Development of Resources for Functional Food and Biological Activity of Lentinus edoes mycelium (표고버섯 균사체의 생리활성과 기능성식품 소재 개발)

  • Objectives : This study was carried out to investigate an antioxidant ability, the change of antioxidant ability, antimicrobial activity and anticancer in functional meat ball and sausage prepared from Lentinus edoes mycelium and texturized soy protein. Methods : This experiments was carried out to investigate antioxidant ability(TBARS, DPPH, SOD-like ability), antibacterial activity and anticancer ability using sarcoma 180 extracts from Lentinus edoes mycelium. Result : Anticancer ability of Lentinus edoes mycelium showed a 28% survival rate and 63% of inhibition rate of tumor, which showed $1.30{\pm}0.4g$ of tumor weight. These results revealed an effective Lentinus edoes mycelium resources as anticancer sources. After heating peoducts prepared from Lentinus edoes mycelium, these products doesn't showed difference between after heating and before heating in measuring of SOD-like activity and DPPH. In DPPH experiment, ethanol extracts showed a high DPPH value as 90.85%, but hot water extracts showed 82.14% in DPPH value. Conclusion : In conclusion, it is very useful resources for preparing functional food on the basis of results from antioxidant(TBARS, DPPH, SOD-like ability), antibacterial activity and anticancer ability using sarcoma 180.

Effect of Dietary Supplementation of Extracts of Mushroom Mycelium on Survival and Growth of juvenile Flounder, Paralichthys olivacceus (버섯균사체 배양액 첨가사료가 넙치 치어의 생존 및 성장에 미치는 영향)

  • The effects of mycelium cultural extract supplemented diet, hematology and disease resistance against Vibrio anguillarum in juvenile flounder, Paralichthys olivaceus were evaluated. Fish were fed the Phellinus linteus with Coriolus militaris versicolor mixed mycelium cultural extract supplemented diet, Phellinus linteus mycelium cultural extract supplemented diet and Coriolus militaris tmycelium cultural extract supplemented diet a commercial diet for 12 week. The body weight and length gain from the fish fed on daily the phellinus with coriolus versicolor mixed mycelium cultural extract supplemented diet, phellinus linteus mycelium cultural extract supplemented diet and Coriolus versicolor mycelium cultural extract supplemented diet of each mycelium cultural extract were higher than the control and the Glutamic oxalacetic transaminase (GPT) were of lower than the control. The relative precent survival rate (RPS) after an artificial challenge with $7{\times}10^5\;CFU$ of Vibrio anguillarum per fish was higher than the control.

Proliferation of Tricholoma matsutake Mycelial Mats in Pine Forest Using Mass Liquid Inoculum

  • Two isolates of Tricholoma matsutake T-008 and T-034, preserved in Entomopathogenic Fungal Culture Collection (EFCC) of Korea, were used in the present study. The isolates had 100% Bootstrap homology with Tricholoma matsutake U62964 and T. matsutake AB188557 and AF309538 preserved in Gene Bank of NCBI. Mycelial growth of T. matsutake was highest in TMM and MYA at $25^{\circ}C$. The highest dry wt. of mycelium was obtained after 65 days of culture, when 6 mycelial discs were inoculated in 100 ml of broth in 250 ml shaking flask. Mycelial mats were observed in clumped condition at the inoculation sites of pine forest after two weeks of inoculation. After 5 months of inoculation, mycelia mats were observed growing inside soil and walls of a few inoculation sites, while mycelial mats growth up to $5{\sim}8$ cm were observed in the roots of pine tree after 6 months. The survival rate of the inoculum was about 40% of the total inoculation sites. The survival rate was found below 20% when the mycelium was inoculated in the summer. The reasons for low survival rates of the mycelium were mainly due to dry season and the soil-borne small animals such as earthworm and mole. After one year of inoculation, no external difference was observed between the artificially inoculated mycelia and the naturally existing mycelia of T. matsutake. The present study showed that fruiting bodies of T. matsutake could be produced by artificial inoculation under the appropriate environmental conditions.

