Microstructure of Faecal Pellets and Silk of the Two Spotted Spider Mite, Tetranychus urticae (Tetranychidae: Acarina) (점박이응애 분비물의 미세구조)

  • Asthma and allergic rhinitis due to outdoor spider mites are major health problems worldwide. The sensitization route to spider mites has not yet been well elucidated. We examined the microstructure of faecal pellets and silk of the two-spotted spider mite, Tetranychus urticae, using a field emission scanning electron microscope. Black and white pellets of the two-spotted spider mites faeces contain a large amount of plant pigment waste products. Black faecal pellets are strawberry-shaped. White faecal pellets are silken threads. These pellets are likely to be the source of allergens of relevant mites because desiccated faeces particles probably disintegrate and become incorporated into dust particles more readily than whole bodies or encased internal organs. We conclude that the importance of spider mites in respiratory allergy needs emphasis.

Detection and genetic characteristic of Yellow-head virus genotype 8 (YHV-8) Cultured Litopanaeus vanamei, in Korea (국내 양식 흰다리새우에서 검출된 노랑머리바이러스(YHV-8)의 유전학적 특성)

  • This study was performed for screening yellow head virus(YHV) complex in 252 including 235 white leg shrimps (Litopenaeus vannamei) and 17 oriental shrimp (Fenneropenaeus chinensis) collected from 18 farms located in southwestern province of Korea. The virus complex was detected by nested reverse-transcriptase polymerase chain reaction (RT-PCR) assay. In the assay, amplicons were resulted in RNAs exracted from 38 shrimps (21 white leg shrimps and 17 oriental shrimps) obtained from 7 farms. In phylogenetic analysis using sequences of ORF1b gene, all 38 sequences obtained in this study formed an independent lineage with YHV-8 genotype firstly isolated in China, belonged to an YHV-8 clade.

  • The transmittance properties of fishing lamp of the squid jigging vessel was investigated during nighttime operations in the Northwest Pacific on 21 and 29 September 2005. The metal halide lamps of white color($2.0kW{\times}168$) in the air and metal halide lamp of white color($10.0kW{\times}1$) in the underwater were used as a fishing lamp for gathering squids. The relative irradiance of metal halide lamp in the air showed peak in 850nm of wave length. The relationship between underwater illuminance(Y) and water depth(X) of metal halide lamp light in the observation areas is represented, $Y=84.137e^{-0.1105X},\;R^2=0.9974$. The distribution of underwater illuminance of measure points St. 1-5 showed low value of 0.11x in 80m depth.

  • Stress-corrosion cracking (SCC) of alloy 600 and alloy 800 in 0.5 mol/L thiosulfate solution during constant strain was investigated using electrochemical noise (EN) combined with 3-D microscope techniques. The in-situ morphology observation and EN results indicate that the SCC process could be divided into three stages: (1) passive film stabilization and growth, (2) crack initiation, (3) and crack growth. Power Spectral Density (PSD) and the probability distribution obtained from EN were used as the "fingerprint" to distinguish the different processes. During passive film stabilization and growth, the current noise signals resembled "white noise": when the crack initiated, many transient peaks could be seen in the current noise and the wavelet energy at low frequency as well as the noise resistance decreased. After crack propagation, the noise amplitudes increased, particularly the white noises at low and high frequencies ($W_L$ and $W_H$) in the PSDs. Finally, the detection of metal structure corrosion in a simulated sea splash zone and pipeline corrosion in the atmosphere are established.

First Record of Gobiid Fish, Astrabe fasciata (Perciformes: Gobiidae) from Dokdo, Korea (독도에서 채집된 망둑어과 어류 1미기록종, Astrabe fasciata)

  • The one specimen of Astrabe fasciata, belonging to the family Gobiidae was collected from the Dokdo of East Sea in Korea, and described as the first record from Korea. The specimen is characterized by having two dorsal fins, pectoral fin with free rays (five upper, two lower), protrusion on upper posterior part of supraorbital dermal fold, white spots dispersed on head and five white transverse bends on the body. We proposed its Korean name "Dok-do-eol-ruk-mang-duk-sok" and "Dok-do-eol-ruk-mang-duk" for the genus and species, respectively.

  • "Sinsulro(신선로)" is the famous soup of Cho-sun Dynasty Royal Cuisine, and the original food name is "Yulgujatang(열구자탕)". The first record is on [Sumunsasul] (1740) and the origin history related with Hirang-Jung is on [Headongjukji] and [Chosunyorihak]. From the analysis with Royal banquet menu and 16 kinds of old culinary literatures, "Sinsulro" are contained 54 and 70 material items and used variety cooking methods. Soup base of "Sinsulro" is well boiled meat, shank, brisket, stomach and intestine of beef with water, Some of beef is made meatball and seasoned raw meat. Beef marrow and tripes and liver are sauted with egg. Dried abalone and sea cucumber are soaked in water and then cutting slices after well boiled. Sliced white fish fillet are sauted with egg. Egg white and egg yolk made to thin sheet on pan with oil. Dropwort made to one sheet with flour and egg. Radish are boiled with meat soup and other vegetable are cooked with boiled or sauted. Seeds of pinenut, gingko, walnut are used of decorative seasonings. Filling mehtods of the Sinsulro casserole, the first layer is seasoning raw beef meat, 2nd layer is cooked slice of meat and radish, 3rd layer is rectangle pieces of egg sheet and sauted fish and intestines of beef. The top layer is decorated with meatball and naked nuts. After hot soup is poured until top of the materials, buning charcoal put into the center fire place and then served.o the center fire place and then served.

