Efficient Procedures for Direct Shoots Regeneration from Leaf Explants of Rehmania glutinosa Lib. (지황 잎조직 절편으로부터 신초 형성)

  • Adventitious shoots were directly induced from leaf explants of R. glutinosa, an important medicinal plant. Proliferating shoot cultures were obtained by culturing leaf discs on Murashige and Skoog(MS) medium alone or combination with auxins and cytokinins. MS medium supplemented with 1 $mg/{\ell}\;BA\;and\;2\;mg/{\ell}$ IAA was the most effective, providing high shoot bud formation frequency without formation of intervening callus. The effect of leaf age on adventitious shoot formation was also investigated. The maximum shoot bud production (93.4%) was achived using 3rd leaf from apex of 6 weeks old plantlets after seed germination. Plantlet were rooted on an half-strength MS (1/2MS) medium containing 0.1 $mg/{\ell}\;IBA$. This prtocol is useful for clonal propagation and Agrobacterium-mediated transforamtion in R. glutinosa.

Neopestalotiopsis Leaf Blight, an Emerging Concern on Leatherleaf Fern in Indonesia

  • Leatherleaf fern (Rumohra adiantiformis) is an important ornamental plant in Indonesia and global. Green fern leaves with bold dark green color with long shelf-life, attract florists as decoration. Indonesia is one important leatherleaf fern exporters, however currently an outbreak of leaf blight decreased production significantly. Initial symptom was reddish brown spots from edge of leaf, which was gradually followed by dark-brown necrotic lesions causing leaf blight and dried. This is a study to do Koch-Postulate approach and molecular identification, to identify the pathogen of the "new emerging disease" reported. Based on multigene analysis using primers from ITS, β-tub and tef1-α gene markers, the pathogen was identified as Neopestalotiopsis sp. All sequences have been deposited in GenBank with accession number of OR905551 (ITS), OR899817 (ßtubulin) and OR899816 (TEF). This Neopestalotiopsis leaf blight causes an emerging concern in leatherleaf fern in Indonesia and global biosecurity because it infected an export commodity.

Effects of Oligosaccharide and Pseudomonas sp. on the Growth of Potted Kalanchoe During Summer Season (천연올리고당 및 Pseudomonas속 길항미생물의 단독 및 혼용처리가 고온기 칼랑코에 생육촉진에 미치는 영향)

  • Most severe problem in production of potted kalanchoe during summer season is retardation of growth caused by high temperature. The aim of this experiment was aimed to investigate the effects of natural products such as algin-oligosacchride and glucosamine oligosaccharide, plant growth promoting rhizovacteria such as Pseudomonas sp. B and Pseudomonas sp. D2, and AG-solution on the growth of potted kalanchoe under the different root zone temperature in the greenhouse. Growth characteristics in terms of plant height, leaf length, leaf width, leaf area, leaf weight, fresh weight of shoot and root and root length were recorded under three root zone temperatures (25$^{\circ}C$, 30$^{\circ}C$, 35$^{\circ}C$). In 25$^{\circ}C$, the mixed treatment of Pseudomonas sp. B and glucosamine oligosaccharide resulted in the best growth in terms of plant height, leaf area and root weight. In 3-$^{\circ}C$, glucosamine oligosaccharide treatment gave fair result in plant height and leaf weight, but the mixed treatemtn of Pseudomonas sp. D2 and algin-oligosaccharide showed better growth on leaf area and root weight. In 35$^{\circ}C$, the mixed treatment of Pseudomonas sp. B and glucosamine oligosaccharide could greatly improve the plant height, leaf area, leaf weight and root weight. These results demonstrated that the mixed treatment of natural products and microorganisms could overcome the detrimental effects caused by high temeprature in the production of kalanchoe.

A Study on Growth and Development Information and Growth Prediction Model Development Influencing on the Production of Citrus Fruits

  • The purpose of this study is to develop the growth prediction model that can predict growth and development information influencing on the production of citrus fruits. The growth model was developed to predict the floral leaf ratio, number of fruit sets, fruit width, and overweight fruits depending on the main period of growth and development by considering the weather factors because the fruit production is influenced by weather depending on the growth and development period. To predict the outdoor-grown citrus fruit production, the investigation result for the standard farms is used as the basic data; in this study, we also understood that the influence of weather factors on the citrus fruit production based on the data from 2004 to 2013 of the outdoor-grown citrus fruit observation report in which the standard farms were targeted by the Agricultural Research Service and suggested the growth and development information prediction model with the weather information as an independent variable to build the observation model. The growth and development model for outdoor-grown citrus fruits was assumed by using the Ordinary Least Square method (OLS), and the developed growth prediction model can make a prediction in advance with the weather factors prior to the observation investigation for the citrus fruit production. To predict the growth and development information of the production of citrus fruits having a great ripple effect as a representative crop in Jeju agriculture, the prediction result regarding the production applying the weather factors depending on growth and development period could be applied usefully.

