  • The production cost of biodiesel from microalgae is still not competitive, compared with that of petroleum fuels. The genetic improvement of microalgal strains to increase triacylglycerol (TAG) accumulation is one way to reduce production costs. One of the most promising approaches is the isolation of starch-deficient mutants, which have been reported to successfully increase TAG yields. To date, such a stable mutant is not available in an oleaginous marine microalga, despite several advantages of using marine species for biodiesel production. Algae in the genus Dunaliella are known to tolerate high salt concentration and other environmental stresses. In addition, the cultivation processes for large-scale outdoor commercialization have been well established for this genus. In this study, Dunaliella tertiolecta was used to screen for starch-deficient mutants, using an iodine vapor-staining method. Four out of 20,016 UV-mutagenized strains showed a substantial reduction of starch content. A significantly higher TAG content, up to 3-fold of the wild-type level, was observed in three of the mutants upon induction by nitrogen depletion. The carotenoid production and growth characteristics of these mutants, under both normal and oxidative stress conditions, were not compromised, suggesting that these processes are not necessarily affected by starch deficiency. The results from this work open up new possibilities for exploring Dunaliella for biodiesel production.

  • This study aimed to obtain equilibrium concentration on adsorption removal of phosphorus by activated carbon, to express the adsorption characteristics following Freundlich isotherm and also, based on the value obtained, to investigate the relationship between physical properties of activated carbon and dynamics of phosphorus removal by obtaining rate constant and effective pore diffusivity. The results summarized from this study are as follows. Phosphorus adsorption equilibrium reaching time of powdered activated carbon was reduced as the dosage of activated carbon increases, while granular activated carbon despite increased dosage did not have influence on adsorption equilibrium reaching times of phosphorus as well, taking more than 10 hours. It was also noted that powdered activated carbon showed better adsorption ability than granular activated carbon. The value of constant (f) of Freundlich isotherm of powered activated carbon on phosphorus was 4.26 which is bigger than those of granular activated carbon. The adsorption rate constant on phosphorus of powered activated carbon with low effective diameter and iodine number was highest as $8.888hr^{-1}$ and the effective pore diffusivity ($D_e$) was lowest as $2.45{\times}10^{-5}cm^2/hr$, and the value of phosphorus adsorption rate constant of granular activated carbon was $0.174{\sim}0.372hr^{-1}$, It was revealed that, with the same amount of dosage, the adsorptive power of activated carbon with lower effective diameter was better and its rate constant was also high.

  • In the fish oil forced oxidized at 6$0^{\circ}C$ for 10 days, changes in the levels of peroxide (POV), anisidine (AnV), total oxidation (Totox), iodine (IV), acid (AV) and fatty acids composition were measured. The levels of POV, AnV and Totox remained unchanged or decreased after reaching the maximum. The concentrations of polyunsaturated fatty acids (PUFA) such as Docosa hexaenoic acid (DHA) or Eicosa pentaenoic acid (EPA) decreased with extended oxidation of fish oil. In saturated fatty acids (SFA) like C16:0, their concentration increased with decreasing PUFA. The ratios of PUFA/SFA and DHA/C16:0 decreased with extended oxidation of fish oil. Using a single parameter of POV, AnV, Totox, AV, IV, or fatty acids for evaluation of the quality of fish oil may prove difficult. Besides other parameters, the ratios of PUFA/SFA and/or DHA/C16:0 could be a good index to evaluate the quality of fish oil.

  • Although soft tunic syndrome (STS) in the ascidian is a serious disease, helpful measures have yet not been established. It was examined in this study by applying aniti-parasitic drugs to eradicate the causative protozoa Azumiobodo hoyamushi from infected ascidians. Formalin was synergistic in killing parasites in vitro when co-treated with hydrogen peroxide ($H_2O_2$) or bronopol, but not with chloramine-T or povidone-iodine (PVP-I), when tested with in vitro parasite culture. The synergistic effects did not change when $formalin-H_2O_2$ (or bronopol) ratios were changed. It was found that treatment periods less than 60 min achieved a sub-maximal efficacy. Increasing drug concentration while keeping 30 min period improved anti-parasitic effects. Anti-parasitic effects of $formalin(F)+H_2O_2$(H) were also assessed in an in vivo STS model infected with cultured parasites. It was observed that combined 50 (40F + 10H) and 100 (80F +20H) ppm were effective in partially preventing STS-caused mortality. In horizontally transmitted artificial STS model, significant prevention of ascidian mortality was also observed after 50 ppm. Marked reduction of living parasites were noted after drug treatments in vivo. The results provide a highly useful basis to develop a preventive or treatment measure against the currently uncontrollable STS in the ascidian.

