Background: Calcineurin plays a crucial role in T cell activation, cell growth, apoptosis, and angiogenesis, and its over-expression has been implicated in the pathogenesis of cardiomyopathy and stroke. However, the expression and function of calcineurin in the pathologic lesion of chronic inflammatory diseases, like rheumatoid synovium, remain to be defined. This study was aimed to determine the role of calcineurin in inflammatory arthritis and investigate the expression and function of calcineurin in the rheumatoid synovium and synoviocytes, the actual site of chronic inflammation. Methods: Immuno-histochemical staining using specific antibody to calcineurin was perfomed in the synovium of rheumatoid arthritis (RA). Fibroblast-like synoviocytes (FLS) from RA and osteoarthritis (OA) patients were isolated from RA and OA patients, and cultured with IL-1${\beta}$ and TNF-${\alpha}$ in the presence or absence of cyclosporin A, a calcineurin inhibitor. The calcineurin expression was assessed by phosphatase assay and Western blotting analysis. IL-6, -10, -17, matrix metalloproteinase (MMP)-1, -2, -3, and -9 released into the culture supernatants were measured by ELISA. After transfection with GFP-Cabin 1 gene into synoviocytes, the levels of IL-6 and MMPs were measured by ELISA. Results: Calcineurin was highly expressed in the lining layer of synovium and cultured synoviocytes of RA patients. The elevated calcineurin activity in the rheumatoid synoviocytes was triggered by proin flammatory cytokines such as IL-1${\beta}$ and TNF-${\alpha}$. In contrast, IL-10, an anti-inflammatory cytokine, failed to increase the calcineurin activity. The targeted inhibition of calcineurin by the over-expression of Cabin 1, a natural calcineurin antagonist, inhibited the production of IL-6 and MMP-2 by rheumatoid synoviocytes in a similar manner to the calcineurin inhibitor, cyclosporin A. Conclusion: These data suggest that abnormal activation of calcineurin in the synoviocytes may contribute to the pathogenesis of chronic arthritis, and thus provide a potential target for controlling inflammatory arthritis.

In this study, we show that Mycobacterium avium subsp. paratuberculosis MAP1305 induces the maturation of bone marrow-derived dendritic cells (BMDCs), a representative antigen presenting cell (APC). MAP1305 protein induces DC maturation and the production of pro-inflammatory cytokines (Interleukin (IL)-6), tumor necrosis factor (TNF)-${\alpha}$, and IL-$1{\beta}$) through Toll like receptor-4 (TLR-4) signaling by directly binding with TLR4. MAP1305 activates the phosphorylation of MAPKs, such as ERK, p38MAPK, and JNK, which is essential for DC maturation. Furthermore, MAP1305-treated DCs transform naive T cells to polarized $CD4^+$ and $CD8^+$ T cells, thus indicating a key role for this protein in the Th1 polarization of the resulting immune response. Taken together, M. avium subsp. paratuberculosis MAP1305 is important for the regulation of innate immune response through DC-mediated proliferation of $CD4^+$ and $CD8^+$ T cells.
Tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and lymphotoxin-$\alpha$ (LT-$\alpha$, TNF-$\beta$) can initiate and perpetuate human diseases such as multiple sclerosis (MS), rheumatoid arthritis (RA), and insulin-dependent diabetes mellitus (IDDM). TNFs can be blocked by the use of soluble TNF receptors. However, since monomeric soluble receptors generally exhibit low affinity or function as agonists, the use of monomeric soluble receptors has been limited in the case of cytokines such as TNF-$\alpha$, TNF-$\alpha$, interleukin (IL)-1, IL-4, IL-6, and IL-13, which have adapted to a multi component receptor system. For these reasons, very high-affinity inhibitors were created for the purpose of a TNFs antagonist to bind the TNFR and trigger cellular signal by using the multistep polymerase chain reaction method. First, recombinant simple TNFR-Ig fusion proteins were constructed from the cDNA sequences encoding the extracellular domain of the human p55 TNFR (CD120a) and the human p75 TNFR (CD120b), which were linked to hinge and constant regions of human $IgG_1$ heavy chain, respectively using complementary primers (CP) encoding the complementary sequences. Then, concatameric TNFR-Ig fusion proteins were constructed using recombinant PCR and a complementary primer base of recombinant simple TNFR-Ig fusion proteins. For high level expression of recombinant fusion proteins, Chinese hamster ovary (CHO) cells were used with a retroviral expression system. The transfected cells produced the simple concatameric TNFR-Ig fusion proteins capable of binding TNF and inactivating it. These soluble versions of simple concantameric TNFR-Ig fusion proteins gave rise to multiple forms such as simple dimers and concatameric homodimers. Simple TNFR-1g fusion proteins were shown to have much more reduced TNF inhibitory activity than concatameric TNFR-Ig fusion proteins. Concatameric TNFR-Ig fusion proteins showed higher affinity than simple TNFR-Ig fusion proteins in a receptor inhibitor binding assay (RIBA). Additionally, concatameric TNFR-Ig fusion proteins were shown to have a progressive effect as a TNF inhibitor compared to the simple TNFR-Ig fusion proteins and conventional TNFR-Fc in cytotoxicity assays, and showed the same results for collagen induced arthritis (CIA) in mice in vivo.
본 연구는 4종의 재래닭인 연산오계, 현인흑계, 횡성약닭, 황봉의 일반성분, 카르노신, 안세린 함량 및 면역활성을 비교하기 위하여 실시하였고, 대조구로 백색 레그혼을 사용하였다. 시험용 닭을 모두 동일한 조건에서 13주간 사육하여 도계한 후 화학적 성분조사를 실시하였다. 일반성분 분석에서는 4종의 재래닭이 백색레그혼에 비하여 조지방 함량이 높았으며, 콜레스테롤 함량은 현인흑계가 다른 품종에 비하여 높았고 횡성약닭이 유의적으로 낮았다(p<0.05). 면역증진 효능을 측정하기 위하여 대식세포를 이용하여 실험한 결과는 재래닭 4종의 가슴육에서만 대식세포 증식 및 NO 억제 효능이 있었다. 대식세포 탐식작용은 현인흑계와 횡성약닭가슴육에서 유의적으로 증가되었고, 염증 관련 사이토카인(IL-4, IL-10, $IFN-{\gamma}$) 유전자 발현은 백색레그혼에 비하여 4종의 재래닭에서 억제효과가 컸다. 결론적으로 4종류의 재래닭이 백색레그혼에 비하여 면역증진 효능이 높았으며, 특히 가슴육에서 효능이 있었다. 재래닭 품종 간에도 일반성분 및 면역조절능에 차이가 있는 것으로 나타났다.
Coptis japonica (C. japonica) is a perennial medicinal plant that has anti-inflammatory activity. C. japonica contains numerous biologically active alkaloids including berberine, palmatine, epi-berberine, and coptisine. The most well-known anti-inflammatory principal in C. japonica is berberine. For example, berberine has been implicated in the inhibition of iNOS induction by cytokines in microglial cells. However, the efficacies of other alkaloids components on microglial activation were not investigated yet. In this study, we investigated the effects of three alkaloids (palmatine, epi-berberine and coptisine) from C. japonica on lipopolysaccharide (LPS)-induced microglial activation. BV2 microglial cells were immunostimulated with LPS and then the production of several inflammatory mediators such as nitric oxide (NO), reactive oxygen species (ROS) and matrix metalloproteinase-9 (MMP-9) were examined as well as the phosphorylation status of Erk1/2 mitogen activated protein kinase (MAPK). Palmatine and to a lesser extent epi-berberine and coptisine, significantly reduced the release of NO, which was mediated by the inhibition of LPS-stimulated mRNA and protein induction of inducible nitric oxide synthase (iNOS) from BV2 microglia. In addition to NO, palmatine inhibited MMP-9 enzymatic activity and mRNA induction by LPS. Palmatine also inhibited the increase in the LPS-induced MMP-9 promoter activity determined by MMP-9 promoter luciferase reporter assay. LPS stimulation increased Erk1/2 phosphorylation in BV2 cells and these alkaloids inhibited the LPS-induced phosphorylation of Erk1/2. The anti-inflammatory effect of palmatine in LPS-stimulated microglia may suggest the potential use of the alkaloids in the modulation of neuroinflammatory responses, which might be important in the pathophysiological events of several neurological diseases including Alzheimer's disease (AD), multiple sclerosis (MS), Parkinson's disease (PD) and stroke.
