제주산 로즈마리 에센셜 오일의 항염 및 피부 상재균에 대한 항균 활성 (Anti-inflammatory and antimicrobial effects of Jeju rosemary essential oil against skin flora)

  • 본 연구는 제주산 자생 로즈마리 에센셜 오일의 항염 및 피부상재균에 대한 항균 활성을 확인하기 위한 것이다. 로즈마리 에센셜 오일은 물 증류법으로 추출하였다. 로즈마리 에센셜 오일의 항염 효능을 확인하기 위하여 RAW 264.7 세포에서 LPS에 의해 유도된 NO와 $PGE_2$의 생성을 농도 의존적으로 억제하는 것을 확인하였다. Western blot 실험을 통해 이들을 생합성하는 효소인 iNOS, COX-2 단백질의 발현이 농도 의존적으로 감소하는 것을 확인하였다. 또한 전염증성 cytokine인 TNF-${\alpha}$와 IL-6의 생성 억제 효능을 확인하였다. 항생제 내성균주 각 2종을 포함한 S. epidermidis 3종, P. acnes 3종에 대한 항균 활성을 실험한 결과, paper disc법에서 저해환이 관찰되었고, MIC, MBC 실험을 통하여 균생육 저해와 사멸를 확인하였다. 위의 실험 결과로부터 로즈마리 에센셜 오일의 항염증, 항균 효능을 확인 하였으며 향후 화장품 및 스킨케어 소재로서 활용 가능성을 확인하였다.

두충나무, 황벽나무 등을 포함하는 수목추출물의 항여드름 및 항아토피 효과 (Anti-acne and Anti-atopic Dermatitis Effect of Plant Extracts Including Eucommia ulmoides Oliv and Phellodendron amurense)

  • 본 연구에서는 두충나무, 황벽나무 등을 포함하는 수목추출물의 소재특성을 시험하여 화장품 및 피부질환 치료소재로서의 응용 가능성을 조사하였다. 수목추출물의 항산화력은 DPPH 자유라디칼 소거법으로 측정되었으며 저농도에서는 항산화력이 낮았으나, $1000{\mu}g/ml$의 고농도에서는 우수한 항산화제인 비타민 C와 유사하였다. 항균력은 원판확산법으로 측정되었으며, 수목추출물은 피부상재균인 Staphylococcus epidermidis에 대해서는 낮은 항균력을 보이지만, 여드름 유발균인 Propionibacterium acne에 대해서는 높은 항균력을 보여주는 선택성 항균력이 있어, 여드름소재로서 매우 우수한 특성을 나타내었다. 무모생쥐를 이용한 항아토피 시험결과, 수목추출물은 DNCB에 의해 유도된 피부염증을 14일 째 거의 정상상태로 회복시켰으며, 피부내 항체인 IgE의 농도도 대조군에 비해 16% 낮추어주어 항아토피 효과가 우수한 것으로 나타났다. 따라서 수목추출물은 항여드름효과 및 항아토피 효과가 뛰어나서 화장품 혹은 피부질환 치료소재로서 응용 가능성이 매우 높음을 알 수 있었다.

Skin Protection Seminars to Prevent Occupational Skin Diseases: Results of a Prospective Longitudinal Study in Apprentices of High-risk Professions

  • Background: Occupational skin diseases (OSDs) are frequent in professions with exposure to skin hazards. Thus, a health educational intervention for apprentices of high-risk professions was conducted. It was the aim of this study to gain insight into possible effects of this intervention. Methods: A one-time skin protection seminar was conducted in 140 apprentices of health-related and non-health-related professions [trained cohort (TC)]. In addition, 134 apprentices of the same occupations were monitored [untrained cohort (UTC)]. The OSD-specific knowledge and the skin condition of the hands were assessed at baseline (T0), after the seminar (T1), and after 6 (T2) and 12 months (T3). Results: The OSD-specific knowledge increased in all cohorts from T0 to T3, but we found a significantly higher knowledge in the TC at T2 (p < 0.001, t = 3.6, df = 196, 95% confidence interval = 0.9, 3.3) and T3 (p < 0.001, t = 3.8, df = 196, 95% confidence interval = 1.0, 3.2) compared to the UTC. Our results indicated a better skin condition of the hands in the TC of the health-related professions but not in the non-health-related professions. Conclusion: The study indicates that an educational intervention may positively influence the disease-specific knowledge and the prevalence of OSD in apprentices. However, definite conclusions cannot be drawn because of the heterogeneous study cohorts and the study design. Future research should aim at tailoring primary prevention to specific target groups, e.g., in view of the duration and frequency of skin protection education, different professions, and gender-specific prevention approaches.

