Production of Free D-amino Acids in Raw Milk Related to Psychrotrophic Bacterial Contamination (원유내 내냉성 미생물의 오염에 따른 유리 D-amino acid의 생성)

  • It is generally believed that amino acids occurring naturally in mammals are of the L-configuration. D-amino acid(DM) are common in nature as constituents of bacterial cell walls and several antibiotics. Recent reports have demonstrated the presence of small amounts of free DM in milk. The presence of free DM may affect the food quality by decreasing the nutritional value. Our objective was to examine whether the free DM carne from psychrotrophic bacteria. Free DM was produced by treating raw milk with Pseudomonas spp. The samples were extracted with sulphosalicylic acid and derivatized with AccQ-$Tag^{TM}$ reagent when the analysis was carried out by reverse-phase HPLC. We tested correlations of the content of free DM with bacterial growth. Significant amounts of free D-a1anine and D-proline have been found in the raw milk inoculated with Pseudomonas spp. The increase of D-alanine and D-proline appeared to be mainly related to the presence of Pseudomonas fluorescens. These results suggest that free DM may be considered as an indicator of psychrotrophic bacterial milk contamination.

A Study on the Nutrients Intakes of Hypertensive Female Farmers (일부 농촌지역 고혈압여성의 영양섭취 상태에 관한 연구)

  • This study was carried out to investigate the nutrients intakes of hypertensive female farmers in Korea. The number of subjects in this survey was 40(normal 22, hypertensive 18). Body weight, height, circumferences of waist, hip and thigh, and systolic and diastolic blood pressure of them were measured, and lipids and mineral levels of venous blood, mineral contents of 24-hour urine and Na or K levels of some sodium sources were analyzed. The evaluation of nutritional status was performed by recording dietary intake for a day and analyzing it with food composition table. Body weight, waist circumference, waist to thigh ratio, and serum triglyceride were higher in hypertensive. The mean values of calcium, vitamin B2 and niacin intakes of the subjects didn't meet the Korean recommended allowances though there was no significant difference in nutrients intake between two groups. The value of sodium intake presumed from content of sodium excreted to urine, and Na content and Na/K in Korean cabbage Kimchi was higher in hypertensive. These results suggest that reducing of serum triglyceride, decreasing of sodium intake and changing the taste for saltiness can help develop the health status of the rural female hypertensive and control the blood pressure of them.

  • Studies on the effect of soil moisture, different seasons, harvesting periods and nitrogen application on the nitrate content of radish(Raphanus sativus L. var. niger(Mill.) S. Kerner) were carried out in pot and green house. The results are as follows; The low level of soil moisture leads to an increased nitrate accumulation in radish root. The nitrate content was found to be higher in spring crops and lower in summer ones. It was decreased during harvesting period. The application of nitrogen fertilizer increased the nitrate content in radish root. We found the highest content in petioles with lesser content in roots and leaf blades in that order. The multiple regression analysis and the nutritional value of nitrate content in radish were discussed.

Improvement on the Quality and Functionality of Red Tanner Crab Cooking Drip Using Commercial Enzymes (효소분해의 의한 붉은 대게 자숙액의 품질 및 기능성 개선)

  • For the improvement on the quality and functionality of red tanner crab cooking drip, the preparation of hydrolysates from red crab cooking drip using commercial enzymes (Alcalase, Flavourzyme, Neutrase and Protamex) was attempted and its taste, nutritional and functional characteristics were also investigated. According to the results of heavy metal contents and proximate composition, red tanner crab cooking drip (RTCCD) could be used as a food resource. From the results of the trichloroacetic acid soluble index (TSI), angiotensin I converting enzyme (ACE) inhibiting activity and antioxidative activity, RTCCD hydrolysates incubated with Alcalase for 2 hrs was superior to the other one-step hydrolysates. There were no differences in the ACE inhibiting activity and antioxidative activity between one-step hydrolysates, which was incubated with Alcalase for 2 hrs, and two-step hydrolysates sequentially incubated with Alcalase and other enzymes. Alcalase-treated hydrolysates was similar in proximate composition and Hunter color value, while high in free amino acid content compared with crab cooking drip. Total amino acid content of Alcalase-treated hydrolysates was 11.9 g/100 mL and the major amino acids were glutamic acid (10.2%), proline (10.1%) and glycine (10.7%).