토양에서 분리한 세포외 다당류 생산균주 PS-12는 형태학적, 생리학적, 화학적 분석에 의하여 Bacillus sp.에 속하는 균주로 동정되었으며, 분리균주 PS-12는 Bacillus sp. PS-12로 명명하였다. Bacillus sp. PS-127가 생산하는 세포외 다당류는 에탄올 침전, cetylpyridinium chloride (CPC) 침전과 gel permeation chromatography를 이용하여 정제하였으며, 정제된 다당류 PS-12의 당조성은 glucose, mannose, galactose와 fucose가 7:3.2:2:1의 몰비로 구성되어있었다. Bacillus sp. PS-12로부터 분리된 다당류 PS-12를 이용하여 면역증강효과를 확인하였다. TNF-${\alpha}$ 및 lL-6 측정은 RAW264.7 대식세포주를 사용하였으며 cytokine 정량을 위하여 ELISA kit를 이용하였다. RAW264.7 세포주에 대한 PS-12의 세포독성을 확인하기 위하여 세포독성이 10% 미만을 나타내는 농도인 2 ${\mu}g$/ml을 PS-12의 최대농도로 측정하였다. PS-12는 ${\mu}g$/ml에서 TNF-${\alpha}$를 정상세포보다 50배 이상 높은 수치로 생산하였다. 또한 lL-6의 생산을 농도 의존적으로 증가시켰다. 이러한 결과로부터 PS-127가 면역세포에 대해 세포독성을 거의 나타내지 않는 농도에서 대식세포로부터 TNF-${\alpha}$와 11-6의 cytokine 생산을 함으로써 면역증강효과를 나타낸다는 것을 확인하였다.
본 논문은 고수 에탄올 추출물의 항산화 활성 및 항염증 효과를 평가하기 위하여 수행되었다. 총 폴리페놀 및 플라보노이드 함량은 1 mg/mL 농도에서 각각 76.03 ± 1.36 mg of Gallic acid equivalents/g, 182.23±4.32 mg of Rutin equivalents/g으로 나타났고 DPPH와 ABTS radical 소거능은 각각 500 ㎍/mL, 1000 ㎍/mL 농도에서 52.8%, 58.3%의 소거율을 확인하였으며 2000 ㎍/mL 농도에서 환원력을 나타났다. 세포독성 검사 결과, LPS와 함께 처리한 세포에서 고수 추출물 (0-500 ㎍/mL)에 의한 독성이 나타나지 않았고 염증 매개인자인 NO, 염증성 cytokine인 TNF-α 및 IL-1β의 생성량을 농도의존적으로 유의하게 감소시켰다. 또한 염증성 단백질인 iNOS 및 COX-2의 발현 수준을 유의하게 감소시켰고 이런 염증성 단백질 발현을 조절하는 전사인자 NF-κB/MAPK signaling pathway의 활성화 수준도 유의하게 억제시킨 것으로 나타났다. 이와 같이 고수 에탄올 추출물의 항산화 및 항염증 활성을 나타냄으로써 활용 가치가 기능성 식품 및 대체 의약 소재로 이용될 수 있다고 사료된다.
