Comparison of Natural Resistance-associated Macrophage Protein (NRAMP)1 Expression between Cows with High and Low Milk Somatic Cells Counts

  • Studies using natural resistance-associated macrophage protein (NRAMP) identification indicated that cattle could be selected for immunity. Several studies performed on intracellular organisms such as Mycobacterium, Salmonella, Brucella and Leishmania in human and mouse revealed that resistance against these bacteria was dependent on high activity of NRAMP1 in macrophages. However, hardly any researches have been done on Staphylococcus aureus in bovine mastitis, which is an intracellular organism and the main cause of bovine mastitis. The objectives of this study were to establish reverse transcriptase polymerase chain reaction (RT-PCR) methods, through which NRAMP1 mRNA expression could be compared and analyzed between mastitis-resistant and -susceptible cows. NRAMP1 gene and its expression were investigated using 20 cows (Holstein Friesian) in Korea. Cows were evenly split into two groups, with and without histories of clinical mastitis. Equivalent numbers of cows were randomly selected from each group. Monocytes were isolated from the bovine peripheral blood of each selected cows and activated with lipopolysaccharide (LPS). mRNA was separated from the monocytes and cDNA of NRAMP1 was synthesized and amplified using RT-PCR with amplification of $\beta$-actin as a control. The difference in NRAMP1 expressions of mastitis-resistant (n=10) and -susceptible (n=10) Holstein cows was analyzed. Results demonstrate that resistant cows produced more NRAMP1 mRNA than the susceptible ones, and ratios of NRAMP1:$\beta$-actin expression were higher in resistant cows with or without LPS activation. Therefore, this study could be applied to select bovine mastitis resistant cows before infection based on the expression of NRAMP1.

Anti-inflammatory Effects of the Methanol Extract of Polytrichum Commune via NF-κB Inactivation in RAW 264.7 Macrophage Cells

  • As an attempt to search for bioactive natural products exerting anti-inflammatory activity, we evaluated the effects of the methanol extract of Polytrichum commune Hedw (PCM) (Polytrichaceae) on lipopolysaccharide (LPS)-induced nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$) and pro-inflammatory cytokines release in murine macrophage cell line RAW 264.7. PCM potently inhibits the production of NO, $PGE_2$, tumor necrosis factor (TNF)-$\alpha$ and interleukin (IL)-6. Consistent with these results, PCM also concentration-dependently inhibited LPS-induced inducible NO synthase (iNOS) and cyclooxygase (COX)-2 at the protein levels, and iNOS, COX-2, TNF-$\alpha$ and IL-6 at the mRNA levels without an appreciable cytotoxic effect on RAW 264.7 macrophag cells. Furthermore, PCM inhibited LPS-induced nuclear factor-kappa B (NF-$\kappa$B) activation as determined by NF-$\kappa$B reporter gene assay, and this inhibition was associated with a decrease in the nuclear translocation of p65 and p50 NF-$\kappa$B. Taken together, these results suggest that PCM may play an anti-inflammatory role in LPS-stimulated RAW 264.7 macrophages through the inhibitory regulation of iNOS, COX-2, TNF-$\alpha$ and IL-6 via NF-$\kappa$B inactivation.

A Basic Study on Strategy Development for Improving the Management of Multicultural Family Support Centers (다문화가족지원센터 운영활성화를 위한 전략개발 기초연구)

  • This paper proposes strategy development to improve the management of Multicultural Family Support Centers by analyzing four Multicultural Family Support Centers (MFSCs) in Chungcheongnam-do. The research was conducted from September 2012 to January 2013. Future directions for the four MFSCs are established based on a SWOT analysis on both the internal and external environments of the centers. Various strategic directions for the four MFSCs were identified based on these analyses. For center A (urban type, college contract type), the strategic directions included the specialization strategy, systemization strategy, empowerment strategy, standardization strategy, and achievement maximization strategy. The strategic directions for center B (urban-rural combined type, social welfare center contract type) included the improvement of multiculture awareness, specialization strategy, human resource development strategy, and networking strategy. For center C (farming and fishing community type, direct management type by local authority), the strategic directions included the specialization strategy, activation strategy, qualification strategy, and networking strategy. For center D (farming and fishing community type, social welfare center contract type), the strategic directions included the specialization strategy, business standardization strategy, human resource development strategy, and network expansion strategy. The suggested management strategy development for the four MFSCs can provide implications for the management of MFSCs in other areas in Korea.

