PEG 스페이서를 통해 Homing 펩타이드를 고정화한 산화철 나노입자의 제조 및 생의학적 응용 (Synthesis of Homing Peptide-Immobilized Magnetite Nanoparticles through PEG Spacer and Their Biomedical Applications)

  • 산화철($Fe_3O_4$은 세포에 의해 섭취된 후 대사반응에 의해 분비되므로 세포독성을 나타내지 않는다. 따라서 산화철 나노입자는 MRI 촬영을 하기에 앞서 조영제로서 널리 사용되고 있다. 본 연구에서는 통상의 공침법으로 산화철 나노입자를 합성하고 폴리에틸렌글리콜을 스페이서로 하여 혈관내피세포 및 방광암 세포막의 IL-4 리셉터에 특이적으로 반응하는 homing 펩타이드(AP)를 고정화하였다. AP를 고정화한 산화철 나노입자의 크기는 수용액 상에서 약 39 nm이었다. 섬유아세포 및 방광암세포를 이용하여 AP고정화 산화철 나노입자의 uptake를 조사한 결과 섬유아세포에는 선택적 uptake를 발견할 수 없었으나 방광암세포에는 선택적으로 uptake됨을 알 수 있었다. 따라서 AP 고정화 산화철 나노입자는 조기 암진단용 조영제로서 가능성을 지니고 있다고 할 수 있다.

  • Iron oxide nanoparticles excite researcher interest in biomedical applications due to their low cost, biocompatibility and superparamagnetism properties. Magnetic iron oxide especially magnetite ($Fe_3O_4$) possessed a superparamagnetic behaviour at certain nanosize which beneficial for drug and gene delivery, diagnosis and imaging. The properties of nanoparticles mainly depend on their synthesis procedure. There has been a massive effort in developing the best synthetic strategies to yield appropriate physico-chemical properties namely co-precipitation, thermal decomposition, microemulsions, hydrothermal and sol-gel. In this review, it is discovered that magnetite nanoparticles are best yielded by co-precipitation method owing to their simplicity and large production. However, its magnetic saturation is within range of 70-80 emu/g which is lower than thermal decomposition and hydrothermal methods (80-90 emu/g) at 100 nm. Dimension wise, less than 100 nm is produced by co-precipitation method at $70^{\circ}C-80^{\circ}C$ while thermal decomposition and hydrothermal methods could produce less than 50 nm but at very high temperature ranging between $200^{\circ}C$ and $300^{\circ}C$. Thus, co-precipitation is the optimum method for pre-compliance magnetite nanoparticles preparation (e.g., 100 nm is fit enough for biomedical applications) since thermal decomposition and hydrothermal required more sophisticated facilities.

산화철 나노입자의 브라운 운동에 대한 자기장 의존성 연구 (Magnetic Field Dependence of Brownian Motion in Iron-oxide Nanoparticles)

  • 유기 용매에 고르게 분산되어 있는 26 nm 크기의 산화철 나노입자를 사용하여 주파수에 따른 교류 자화율을 측정하였다. 자기장이 없는 조건에서 측정한 나노입자의 자화율은 Debye 완화 모델로 계산한 결과와 일치하였으며, 완화 주파수(relaxation frequency)는 370 Hz였다. 나노입자의 완화 주파수는 용매의 점성에 의한 브라운 운동(Brownian motion)의 완화 시간과 일치하였다. 브라운 운동에 의한 나노입자의 완화 주파수는 자기장의 세기에 따라 선형적으로 증가하는 특성을 보였다.

One-pot 합성 방법을 이용한 나노 철입자가 담지된 폐목재 기반 하이드로차의 제조 및 특성 평가 (Preparation of Iron Nanoparticles Impregnated Hydrochar from Lignocellulosic Waste using One-pot Synthetic Method and Its Characteristics)

  • In this study, iron nanoparticles impregnated hydrochar (FeNPs@HC) was synthesized using lignocellulosic waste and simple one-pot synthetic method. During hydrothermal carbonization (HTC) process, the mixture of lignocellulosic waste and ferric nitrate (0.1~0.5 M) as a precursor of iron nanoparticles was added and heated to 220℃ for 3 h in a teflon sealed autoclave, followed by calcination at 600℃ in N2 atmosphere for 1 h. For the characterization of the as-prepared materials, X-ray diffraction (XRD), cation exchange capacity (CEC), fourier transform infrared spectrometer (FT-IR), Brunauer-Emmett-Teller (BET), transmission electron microscope (TEM), Energy Dispersive X-ray Spectroscopy (EDS) were used. The change of Fe(III) concentration in the feedstock influenced characteristics of produced FeNPs@HC and removal efficiency towards As(V) and Pb(II). According to the Langmuir isotherm test, maximum As(V) and Pb(II) adsorption capacity of Fe0.25NPs@HC were found to be 11.81 and 116.28 mg/g respectively. The results of this study suggest that FeNPs@HC can be potentially used as an adsorbent or soil amendment for remediation of groundwater or soil contaminated with arsenic and cation heavy metals.

