Separation and Purification of Lipase Inhibitory Peptide from Fermented Milk by Lactobacillus plantarum Q180

  • In this study, we separated and purified lipase inhibitory peptide from fermented milk by Lactobacillus plantarum Q180 with the aim of developing a new functional anti-lipase activity yogurt product. L. plantarum 180 was inoculated into 10% reconstituted skimmed milk and incubated at 37℃ until the pH of the culture reached pH 4.4. The lipase activity was measured using porcine pancreatic lipase. The lipase inhibitory peptides were gradually isolated by ultrafiltration, reversed phase column chromatography (RPC), reversed phase high-performance liquid chromatography (RP-HPLC), and gel permeation high-performance liquid chromatography (GP-HPLC) from the fermented milk by L. plantarum Q180. An ODS-AQ column was used for the RPC, a Vydac C18 column for the RP-HPLC, and a Superdex Peptide HR column for the GP-HPLC. The peptide was composed of Asp, Thr, Ile, Ser, Ala, and Gln, and the anti-lipase activity (IC50) was 2,817 ㎍/mL.

Determination of Chiisanoside in Acanthopanax Species by High Performance Liquid Chromatography

  • The content of chiisanoside in the Acanthopanax Species was determined by reversed-phase high performance liquid chromatographic method. Chiisanoside was separated from the other components in the plant extracts using Zorbax 300 SB $C_{18}$ column with gradient elution of acetonitrile. Identification of chiisanoside was carried out by comparison in the LC/MS spectrum of separated peak from extract with that of standard. By HPLC analysis in this experiment, Acanthopanax species could be classified into two groups based upon the content of chiisanoside-one with low concentration of chiisanoside, such as A. senticosus and A. koreanum, and another with high concentration of chiisanoside, such as A. senticosus f. inermis, A. Divaricatus var. albeofructus, and A. chiisanensis.

  • Complex, ill-defined mixtures of natural origin are often used as nutrients in the production of biological products through microbial fermentation. Product yields are affected by variation in these natural products. Yeast extract is a typical example of these natural products. Since it is a mixture of amino acids, peptides and nucleic acids, its composition is not well characterized. In this study, we investigated the properties of thiamine hydrochloride, riboflavin and pyridoxine hydrochlride in yeast extract by using a gel filtration chromatography, ion exchange chromatography and high performance liquid chromatography. Yeast extract solution was fractionated by gel filtration chromatography and ion exchange chromatography, and then, each fraction was analyzed by using a high performance liquid chromatography.

팜유(Plam Oil)의 탈산소 공정 중 운전 조건이 생성물의 조성에 미치는 영향 (Effects of reaction conditions on composition of the organic liquid product during the deoxygenation process of palm oil)

  • 식물성 오일을 이용한 바이오 항공유의 제조공정에서 탈산소 반응의 적절한 운전조건 선정을 통한 생성물 물성 최적화는 최대의 바이오항공유 수율을 얻기 위해 필수적인 요소이다. 이에 따라 팜유의 탈산소화 반응이 1 wt.% $Pt/Al_2O_3$촉매가 장입된 내경이 1인치인 고정층 반응기에서 수행되었다. 업그레이딩 공정을 통하여 수송 연료로 활용될 수 있는 액체 생성물(organic liquid product)은 가스크로마토그래피 방법으로 그 조성을 분석하였다. 피드 내의 팜유/수소 비율과 수소 압력은 탈카르복실레이션과 수첨탈산소 반응에 영향을 주어 생성물의 조성 변화를 초래하였다. 반응 온도가 증가함에 따라 탈산소 생성물의 연속적 크래킹 반응이 촉진되어 $C_5{\sim}C_{14}$영역의 생성물 조성이 증가하였다. 본 연구의 결과는 팜유의 탈산소화 반응 특성의 이해 뿐 아니라 연속 공정인 수첨 업그레이딩 공정을 통한 바이오 항공유의 제조에 도움을 줄 수 있다.

홍삼 및 기능성 소재를 이용한 주류첨가용 액상제제 개발 (Development of Liquid Phase Product from Red Ginseng and Medicinal Herbs for Alcoholic Beverage)

  • 본 연구에서는 홍삼 농축액의 crude saponin과 생약재의 물추출물이 ADH와 ALDH의 활성에 미치는 영향 및 이들 생약재를 이용한 주류첨가용 액상제제 개발을 위한 모델을 제시하였다. 재료자체의 비중이 커서 용매비율을 낮게 한 갈근의 경우, 추출에 따른 고형분의 함량이 매우 낮았고, 3시간이후 고형분의 함량에는 변화가 없었다. 갈화, 지구자, 상엽의 경우는 4시간이후 고형분 함량의 변화가 없었다. 홍삼의 crude saponin, 상엽, 갈화, 갈근, 지구자의 경우 적정농도에서 ADH의 상대활성을 104-114%까지 증가함을 보였고, 상엽, 지구자는 ALDH의 상대활성을 102∼106%까지 증가시켰다. 알코올 분해에 영향을 미치는 생약재를 원료로 액상제제를 제조하여 소주에 혼합한 후 1차 소비자 기호도 조사 결과, 경옥고향, 쌍화향, 칡향을 첨가한 세가지 시료 상호간에 유의적인 차이를 나타냈고, 쌍화향을 첨가한 시료가 가장 양호한 것으로 나타났다. 1차 기호도 조사결과 가장 양호한 것으로 나타난 쌍화향을 첨가한 시료와 향 무첨가 시료, 시판중인 K사제품간의 2차 기호도 조사 결과, 세가지 시료 상호간에 유의적인 차이를 보이지 않았다.