  • Peroxide values (PV) of fat from fallow up formula were determined using redox-potentiometric titration method and standard method, which is based on KI oxidation by hydroperoxides and volumetric titration of liberated iodine, and their uncertainties were compared. Uncertainty sources in measurement, such as sample weight, sodium thiosulfate concentration, and titer, were identified and used as parameters for combined standard uncertainty based on Guide to the expression of uncertainty in measurement and Draft EURACHEM/CITAC Guide. Analytical results and combined standard uncertainties of peroxide values (PV) determined by standard burette and potentiometric titrations were $2.05{\pm}0.17\;and\;1.96{\pm}0.07\;meq/kg$, respectively, suggesting potentiometric titration method is suitable for determining PV of fat with low PV, because uncertainty of PV determination obtained by potentiometric titration was lower than that obtained by burette titration.

  • $\alpha$-Amylase producing bacteria were isolated from activated sludge of corn processing wastewater plant and paddy field soil samples and selected by the direct iodine reaction. The isolate was identified as Bacillus sp. after morphology, API system and fatty acid analyses. To enchance $\alpha$-amylase productivity, a successive mutation of Bacillus sp. AIV 19 was performed using the treatment of nitrosoguanidine(NTG).The mutant, Bacillus sp. AIV 1940, showed about 1.8-fold level of amylase activity compared with parental strain. The isolate was Gram-positive and rod (2.8-3.0 $\mu$m long, 0.5-0.6 $\mu$m wide) type. The strain increased the bacterial mass at 3000 mg/l starch concentration. Organic substance removal rate was 40.2, 72.3% respectively after 1 and 3 day reaction using starch synthetic wastewater (intial CODcr was 4,455 mg/l).

  • The methods for determining C-14 and tritium contents in the spent nuclear fuel sample were developed. The carbon-14($^{14}CO_2$) released during the dissolution of the spent fuel sample and $CaCO_3$ ($CO_2$ carrier) with 8 M $HNO_3$ at $90^{\circ}C$ was collected in trap containing 1.5 M NaOH. The volatile radioactive iodine evolved when the spent fuel was dissolved, was trapped on to Ag-silicagel (Ag-impregnated silicagel) adsorbent in column which is connected to two NaOH traps. The solutions which contain tritium as HTO after fuel dissolution were decontaminated by deionization with a mixture of cation and anion exchange resins and inorganic ionexchangers. The amount of C-14 in the trap solutions and the HTO concentration in the resulting deionization water were then determined by liquid scintillation counting.

  • In this study, the changes in collection efficiencies due to the time changes of activated carbons were ascertained, and in order to identify the magnitude of adsorption, the before-use and after-use iodine adsorption values were analyzed. In addition, as a result of examining the characteristics of continuous process and non-continuous process and as a result of investigating whether the emission standards would be maintained, the continuous process and printing facilities were seen as not being able to maintain the emission standards. Also were found, in the case of non-continuous process,-taking into consideration the special nature of the job -for 4${\o}$ palletized charcoal, a collection efficiency near 50% was shown even after 96 hours. Also, when the inlet concentration was about 300ppm, it is thought that the emission standards would be maintained if the activated carbons are replaced within at least 96 hours in the case of 4${\o}$ palletized charcoal and the use was deemed pointless in the case of carbon. The results of this study are expected to provide assistance in selecting replacement periods for activated carbons and in selecting absorbents at the project sites, and are expected to be of significant help in the selection of precipitators that can collect total hydrocarbons for compliance of the emission standards.

  • In this study, we have investigated the removal of the low level radioactive ions of Cs and I in water by the reverse osmosis (RO) process. The two RO modules produced in domestic region and the waste RO module after the cleaning process were selected. Then we compared removal performance of both Cs and I. The experiments are conducted by varying the concentration of feed, the pressure. As a results, it was confirmed that all three modules are higher I decontamination factor than Cs. And particularly, for the cleaned RO module, its decontamination factor of I was 1140. Since the results at low pressure condition were better than that at high pressure conditions, the use of the direct installation of RO modules on the tap water might be possible. In addition, it was confirmed that the waste RO module after cleaning process using EDTA, SBS and NaOH, increased the decontamination performance better than before cleaning, in particular, the recovery ratio after cleaning was 6.3% higher.

  • The Sulfur-Iodine thermochemical hydrogen production process (SI process) consists of the Bunsen reaction section, the $H_2SO_4$ decomposition section, and the HI decomposition section. The $HI_x$ solution ($I_2-HI-H_2O$) could be recycled to Bunsen reaction section from the HI decomposition section in the operation of the integrated SI process. The phase separation characteristic of the Bunsen reaction using the $HI_x$ solution was similar to that of $I_2-H_2O-SO_2$ system. On the other hands, the amount of produced $H_2SO_4$ phase was small. To investigate the effects of $SO_2$ solubility on Bunsen reaction, the continuous Bunsen reaction was performed at variation of the amounts of $SO_2$ gas. Also, it was carried out to make sure of the effects of partial pressure of $SO_2$ in the condition of 3bar of $SO_2-O_2$ atmosphere. As the results, the characteristic of Bunsen reaction was improved with increasing the amounts and solubility of $SO_2$ gas. The concentration of Bunsen products was changed by reverse Bunsen reaction and evaporation of HI after 12 h.
