청열소독음(淸熱消毒飮)과 구성약재의 암세포 독성, 항균 및 항산화 효과 (Cytotoxicity, Antibacterial and Antioxidant Activities of the Prescription Cheongyeolsodokum and Its Constituent Herbs)

  • Objectives : The purpose of this research was physiological activities, cytotoxicity, and antibacterial activities of the Cheongyeolsodokum and its constituent herbs. Methods : Physiological activities, cytotoxicity, and antibacterial activities were examined through the Cheongyeolsodokum and its constituent herbs. Results : In the physiological activities, the electron donating ability(EDA) of the water and ethanol extracts from the Cheongyeolsodokum were over 60% and 80% at 100 ppm, respectively. The EDA of the water and ethanol extract from the Cheongyeolsodokum ingredients were gradually increased as well. Water and ethanol extracts from the Cheongyeolsodokum and its constituent herbs inhibited xanthine oxidase activity, they showed superoxide dismutase(SOD)-like activity. The Cheongyeolsodokum and its constituent herbs inhibited cancer cell growth in a dose-dependant manner. Also, the clear zones against Staphylococcus aureus and S. epidermidis were clear shown at 2.5 and 5 mg/disc. Its constituent herbs showed the clear zone against various bacteria such as Candida albicans, S. aureus, S. epidermidis, and S. mutans were shown at 0.5 and 1mg/disc. Conclusions : We observed physiological activities, cytotoxicity, and antibacterial activities of the Cheongyeolsodokum and its constituent herbs. The results also indicated that water extract and ethanol extract of Cheongyeolsodokum and its constituent herbs can be used as a natural ingredient in food or cosmetic industry.

  • Objectives : The purpose of this research was physiological activities and investigate cytotoxicity of Sunbanghwalmyung-um extract. Methods : Physiological activity and a cytotoxicity were examined through the hot water and ethanol extracts from Sunbanghwalmyung-um and its ingredient. Results : The electron donating ability(EDA) was 66.9%, 71.3% in 1000 ppm water extract and ethanol extract. Above 90% from 1000 ppm ethanol extract showed a higher activities and it is Rhei Radix et Rhizoma, Lonicerae Flos, Gleditsiae Spina, Glycyrrhizae Radix, Myrrh. SOD-like activity was weak as 12.24%, 16.62% in 700 ppm. In water and ethanol extracts cytotoxicity were against G361, B16F10, MDA, A549, high cytotoxity over 70%. Rhei Radix et Rhizoma, Gleditsiae Spina, Trichosanthis Radix, Paeoniae Radix Rubra showed high cytotoxicity in water and ethanol extracts. Conclusions : We observed physiological activities and investigated cytotoxicity of Sunbanghwalmyung-um and its ingredients. The results also demonstrated in food or cosmetic industry.

  • Objectives : In this paper, we tested the applicability of shell from Persicae semen in cosmetics through cosmeceutical activities including anti-oxidant, tyrosinase inhibition and anti-inflammatory effects. Methods : Persicae semen, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect have been investigated in the cosmeceutical activity measurement of function experiment. Results : In the electron donating ability test, 1,000ppm of EPS (ethanol extract of shell from Persicae semen) showed an effect of 87%. SOD-like activities showed 93% at the 10,000ppm of WPS (water extract of shell from Persicae semen). In the xanthine oxidase inhibition test, 1,000ppm of BHA showed an effect of 27%, while EPS showed an effect of 62%. We were able to get an effect of 95% from EPS at 10,000 ppm in the tyrosinase inhibition test. In the anti-inflammatory test, the EPS inhibited the generation of nitric oxide. In the case of the EPS, there were no signs of cytotoxicity against raw 264.7 and anti-inflammatory effects could be identified when the manifestation of iNOS was decreased. Conclusion : Therefore, the EPS has potential as an effective raw materials for cosmetic.