하이퍼써미아 응용을 위한 하이브리드 에어로젤 내 분산된 마그네타이트 나노입자 (Magnetite Nanoparticles Dispersed in Hybrid Aerogel for Hyperthermia Application)

  • Magnetite nanoparticles(NPs) have been the subject of much interest by researchers owing to their potential use as magnetic carriers in drug targeting and as a tumor treatment in cases of hyperthermia. However, magnetite nanoparticles with 10 nm in diameter easily aggregate and thus create large secondary particles. To disperse magnetite nanoparticles, this study proposes the infiltration of magnetite nanoparticles into hybrid silica aerogels. The feasible dispersion of magnetite is necessary to target tumor cells and to treat hyperthermia. Magnetite NPs have been synthesized by coprecipitation, hydrothermal and thermal decomposition methods. In particular, monodisperse magnetite NPs are known to be produced by the thermal decomposition of iron oleate. In this study, we thermally decomposed iron acetylacetonate in the presence of oleic acid, oleylamine and 1,2 hexadecanediol. We also attempted to disperse magnetite NPs within a mesoporous aerogels. Methyltriethoxysilicate(MTEOS)-based hybrid silica aerogels were synthesized by a supercritical drying method. To incorporate the magnetite nanoparticles into the hybrid aerogels, we devised two methods: adding the synthesized aerogel into a magnetite precursor solution followed by nucleation and crystal growth within the pores of the aerogels, and the infiltration of magnetite nanoparticles synthesized beforehand into aerogel matrices by immersing the aerogels in a magnetite nanoparticle colloid solution. An analysis using a vibrating sample magnetometer showed that approximately 20% of the magnetite nanoparticles were well dispersed in the aerogels. The composite samples showed that heating under an inductive magnetic field to a temperature of $45^{\circ}C$ is possible.

  • Superparamagnetic iron oxide nanoparticles (SPIONs) have been prepared without using surfactants to assess their stability at different time intervals. The synthesized particles were characterized by X-ray diffraction, Fourier-transform infrared spectroscopy, ultraviolet-visible-near infrared spectroscopy, and energy dispersive spectroscopy. Field emission scanning electron microscopy and high-resolution transmission electron microscopy images of the samples were also investigated. The average particle size was measured to be 12.7 nm even in the polydispersed form. The magnetic and dielectric characteristics of the $Fe_3O_4$ nanoparticles have also been studied and discussed in detail.

  • A new and simple method has been developed to synthesize highly crystalline and monodisperse maghemite (γ-Fe₂O₃) and zirconia (ZrO₂) nanocristallites. High temperature aging of metal-surfactant complex was founded to generate monodisperse nanoparticles, wherein the nuclei were prepared by the thermal decomposition of iron-oleate complex in case of iron oxide and nonhydrolytic sol-gel reaction in case of zirconia respectively. By varying the experimental conditions, in other words concentration of surfactants, kind of metal precursor, reaction temperature and so on, the diameter of spherical nanoparticles could be controlled at various size. The synthesized nanoparticles were characterized by electron diffraction, X-ray diffraction, and low- and high-resolution transmission electron microscope.

Fabrication and Characterization of Immiscible Fe-Cu Alloys using Electrical Explosion of Wire in Liquid

  • Iron and copper are practically immiscible in the equilibrium state, even though their atomic radii are similar. As non-equilibrium solid solutions, the metastable Fe-Cu alloys can be synthesized using special methods, such as rapid quenching, vapor deposition, sputtering, ion-beam mixing, and mechanical alloying. The complexity of these methods (multiple steps, low productivity, high cost, and non-eco-friendliness) is a hinderance for their industrial applications. Electrical explosion of wire (EEW) is a well-known and effective method for the synthesis of metallic and alloy nanoparticles, and fabrication using the EEW is a simple and economic process. Therefore, it can be potentially employed to circumvent this problem. In this work, we propose the synthesis of Fe-Cu nanoparticles using EEW in a suitable solution. The powder shape, size distribution, and alloying state are analyzed and discussed according to the conditions of the EEW.

Photo-triggered Theranostic Nanoparticles in Cancer Therapy

  • In cancer therapy, it is often desirable to use precision medicine that involves treatments of high specificity. One such treatment is the use of photo-triggered theranostic nanoparticles. These nanoparticles make it possible to visualize and treat tumors specifically in a controlled manner with a single injection. Several novel and powerful photo-triggered theranostic nanoparticles have been developed. These range from small organic dyes, semiconducting and biopolymers, to inorganic nanomaterials such as iron-oxide or gold nanoparticles, carbon nanotubes, and upconversion nanoparticles. Using photo-triggered theranostic nanoparticles and localized irradiation, complete tumor ablation can be achieved without causing significant toxicity to normal tissue. Given the great advances and promising future of theranostic nanoparticles, this review highlights the progress that has been made in the past couple of years, the current challenges faced and offers a future perspective.

Removal of Aqueous Cr(VI) using Magnetite Nanoparticles Synthesized from a Low Grade Iron Ore

  • We demonstrated the efficacy of magnetic nanoparticles (MNPs) produced from a low grade iron ore as an adsorbent for the removal of Cr(VI), a toxic heavy metal anion present in wastewater. The adsorption of Cr(VI) by these MNPs strongly depended on the dosage of MNPs, the initial concentration of the Cr(VI) solutions, and pH. The highest Cr(VI) adsorption efficiency of 22.0 mg/g was observed at pH 2.5. The adsorption data were best fit with the Langmuir isotherm and corresponded to a pseudo-second-order kinetic model. The used adsorbent was regenerated by eluting in highly alkaline solutions. Sodium bicarbonate showed the highest desorption efficiency of 83.1% among various eluents including NaOH, $Na_2HPO_4$, and $Na_2CO_3$. Due to the high adsorption capacity, the simple magnetic separation, and the high desorption efficiency, this nano-adsorbent produced from inexpensive and abundant resources may attract the attention of the industries to apply for removing various metal anionic contaminants from wastewater.
