본 연구는 아까시나무의 물 추출물, 에탄올 추출물 및 고온 추출물을 이용하여 마우스 대식세포주인 Raw 264.7 세포에 대해 염증억제 효과가 있는지 알아보고자 수행하였다. RP1(아까시 나무 물 추출물), RP2(아까시 나무 에탄올 추출물) 및 RP3(아까시 나무 고온 추출물)은 세포생존율 분석에서 $200{\mu}g/m{\ell}$의 농도까지 Raw 264.7 세포에 세포독성을 나타나지 않았다. NO 생성 억제효과를 분석하였을 때 LPS 처리군과 비교하여 RP3는 약 87% 정도의 억제효과를 나타내 RP1과 RP2에 비해 NO 억제활성이 가장 높았다. 뿐만 아니라 RP3는 RP1과 RP2와 비교하여 염증매개인자의 억제율이 각각 $PGE_2$ (86.3%), $TNF-{\alpha}$ (64.1%), IL-6 (65.1%) 및 $IL-1{\beta}$ (63.3%)로 가장 높았다. 이는 RP3의 처리가 LPS에 의해 증가된 염증매개인자의 분비를 억제함을 통해 항염증 효과가 있을 것으로 생각되며, 염증관련 신호전달경로에 직접적으로 작용할 가능성이 있는 것으로 판단된다. 하지만 Raw 264.7 세포주는 염증조절복합체를 구성하는 ASC 단백질이 발현되지 않아 다른 신호전달 경로를 통해 염증매개인자를 분비하기 때문에, 설치류의 대식세포를 직접 일차배양(primary culture)하여 이에 관련된 신호전달경로를 확인하는 추가 실험이 필요하다고 사료된다.
동충하초균으로 발효한 인삼잎의 면역활성과 구조를 규명하기 위하여 조다당(GLF)를 분리하고 구성당과 당쇄 결합양식을 확인한 결과 C. sinensis 유래의 glucan이 주를 이루며 소량의 인삼잎 유래 pectic substances가 혼재되어 있을 것이라 추정하였다. 이온 교환 수지를 이용해 GLF로부터 중성다당체(GLF1)를 분리하였으며, 이를 lyticase, β-glucosidase 및 α-glucoamylase 효소를 처리한 결과, GLF1은 α-glucoamylase에 의해 가수분해되는 것을 확인함으로써 주로 α-glucan을 함유하고 있음을 추정할 수 있었다. 대식세포 분비능을 측정한 결과, 동충하초균으로 인삼잎을 발효하여 얻은 조다당 GLF가 단순 열수추출 조다당인 GLW보다 더 우수한 활성(Data not shown)과 수율을 보여주었으며, GLF와 GLF1 모두 농도 의존적으로 면역활성이 증가하는 경향과 동일 농도에서 유사한 높은 활성을 나타냈다. 한편 GLF1의 전체구조의 특성을 확인하기 위하여 isoamylase 및 α-amylase 효소를 처리한 결과, isoamylase 처리 획분인 PHI가 72.8%의 glucose로 구성되며 iodine-starch 반응이 증가하는 결과를 보였다. 반면, PHI의 αamylase 처리 후 분리한 PHIA1, PHIA2 및 PHIA3 획분에서는 iodine-starch 반응이 나타나지 않았으며, 이들의 대식세포 분비능을 확인한 결과 세 가지 획분 모두 어떠한 활성도 나타내지 않음을 확인하였다. 이상의 결과를 종합하면 동충하초 발효 인삼잎유래 중성다당은 α-(1→4)-glucan을 주쇄로 존재하며 C(O)-6 위치에서 측쇄가 연결되어 존재하는 α-glucan의 구조를 이루고 있으며, 이들의 면역활성은 α-glucan 전체 구조에서 기인하는 것임을 최종 확인하였다.

