연교추출물의 피부 항노화 효과 (Skin Anti-aging Effect of Forsythia viridissima L. Extract)

  • 본 연구에서는 새로운 주름개선 소재로서 phytoestrogen의 일종인 lignans를 각각 함유하는 연교추출물을 추출 분리하여 노화와 관련된 생리활성 효과를 확인하였다. Arctigenin과 matairesinol을 각각 함유하는 연교추출물은 처리농도가 증가함에 따라 농도 의존적으로 DPPH와 superoxide radical에 대한 소거효과를 나타내었다. DPPH radical 소거효과에서 arctigenin-rich와 matairesinol-rich는 각각 $IC_{50}$ 값이 $5.747{\mu}g/ml,\;4.450\;{\mu}g/ml$를 나타내었으며, superoxide radical 소거효과에서는 각각 1.634 mg/m1, 542.428 mg/ml로 양성 대조군으로 사용한 BHT보다 우수한 항산화 효과를 보였다. Type I collagen 합성효과도 arctigenin 및 matairesinol을 각각 함유한 연교추출물 그리고 양성 대조군 vitamin C를 $100\;{\mu}g/ml$을 처리하였을 때 각각 10.5, 26.4, 15.7%의 합성율을 보임으로써 vitamin C와 유사하거나 우수한 효과를 나타내었다. 또한, UVA에 의해 발현이 증가되는 MMP-1의 발현저해 효과는 matairesinol을 함유한 연교추출물의 경우 oleanolic acid와 유사한 저해 효과를 보였다. Matairesinol을 함유한 연교추출물을 함유한 크림의 피부 주름개선 효과를 평가하기 위하여, 눈가 주름 부위의 제품 도포 후 피부 주름의 변화와 피부거침 개선 효과를 평가하였다. 연교추출물을 함유한 크림을 약 12주간 도포하였을 경우, 깊은 주름과 총 주름수의 감소효과를 관찰할 수 있었으며, 연교추출물 2%를 적용한 인체 안전성시험에서 무자극으로 판명 되 었다. 본 연구를 통하여 lignans를 각각 함유하는 연교추출물은 항산화 효과와 생체 단백질의 30%를 구성하는 collagen의 합성효과 및 UVA에 의한 MMP-1의 발현저해 효과를 보였다. 인체에 무자극성으로 제품에 적용시 관찰되는 피부 주름개선 효과는 lignans를 각각 함유한 연교추출물을 새로운 주름개선을 위한 천연소재로 이용될 수 있을 것으로 사료된다.

Schizandra chinensis Alkaloids Inhibit Lipopolysaccharide-Induced Inflammatory Responses in BV2 Microglial Cells

  • Schizandra chinensis (S. chinensis) exhibits a harmless, 'adaptogen-type' effect leading to improvements in mental performance and learning efficacy in brain. Activated microglia contributes to neuronal injury by releasing neurotoxic products, which make it important to regulate microglial activation to prevent further cytological as well as functional brain damage. However, the effect of S. chinensis on microglial activation has not been examined yet. We have investigated the effects of four compounds (Gomisin A, Gomisin N, Schizandrin and Schizandrol A) from S. chinensis on lipopolysaccharide (LPS)-induced microglial activation. In this study, BV2 microglial cells were activated with LPS and the microglial activation was assessed by up-regulation of activation markers such as nitric oxide (NO), reactive oxygen species (ROS), and matrix metalloproteinase-9 (MMP-9). The results showed that all four compounds significantly reduced the intracellular level of ROS, the release of NO and MMP-9 as well as LPS-induced phosphorylation of ERK1/2. These results strongly suggested that S. chinensis may be useful to modulate inflammation-mediated brain damage by regulating microglial activation.

