Effect of Plant Growth Regulators on Plant Regeneration from Leaf and Stem Explant Cultures of Sedum erythrostichum Miq. (꿩의비름(Sedum erythrostichum Miq.)의 잎과 줄기 절편으로부터 식물체의 재분화에 미치는 생장조절제의 영향)

  • Leaf and stem explants of Sedum erythrostichum Miq. were cultured on MS medium supplemented with various combinations of growth regulators. After two weeks of culture, 100% of the leaf explants formed calli on medium containing 2.0 mg/L 2, 4-D and 1.0 mg/L BA. Callus proliferated when subcultured on medium containing 2.0 mg/L 2, 4-D and 1.0 mg/L BA. Numerous adventitious buds were regenerated from callus cultured on the medium containing 2.0 mg/L NAA and 1.0 mg/L BA. Root formation from shoot was occurred on the MS basal medium containing 1.0 mg/L IAA.

  • Efficient plant regeneration system of birdsfoot trefoil (Lotus corniculatus L.) was development. The factors affecting the somatic embryo formation, its proliferation and regeneration capacity of leaf and stem explants of Empire cultivar was investigated. The highest number of somatic embryos was obtained on B5 medium supplemented with 1 mg/L NAA and 1 mg/L BA. Depending on different explants, highest frequency of embryogenic callus and regeneration were observed in Empire with leaf explants. The response from stem explants was slower and callus induction was less than that from leaf explants. Regenerated shoots formed complete plantlets in on 1/2 MS medium supplemented with 1 mg/L IBA. Regenerated plants were morphologically uniform with normal shape and growth pattern.

  • An efficient system for high frequency plant regeneration was established through investigating various factors such as plant growth regulator combinations, explant types and ages, and addition of $AgNO_3$ influenced on shoot regeneration in Brassica juncea L. cv. BARI sarisha-10. Murashige and Skoog (MS) medium supplemented with 0.1 mg/L NAA (1-naphthaleneacetic acid) and 1 mg/L BA (6-benzyladenine) showed the maximum shoot regeneration frequency (56.67%) among the different combinations of NAA and BA. Explant type, explant age, and addition of $AgNO_3$ also significantly affected shoot regeneration. Of the four type of explants (cotyledon, hypocotyl, root, and leaf explants)- cotyledon explants produced the highest shoot regeneration frequency and hypocotyls explants produced the highest number of shoots per explant, whereas root explants did not produce any shoot. The cotyledonary explants from Four-day-old seedlings showed the maximum shoot regeneration frequency and number of shoots per explant. Shoot regeneration frequency increased significantly by adding $AgNO_3$ to the medium. Two mg/L $AgNO_3$ appeared to be the best for shoot regeneration with the highest shoot regeneration frequency (86.67%) and number of shoots per explant (7.5 shoots). Considerable variation in shoot regeneration from cotyledonay explants was observed within the B. juncea L. genotypes. The shoot regeneration frequency ranged from 47.78% for cv. Shambol to 91.11% for cv. Rai-5. In terms of the number of shoots produced per explant, B. juncea L. cv. Daulot showed the maximum efficiency. MS medium supplemented with 0.1 mg/L NAA showed the highest frequency of rooting. The regenerated plantlets were transferred to pot soil and grown to maturity in the greenhouse. All plants were fertile and morphologically identical with the source plants.

  • This study was conducted to determine the optimal concentrations of plant hormones (2,4-D, picloram, dicamba, NAA, kinetin) and the suitable explants among seeds, hypocotyl, and cotyledons on calls formation and plant regeneration of stevia(Stevia rebaudiana Bertoni). The frequency of cellus formation was higher in the young leaf-explants then the older ones, and in the seeds then the hypocotyls and cotyledons on MS medium with 1mg/L 2,4-D. After transfer of seed-derived stevia callus producing embryogenic callus on plant-regeneration medium, the frequency of plant regeneration from callus was 23.8% in MS medium with 1mg/L NAA and 3mg/L kinetin.