BACKGROUND/OBJECTIVES: Schisandrae Fructus, the fruit of Schisandra chinensis Baill., has traditionally been used as a medicinal herb for the treatment of various diseases, and has proven its various pharmacological effects, including anti-inflammatory and antioxidant activities. In this study, we investigated the inhibitory effect of Schisandrae Fructus ethanol extract (SF) on inflammatory and oxidative stress in particulate matter 2.5 (PM2.5)-treated RAW 264.7 macrophages. MATERIALS/METHODS: To investigate the anti-inflammatory and antioxidant effects of SF in PM2.5-stimulated RAW 264.7 cells, the levels of pro-inflammatory mediator such as nitric oxide (NO) and prostaglandin E2 (PGE2), cytokines including interleukin (IL)-6 and IL-1β, and reactive oxygen species (ROS) were measured. To elucidate the mechanism underlying the effect of SF, the expression of genes involved in the generation of inflammatory factors was also investigated. We further evaluated the anti-inflammatory and antioxidant efficacy of SF against PM2.5 in the zebrafish model. RESULTS: The results indicated that SF treatment significantly inhibited the PM2.5-induced release of NO and PGE2, which was associated with decreased inducible NO synthase and cyclooxygenase-2 expression. SF also attenuated the PM2.5-induced expression of IL-6 and IL-1β, reducing their extracellular secretion. Moreover, SF suppressed the PM2.5-mediated translocation of nuclear factor-kappa B (NF-κB) from the cytosol into nuclei and the degradation of inhibitor IκB-α, indicating that SF exhibited anti-inflammatory effects by inhibiting the NF-κB signaling pathway. In addition, SF abolished PM2.5-induced generation of ROS, similar to the pretreatment of a ROS scavenger, but not by an inhibitor of NF-κB activity. Furthermore, SF showed strong protective effects against NO and ROS production in PM2.5-treated zebrafish larvae. CONCLUSIONS: Our findings suggest that SF exerts anti-inflammatory and antioxidant effects against PM2.5 through ROS-dependent down-regulating the NF-κB signaling pathway, and that SF can be a potential functional substance to prevent PM2.5-mediated inflammatory and oxidative damage.
Schisandra chinensis (SC) and Lycium chinense (LC) were widely distributed in Asia and the fruit has been used traditionally for medicinal herbs. The processing method was solid-state fermentation using Aspergillus oryzae for 48 h after stir-frying treatment at $220^{\circ}C$ for 12 min. In this study, in vitro the anti-inflammatory effect and in vivo hangover reduction were compared to unprocessed SC and LC water extract. Anti-inflammatory effects have been evaluated in pro-inflammatory mediators which were secreted by lipopolysaccharide (LPS)-induced RAW 264.7 macrophages. Nitric oxide (NO) was determined using Griess reaction. Proinflammatory cytokines such as tumor necrosis factor $(TNF)-{\alpha}$ and interleukin $(IL)-1{\beta}$ were measured by enzyme-linked immunosorbent assays (ELISA). Alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities were compared to processed SC or LC and mixtures thereof (1:1). In vivo study was compared to hangover relief in alcohol-fed mice. After administering a mixture of SC and LC (300 mg/kg) water extract (1:1), mice were fed 3 g/kg of ethanol. Serum was collected at 1, 3, and 5 h intervals to analyze ethanol and acetaldehyde levels using a colorimetric assay kit. The processed SC and LC water extracts compared to raw materials significantly inhibited LPS-induced NO and inflammatory cytokine production in RAW 264.7 cells. The results of the hangover mouse model are also consistent with anti-inflammatory effects. These results suggest that processed SC and LC extracts may be functional materials for the treatment of inflammation and hangover.
뇌신경 질환의 주요 원인이 되는 것으로 알려진 미세아교세포의 과도한 활성화에 의한 신경염증반응에서 풀무치 에탄올 추출물이 미세아교세포의 염증 반응에 미치는 영향을 검토하였다. 미세아교세포의 활성화를 유도하기 위해 LPS를 사용하였으며, LPS 처리에 의해 신경염증반응의 지표인 NO의 생성량과 이들을 조절하는 iNOS, COX-2의 발현이 증가됨을 확인 할 수 있었다. 그러나 풀무치 에탄올 추출물을 1시간 전처리 한 후 LPS를 처리한 경우 추출물의 농도에 의존적으로 이들의 발현량이 현저히 감소되는 것을 확인 하였다. 또한 LPS 처리로 인해 분비되는 염증성 cytokine들의 생성량도 풀무치 에탄올 추출물에 의해 현저히 억제 됨을 확인 할 수 있었다. 따라서 본 연구의 결과는 미세아교세포의 과도한 활성화로 인해 발생되는 뇌 신경질환의 치료 소재로서 풀무치 에탄올 추출물의 활성 가능성을 제시하였다.