Effects of nitrite exposure on survival and physiology of white leg shrimp, Litopenaeus vannamei (아질산 노출이 흰다리새우, Litopenaeus vannamei의 생존율 및 독성 생리에 미치는 영향)

  • Juveniles of the white leg shrimp Litopenaeus vannamei (Weight 0.18±0.08 g) were exposed to nitrite-N at 0, 25, 50, 100, 200 and 400 mg/L for 72 hours, and the lethal concentration, heamolymph and genes regulation were evaluated. The lethal concentration 50 (LC50) of L. vannamei exposed to nitrite-N was 141.2 mg/L at 25℃ and 33 psu. In Total protein, total cholesterol, and BUN in heamolymph temporarily increased after the start of the experiment and then stabilized, but glucose, an indicator of stress, decreased over time in the entire experimental group, and creatines, an indicator of tissue damage, decreased with nitrite concentration until the first 12 hours. The genes of immune-related showed that masquerade-like serine proteinase(Mas) increased at 50 and 400 ppm for 24 hours, and then gradually decreased depending on concentration. In the case of prophenoloxidase, it was highest at 400 ppm for 40 hours, and other genes(Ras-related nuclear protein, Masquerade-like serine proteinase, proPO-activating enzyme) showed a response for 48 hours and then gradually decreased. The results of this study indicate that exposure to nitrite can affect the survival and hematological physiology of L. vannamei.

  • The author carried out an experiment to find out the response of sea-bass, Epinephelus septemfasciatus (Thunberg) to the color lights. The experimental tank (360L$\times$50W$\times$55H cm) was set up in a dark room. Six longitudinal sections with 60 cm intervals are marked in the tank to observe the location of the fish. Water depth in the tank was kept 50 cm level. Light bulbs of 20 W at the both ends of the tank projected the light horizontally into the tank. Two different colored filters were selected from four colors of red, blue, yellow, and white, and they were placed in front of the light bulbs to make different colors of light. Light intensity were controlled by use of auxiliary filters intercepted between the bulb and the filter. The fishes were acclimatized in the dark for 50 minutes before they were employed in the experiment. Upon turning on the light, the number of fish in each section was counted 40 times in 30 second intervals, and the mean of the number of fish in each section was given as the gathering rate of the fish. The colors favourited by the fish was found in the order of blue, red, white and yellow in day time, and red, blue, yellow and white at night time. The gathering rate of fish on illumination period was not constant and fluctuated with irregularity. The difference of the gathering rate on two different colors of light was small and the difference was larger in night time then in day time.

Variation of Shell Color in Three Geographic White Clam ($Meretrix$ $lusoria$) Populations of the Yellow Sea

  • Genomic DNAs (gDNAs) were isolated from the hard clam ($Meretrix$ $lusoria$, Roding, 1798) populations of Gunsan located in the Yellow Sea of the Korean peninsula. Genetic distances among different individuals of the LSCP (light shell color population) population of the hard clam (lane 1-11), GSCP (grey shell color population) population of the hard clam (lane 12-22) and DSCP (dark shell color population) population of the hard clam (lane 23-33), respectively, were generated using Systat version 10 according to the bandsharing values and similarity matrix. The dendrogram, generated by seven reliable oligonucleotides primers, indicates 3 genetic clusters. LSCP population could be evidently discriminated with the other two populations among three populations. The longest genetic distance (0.801) was found to exist between individuals in the two populations, between individuals' no. 33 of the DSCP population and no. 06 of the LSCP population. The higher fragment sizes (>2,000 bp) are much more observed in the GSCP population. Three hard clam populations can be clearly distinguished, especially, by their morphological characters and PCR-based approach.

Purification, Characterization and Application of a Cold Active Lipase from Marine Bacillus cereus HSS

  • Lipases (triacylglycerol acylhydrolases [EC 3.1.1.3]) are water-soluble enzymes. They catalyze the hydrolysis of fats and oils. A cold-active lipase from marine Bacillus cereus HSS, isolated from the Mediterranean Sea, Alexandria, Egypt, was purified and characterized. The total purification depending on lipase activity was 438.9 fold purification recording 632 U/mg protein. The molecular weight of the purified lipase was estimated to be 65 kDa using sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis. The optimum substrate concentration, enzyme concentration, pH, and temperature were 1.5 mM, 100 µl, pH 6 and 10℃, respectively. The lipase was tolerant to NaCl concentrations ranging from 1.5 to 4.5%. The lipase was affected by the tested metal ions, and its activity was inhibited by 16% in the presence of 0.05 M SDS. The application of the cold-active lipase for the removal of an oil stain from a white cotton cloth showed that it is a promising biological agent for the treatment of oily wastes and other related applications. To the best of our knowledge, this is the first report of the purification and characterization of a lipase from marine B. cereus HSS isolated from the Mediterranean Sea.