  • 1-Deoxynojirimycin (DNJ) have been extensively investigated for their α-glucosidase inhibitor on postprandial hyperglycemia, and applied in nutraceuticals and medicine for preventing or delaying progression of type 2 diabetes. However, the amount of DNJ in mulberry leaves is low (about 0.1%), therefore, more effective extraction method is needed. This study was performed to develop microbial DNJ for biological methods of DNJ as an alternative to the chemical methods. In this study, we obtained evidence for Bacillus subtilis that produce DNJ in large quantities by high performance liquid chromatography. Inhibition of α-glucosidase activity was determined to DNJ production or non-production. Investigation of the effect of mulberry leaves powder concentration (1~5%), using the DNJ high-production bacteria, provided evidence for microbial mass production of DNJ. When the 4% mulberry leaf powder for 9 days was used, the α-glucosidase inhibitory activity was over the 85%. Also, the results presented in this study confirm DNJ yield's increasement in microbes using the various of nutrients and provide insight of ways to improve DNJ yields in microorganisms.

Analysis of Productivity in Rice Plant -IV. Sink-filling rate and sink-source relation (벼의 생산력 분석(分析) -IV. 수기(受器)의 충전속도(充塡速度)와 수기(受器)-급기(給器) 관계(關係))

  • Sink (grain)-filling rates of IR667 line (Suwon 213 and 214) and local leading varieties (Jinheung and Paldal) were investigated in relation to sink-source ratio, sink-source distance, forms of photosynthates, and weather factors. The results are as follows. 1. IR667 line have higher maximum filling rate (g. grain/day. ear) and shorter duration to reach maximum filling rate than local varieties. The curve pattern of sink-filling rate was high and steep type in IR667 line and low and broad type in local varieties. 2. Weather factors seem to give little effect on the curve pattern of sink-filling rate. 3. The functional sink-source distance (actual sink-source distance multiplied by the contribution rate of leaf for grain production) was shorter in upper leaves (flag and 2nd leaf) for IR667 line, the upper leaf-dependent type in grain production, and was longer in lower leaves (3rd and 4th) for local varieties, the lower leaf-dependent type. Specially short first internode from top may contribute to the upper leaf depencency of IR667 line. 4. According to free sugar-starch ratio (sugar/starch) in the culm and leaf sheath IR667 line could be classified as the high sugar type and the local varieties as the high starch type. The ratio of transportable form (sugar) to non transportable photosynthate (starch) seems to relate with sink-filling rate. And high sugar type is expected to have higher efficiency for grain production in view of energy balance. 5. A hypothesis that the higher in the uniformity within the series of productive structure the more efficient in grain production is proposed and discussed in relation to productivity. 6. According to the pattern of percent nutrient retention of each leaf blade IR667 showed the central retention type and Jinheung showed the apical retention type and each retention type appears to be a cause of each canopy conservation pattern. 7. From the content and percent distribution of nutrient in various organ IR667 could be classified as the leaf sheath dominant type and Jinheung as the leaf blade dominant type. 8. The fact that the greater the percent translocation of nutrient into grain the greater the percent nutrient retention in leaf blade was held between nitrogen and phosphorus within a variety and between varieties within a nutrient (N, P or K).

Enhancement of γ-aminobutyric Acid Production by Combination of Barley Leaf and Corn Silk and Its Fermentation with Lactic Acid Bacteria (보리 잎과 옥수수 수염의 혼합과 유산균 발효를 이용한 γ-aminobutyric acid 생산 증진)

  • ${\gamma}$-aminobutyric acid (GABA) is a non-proteinogenic amino acid biosynthesized through decarboxylation of L-glutamic acid by glutamic acid decarboxylase. GABA is believed to play a role in defense against stress in plants. In humans, it is known as one of the major inhibitory neurotransmitters in the central nervous system, exerting anti-hypertensive and anti-diabetic effects. In this report, we wanted to enhance the GABA production from the barley leaf and corn silk by culturing them with lactic acid bacteria (LAB). The barley leaf and corn silk were mixed with various weight combinations and were fermented with Lactobacillus plantarum in an incubator at $30^{\circ}C$ for 48 h. After extracting the fermented mixture with hot water, we evaluated the GABA production by thin layer chromatography and GABase assay. We found that the fermented mixture of the barley leaf and corn silk in a nine to one ratio contained a higher level of GABA than other ratios, meaning that the intermixture and fermentation technique was effective in increasing the GABA content. We also tested several biological activities of the fermented extracts and found that the extracts of the fermented mixture showed improved antioxidant activities than the non-fermented extracts and no indication of cytotoxicity. These results suggest that our approach on combining the barley leaf and corn silk and its fermentation with LAB could lead to the possibility of the development of functional foods with high levels of GABA content and improved biological activities.