본 연구에서는 홍삼 비사포닌 분획(NSF)의 항 염증 효과를 마우스 대식세포와 인간유래 단핵세포에서 확인하였다. NSF는 마우스 대식세포에서 LPS로 유도된 NO, iNOS 그리고 COX-2의 양 뿐만 아니라 IL-6, $TNF-{\alpha}$, MCP-1과 같은 염증성 싸이토카인의 생성량을 유의적으로 감소시켰다. 인간 유래 단핵세포에서는 PMA에 의해 유도되는 대식세포로의 분화를 효과적으로 억제하면서 분화인자인 $CD11{\beta}$와 CD36의 발현을 유의적으로 감소시켰다. 마우스 대식세포에서와 마찬가지로 염증성 싸이토카인들의 생성량 또한 감소하였는데, 이러한 NSF의 항 염증 효과는 두 전사인자의 조절작용에 의한 것으로 사료된다. 즉 NSF는 $NF-{\kappa}B$의 핵으로 이동을 감소시킴으로써 전사활성을 억제하여 염증성 싸이토카인들의 발현을 저해하고 이와 반대로 Nrf2의 발현과 핵으로의 이동을 증가시켜 항산화 효소이면서 항 염증 작용을 나타내는 HO-1의 발현을 촉진하는 것으로 관찰되었다. 따라서 NSF는 $NF-{\kappa}B$와 Nrf2의 두 가지 신호전달체계를 조절함으로써 항 염증 작용을 나타냈으며 이를 홍삼 NSF의 항 염증 기작으로 보고하는 바이다.
상엽 추출물의 항염 효능을 연구하고자 하였다. 상엽을 열수 추출하여 제조한 시료를 대상으로 RAW 264.7 macrophage cells를 24시간 배양하여 세포생존율과 LPS로 유발된 RAW 264.7 macrophage cells의 일산화질소(NO) 생성증가, $IL-1{\alpha}$, IL-6 그리고 IL-10 등의 사이토카인 생성증가에 미치는 영향을 측정하였다. 상엽 추출물의 MTT assay를 수행한 결과 상엽 추출물은 대식세포의 생존율에는 크게 영향을 주지 않았다. LPS로 활성화된 대식세포의 일산화질소(NO)의 생성증가를 $25{\mu}g/mL$ 이상의 농도에서 농도 의존적으로 유의하게 억제시켰다. 또한 LPS로 유발된 대식세포에 염증과 연관된 사이토카인인 $IL-1{\alpha}$, IL-6의 생성증가를 $50{\mu}g/mL$ 이상의 농도에서 농도 의존적으로 유의하게 억제하는 등 항염효과를 가지고 있는 것으로 나타났다. 본 연구 결과는 상엽이 건강 친화적인 기능성 소재로 활용도가 높을 것으로 기대된다. 또한 향후 지속적인 연구를 통한 대식세포를 항염증과 관련된 신호전달경로를 확인하는 추가 실험이 필요하다고 사료된다.
본 연구는 표고버섯 균사체를 이용한 사과, 배 부산물의 기능성소재로의 활용가능성을 파악하고자 진행하였다. 사과/배 부산물 분말 농도 30% 및 미강, 대두박의 영양성분을 비율별로 첨가하고 Lentinula edodes KCCM 12339를 접종하여 온도 $24^{\circ}C$, 습도 80%에서 발효하였다. 사과/배 부산물 및 표고버섯 발효물의 효소활성은 전분분해 및 단백분해활성은 발효전과 발효후의 차이는 보이지 않았으며, cellulase 및 pectinase 활성에서 표고버섯 균사체를 이용한 사과/배 부산물 발효물이 더 높은 활성을 나타내어 가수분해 작용에 균사체가 작용함을 알 수 있었다. 사과/배 부산물 및 표고버섯 발효물의 생리활성은 DPPH radical 소거능, SOD-like activity, total polyphenol, ABTS radical 소거능, 베타글루칸 함량을 측정한 결과 사과/배부산물 보다 사과/배부산물 표고버섯 균사체 발효물이 더 높은 함량을 나타내었다. Raw 264.7(대식세포)cell 배양 중 LPS를 처리하여 염증반응을 유도시켜 사과/배 부산물 및 표고버섯 균사체 발효물의 NO 생성능을 측정한 결과 발효하지 않은 사과/배 부산물보다 사과/배 부산물 표고버섯 균사체 발효물이 더 낮은 생성량을 보여 발효물 처리 시 염증유발을 억제하는 것으로 판단되었다. 사과/배 부산물의 표고버섯 균사체 발효를 통하여 다양한 활용이 가능할 것으로 기대되며, 천연 기능성 물질소재로 이용되기 위해서는 유효성분에 대한 균사체 발효 최적화 조건의 구체적인 연구가 더 필요할 것으로 사료된다.