악관절질환에서 MRI 상 관절원판 후조직의 상대적 신호강도와 관절액의 Interleukin-6, MMP-2 및 MMP-9 농도 (RELATIVE SIGNAL INTENSITY OF RETRODISCAL TISSUE IN MRI, AND SYNOVIAL FLUID CONCENTRATION OF INTERLEUKIN-6, MMP-2 AND MMP-9 IN TEMPOROMANDIBULAR JOINT DISORDER)

  • In the progression of the Temporomandibular Joint Disorder(TMD), not only deformation and perforation of disc occur. But also fibrotic adhesion and inflammatory changes to the retrodiscal tissue can be seen in addition to the condylar degenerative change (e.g. osteoarthritis). However, the correct diagnosis,?planning for appropriate treatment, and prediction of prognosis are limited, because there are no means to stage the progression of the disorder. In this study relative signal intensity of retrodiscal tissue in MRI and the synovial fluid concentration of matrix metalloproteinase-2 (MMP-2), MMP-9, and Interleukin-6(IL-6) in the 23 temporomandibular joints(TMJ), from 17 patients with TMD were evaluated as a possible diagnostic marker. The relative signal intensity of retrodiscal tissue was referenced to brain gray matter with same region of interest(ROI) size. The concentrations of MMP-2, MMP-9, and IL-6 were evaluated by Enzyme Linked Immunosorbent Assay (ELISA). The collected data were compared with condylar degenerative change, joint effusion and disc position observed in MRI. The relative signal intensity of the retrodiscal tissue was increased significantly when degenerative changes were present. In addition, there was significantly high signal intensity in the presence of a disc displaced without reduction. The concentration of IL-6 was significantly increased when condylar degenerative change was no observed. And there were no changes in the levels of IL-6 according to disc position and joint effusion measurement. Moreover, there were no significant relevance between the concentration of total MMP-2 and active MMP-9 in synovial fluid, relative to degenerative changes in the mandibular condyle, to joint effusion, and to disc position observed on MRI images. In conclusion, the relative signal intensity of the retrodiscal tissue can be regarded as a mean of diagnosing the procession of TMD in a non-invasive manner. But more additional studies are required for the levels of MMP-2. MMP-9, and IL-6 to determine their potentials as a diagnostic marker for TMD.

Cell clusters in intervertebral disc degeneration: an attempted repair mechanism aborted via apoptosis

  • Cell clusters are a histological hallmark feature of intervertebral disc degeneration. Clusters arise from cell proliferation, are associated with replicative senescence, and remain metabolically, but their precise role in various stages of disc degeneration remain obscure. The aim of this study was therefore to investigate small, medium, and large size cell-clusters. For this purpose, human disc samples were collected from 55 subjects, aged 37-72 years, 21 patients had disc herniation, 10 had degenerated non-herniated discs, and 9 had degenerative scoliosis with spinal curvature <45°. 15 non-degenerated control discs were from cadavers. Clusters and matrix changes were investigated with histology, immunohistochemistry, and Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). Data obtained were analyzed with spearman rank correlation and ANOVA. Results revealed, small and medium-sized clusters were positive for cell proliferation markers Ki-67 and proliferating cell nuclear antigen (PCNA) in control and slightly degenerated human discs, while large cell clusters were typically more abundant in severely degenerated and herniated discs. Large clusters associated with matrix fissures, proteoglycan loss, matrix metalloproteinase-1 (MMP-1), and Caspase-3. Spatial association findings were reconfirmed with SDS-PAGE that showed presence to these target markers based on its molecular weight. Controls, slightly degenerated discs showed smaller clusters, less proteoglycan loss, MMP-1, and Caspase-3. In conclusion, cell clusters in the early stages of degeneration could be indicative of repair, however sustained loading increases large cell clusters especially around microscopic fissures that accelerates inflammatory catabolism and alters cellular metabolism, thus attempted repair process initiated by cell clusters fails and is aborted at least in part via apoptosis.