Production and Transplanting of Ectomycorrhizal Pine Seedlings Using the Old Fairy Ring of Tricholoma matsutake (기존 송이 균환(菌環)을 이용한 송이균 감염 소나무의 생산 및 이식)

  • To make a new fairy ring of Tricholoma matsutake in situ, the way of production and transplanting of ectomycorrhizal seedlings of T. matsutake using Pinus densiflora was investigated after transplanting from 2000 to 2005 as well as the method to improve their survival rate for the fungus. For the production of ectomycorrhizal pine seedlings, the seedlings planted at the edge of fairy ring of T. matsutake in November showed 97% of the survival rate, while those planted in April showed 80% of the rate. For the complete infection of the T. matsutake, it required more than two years after planting. The infection rate of mycelia for the ectomycorrhizal seedlings was 17.6% when the natural seedlings were used, whereas it was relatively low when the seedlings prepared from the nursery were used. The survival of T. matsutake mycelium reached up to 22% by the transplanting in April, while the mycelium transplanted in October and November showed less than 5% of the survival. The survival of T. matsutake on the transplanted seedlings was the highest in the seedlings having 50% of infection rate before transplanting. Excavation of the ectomycorrhizal seedling to examine the vitality of ectomycorrhizal roots of T. matsutake resulted in the perishing of them. Therefore, the method using a 'rice bag triers' to check living mycelium of T. matsutake without digging of transplanted seedlings was introduced in this study. In addition, it is recommended that the examination has to be conducted at least two years after transplanting.

Proton Beam Sensitivity of Basidiospore and Mycelium in Pleurotus ostreatus (느타리버섯 포자와 균사체의 양성자 빔 조사에 대한 민감도)

  • To assess the effects of proton beam on radiation sensitivity of the basidiospore and mycelium of oyster mushroom (Pleurotus ostreatus), the $D_{10}$ values and $L_{50}$ (lethal 50%) values were analysed. By the proton beam radiation, the survival rate and germination rates increased at the dose of $10\;Gy{\sim}100\;Gy$ and then decreased significantly over 500 Gy. $L_{50}$ values of basidiospore and mycelium of Pleurotus ostreatus were over 500 Gy and 400 Gy, respectively. $D_{10}$ values were calculated from linear regression formulae ($D_{10}\;=\;-1/slope(b)$, y = a + bx) as 750 Gy and 1,250 Gy, respectively. Based on our experiment, the optimum dose of proton beam as a mutation source would be between from 500 Gy to 750 Gy for basidiospores and from 400 Gy to 1000 Gy for mycelium of oyster mushroom.

The Possible Role of SCO3388, a tmrB-like Gene of Streptomyces coelicolor, in Germination and Stress Survival of Spores

  • The SCO3388 gene from Streptomyces coelicolor is homologous to tmrB, the tunicamycin resistance gene of Bacillus subtilis. The SCO3388-inactivation strain (SY-tbl-1) was generated by replacing SCO3388 with thiostrepton resistance gene. Spores of S. coelicolor derivatives were prepared on mannitol-soy flour (MS) agar on which SY-tbl-1 displayed no significant defect in growth and development. When plated on R4 agar, spores of SYtbl-1 displayed retardation in growth and sporulation, whereas its mycelium gave rise to normal growth. Thus, SCO3388 is suggested to be involved in the dormant spore germination. Expression of SCO3388 under the ermE1 promoter restored but only partially the ability to sporulate in SY-tbl-1. Neither SY-tbl-1 nor SY-tbl-1/ermE1p-SCO3388 showed a difference in tunicamycin resistance to the wild type whereas, interestingly, the introduction of ermE1p-SCO3388 dramatically enhanced spore survival to heat and detergent treatments, suggesting that SCO3388 might play a role in the maintenance of spore cell wall integrity.

Studies on the Possidility of Oak Mushroom(Shiitake) Cultivation on Ban Oak (Quercus incana) of India (인도산(印度産) 참나무(Quercus incana)를 이용한 표고재배(栽培)의 가능성(可能性)에 관한 연구(硏究))

  • The study has been carried out to invetigate the possibility of oak mushroom (shiitake) cultivation on Ban Oak (Quercus incana) growing naturally in Simlaregion, India. The survival and growth of oak mushroom mycelium and fruit body formation on Q. incana as well as the composition of the log were compared with that of Bristle-tooth oak (Q. serrata) which are being used for mushroom cultivation in Korea. The results are summarized as follows. 1. The content of alcohol-benzen extract, NaOH extract, NaOH extract and ash were higher in Q. incana than in Q. Serrata. While cellulose and pentosan were less in the formers. 2. The mycelial growth of oak mushroom were more rapid on sawdust medium of Q. Serrata than on Q. incana. However the mycelial growth on the later were more compact. 3. The mycelial growth of oak mushroom were more rapid on the logs of Q. serrata than Q. incana. The mycelium survived well on both two species, and no difference in the survival rate of mycelium were observed. 4. The first fruit bodies on logs of Q. Serrata and Q. incana were appeared 16 months inoculation of spawn. 5. In view of the above results. it seems that the cultivation of oak mushroom (shiitake) on Ban oak (Q. incana), growing in India, is possible.