초음파 검사용 젤과 젤 용기의 세균증식 원인 (Causes of Bacterial Growth in Gels and Gel Containers Used for Ultrasonography)

  • This study aimed to investigate the causes of bacterial growth to prevent infection caused by ultrasound gel and gel containers in contact with patients during ultrasonography. To investigate bacterial contamination during manufacturing or storage, we cultured ultrasound gels originally supplied from three manufacturers. To analyze bacterial growth according to the lapse of time and frequency of use of the ultrasound gel container, the gel and container were cultured at regular intervals every week for 4 weeks. In addition, to determine the source of infection, the examiner's hand was inspected with hand plate and the degree of bacterial contamination was measured before the test. As a result of the study, bacteria were not detected in the gel provided at the initial supply, and in the gel and gel container used repeatedly for 4 weeks, the same bacteria residing on the skin were identified in the examiner's hand, such as Staphylococcus epidermidis, Micrococcus luteus, Leuconostoc mesenteroid spp cremoris, Kocuria rhizophila, and etc. Separated strains were classified as those of the low- or non-pathogenicity; however, most of these strains may render fatal consequences to patients of lower level of immunity due to acquired tolerance to antibiotics. At week 1, when the number of tests was the highest, 44 colonies were identified, and at week 4, when the number of tests was the lowest, 4 colonies were identified. As r=0.994, it was found that the number of colonies increased as the number of tests increased. In conclusion, it was confirmed that the cause of the infection was not the ultrasound gel, but the examiner's hand. The ultrasound gel or gel container may be contaminated by skin flora of examiner's hands, which can cause opportunistic infection in patients with low immunity. 부산 해운대 백사장에서 분리한 각질친화성 피부사상균의 분포 (A Distribution of Keratinophilic Fungi Isolated from the Soil of Haeundae Beach in Korea)

  • 각질친화성 진균(keratinophilic fungi)은 대부분 토양에 분포하면서 사람이나 동물의 피부에 감염되어 상재균으로 존재하고, 각질에 침입하여 백선이라는 피부질환을 유발하기도 한다. 전세계적으로 광범위하게 분포되어 있으나 발생 양상은 기후나 지리적인 요건 등에 뚜렷한 차이를 보인다. 이 균주들은 피부나 손톱, 모발 등에 풍부한 케라틴을 분해하면서 영양분을 공급받으며 증식한다. 이 논문에서는 우리나라 여름철 휴가지로 가장 많은 인파가 몰리는 해운대를 선택하게 되었으며 매년 많은 사람들이 모이는 해운대 백사장에 피부사상균의 존재와 검출되는 균의 종에 대해 조사하고 고찰하였다. 토양시료는 무작위로 총 100개를 채취하였고 채취된 검체는 hair-baiting technique을 이용하여 분석하였으며 100개 시료 중 21곳의 모래시료에서 23종의 피부사상균이 검출되었다. 검출된 균들은 Microsporum gypseum (43%), Chrysosporium spp. (35%), Trichophyton ajelloi (13%), Microsporum cookie (9%) 순이었다. 본 연구를 통해 많은 사람들이 모이는 해변 모래사장에 다양한 병원성 진균들이 분포함을 확인하였다. 향후 지속적인 자료수집을 통해 각질친화성 피부사상균의 분포 추이를 조사할 수 있을 것으로 사료된다.

오배자의 항균활성 및 세포독성에 미치는 영향 (Antimicrobial Effect and Cytotoxicity of Chinensis Galla)

  • Chinensis galla has been used as an multi-functional herb, such as anti-inflammatory, anti-virus, and antitumor agent. This study was performed to antimicrobial and cytotoxicity effect in vitro. The results were summarized as follows : Chinensis galla was antimicrobial effect on Staphylococcus aureus subsp. aureus, Bacillus cereus, Bacillus subtilis, Staphylococcus epidermidis. Chinensis galla was antimicrobial effect on Kocuria rhizophila, Corynebacterium ammoniagenes. The extracts of Chinensis galla exhibited cytotoxicity on human dermal fibroblast at $10\;{\mu}{\ell}$ but not at $5\;{\mu}{\ell}$, and the same results was known under a microscope. Accordingly the results show Chinensis galla could antimicrobial effect but exhibited cytotoxicity on human dermal fibroblast at high concentration and it needs more research.

Lactobiocin의 피부 염증 및 여드름 저해효과에 관한 연구 (Inhibitory activity of Lactobiocin on the skin inflammation and acnes)

  • The purpose of this study was to evaluate bacteriocin activity against human flora. Lactobiocin, a bacteriocin produced by Lactococcus sp. HY 449, inhibited the growth of Starphylococcus epidermidis, Starphylococcus aureus, Streptoccoccus pyogenes and Propionibacterium acnes. When crude bacteriocin was added to indicator cells during logarithmic growth, the optical density(O.D 650nm) of cells without bacteriocin increase after 5h of incubation. Whereas in the presence of bacteriocin, the O.D of cell suspensions decreased. The similar patterns were observed for absorbance readings at 280 nm and 260 nm. The release of cellular components when cell were treated with Lactobiocin suggests some degree of membrane damage or cell lysis. Scanning electron microscopy of cells following treatments with Lactobiocin in PBS buffer revealed disruptures of cell morphology. These results indicate that bacteriocin appears to cause cell lysis of tested strains. In cytotoxicity on human fibroblast, LD$\_$50/ of Lactobiocin was ca. 50 mg/ml and no change was observed cell proliferation at the same concentration. Any irritation and allergic reaction did not observed when evaluated by human patch test for Lactobiocin.