Prevotella nigrescens lipopolysaccharides로 자극된 MG63 세포에서 분비되는 기질금속단백질 MMP-1과 TIMP-1의 수준에 관한 연구 (MMP-1 and TIMP-1 production in MG-63 cells stimulated with Prevotella nigrescens Lipopolysaccharide)

  • 본 연구의 목적은 Prevotella nigrescens lipopolysaccharides (LPS)로 자극된 MG63 osteosarcoma 세포에서 생성, 분비되는 기질금속단백효소인 MMP-1과 그 억제제인 TIMP-1을 측정할 뿐아니라 수산화칼슘으로 처리한 P. nigrescens LPS에 의한 기질금속단백효소와 그 억제제의 분비수준의 변화를 알아보는데 있다. 혐기성 조건에서 배양한 P. nigrescens로부터 LPS를 추출하여 순수정제한 다음 0, 1 그리고 $10{\;}\mu\textrm{g}/ml$의 LPS 농도로 MG63 세포를 자극하거나 또는 수산화칼슘으로 처리한 $10{\;}\mu\textrm{g}/ml$의 LPS로 세포를 다양한 자극하여 다양한 시간이 경과한 다음 세포로부터 분비되는 MMP-1과 TIMP-1의 RNA 수준을 real time-PCR 방법으로 측정하였다. 실험결과 MMP-1의 mRNA수준은 48시간에서 최고에 달하였고 그 분비정도는 LPS의 농도에 비례하였다. TIMP-1 mRNA는 $1{\;}\mu\textrm{g}/ml$의 세균성 LPS 자극시 24시간 및 48시간에서 높은 증가를 보였으나 고농도인 $10{\;}\mu\textrm{g}/ml$의 LPS로 자극한 경우 오히려 그 발현이 억제되었다. 또한 수산화칼슘으로 전처리한 P. nigrescens LPS로 자극한 MG 63 세포에서는 MMP-1과 TIMP-1의 분비가 억제되었다. 이러한 결과를 통해 볼 때 P. nigrescens LPS에 의한 MMP-1과 TIMP-1의 발현조절이 치근단 질환에서 발생하는 치조골 흡수 기전중 하나로 사료된다. 뿐만 아니라 P. nigrescens에 의해 분비되는 기질금속단백효소를 매개로 하는 염증반응 감소에 수산화칼슘이 효과적으로 작용하는것으로 확인되어 치근단 질환에 관여하는 세균성 LPS를 제거하기 위해 임상적으로 사용되는 근거가 될 수 있다.

Sulforaphane controls TPA-induced MMP-9 expression through the NF-κB signaling pathway, but not AP-1, in MCF-7 breast cancer cells

  • Sulforaphane [1-isothiocyanato-4-(methylsulfinyl)-butane] is an isothiocyanate found in some cruciferous vegetables, especially broccoli. Sulforaphane has been shown to display anti-cancer properties against various cancer cell lines. Matrix metalloproteinase-9 (MMP-9), which degrades the extracellular matrix (ECM), plays an important role in cancer cell invasion. In this study, we investigated the effect of sulforaphane on 12-O-tetradecanoyl phorbol-13-acetate (TPA)-induced MMP-9 expression and cell invasion in MCF-7 cells. TPA-induced MMP-9 expression and cell invasion were decreased by sulforaphane treatment. TPA substantially increased NF-${\kappa}B$ and AP-1 DNA binding activity. Pre-treatment with sulforaphane inhibited TPA-stimulated NF-${\kappa}B$ binding activity, but not AP-1 binding activity. In addition, we found that sulforaphane suppressed NF-${\kappa}B$ activation, by inhibiting phosphorylation of $I{\kappa}B $ in TPA-treated MCF-7 cells. In this study, we demonstrated that the inhibition of TPA-induced MMP-9 expression and cell invasion by sulforaphane was mediated by the suppression of the NF-${\kappa}B$ pathway in MCF-7 cells.

The effect of baicalin in a mouse model of retinopathy of prematurity

  • Baicalin is a flavonoid derived from the dried root of Scutellaria baicalensis. In this study, oxygen-induced retinopathy was used to characterize the anti-angiogenic properties of baicalin in mice. Pups were exposed to a hyperbaric oxygen environment to induce retinal angiogenesis and were subjected to intraperitoneal injection of baicalin. Avascular area, neovascular tufts, and neovascular lumens were quantified from digital images. Compared to the vehicle, baicalin clearly reduced the central avascular zone and the number of neovascular tufts and lumens. High-dose baicalin (10 mg/kg) significantly reduced the expression of matrix metalloproteinase-2 (MMP-2), MMP-9, angiotensin II, and vascular endothelial growth factor (VEGF). These results show that baicalin is a powerful antiangiogenic compound that attenuates new vessel formation in the retina after systemic administration, and is a candidate substance for therapeutic inhibition of retinal angiogenesis. Development of Vaccinium uliginosum L. extracts for whitening & anti-wrinkle functional food