벨벳콩(Mucuna pruriens) 추출물의 항산화 효능 검증 (Verification of Antioxidant Activity from Velvet Bean (Mucuna pruriens) Extracts)

  • 본 연구에서는 벨벳콩(Mucuna pruriens, M. pruriens) 추출물을 활용하여 천연소재 기반 항산화 기능성 화장품 소재로서의 활용가치를 검증하고자 하였다. 벨벳콩을 물과 70% 에탄올, 70% 아세톤을 용매로 하여 추출, 농축, 동결건조한 다음 각각의 분말화된 벨벳콩 열수 추출물(MW), 벨벳콩 70% ethanol 추출물(ME), 벨벳콩 70% acetone 추출물(MA) 시료를 확보하였다. MW, ME, MA를 활용하여 DPPH 라디칼 소거 활성, ABTS+ 라디칼소거 활성, SOD 유사 활성, 환원력, 총 페놀 및 플라보노이드 함량 분석을 통해 항산화 활성을 분석한 결과, 모두 우수한 항산화 효능을 나타내었다. 따라서 벨벳콩 추출물은 항산화 관련 기능성 화장품 천연소재로서의 활용 가능성이 높을 것으로 사료된다.

  • The aim of this study was to assess the cosmeceutical activies of four kinds of Korea herb medicine extracts using in cosmetics and related industries. The cosmeceutical activities of extracts were investigated by tyrosinase inhibition, astringent, anti-bacterial and MTT assay for cell viability. In the whitening effect, PA that the highest tyrosinase inhibition activity showed 56% at 10 ppm in ethanol extract. Also water and ethanol extract of RE showed 54%, 68% at 1,000 ppm, respectively, but LE and CA showed lower effect. Astringent effect of water and ethanol extract of PA appeared over 60% at 1,000ppm concentration but other extracts showed no astringent effect. In the anti-bacterial test, water and ethanol extract of PA showed no anti-bacterial effect against all microorganisms. But water and ethanol extract of RE showed anti-bacterial effect on Staphylococcus aureus, ethanol extract of showed on Staphylococcus aureus and Propionibacterium acnes, ethanol extract of CA showed on Candida albicans. The resUlt of stability test showed that the emulsion of containing PA were very stable at various temperature and sun-light test. Viscosity and pH of emulsion did not change. From the results of human patch test to assess the safety of cosmetics containing PA there was no negative reaction on skin was found.

화피(樺皮)의 항산화 효과 및 tyrosinase 저해활성 (Antioxidant Effects and Tyrosinase Inhibition Activity of Extract of Prunus sargentii Rehder)

  • 본 연구는 화피의 효소학적 생리활성을 검토하였다. 효소학적 생리활성 실험에서의 전자 공여능은 100 ppm에서 에탄올추출물은 85% 이상의 효능을 나타냈으며, SOD-유사활성능은 1,000 ppm에서 50%의 활성을 나타내었으며, 농도가 증가함에 따라 유의적인 차이를 나타내었다. Xanthine oxidase의 저해활성을 관찰한 결과 1,000 ppm에서는 70% 이상으로 저해효과를 나타내었다. 피부 미백과 관련 있는 tyrosinase 저해효과를 측정한 결과 1,000 ppm 이하의 농도에서 20%의 저해효과를 나타내었다.

  • Objectives : This is the study of the application as the ingredients of cosmetics through the examination of the function for physiological activity of Chestnut inner shell. Methods : Chestnut inner shell, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect, nitric oxide inhibition have been investigated in the physiological activity measurement of function experiment. Results : We used BHA and kogic acid for the comparative. As a result of testing electron donating ability, at over 100ppm of water extract and ethanol extract, BHA showed relatively high donating ability by more than 90%. And as a result of measuring SOD like activity, 1000ppm of water extract showed an effect of 30% and ethanol extract showed an effect of 40%, BHA showed an effect of 30%. In the xanthine oxidase inhibition test, 1000ppm of water extract showed an effect of 70% and ethanol extract showed an effect of 63%, BHA showed an effect of 100%. In the tyrosinase inhibition test, 1000ppm of water extract showed an effect of 55% and ethanol extract showed an effect of 87%, Kogic acid showed an effect of 98%. In the anti-inflammatory test, the water extract and ethanol extract inhibited the generation of nitric oxide. Conclusions : The results indicated that extract of Chestnut inner shell can be used as a natural ingredients with biological function in cosmetics ingredients.