Physicochemical Properties of Rice Extrudate Added with Onion Kimchi Powder (양파김치 분말을 첨가한 쌀 압출팽화물의 이화학적 특성)

  • An extrusion process was to make an onion kimchi snack from rice grit and onion kimchi powder, in an effort to enhance the nutritional value, flavor, and physicochemical properties of the extrudate. This study investigated optimum conditions (moisture content, barrel temperature, and the content of onion kimchi powder) for the production of high-quality rice extrudate products, and measured quality properties (water absorption index, texture, expansion ratio, and color) of rice extrudate to which onion kimchi powder had been added. Onion kimchi powder at 3%, 5%, 7%, and 10% (all w/w) was mixed with rice grit and the mixture then extruded in a twin-screw extruder. The texture of onion kimchi mack became softer as onion kimchi powder level rose, and water absorption ability increased. The expansion ratio and the lightness of extrudates decreased with increases in onion kimchi powder levels. The maximum water absorption index am the minimum hardness were obtained with 10% onion kimchi powder. Rice extrudate with 10% onion kimchi powder was suitable for extrusion cooking md obtained the highest score for overall acceptability by sensory evaluation.

Comparison of Physicochemical Properties of Wild and Cultivated Lactuca indica (야생 및 재배 왕고들빼기(Lactuca indica)의 이화학적 특성 비교)

  • This study was conducted to investigate chemical properties of wild $Lactuca$ $indica$ (WL) and cultivated $Lactuca$ $indica$ (CL). The proximate composition, reducing sugar, free amino acids, organic acid, vitamin C, minerals, chlorophyll, and crude saponin were analyzed. WL and CL contained high levels of carbohydrate. The leaves and roots of CL contained higher levels of free amino acid than those of WL. Especially, the proline content of CL leaf was 12 times higher than that of WL leaf, and the arginine content of CL root was 100 times higher than that of WL root. The major organic acid and mineral of $Lactuca$ $indica$ were tartaric acid and potassium, respectively. CL showed significantly higher value of reducing sugar than WL. The vitamin C content of the samples ranged from 0.4 to 24.1 mg%, and CL leaf was the highest amount of vitamin C among the samples. CL leaf had a higher amount of chlorophyll than WL leaf, but WL root contained a higher amount of crude saponin than CL root. As in this study, CL showed better nutritional properties than WL, and these results will provide fundamental data in order to activate the cultivation of wild plants.

Monitoring of Commercial Cephalopod Products Sold on the South Korea Market using DNA Barcode Information (DNA 바코드를 이용한 국내 유통 두족류 제품의 원재료 모니터링 연구)

  • Cephalopods are one of the most important fishery resources in the world because of their desirable taste and nutritional value. In south Korea, one of the countries in which a large amount of seafood is consumed, cephalopods (e.g., octopus, squid, and cuttlefish) have an annual consumption rate of over 400,000 metric tons. In this study, octopus and squid products (n=28) sold on the market were monitored by analyzing sequences of DNA barcode markers (cytochrome c oxidase subunit I and 16S ribosomal RNA genes). For species identification, the NCBI BLAST database was screened with the sequences and analyzed as a query. In this BLAST search, twelve squid products showed 99-100% sequence identity to Dosidicus gigas (n=3) and Todarodes pacificus (n=9). In the case of the other 16 products that were declared using octopus as raw materials on the labels, six products were identified as Cistopus taiwanicus (n=1), Amphioctopus marginatus (n=1), Scaeurgus unicirrhus (n=1), and Dosidicus gigas (n=3). Monitoring results indicated that a significant percentage (37.5%) of mislabeling was present in octopus products sold on the South Korean market.