Enhancement of Skin Immune Activities of Curcuma longa L. Leaf Extract by Ultra High Pressure Process (초고압 공정을 이용한 강황 잎 추출물의 피부 면역 활성 증진)

  • Curcuma longa L. leaf was extracted by water at $60^{\circ}C$ for 12 hours after being treatment of Ultra High Pressure under 500 MPa for 5-15 minute. The high pressure extraction for 15 minute (HPE15) was measured the highest extraction yield of 25.48% (w/w), compared to those from conventional extraction methods. The HPE15 showed the lowest cytotoxicity as 11.97% in adding $1.0mg/m{\ell}$ of concentration. Also, HPE15 was measured the highest inhibition of hyaluronidase as 44.48% in adding $1.0mg/m{\ell}$. In addition, The production of NO from macrophages was measured as $7.06{\mu}M$ in adding $1.0mg/m{\ell}$ of HPE15, which was lower than the those from others processes. Finally, HPE15 significantly reduced up to $649.44pg/m{\ell}$ of $ProstaglandinE_2$ production from UV-irradiation. These results suggest that the Curcuma longa Linne leaf extract from high pressure process might enhance the skin immune activities possibly by high elution of active components than other processes.

  • A three year's field experiment was carried out on newly reclaimed tidal saline soils to evaluate the salt tolerance and growht characteristics, and their relationship to dry matter production and nutrient quality of main selected pasture species. Nine temperate grasses (14 varieties) and two forage crops (sorghum and pearl millet) were grown under different mulching treatments with medium sand and red-yellow soils (fine loamy materials of Typic Hapludults) from 1986 to 1988. Tall wheatgrass, tall fescue, reed canarygrass and alfalfa showed a good tolerance to soil salinity, especially tall wheatgrass (cv. Alkar) produced 19.6 ton/ha dry matter yield annualy under mulching treatment with medium sand depth in lcm. Pearl millet (cv. Gahi-3) was also evaluated as a salt tolerable forage species. Under salt stress in newly reclaimed tidal lands, plant showed a decrease in the assimirable leaf area (LA) as well as specific leaf area (SP. LA) and a low leaf weight ratio(LWR), and it resulted in a low concentration of crude protein and low digestible dry matter contents. Absorption of macro and micro elements in the plant on tidal lands was increased markedly.

Protective effects of quercetin-3-glucosyl-(1-2)-rhamnoside from Schizophragma hydrangeoides leaves on ultraviolet A-induced photoaging in human dermal fibroblasts

  • Schizophragma hydrangeoides (S. hydrangeoides) is a vine endogenous to Jeju Island and Ulleungdo, where it grows attached to the foothills and rock surfaces. Previous research has mostly focused on the whitening effect of S. hydrangeoides leaf extract. In this study, we investigated S. hydrangeoides leaf extract further, and detected four phytochemicals in the extract: chlorogenic acid, quercetin-3-O-glucosyl-(1-2)-rhamnoside, quercetin-3-O-xylosyl-(1-2)-rhamnoside, and quercitrin. We pretreated human dermal fibroblast (HDFn) cells with previously established concentrations of the four compounds for 1 h before ultraviolet A (UVA) irradiation. Among the four compounds, quercetin-3-O-glucosyl-(1-2)-rhamnoside (Q-3-GR) best inhibited matrix metalloproteinase-1 (MMP-1) levels. Thus, we investigated the protective effects of Q-3-GR on photoaging and its underlying mechanisms. Q-3-GR significantly reduced MMP-1 production and inhibited MMP-1 protein expression in UVA-irradiated HDFn cells. Furthermore, Q-3-GR increased procollagen type I production and protein expression. Q-3-GR exerted its anti-photoaging effects by downregulating the mitogen-activated protein kinase/ activator protein-1 signaling pathway, and upregulating the transforming growth factor-β/Smad signaling pathway. Thus, S. hydrangeoides leaf-derived Q-3-GR is a potential potent cosmetic ingredient for UV-induced skin aging.