An Ethnographic Research on the Practice of Maeul Community Media (마을공동체미디어 실천에 대한 민속지학적 연구)

  • This study examines the practice of maeul community media as follows; Who, why, and how the role of participants is related to the operation of maeul community media, and the participation requirements. For this purpose, the maeul newspaper published in rural areas was approached by ethnographic Research, and participation observation, in-depth interview, and content analysis were performed. In conclusion, it was confirmed that harmonious participation of residents is the core condition of maeul community media practice. Residents' participation is based on the requirements of spontaneity, openness, talent donation, and local relationship. Spontaneity is a desire and effort for the activation of the community as a member of the local community. Openness is related to the operation structure of maeul community media where anyone can freely participate. Talent donation is based on their experience, knowledge, and expertise as a concrete practice of resident participation. Local relationship is the network of community that connects spontaneity and practice, and the operating structure of maeul community media. These requirements should be operated positively to ensure the function of maeul community media and sustainability.

Research on Revitalizing Public Libraries Reflecting Community Characteristics in Yangpyeong-gun (지역사회 특성을 반영한 공공도서관 활성화 방안 연구 - 경기도 양평군을 중심으로 -)

  • This study's purpose is to suggest ways to improve and revitalize public libraries' operations to reflect the Yangpyeong-gun community's characteristics. To this end, the Yangpyeong-gun region's characteristics and the current state of public library operation were analyzed, and a survey was conducted targeting Yangpyeong-gun residents (library users and nonusers). Additionally, interviews were conducted to investigate the library's operating status and limitations and librarians' perceptions of library revitalization plans. As a result of the analysis, in Yangpyeong-gun, improvement points such as library operation improvement and library accessibility improvement reflecting local community changes such as urban development, population change, manpower and collections expansion, complex cultural space strengthening, reading culture programs, and user services were derived.

Changes in Free and Bound Forms of Bioactive Compound Profiles of Adzuki Bean with Germination (발아에 따른 팥의 유리형 및 결합형 기능성분 변화)

  • This study was performed to investigate the changes between free and bound forms of bioactive compounds in germinated adzuki bean. Adzuki bean was germinated at $25^{\circ}C$ for 6 days, and then free and bound forms of bioactive compounds were extracted. Total free polyphenol and flavonoid contents in of raw adzuki bean increased from 0.91 mg/g and 0.60 mg/g in before germination to 2.37 mg/g and 3.03 mg/g in at 6 days after germination, respectively. Bound polyphenol, flavonoid, and phenolic acid contents also increased with increasing germination periods, whereas bound polyphenol and flavonoid contents slightly reduced in at 2 days after germination. The total phenolic acid contents, including ferulic acid, veratric acid, hesperidin, salicylic acid, naringenin, and hesperidin, increased during germination, and germination process can convert compounds to phenolic acid via anabolism and catabolism. Total anthocyanin and anthocyanidin contents of adzuki bean decreased during germination due to hydration by water during soaking or enzyme activation of anthocyanase during germination. These results suggest that the germination process increased utilization of functional compounds such as phenolic compound and isoflavones in black soybean.

In vitro Biological Activity Assay of Ethanol Extract of Radish (무 에탄올 추출물의 in vitro 생리활성 분석)

  • In vitro biological activities of ethanol extract of radish including whitening, hangover removal, antimicrobial and antioxidant activities were analyzed. For whitening activity assay, tyrosinase inhibition rate was measured as $IC_{50}$ (50% inhibitory concentration). The $IC_{50}$ values of radish trunk and root extracts were estimated as 0,9 mg/ml and 2.1 mg/ml, respectively. Radish trunk extract showed 2.5-fold tyrosinase inhibition activity of radish root extract, however, there was no significant difference according to radish species. By alcohol dehydrogenase (ADH) activity assay as a hangover removal activity assay, radish trunk extract (2.5 mg/ml) and root extracts (8 mg/ml) showed ]50% activation of ADH. TBA values of radish trunk and root extracts (1% of each) were 43-61 % level of ${\alpha}-tochoperol$ (2.2%). From the analysis of in vitro biological activities of radish, it was suggested that radish could be used in functional food or cosmetics containing hangover removal, whitening and antioxidant activities.