Reference intervals for blood metabolic profiles of Holstein cows in Korea (국내 젖소의 혈액 대사인자 프로파일 분석)

  • Metabolic profile test is used to evaluate nutritional imbalance and metabolic disease in dairy cows. The reference intervals of metabolic parameters may change according to nation, region, decades, and maintenance system. Despite the need to be periodically updated for the reference intervals of metabolic parameters, it has rarely been investigated in Korea. Therefore, this aim of study was to provide the reference intervals of metabolic parameters using dairy cows surveyed in Korea during recent years. A metabolic profile test was conducted for 2,976 clinically healthy dairy cows in Korea. Blood samples were collected for the analysis of serum metabolites. This study provided reference intervals of thirteen metabolic parameters (${\beta}$-hydroxybutyrate [${\beta}-HB$], non-esterified fatty acids [NEFA], glucose, total cholesterol [T-COL], total protein, albumin, globulin, blood urea nitrogen [BUN], aspartate aminotransferase [AST], gamma-glutamyl transferase [GGT], calcium, phosphorus, and magnesium). BUN and AST values of the current study were higher than those of previous studies. In the present study, the other metabolic parameters showed low or similar value compared to previous results. Moreover, ${\beta}-HB$, NEFA, T-COL, ALB, BUN, AST, and GGT values were affected by lactation period. This study provided information on the reference intervals of metabolites in healthy dairy cows in Korea. The reference intervals from the present study would be useful in managing and diagnosing disease of dairy cows. However, careful attention should be given in interpreting disease condition for metabolites affected by lactation.

Topographical Chemical Properties of Paddy Soils in Gyeongnam Province (경남지역 논토양 지형과 화학성의 관계)

  • Sustainable agriculture requires reliable information on the spatial distribution of the soil chemical properties for effective management of paddy fields. In order to provide adequate soil nutritional information for environmental-friendly agriculture, we investigated the soil chemical properties for 260 paddy soils sampled in Gyeongnam Province from 1999 to 2011 by four years of interval. Soil chemical properties of pH, organic matter, available phosphate and silicate, exchangeable potassium, calcium, and magnesium contents were analyzed. The pH value, organic matter, available phosphate and silicate, and exchangeable calcium and magnesium in paddy soils were significantly elevated in 2011 compared to 1999. The excessive levels of organic matter and available phosphate in paddy soils were detected in 2011. The soil available phosphate was highest in mountain foot-slope (p<0.05) and the soil organic matter was highest in diluvial terrace (p<0.05). It revealed that the soil available phosphate was the responsible factor for the differentiation of soil chemical properties by the topography in paddy soils.

Chemical Components and Physiological Activities of Neungee Mushroom (Sarcodon aspratus) (능이버섯(Sarcodon aspratus)의 화학성분과 생리활성)

  • The chemical components and physiological activities of Neungee mushroom (Sarcodon aspratus) were investigated to assess its nutritional and functional value. The moisture, total protein, crude fat and ash contents of Neungee mushroom were 85.73%, 1.78%, 1.87% and 1.27%, respectively. The alanine, linoleic acid, tartaric acid and glucose concentrations in Neungee mushrooms were 90.11, 39.09, 75.47 and 1,680 mg%, respectively. The radical and nitrate scavenging activities in Neungee mushroom extracts were 46.2% and 77.8% on $800{\mu}g/mL$ depending on the extract concentration. The lipid peroxidation inhibitory effect of Neungee mushroom extract ($1,500\;{\mu}g/mL$) was $2,347\;{\mu}mol$ MDA/g liver. We also observed that an extract concentration of $1,500\;{\mu}g/mL$ was more effective than the control at 7 d. The cytotoxicity of the Neungee mushroom extract ($100\;{\mu}g/mL$) for the A549 (lung carcinoma) cells was 96.0%.