  • 열화학적 물분해에 의한 수소 생산 공정 중의 하나인 IS(Iodine-sulfur) 사이클에서 요오드와 이산화황, 그리고 물을 반응물로 하여 요오드화수소와 황산을 제조하는 분젠 반응에 대한 연구를 수행하였다. 요오드의 투입 몰수에 관계없이 황산의 생성량은 일정하였으나 요오드화수소의 생성량은 요오드의 투입 몰수가 증가함에 따라 감소하는 경향을 나타내었다. 이는 생성된 요오드화수소와 미 반응된 요오드가 $HI_3$ $HI_5$ or $HI_7$와 같은 착화합물인 polyiodide를 형성하기 때문인 것으로 생각된다. 이들 착화합물의 형성은 생성물의 2액상 분리 특성의 향상을 가져온다. 또한 요오드 투입 몰수의 증가함에 따라 반응 속도는 향상되었다. 반응물인 요오드의 투입 몰수 및 반응 온도가 증가함에 따라 생성 용액의 2액상 분리 특성이 향상되었으며 모든 실험의 조건 하에서 부반응은 발생 되지 않는 것으로 나타났다.

  • Hydrogen is currently produced from natural gas reforming or industrial process of by-product over than 90%. Additionally, there are green hydrogens based on renewable energy generation, but the import of green hydrogen from other countries is being considered due to the output variability depending on the weather and climate. Due to low density of hydrogen, it is difficult to storage and import hydrogen of large capacity. For improving low density issue of hydrogen, the gaseous hydrogen is liquefied and stored in cryogenic tank. Density of hydrogen increase from 0.081 kg/m3 to 71 kg/m3 when gaseous hydrogen transfer to liquid hydrogen. Density of liquid hydrogen is higher about 800 times than gaseous. However, since density and boiling point of liquid hydrogen is too lower than liquefied natural gas approximately 1/6 and 90 K, to store liquid hydrogen for long-term is very difficult too. To overcome this weakness, this paper introduces storage method of hydrogen based on liquid/solid (slush) and facilities for producing slush hydrogen to improve low density issue of hydrogen. Slush hydrogen is higher density and heat capacity than liquid hydrogen, can be expected to improve these issues.

  • Sterile Products may be broadly classified into two main categories, according to the manner in which they are produced: those which are sterilized after the product has been filled and sealed in the final container(s) ("terminally sterilized" products) and those where the sterilization stage (or stages) takes place is it before or after the bulk product filled in to final container. In this latter instance, all subsequent processing (typically, the filling and sealing operations) must be conducted aseptically in order to prevent recontamination of the sterilized product. The two most common pharmaceutical applications of aseptic processing methods are (a) the filling of liquid products following sterilization by filtration and (b) the filling of previously sterilized bulk powder products. An aseptic processing operation should be tested using a microbiological growth medium (media fill) during lyophilized injection formulation, filling, loading, lyophilisation, stoppering, and unloading activities.

Biotransformation of Ginsenoside Rb1 to Prosapogenins, Gypenoside XVII, Ginsenoside Rd, Ginsenoside F2, and Compound K by Leuconostoc mesenteroides DC102

  • Ginsenoside $Rb_1$ is the main component in ginsenosides. It is a protopanaxadiol-type ginsenoside that has a dammarane-type triterpenoid as an aglycone. In this study, ginsenoside $Rb_1$ was transformed into gypenoside XVII, ginsenoside Rd, ginsenoside $F_2$ and compound K by glycosidase from Leuconostoc mesenteroides DC102. The optimum time for the conversion was about 72 h at a constant pH of 6.0 to 8.0 and the optimum temperature was about $30^{\circ}C$. Under optimal conditions, ginsenoside $Rb_1$ was decomposed and converted into compound K by 72 h post-reaction (99%). The enzymatic reaction was analyzed by highperformance liquid chromatography, suggesting the transformation pathway: ginsenoside $Rb_1$ ${\rightarrow}$ gypenoside XVII and ginsenoside Rd${\rightarrow}$ginsenoside $F_2{\rightarrow}$compound K.

  • Difficulties in developing process of Liquid Crystal Display(LCD) products such as frequent design modifications, various design requirements, and short-term development period bring on the need of integrated design approach that is efficient and easy to handle. Back Light Unit(BLU) of the LCD, which drastically affects the optical performance of LCD products, is divided into in-coupling part and out-coupling part. Serration of the in-coupling part flattens the light received from point light sources and dot pattern of the out-coupling part regularizes the light sent to screen. Therefore, the optical performance of a LCD product is largely influenced by the shape of serration and the arrangement of dot pattern. In this research, a new design approach which enables to improve the optical performance of LCD products and overcome the prementioned difficulties in developing process of LCD products is proposed. The shape of serration is parameterized to 3 parameters and out-coupling part is partitioned into 10 partitions to apply the optimization technique to this problem. 3 parameters for the shape of serration and densities of 10 partitions are used as design variables in the design optimization. Optical simulation tool named SPEOS is used to evaluate the optical performance of the LCD product. Since the optical simulation uses the random ray tracing technique, numerical noise may possibly be included in the simulation process. To solve the problem caused by numerical noise, the PQRSM which can stably find the solution of the noise problem is used in this research.
