양액(Fermented Red Ginseng; FRG)의 항염, 세포 이동, 항산화, 콜라겐 타입 I 합성과 MMP-1 억제효능에 대한 생물학적 효능을 확인하였다. 그 결과, FRG는 RG보다 항염 및 cell migration 촉진효과가 더 우수하였다. FRG는 LPS로 유도된 RAW 264.7 대식세포의 NO 생성을 억제하였으며, iNOS와 IL-6의 발현을 mRNA 수준에서 억제하였다. 이 결과로 FRG는 새로운 항염소재로서 제안이 가능하다고 사료된다.

Quality and characteristics of fermented ginseng seed oil based on bacterial strain and extraction method

  • Background: In this study, the fermentation of ginseng seeds was hypothesized to produce useful physiologically-active substances, similar to that observed for fermented ginseng root. Ginseng seed was fermented using Bacillus, Pediococcus, and Lactobacillus strains to extract ginseng seed oil, and the extraction yield, color, and quantity of phenolic compounds, fatty acids, and phytosterol were then analyzed. Methods: The ginseng seed was fermented inoculating 1% of each strain on sterilized ginseng seeds and incubating the seeds at $30^{\circ}C$ for 24 h. Oil was extracted from the fermented ginseng seeds using compression extraction, solvent extraction, and supercritical fluid extraction. Results and Conclusion: The color of the fermented ginseng seed oil did not differ greatly according to the fermentation or extraction method. The highest phenolic compound content recovered with the use of supercritical fluid extraction combined with fermentation using the Bacillus subtilis Korea Food Research Institute (KFRI) 1127 strain. The fatty acid composition did not differ greatly according to fermentation strain and extraction method. The phytosterol content of ginseng seed oil fermented with Bacillus subtilis KFRI 1127 and extracted using the supercritical fluid method was highest at 983.58 mg/100 g. Therefore, our results suggested that the ginseng seed oil fermented with Bacillus subtilis KFRI 1127 and extracted using the supercritical fluid method can yield a higher content of bioactive ingredients, such as phenolics, and phytosterols, without impacting the color or fatty acid composition of the product.