Effects of Plant Growth Regulators on Callus Formation and Organogenesis of Sicyos angulatus L. (야생식물 안동오이의 기내배양시 캘러스형성과 기관분화에 미치는 생장조절제의 영향)

  • This study was carried out to determine the effects of plant growth regulators on cell culture and organogenesis from Sicyos angulatus L. using explants of leaves, stems and cotyledons. Optimal callus induction for S. angulatus was obtained on MS medium with 0.1mg/$\ell$ BA and 2.0mg/$\ell$ 2,4 -D from cotyledons, 0.1mg/$\ell$ BA and 5.0mg/$\ell$ NAA from leaves explants, Optimal media for subculture and growth of S. angulatus callus were 1/2 MS medium with 0.1mg/$\ell$ BA and 1.0mg/$\ell$ 2,4 -D for solid culture, and 0.1mg/$\ell$ 2,4-D for suspension culture. Many adventitious roots with some shoots were formed were formed from leaf and cotyledon explants of S. angulatus during callus induction with optimal combinations of plants growth regulators.

  • Efficient plant regeneration via shoot organogenesis from in vitro cultured leaf segments of chrysanthemum (Dendranthema grandiflora Tzvelev cv. Namjeon) was achieved. Adventitious shoot formation from leaf explants was greatly influenced by plant growth regulator, leaf age, light condition, explant number per culture vessel, and explant orientation. Leaf segments, obtained from fully expanded young 1-2nd leaves and inoculated 8 explants per petri-dish with adaxial surface contact with MS medium supplemented with 0.5 mg/L BA and 2.0 mg/L NAA, produced 100% regeneration frequency and 13.7 shoots per explant. Regenerated adventitious shoots were successfully rooted in MS medium with 0.1 mg/L NAA. The plantlets were acclimatized in artificial soil mixtures (Vermiculite:Perlite=1:1), and transferred to greenhouse for flowering. The regenerated plants showed normal phenotypes.

  • Genetic transformation in dandelion(Taraxacum mongolicum Hand). was studied. We used for transformation by Agrobacterium tumefaciens strian LBA4404 harboring a binary vector pBI121 carrying the CaMV 35S promoter-GUS gene fusion used as a reporter gene and NOS promoter-NPTII gene as a positive selection marker. To obtain transformed plants, leaf explants of dandelion were cocultured with Agrobacterium tumefaciens LBA4404 for 10 mins, then transferred to MS medium containing 1 $\mu$M IAA, 1$\mu$M BA, 100$\mu$g/ML carbenicillin and 50 $\mu$g/ML kanarmycin sulfate. After two weeks of subculture of the explants, Kanamycin-resistant shoots were formed on explants survived. When subjected to GUS histochemical assay, all of the regenerants showed the GUS-positive responses. Plantlets were be be transformed to soil for further growth.

  • Leaf and petiole explants of Pulsatilla koreana NAKAI were cultured on MS medium supplemented with various concentrations of zeatin, kinetin or BAP combined with 0.05 mg/L IAA. After 6 weeks of culture, effects of cytokinin on adventitious shoot formation from explants were investigated. The highest frequency of shoot formation was obtained when petiole explants were cultured on medium with 0.5 mg/L zeatin and 0.05 mg/L IAA. Regenerated shoot were transferred on to root induction medium. The best root formation was observed at 1/2 MS medium with 1.5 mg/L NAA. Rooted plantlets were transplanted to a mixture of perlit and soil (1:3), where they were successfully acclimatized.

  • Calli were induced from the explants of infloresence, petiole and leaf blade of Cnidium officinale on MS medium with 2.4-D, while embryogenic callus was induced only from inflorescence explants. Somatic embryos of 78 per explant were formed during subculture of the explants on medium without 2.4-D after culture on medium with 2 mg/L 2.4-D. Cotyledonary variation, cup-shaped cotyledon of 49% and other abnormal cotyledons of 13.5 % was observed on the somatic embryos. However this variation could be overcomed by the addition of activated charcoal onto culture medium. Somatic embryos at cotyledonary stage germinated on MS basal medium but the germination rate was very poor, blow 50 %. Somatic embryos on the medium with activated charcoal showed improved germinaton.

  • Shoot formation was investigated from in vitro cultivation of exotic hybrid poplar (Populus koreana ${\times}$ P. trichocarpa) with a specific stomatal character occurring both upper and lower surface of leaves. Two different explants (stem and leaf segment) of Peace poplar were cultured on half strength Murashige and Skoog (MS) basal medium supplemented with the various concentrations of thidiazuron as a plant growth regulator. Most adventitious shoots were produced from excised ends of stem or mid-veins of leaf segments. The highest average numbers of shoots were 7.1 and 5.3 with the treatments of 0.02mg/L TDZ in both explants of stem and leaf segment. The highest shooting rates were achieved to 83.3% and 47.6% with the concentrations of 0.01mg/L and 0.02mg/L TDZ by axillary bud and leaf cultures, respectively.