In Vitro Developmental Competence of Porcine SCNT Embryos is improved by m-Carboxycinnamic Acid Bishydroxamide, Histone Deacetylase Inhibitor

  • Differentiated nuclei can experimentally be returned to an undifferentiated embryonic status after nuclear transfer (NT) to unfertilized metaphase II (MII) oocytes. Nuclear reprogramming is triggered immediately after somatic cell nucleus transfer (SCNT) into recipient cytoplasm and this period is regarded as a key stage for optimizing reprogramming. In a recent study (Dai et al., 2010), use of m-carboxycinnamic acid bishydroxamide (CBHA) as a histone deacetylase inhibitor during the in vitro early culture of murine cloned embryos modifies the acetylation status of somatic nuclei and increases the developmental competence of SCNT embryos. Thus, we examined the effects of CBHA treatment on the in vitro preimplantation development of porcine SCNT embryos and on the acetylated status of histone H3K9 on cloned embryos at the zygote stage. We performed the three groups SCNT: SCNT (NT), CBHA treatment at the porcine fetus fibroblast cells (PFFs) used as donor cells prior to SCNT (CBHA-C) and CBHA treatment at the porcine SCNT embryos during the in vitro early culture after oocyte activation (CBHA-Z). The PFFs were treated with a $15{\mu}M$ of CBHA (8 h) for the early culture and the porcine cloned embryos were treated with a $100{\mu}M$ concentration of CBHA during the in vitro early culture (10 h). Cleavage rates and development to the blastocyst stage were assessed. No significant difference was observed the cleavage rate among the groups (82.6%, 76.4% and 82.2%, respectively). However, the development competence to the blastocyst stage was significantly increased in CBHA-Z embryos (22.7%) as compared to SCNT and CBHA-C embryos (8.6% and 4.1%)(p<0.05). Total cell numbers and viable cell numbers at the blastocyst stage of porcine SCNT embryos were increased in CBHA-Z embryos as compared to those in CBHA-C embryos (p<0.05). Signal level of histone acetylation (H3K9ac) at the zygote stage of SCNT was increased in CBHA-Z embryos as compared to SCNT and CBHA-C embryos. The results of the present study suggested that treatment with CBHA during the in vitro early culture (10 h) had significantly increased the developmental competence and histone acetylation level at the zygote stage.

  • Currently, our country has promoted sustainable growth in agriculture field by expanding the growth engine which is going to creat new value through agrifood industry & ICT convergence, the deployment of computerization in rural areas and the efficiency increase of agricultural administration system. Since the level of domestic agriculture-ICT convergence technology focusing on production areas is at early stage, it is necessary to deploy the successful models through the systematic development of technology and standardization including production, distribution and consumption phase. In addition, because the management and control systems of large glass greenhouse are mostly foreign products with no standardization and related small domestic companies, there is a limit to agri-food & ICT convergence activation led by the agri-food private sector. Also, it is vital to increase productivity & efficiency and improve quality throughout the entire agricultural process including production, distribution and consumption by the fusion of information technology, automatic control technology and unique ICT on existing agricultural technology, Therefore, in this paper we propose the agricultural-ICT convergence technology fields in which our country can lead technology and the standardization plans through analyzing the development, policy and standardization trends on agricultural-ICT convergence technology.

Eleocharis kuroguwai Ohwi Ameliorates LPS-mediated Inflammation by Suppressing MAPKs Signaling (LPS로 유도된 염증 상태 내에서 MAPKs 세포신호 전달체계를 저해하는 올방개의 항염증 효과)

  • In rice agriculture, Eleocharis kuroguwai Ohwi (olbanggae) is a target for herbicidal intervention as a problem weed although it has also long been used clinically as a traditional medicine for jaundice, fever, and blood flow. E. kuroguwai has been evaluated in many clinical trials, but its molecular biological advantages are still unknown. Here, we investigate the anti-inflammatory effects of E. kuroguwai 80% ethanol extracts by screening NO production in LPS-induced macrophage activation. To find the most effective fractions, we partitioned five sub-fractions using HP20 column chromatography, namely 20%, 40%, 60%, 80%, and 100%. Of these, the 60% and 80% sub-fractions were found to significantly inhibit NO production; there were no toxicological effects at any concentration. In addition, the 80% sub-fraction inhibited significantly the iNOS and the mRNA of the pro-inflammatory mediators IL-6, TNF-α, and IL-1β by inhibiting the phosphorylation of JNK and ERK pathways associated with MAPKs signaling. Our results suggest that the 80% E. kuroguwai sub-fraction has the most significant anti-inflammatory effects of inhibiting iNOS and pro-inflammatory mediators and suppressing the phosphorylation of JNK and ERK. Therefore, the 80% sub-fraction of E. kuroguwai extract may be a therapeutic candidate for inflammatory diseases associated with the overexpression of MAPKs.