  • This study is peformed to investigate the effect of water extract from Vaccinium uliginosum L., on melanin production in B 16 melanoma cells, procollagen production and matrix metalloproteinase-1(MMP-1) inhibition in human fibroblast cells. One hundred grams of the Vaccinium uliginosum L. was extracted with 2000 mL of water($90^{\circ}C$, 16h, 2times). The water extracts were lyophilized and stored at $4^{\circ}C$ until used. Dry weight yields of extracts of Vaccinium uliginosum L. were $3\%$(w/w). Extracts from Vaccinium uliginosum L. showed scavenger activities on DPPH radical, superoxide anion radical, hydroxyl radical, hydrogen peroxide and singlet oxygen radical. And these substances inhibited release of cyiokines from human keratinocyte after UV B exposure. Therefore we confirmed that extracts from Vaccinium uliginosum L. had antioxidative effect. These substances inhibited purified tyrosinase activity and melanogenesis in B 16 melanoma cells treated/untreated IL-$1{\alpha}$. Moreover this extract stimulated procollagen production and inhibited MMP-1 production in human fibroblast cells treated/untreated IL-$1{\beta}$. Therefore we confirmed that extracts from Vaccinium uliginosum L. had whitening effect. And these substances decreased degree of wrinkle in hairless mouse skin that induced by UV B irradiation. Therefore we confirmed that extracts from Vaccinium uliginosum L. had anti-wrinkle effect. From the above results, it is possible that Vaccinium uliginosum L. may be developed to be an anti-melanogenesis agent and anti-wrinkle agent.

유근피 추출물의 피부개선효과 (Anti-Wrinkle Effect of Ulmus davidiana Extracts)

  • The bark of the root and stem of Ulmus davidiana var. japonica has been used as a traditional Korean medicine to treat inflammatory disorders. This plant reportedly shows antioxidant, anticancer, and anti-inflammatory effects. In this study, we investigated the protective effects of Ulmus davidiana var. japonica ethanolic extract (UDE) on UVB irradiation-induced wrinkle in hairless mice. We evaluated for their free radical-scavenging activities against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the anti-elastase activities, and for their anti-matrix metalloproteinase-1 (MMP-1) activity in human skin fibroblast cells. In the wrinkle measurement and image analysis of skin replicas, the results showed that UDE significantly inhibited wrinkle formation caused by chronic UVB irradiation. These results suggest that UDE has anti-wrinkle activity.

Ellagic acid plays a protective role against UV-B-induced oxidative stress by up-regulating antioxidant components in human dermal fibroblasts

  • Ellagic acid (EA), an antioxidant polyphenolic constituent of plant origin, has been reported to possess diverse pharmacological properties, including anti-inflammatory, anti-tumor and immunomodulatory activities. This work aimed to clarify the skin anti-photoaging properties of EA in human dermal fibroblasts. The skin anti-photoaging activity was evaluated by analyzing the reactive oxygen species (ROS), matrix metalloproteinase-2 (MMP-2), total glutathione (GSH) and superoxide dismutase (SOD) activity levels as well as cell viability in dermal fibroblasts under UV-B irradiation. When fibroblasts were exposed to EA prior to UV-B irradiation, EA suppressed UV-B-induced ROS and proMMP-2 elevation. However, EA restored total GSH and SOD activity levels diminished in fibroblasts under UV-B irradiation. EA had an up-regulating activity on the UV-B-reduced Nrf2 levels in fibroblasts. EA, at the concentrations used, was unable to interfere with cell viabilities in both non-irradiated and irradiated fibroblasts. In human dermal fibroblasts, EA plays a defensive role against UV-B-induced oxidative stress possibly through an Nrf2-dependent pathway, indicating that this compound has potential skin antiphotoaging properties.