결명자(Cassia obtusifolia) 및 포공영(Taraxacum platycarpum), 유근피(Ulmus macrocarpa)로 제조된 생약재 추출혼합물이 in vitro와 in vivo상에서 아토피 피부염에 미치는 영향을 실험하였다. HMC-1 cell을 이용하여 비만세포의 $\beta$-hexosaminidase 방출량을 측정한 결과, 단일 추출물보다 추출복합물이 탈과립을 억제하는 효과가 우수한 것으로 측정되었다. 그리고 HMC-1 cell에서 PMA와 A23187로 자극하였을 때 생성되는 염증성 cytokine의 분비에서도 추출혼합물의 저해 효과가 가장 높게 나타났다. 아토피 질환모델인 NC/Nga 마우스를 사용하여 생약재 추출혼합물의 아토피 저감효과를 측정한 결과, 진드기 추출물에 의해 발진된 마우스의 귀두께 및 조직학적인 부종 관찰을 통해 생약재 추출혼합물 농도가 증가함에 따라 대조군에 비해 귀 두께 및 부종의 감소가 관찰되었다. 또한 마우스의 혈중 IgE 농도와 이와 관련된 cytokine의 농도를 측정한 결과, 생약재 추출혼합물 투여농도가 증가함에 따라 IgE의 농도가 유의적으로 감소하였고, IgE의 과발현을 유도하는 IL-4의 분비 억제 및 IFN-$\gamma$의 분비가 증가되는 것으로 나타나 생약재 혼합추출물의 섭취가 아토피 피부염 증세 완화 효과에 도움이 되리라 사료된다.
고혈당으로 유도된 염증반응 모델에서 유황양파즙(onion juice)의 항염증 효능을 평가하였다. 고혈당(HG, 25 mM glucose) 조건하에서 48시간 배양된 THP-1 세포들은 정상혈당(NG, 5.5mM glucose) 조건의 세포들에 비해 유의적으로 낮은 세포 생존율을 보였으나, $50-150{\mu}L$ 유황양파즙을 함께 처리하자 NG군 수준으로 회복되었다. 이는, 유황양파즙이 고농도 당에 의한 세포독성을 완화시켜 주었음을 시사한다. 고혈당으로 유도된 HG군에서 염증성 사이토카인 TNF-${\alpha}$는 NG군의 290% 수준까지 분비되어 유의적으로 증가하였으나, $50{\mu}L$ 유황양파즙 처리로 TNF-${\alpha}$의 분비 수준이 전반적으로 저해되었으며, TNF-${\alpha}$의 발현 수준 역시 낮아져, 유황양파즙이 고혈당에 의한 염증반응을 저해하였음을 시사해 주었다. 이는 유황양파즙에서 보고된 높은 수준의 플라보노이드에 의한 효과로 일부 해석되었다. 본 연구는 식품 섭취를 통한 당뇨 및 당뇨합병증 예방의 가능성을 보여줌으로써, 식이인자의 항염증 효능에 관한 향후 연구들의 기초적 근거 자료로 활용될 수 있다.
Objective : This study was performed to estimate the effects of OMC-2010 extract on cytokine production in mouse spleen cells. Methods : Mouse spleen cells were pre-treated with ethanol and water extract of OMC-2010 for 1 h, then stimulated with lipopolysaccharide (LPS, 1 ${\mu}g/ml$) for 48 h. Then the cells were harvested for real-time reverse transcription polymerase chain reaction to detect cytokines. Results : OMC-2010 ethanol extract significantly inhibited the LPS-induced interleukin (IL)-1beta, tumor necrosis factor (TNF)-alpha and IL-5 mRNA expressions, but not shown such changes in IL-6, IL-4, IL-13. OMC-2010 water extract significantly inhibited the LPS-induced TNF-alpha, and IL-5 mRNA expressions, but not shown such changes in IL-1beta, IL-6, IL-4, IL-13. Conclusions : Theses results could suggest that both ethanol and water OMC-2010 extract could inhibit the TNF-alpha and IL-5 mRNA expression.