Characteristics of Phytophthora capsici Causing Pepper Phytophthora Blight Resistant to Metalaxyl (Metalaxyl에 대한 저항성 고추 역병균의 특성)

  • Isolation frequency of resistant isolates of Phytophthora capsici to metalaxyl was reported to be 38.9% through the resistance monitoring for metalaxyl in P. capsici causing pepper Phytophthora blight. Metalaxyl was very effective to mycelium growth, while not to zoosporangium germination and zoospore release. $EC_{50}$ values of metalaxyl in the inhibition of mycelium growth were 0.204, 0.151, 0.379, and $0.215\;{\mu}g\;mL^{-1}$ against each isolate sensitive to the fungicide as P. capsici 06-119, 06-143, P08-7, and P08-31, respectively, whilst those were 5.242, 5.724, 6.621, and $5.377\;{\mu}g\;mL^{-1}$ in P. capsici 06-125, 06-155, P08-50, and P08-60. For the field fitness, several factors, which were mycelium growth, zoosporangium germination, zoospore release, virulence to pepper plants, and the zoosporangium and the oospore production, were investigated with 4 sensitive isolates and 4 resistant isolates. Between 2 groups differentiated by the sensitivity of metalaxyl, there was no significance in mycelium growth, zoosporangium germination, zoospore release, and virulence to pepper plants. However, the zoosporangium and the oospore production in each resistant isolate, which were related to survival of P. capsici in fields, were superior to those of sensitive isolates. Based on results of this study, it was suggested that the increase of the percentage of resistant isolates to metalaxyl resulted from the high capacities of the zoosporangium and the oospore production.

Determination of the Minimum Size of Seedlings with Matsutake Mycelia That Can Survive in the Field for Matsutake-infected Pine Tree Production (송이 감염묘 육성을 위해 야외 조건에서도 송이균 생존이 가능한 소나무의 최소 크기 결정)

  • Despite the high commercial value of the pine mushroom (Tricholoma matsutake) around the world, its production still depends upon natural harvesting. In recent years, mushroom researchers in Korea and Japan have been successful in artificially cultivating T. matsutake by producing single mushroom through matsutake-infected pine seedlings. In this study, we determined the minimum seedling height required for the survival of matsutake mycelia in the infected pine seedlings in the field. The survival rate of matsutake mycelia in the matsutake-infected pine seedlings was 50% (14) in two years and 71% (10) in one year after their transplantation. The average height at time of transplantation of the matsutake-infected pine seedlings that bore surviving mycelia after transplantation was 25 cm (minimum 12 cm to maximum 40 cm). In conclusion, the matsutake mycelium of matsutake-infected pine seedlings was able to survive in field conditions if the height of the seedling at the time of T. matsutake infection was at least 12 cm. These results suggest that the height of the host plants used in conventional matsutake-infected pine seedling production should be greatly reduced to improve the matsutake cultivation. Therefore, standardization of the seedling height for artificial cultivation of pine mushrooms by the matsutake-infected pine seedling method is suggested.

The Outbreak and Propagule formation of black root rot caused by Calonectria crotalariae in Korea (콩 흑색뿌리썩음병의 발생과 Propagule의 형성)

  • The infection rate of soybean black root rot disease caused by Calonectria crotalariae was about $14\%$. The isolated fungi from the infected soybean roots and stems were Calonectria crotalariae, Fusarium solani, F. roseum, Phomopsis sojae, Pythium aphanidermatum, Rhizoctonia solani and Macrophomina sp. Among them, C. crotalariae was the most virulent pathogen under the laboratory conditions. Mycelial growth and microsclerotial formation were good on PSA containing 1000cc of water, 100g of potato and 20g of sugar. Mycelial growth, sporulation and microsclerotial formation were good on sterilized root. Perithecial formation was better in the dark condition than in the light. Survival of macroconidia was not available between $0\~25\%$ soil water content. Microsclerotia and mycelium in infected plant debris were survived for 4 months at to $8\%\;50\%$ soil water content. The plant height, when inoculated with $1.2\%$ inoculum density, reached approximately half of uninoculated plants. Disease severity was much higher at nonsterilized soil than completely sterilized soil. It was determined that the host range of this pathogen includes soybean, peanut, green bean and red bean.