Objectives: This study was performed to identify the anti-inflammatory effects of Salvia miltiorrhizae radix Water extract (SMW) on lipopolysaccharide (LPS) induced inflammation. Methods: RAW 264.7 cells were treated with 500 ng/ml of LPS. SMW (0.1, 0.25, 0.5 mg/ml) was treated 1 h prior to LPS. Cell viability was measured by MTT assay. Levels of nitric oxide (NO) were measured with Griess reagent and pro-inflammatory cytokines were measured by enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (PCR). We also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-kappa B ($NF-{\kappa}B$) activation by western blot. In addition, we observed mice survival rate after LPS and examined their cytokine levels of serum and liver tissue. Results: SMW itself did not have cytotoxic effects in RAW 264.7 cells less than 0.5 mg/ml. SMW treatment inhibited the production of NO, and interleukin $(IL)-1{\beta}$ which is pro-inflammatory cytokine. And SMW treatment inhibited the LPS-induced activation of MAPKs such as extracellular signal-regulated kinase1/2 (ERK1/2), p38 kinases (p38), c-Jun NH2-terminal kinase (JNK) and $NF-{\kappa}B$. In addition, it also showed reducing the level of $IL-1{\beta}$ on the serum and liver tissue of mice. Also, death of LPS-induced mice was inhibited by SMW. Conclusions: The result suggests that treatment of SMW could reduce the LPS-induced inflammation. Thereby, SMW could be used as a protective agent against inflammation. Also, this study could give a clinical basis that SMW could be a drug or agent to prevent inflammatory diseases.
Objective: Gram-negative bacteria lipopolysaccharide (LPS) has been reported to be associated with uterine impairment, embryonic resorption, ovarian dysfunction, and follicle retardation. Here, we aimed to investigate the toxic effects of LPS on the maturation ability and parthenogenetic developmental competence of bovine oocytes. Methods: First, we developed an in vitro model to study the response of bovine cumulusoocyte complexes (COCs) to LPS stress. After incubating germinal vesicle COCs in $10{\mu}g/mL$ of LPS, we analyzed the following three aspects: the expression levels of the LPS receptor toll-like receptor 4 (TLR4) in COCs, activities of intracellular signaling protein p38 mitogen-activated protein kinase (p38 MAPK) and nuclear factor-kappa B (NF-${\kappa}B$); and the concentrations of interleukin (IL)-$1{\beta}$, tumor necrosis factor (TNF)-${\alpha}$, and IL-6. Furthermore, we determined the effects of LPS on the maturation ability and parthenogenetic developmental competence of bovine oocytes. Results: The results revealed that LPS treatment significantly elevated TLR4 mRNA and protein expression levels in COCs. Exposure of COCs to LPS also resulted in a marked increase in activity of the intracellular signaling protein p-p38 MAPK and NF-${\kappa}B$. Furthermore, oocytes cultured in maturation medium containing LPS had significantly higher concentrations of the proinflammatory cytokines IL-$1{\beta}$, TNF-${\alpha}$, and IL-6. LPS exposure significantly decreased the first polar body extrusion rate. The cytoplasmic maturation, characterized by polar body extrusion and distribution of peripheral cortical granules, was significantly impaired in LPS-treated oocytes. Moreover, LPS exposure significantly increased intracellular reactive oxygen species levels and the relative mRNA abundance of the antioxidants thioredoxin (Trx), Trx2, and peroxiredoxin 1 in oocytes. Moreover, the early apoptotic rate and the release of cytochrome C were significantly increased in response to LPS. The cleavage, morula, and blastocyst formation rates were significantly lower in parthenogenetically activated oocytes exposed to LPS, while the incidence of apoptotic nuclei in blastocysts was significantly increased. Conclusion: Together, these results provide an underlying mechanism by which LPS impairs maturation potential in bovine oocytes.

② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
