Effects of Cadmium Exposure on Tissues of Carassius auratus (카드뮴 노출이 붕어(Carassius auratus) 조직에 미치는 영향)

  • The present study aimed to investigate the level of accumulated heavy metal in various tissues of Carassius auratus after exposure to Cadmium (Cd), histologically and physiologically. After treating C. auratus with Cd, the accumulated Cd in gill tissues was detected to be of the highest content, and showed the lowest content in integument tissues. Also, Cd content increased in a time dependent manner and showed the highest accumulation in the tissues exposed for 20 days. Antioxidant enzyme activities showedhigher activity in the gill and integument than in the kidney and liver tissues. In the case of SOD, antioxidation activity of SOD in all Cd exposed tissues was higher than in unexposed tissues. The activities of SOD and CAT also became higher after Cd exposure. Gill tissues exposed to Cd showed an increased number of mucous cells between lamella in a time dependent manner. In addition, the gills showed morphological changes such as edema, exfoliation of epithelial cells, and fusion of the secondary lamellae. Also, exposure to Cd for 20 days had an effect on gill tissues, causing membrane damage in the mitochondria and nucleus. In kidney tissues, atrophied glomerulus was observed, and the empty space in Bowman's capsule was wider.

Screening of Phenolic Compounds with Inhibitory Activities against HMG-CoA Reductase (페놀 화합물로부터 HMG-CoA reductase 저해 활성 물질 탐색)

  • High level of plasma cholesterol is strongly associated with the development of atherosclerosis and coronary heart disease. Clinical trials designed to reduce plasma cholesterol level by diet or pharmacological intervention have resulted in marked reduction of disease incidence. The enzyme 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase which reduces cholesterol biosynthesis in the liver is the key enzyme of the mevalonate pathway that produces cholesterol. In this study, 71 naturally occurring phenolic compounds were tested for inhibitory activities against HMG-CoA reductase. Eleven compounds out of 71 showed inhibitory activities: three hydrolyzable tannin (geraniin, acetonyl geraniin and pentagalloyl ${\beta}-D-glucose$), four benzoic acid derivatives (benzoic acid, trans-cinnamic acid, 2,4-dihydroxybenzoic acid and 2,5-dihydroxybenzoic acid), and four naphthoquinone derivatives (1,2-naphthoquinone, 1,4-naphthoquinone, plumbagin and shikonin). At the concentration of $10{\mu}g/ml$, 1,4-naphthoquinone inhibited HMG-CoA reductase by 99.4%, and then plumbagin 91.4%, pentagalloyl ${\beta}-D-glucose$ 46.6%, 2,4-dihydroxybenzoic acid 40.9%, shikonin 37.7%, 1,2-naphthoquinone 36.6%, trans-cinnamic acid 32.0%, acetonyl geraniin 30.2%, benzoic acid 28.5%, geraniin 28.3% and 2,5-dihydroxybenzoic acid 22.3%, respectively. $IC_{50}$ values of 1,4-naphthoquinone and plumbagin was $2.1{\mu}g/ml$ and $5.8{\mu}g/ml$, respectively.

Effects of Extracts of Monascus-fermented Angelica gigas Nakai in High-fat Diet-induced Obesity in Rats (고지방 식이 유도 비만 흰쥐에서 홍국발효 참당귀 추출물의 항비만 효과)

  • This study aimed to identify the effects of extracts of fermented Angelica gigas Nakai (A. gigas) with a Monascus purpureus strain on anti-obesity in a high-fat diet (HFD)-induced obesity rat model. Male Sprague-Dawley rats were randomly divided into seven dietary groups (n=8 per group), as follows: a normal diet group (N) and six HFD groups (C: control, HFD and no treatment; AG: HFD +10% A. gigas extracts; FAG2.5: HFD +2.5% fermented A. gigas extracts; FAG5: HFD +5% fermented A. gigas extracts; FAG10: HFD +10% fermented A. gigas extracts; GC: HFD + Garcinia cambogia extracts). FAG-fed rats exhibited effectively lowered rates of increasing body weight and visceral fat accumulation in the HFD-induced obesity model. The activities of several hepatic marker enzymes, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase (LDH), and alkaline phosphatase (ALP), were increased with HFD-induced obesity, but levels of these marker enzymes were significantly decreased in FAG-fed rats. The consumption of FAG reduced triglyceride concentrations in serum to normal levels. FAG-fed rats showed effectively increased leptin concentrations in the HFD-induced obesity model. HFD ingestion induced a significant increase in the thiobarbituric acid reactive substances (TBARS) levels, which was decreased in FAG-fed rats. Hematoxylin and eosin staining and Oil Red O staining of the liver showed that the lipid deposits were decreased via FAG feeding. Moreover, hematoxylin and eosin staining of epididymal adipose adipose tissue showed that the adipocyte were decreased by feeding FAG.