진달래꽃(Rhododendron mucronulatum Turczaninow) 추출물의 항산화, 항암 효과 및 tyrosinase 저해활성 (Antioxidant, Anticancer and Tyrosinase Inhibition Activities of Extracts from Rhododendron mucronulatum T.)

  • Several biological activities of Rhododendron mucronulatum were investigated. The electron donating abilities of ethanol extracts of Rhododendron mucronulatum were more than 90% at 100 ppm, 500 ppm and 1,000 ppm. Xanthine oxidase was inhibition about 46% by the ethanol extracts of R. mucronulatum at 500 ppm 48% of tyrosinase activity relating to skin-whitening was shown at 1,000 ppm. Uniquely, the anti-microbial effects of water extract and ethanol extract were shown only on Staphylococcus aureus. The water extract 1 mg/disc showed the higher activity than ethanol extract. The growth inhibition effect of each sample on lung cancer (A549) and melanoma (B16F10) cell lines were over 70% at 1,000 ppm, while the effects on the melanoma (G361) and liver cancer (HepG2) were about 50% at the same concentration.

  • Objectives : The antioxidant activities of Yeonkyo-san and its ingredient were investigated for industrial application. Methods : We experimented radical scavenging effect, superoxide dismutase and cytotoxicity effect of various cancer cell. Results : The result were obtained as follows : Primary testing of cytotoxicity of Yeonkyo-san and ingredient was done for G361. B16F10, MDA and A549 cell lines. Yeonkyo-san and ingredient water extract, ethanol extract showed cytotoxicity of over 90% respectively against cell line. The electron donating ability(EDA) of water extract from the Yeonkyo-san was higher than 60%, ethanol extract from the Yeonkyo-san was over 80% at 1000 ppm. The electron donating ability(EDA) of water extract and ethanol extract from the Yeonkyo-san ingredients were increased as well. SOD-like activity was high as 75% in 700 ppm, it was increased. All the other samples showed less than 40% SOD-like activity. Conclusions : The results indicated that water extract and ethanol extract of Yeonkyo-san and ingredient can be used as a natural ingredient with biological functions in cosmetic and food composition.

산벚나무 수피추출물의 생리활성 및 페놀성 화합물 분리 (Biological Activity Investigation, and Phenol Compounds Isolation from Barks of Prunus Sargentii R.)

  • Prunus sargentii of Rosaceae familiy, has been reported to have radical scavenging activity and anti-inflammatory effect. On these facts, this research was conducted to evaluate pharmaceutical activities of the bark extracts P. sargentii. Free radical scavenging activities of fraction (Fr) -5${\sim}$10 isolated from P. sargentii was higher than 80% respectively at 10ppm. The superoxide dismutase (SOD)-like activity of Fr-5, 9 were about 97, 84%, respectively at 1,000 ppm. Xanthine oxidase inhibitory effect of Fr-9, 10 were about 75, 78%, respectively at 1,000 ppm. The tyrosinase inhibitory effect related to skin-whitening was 72, 68% in Fr-2, 9 isolated from Prunus sargentii R. at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 98% for Fr-8 at 500 ppm. Isolation of the methanol soluble fraction from P. sargentii yielded two major phenol compounds, (-)-epicatechin and taxifolin. The structure of the compound was certainly determined by chemical analyses, as well as NMR and Mass spectroscopy. The present study was carried out in a search for new cosmetic material from the bark from P. sargentii. and, (-)-epicatechin and taxifolin were isolated as active principles. So P. sargentii R. methanolic extracts may be used for the cosmetic material.