구아바 잎 추출물의 항산화 활성 및 항균력 분석 (Antioxidant and Antimicrobial Activities of Psidium guajava leaf extract)

  • 본 연구는 ABTS radical 소거 활성을 통한 항산화 활성, RAW 264.7 대식 세포에서의 세포 독성, DCF-DA assay을 통한 세포 내 ROS 생성 억제 효과, 항균력을 측정하여 구아바 잎 추출물이 화장품 소재로써의 활용 가능성을 검토하고자 하였다. 실험 결과, 구아바 잎 추출물의 우수한 ABTS radical 소거능을 확인하였다. RAW 264.7 대식세포에서의 세포 독성은 나타나지 않았으며 세포 내에서 ROS 생성량은 농도 의존적으로 감소하는 억제효과를 확인하였다. 또한 구아바 잎 추출물의 항균력 분석에서는 피부상재균인 S. aureus, E. coli, C. albicans, P. acnes 균주들에서 항균 활성이 확인되었으며, 각 균주에 대한 최소 저해 농도(MIC)는 대체로 0.25 - 1 mg/mL의 수준으로서 P. acnes ≃ S. aureus < E. coli < C. albican 순의 낮은 농도로 측정되었다. 이와 같은 결과를 통해 구아바 잎 추출물의 항산화 활성과 세포 내 ROS 생성 억제 효과, 피부에 염증을 유발하는 피부상재균에 대한 항균력이 확인됨에 따라 독성이나 부작용이 없는 기능성 화장품 소재로써의 활용이 가능할 것으로 사료된다.

In vitro screening of elastase, collagenase, hyaluronidase, and tyrosinase inhibitory and antioxidant activities of 22 halophyte plant extracts for novel cosmeceuticals

  • Background: Halophyte plant (HPs), a salt-resistant flora, has been reported to provide several health benefits, but the knowledge of its cosmeceutical potential is still ambiguous. Here, 70% ethanol extracts of 22 HPs collected from along the coast of South Korea were investigated for their potentials of antioxidant, anti-aging, and whitening properties for use as materials in novel cosmeceuticals. Methods: Antioxidant activities were determined by DPPH (1,1-diphenyl-2-pricrylhydrazyl) free radical and hydrogen peroxide scavenging assays, and skin aging-related enzyme activities (anti-elastase, anti-collagenase, anti-hyaluronidase, and anti-tyrosinase) were evaluated by using the spectrophotometric method. Results: Among the 22 HPs, we found that Ischaemum antephoroides f. coreana and Atriplex gmelinii extracts presented the strongest scavenging effects against DPPH free radical and hydrogen peroxide, respectively. Our finding additionally suggested that Salicornia europaea extract might provide a major source of anti-elastase and anti-hyaluronidase; meanwhile, Rosa rugosa extract showed the highest anti-collagenase effect. Furthermore, the highest tyrosinase inhibitory activity was possessed by Spartina anglica extract. Conclusion: These findings may suggest that halophyte plants showing biological activities may be potent inhibitors of tyrosinase, elastase, collagenase, and hyaluronidase and could be useful for application in cosmeceuticals.

가축사료에서 병원성 진균 분리 동정 (Isolation and identification of pathogenic fungi from animal feedstuffs)

  • Fugi are eukaryotic, nonphotosynthetic, filamentous or unicellular organisms, most of which grow on nonliving materials as saphrophytes. The majority are therefore opportunistic pathogens and predisposing factors often contribute to the establishment of fungal infection. These include an alteration in the normal flora of the host by prolonged administration of antibiotics, immunosup-pression, concurrent infections, damage to the skin or mucous membranes, constantly moist areas of skin or the exposure to a large infective dose, and as with fungal spores. Fungi may cause a variety of diseases which may be due directly to fungal invasion of tissue or more often to the ingestion of toxins produces by fungi in growing, standing or stored grains and other animals feeds. In this experiment, contaminated fugi were isolated and identified from animal feedstuffs such as Korean cattle, milking cows, pigs and chickens. Twelve genues were isolated from animal feeds, they are 9 from Korean cattle and milking cows feeds, 6 from pigs feeds, and 10 from chickens feeds. Among them, most frequently encountered species was Yeast(56 strains), followed by Fusarium sp(41 strains), Aspergillus sp(20 strains), each of Micorsporum sp and Trichophyton sp(17 strains), Penicilium sp(12 strains), in order. And also minority was isolated as Candide sp(4 strains), Trichoderma sp(3 strains), each of Epidermophytom sp and Absida sp(2 strains), and each of Sporothrix sp and Maduromyces sp(1 strain). Among the Aspergillus sp 20 isolates, A flavus(5 strains), A nidulans(4 strains), A fumigatus(3 strains), A glucans(3 strains), A niger(3 strains) and A terreus(2 strains) were identified.