Biochemical Responses in Olive Flounder, Paralichthys olivaceus Fed Diet Supplemented with Fermented Aquaculture Sewage (양식장 배출물 발효물의 사료첨가에 따른 넙치, Paralichthys olivaceus의 생화학적 반응)

  • Effluent of aquaculture industry has caused a growing concern regarding its environmental impact. We assessed the use of flounder farming sewage as supplement of diet, to minimize the impact of aquaculture on the environment or also establish the technique for the recycling of effluent sediment derived from land-based seawater fish farm. In order to investigate the effects of a fermented aquaculture waste on biochemical responses of olive flounder (Paralichthys olivaceus), fermented products of aquaculture wastes were used as test compounds that cause hepatic and renal stress through the induction of oxidative stress in liver and kidney. Hepatosomatic index (HSI), glutathione content and glutathione dependent enzyme were not significantly different and no correlation was found within the different types of fermentation condition or supplement concentration, except for significant increases in 50% fermentation group and 50% concentration group in case of glutathione peroxidase activity and HSI value, respectively. These results showed addition of fermented aquaculture sewage may be an economic artificial sources of diet for fish aquaculture practices without affecting the function and safety in view of biochemical examination.

Effects of Chicken Treated with Hwangki-Beni Koji Sauces on ROS Generating and Scavenging Related Enzyme Activities in Rats Fed with a High Fat and High Cholesterol Diet (황기홍국소스를 처리한 계육이 고지방 및 고콜레스테롤 식이 흰쥐의 ROS 생성 및 소거계 관련 효소의 활성에 미치는 영향)

  • The dietary effects of chicken treated with Hwangki-Beni Koji sauce (HBS) on reactive oxygen species (ROS) generating and scavenging related enzyme activities in rats fed with a high-fat and high-cholesterol diet were investigated. The rats (five rats per group) were divided into a normal control diet group (NC), a high-fat and high-cholesterol diet control group (HFC), HFC plus base sauce-treated chicken supplemented diet group (HFC-BS), and a HFC plus HBStreated chicken supplemented diet group (HFC-HBS), and fed for 5 weeks. Total type T (T) and type O (O) hepatic xanthine oxidoreductase in HFC-HBS were 27.91-35.78% and 24.57-31.84% lower than those of HFC and HFC-BS, respectively. In HFC-HBS compared with HFC and HFC-BS, superoxidase dismutase activity was 62.89-64.50% higher, glutathione S-transferase activity was 19.29-25.17% higher, glutathione content was 25.11-53.30% higher, and lipid peroxide content was 20.29-24.19% lower. Therefore, chicken treated with HBS may prevent liver damage by the ROS formed from a high-fat and high -cholesterol diet.

Effect of fermented soybean curd residue (FSCR; SCR-meju) by aspergillus oryzae on the anti-obesity and lipids improvement (Aspergillus Oryzae로 발효시킨 비지메주의 항비만 및 지질개선 효과)

  • In this study, we designed to confirm the dietary effect of anti-obesity of fermented soybean curd residue (FSCR; SCR-Meju; Biji-meju) by A. oryzae, which is well known as a Korean traditional meju microbe. We observed that body weight gain, serum and hepatic lipid profile, as well as the activity of ROS generating enzyme and ROS scavenging enzyme in high-fat diet induced obese mice fed experimental diet (SCR and SCR-meju). Body weight gain and epididymal fat weight of HC (high-fat diet control) was markedly higher than that of NC (Normal control). Conversely, body weight gain and epididymal fat weight of the SCR (Biji) and SCR-meju (Biji-meju) group was significantly lower than that of HC; these of the SCR-meju group was lower than that of the SCR group. Furthermore, serum TG and total-cholesterol and LDL-cholesterol contents of SCR and SCR-meju groups were lower than that of HC, and HDL-cholesterol level of the SCR-meju group was significantly higher than that of HC. In conclusion, although precise mechanisms of the antiobese effects of SCR-meju in this study are unknown, the present study provides an experimental evidence that SCR-meju may prevent obesity and obesity related metabolic syndromes, such as hyperlipidemia, hypertension and diabetes, and liver disease by high-fat diet. Nevertheless, further study in this filed will be needed.

The Antioxidative Effect of Black Garlic Extract on Paraquat-induced Oxidative Stress in ICR Mice (Paraquat에 의해 유발된 산화적 스트레스에 의한 흑마늘 추출물의 항산화 효과)

  • We investigated the antioxidative effect of black garlic extract (BGE) on paraquat (PQ)- induced oxidative stress in mice. A DPPH radical scavenging activity assay showed that BGE had potent free radical scavenging activity, comparable to that exhibited by vitamin C. Mice were administered with either vitamin C or two levels of BGE by oral gavage for 10 days, with PQ being injected intraperitoneally on day five. It was found that BGE reduced the liver enzyme levels induced by PQ injection in mice. The concentrations of plasma and hepatic malonedialdehyde were both significantly reduced in the BGE groups compared with the levels in the PQ group, whereas the hepatic superoxide dismutase and catalase activities were significantly increased in the BGE groups compared with the PQ group. These findings suggest that BGE has potent antioxidative activities in vivo and thus could prevent the oxidative stress induced by PQ injection in mice by two mechanisms: the induction of antixoxidative enzyme systems and direct scavenging of reactive oxygen species.

Studies on the Influence of Certain Heavy Metals on Acid Phosphatase Activities (수종 중금속에 의한 Acid Phosphatase의 영향에 관한 연구)

  • Mice were dosed with $HgCl_2$ (5, 10 and 20 mg per kilogram body weight) and $CdCl_2$ (10, 15 and 20 mg per kilogram body weight) by the abdominal injection. Acid phosphatase activites of the liver at 24, 48 and 72 hours following the injection were measured by the Mundry colorimetric method using disodium p-nitrophenyl phosphate as the substrate, and the following results were obtained. The enzyme activities measured were 3.47 mg Pi/ml/0.5 hr at 24 hr and 5.00mg/ml/0.5 hr at 72hr respectively following the injection of 5 mg/kg body weight of $HgCl_2$ and 6.79 mg Pi/ml0.5 hr at 24 hr and 3.47 mg Pi/ml/0.5 hr respectively following the injection of 20 mg/kg body weight of the mercury compound, as compared with the activity of 8.3 mg Pi/ml/0.5 hr for the control. With the cadmium treatment, about 50% of the animals injected with 10mg/kg body weight, and none of the animals injected with 15 and 20mg/kg body weight, survived. Of the surviving animals, the sublethal concentration of cadmium was shown to activate the enzyme: the activities at 24, 48 and 72hr following the injection were around 11.2 mg Pi/ml/0.5 hr as compared with 8.63 mg Pi/ml/0.5 hr for the control.

Protective Effect of 3,5-Dicaffeoylquinic Acid Isolated from Ligularia fischeri against Oxidative Damage in HepG2 Cells (HepG2 세포에서 산화적 손상에 대한 곰취 유래 3,5-Dicaffeoylquinic Acid의 보호 효과)

  • This study was conducted to investigate the hepatoprotective effects of 3,5-dicaffeoylquinic acid (3,5-DCQA) isolated from Ligularia fischeri against hydrogen peroxide-induced oxidative stress in HepG2 cells. Antioxidative effects of 3,5-DCQA were determined by measuring antioxidant enzyme [superoxide dismutase (SOD), catalase (CAT) glutathione peroxidase (GPx)] expression levels against hydrogen peroxide-induced oxidative stress using real-time PCR analysis. 3,5-DCQA treatment significantly increased gene expression levels of SOD, CAT, and GPx in a dose-dependent manner ($10{\sim}30{\mu}g/mL$) in HepG2 cells. Hepatoprotective effects were analyzed by measuring glutamic oxaloacetic transaminase (GOT), lactate dehydrogenase (LDH), and gamma-glutamyl transferase (GGT) activities using a biochemistry analyzer in hydrogen peroxide-treated HepG2 cells. 3,5-DCQA treatment significantly reduced GOT, LDH, and GGT activities in a dose-dependent manner ($10{\sim}30{\mu}g/mL$) against increased liver function index enzyme activities induced by hydrogen peroxide oxidative stress in HepG2 cells. The results reveal that 3,5-DCQA compound isolated from Ligularia fischeri can be useful for the development of an effective hepatoprotective agent.

Effects of Cudrania Tricuspidata Root Extract (CTE) on Ethanol-Induced Hangover via Modulating Alcohol Metabolizing Enzyme Activities and Blood Gas Levels in Rats (꾸지뽕나무 뿌리 추출물의 알코올 대사 효소 활성 및 혈액 산성화 기전 조절을 통한 숙취해소 효과)

  • To investigate the anti-hangover effects of Cudrania tricuspidata root extract (CTE), the blood alcohol metabolism and blood gas imbalance of CTE in rats treated with 10 ml/kg alcohol were examined. CTE (500 mg/kg and 750 mg/kg) was administrated after 30 minutes of alcohol consumption (10 ml/kg). Blood collection was implemented from the tails of the animals after 1, 3, and 5 hours post alcohol consumption. The Condition drink (a commercial anti-hangover beverage) was used as a positive control. Single administration by the oral route was performed. The consumption of CTE (500 mg/kg and 750 mg/kg) decreased the serum alcohol concentration by increasing and maintaining both the alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) enzyme activity levels in the blood and liver. In addition, CTE led to recovery from the imbalances in the blood gas levels, including carbon dioxide ($CO_2$) and changes in pH, bicarbonate ($HCO_3{^-}$) and lactic acid levels due to alcohol ingestion. In conclusion, CTE exerted a more pronounced anti-hangover effect than a commercial anti-hangover drink. Therefore, CTE can be a novel and safe anti-hangover natural product agent for the prevention or treatment of symptoms caused by excessive alcohol consumption.
