Effects of Monovalent Cations on the βReaction Kinetics of Tryptophan Synthase (트립토판 합성효소의 β반응속도에 미치는 일가양이온의 영향)

  • Effects of monovalent cations were examined on the fast $\beta$reaction of $\alpha$D56N and $\alpha$D56G mutant tryptophan synthase. Reaction rates for the production and degradation of intermediates in the reaction were changed in the presence of cathons. The mutant proteins showed different reaction rates from those of wild-type protein, and additional changes occurred in the presence of cations. The results showed that monovalent cations and $\alpha$D56 are important in allosteric properties of this protein.

Appraisal of re-irradiation for the recurrent glioblastoma in the era of MGMT promotor methylation

  • Despite recent innovation in treatment techniques and subsequently improved outcomes, the majority of glioblastoma (GBL) have relapses, especially in locoregional areas. Local re-irradiation (re-RT) has been established as a feasible option for recurrent GBL of all ages with safety, tolerability, and effectiveness both in survival and quality of life regardless of fractionation schedule. To keep adverse effects under acceptable range, cumulative dose limit in equivalent dose at 2 Gy fractions by the linear-quadratic model at α/β = 2 for normal brain tissue (EQD2) with narrow margin should be observed and single/hypofractionated re-RT should be undertaken very carefully to recurrent tumor with large volume or adjacent to the brainstem. Promising outcome of re-operation (re-Op) plus re-RT (re-Op/RT) need to be validated and result from re-RT with temozolomide/bevacizumab (TMZ/BV) or new strategy is expected. Development of new-concept prognostic scoring or risk group is required to select patients properly and make use of predictive biomarkers such as O(6)-methylguanine-DNA methyltransferase (MGMT) promotor methylation that influence outcomes of re-RT, re-Op/RT, or re-RT with TMZ/BV.

Characterization of a Glycoside Hydrolase Family 50 Thermostable β-agarase AgrA from Marine Bacteria Agarivorans sp. AG17

  • An agar-degrading Agarivorans sp. AG17 strain was isolated from the red seaweed Grateloupia filicina collected from Jeju Island. A beta-agarase gene from Agarivorans sp. AG17 was cloned and designated as agrA. agrA has a 2,985 bp coding region encoding 995 amino acids and was classified into the glycoside hydrolase family (GHF)-50. Predicted molecular mass of the mature protein was 105 kDa. His-tagged agrA was overexpressed in Escherichia coli and purified as a fusion protein. The enzyme showed 158.8 unit/mg specific activity (optimum temperature at $65^{\circ}C$ and pH 5.5 in acetate buffer) with unique biochemical properties (high thermal and pH stabilities). Enzyme produced neoagarohexaose, neoagarotetraose and neoagarobiose by degrading agar, and hydrolyzed neoagaro-oligosaccharides were biologically active. Hence the purified enzyme has potential for use in industrial applications such as the development of cosmetics and pharmaceuticals.

Immune Regulatory Effect of Newly Isolated Lactobacillus delbrueckii from Indian Traditional Yogurt

  • Lactic acid bacteria (LAB) are microorganisms that are believed to provide health benefits. Here, we isolated LAB from Indian fermented foods, such as traditional Yogurt and Dosa. LAB from Yogurt most significantly induced TNF-α and IL-1β production, whereas LAB from Dosa induced mild cytokine production. After 16S rRNA gene sequencing and phylogenetic analysis, a Yogurt-borne lactic acid bacterium was identified and classified as Lactobacillus delbrueckii subsp. bulgaricus, and it was renamed L. delbrueckii K552 for the further studies. Our data suggest that the newly isolated L. delbrueckii can be used for the treatment of immune deficiency disorders.

Effect of Theobroma cacao L. Extracts on Mouse Spleen Immune Cells Activation (카카오 열수 추출물 투여의 마우스 면역세포 활성효과)

  • Theobroma cacao L., a fruit of cacao trees, is a perennial plant, which belongs to Sterculiaceae, and is native to the Amazon in South Africa. It also has been known for its various biologically active effects, such as anti-oxidation, anti-cancer, and anti-bacterial. The spleen cell proliferations of mice were measured at 48 hours after treatment of Theobroma cacao L. water extracts in seven concentrations(0, 5, 10, 50, 100, 250, 500 and 1,000 ㎍/mL) an ELISA assay. The production of cytokine (IL-1β, TNF-α, IFN-γ), is secreted by macrophages stimulated with LPS, was detected by ELISA assay using the cytokine kit. From the results of in vitro study, both splenocytes and cytokine production activated by peritoneal macrophages have increased when water extracts were supplemented in the range between 250 and 500 ㎍/mL concentration. Notably, splenocytes production has a signigicant proliferation at 500 ㎍/mL concentration. The result from this research suggests that supplementation with Theobroma cacao L. water extracts may enhance the immune function by stimulating the splenocyte proliferation and improving the cytokine production activating macrophage in vitro.

Physicochemical Characteristics and Antioxidant Activity of Cereal Bars Containing Apple Pomace (사과박 첨가 시리얼바의 이화학적 특성 및 항산화 효과)

  • This study aimed to investigate the physicochemical characteristics and antioxidant activity of cereal bars containing various concentrations (0, 10, 20, or 30%) of apple pomace (AP). Highest vitamin B1 and C contents were observed in 30% AP-containing bars, but vitamin B2 and B3 contents were highest in the control (0.347 and 3.566 mg/100 g, respectively). Cutting strength decreased significantly in an AP concentration-dependent manner. Total polyphenol and flavonoid contents in 10% AP bars were 2.949±0.157 mg of GAE/g and 1.001±0.009 mg of CE/g, respectively. Antioxidant activities were assessed by measuring free radical scavenging activities, and 10% AP bars had the highest activities, as determined by ABTS (2,2'azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) and DPPH (α-α-diphenyl-β-picrylhydrazyl) assays (0.412±0.008 and 0.492±0.021 mg GAE/g, respectively). These results suggested that apple pomace should be regarded a potential nutritional and antioxidant source.

Improvement of Anti-Inflammation Activity of Gardeniae fructus Extract by the Treatment of β-Glucosidase (β-Glucosidase 처리에 의한 치자추출물의 항염증 활성 증진)

  • In this study, we selected Gardeniae fructus (GF) as an anti-inflammatory functional material and improved the biological activity of GF through the treatment of ${\beta}$-glucosidase. For the simple evaluation of anti-inflammatory activity, the inhibitory activity of GF extract (GFE) on the production of NO by RAW264.7 cells in the presence of LPS was examined. ${\beta}$-glucosidase originating from Aspergillus niger or Aspergillus fumigatus has effectively improved the anti-inflammatory activity of GFE. The enzyme treatment raised the activity of GFE by more than 10 times. The optimum conditions for the enzyme reaction were at pH 4.6, $45^{\circ}C$, and 20 U/mL for 24 h with agitation. In addition, in vitro production of cytokines (IL-$1{\beta}$, IL-6, TNF-${\alpha}$), COX-2, and the NF-${\kappa}B$ activation of RAW264.7 cells decreased more in the presence of GFE treated with ${\beta}$-glucosidase originating from Aspergillus niger (GFAN) than in the presence of GFE. These results suggest that enzyme-treated GFE might be a potential candidate for natural anti-inflammatory food materials.

Ethyl acetate fraction of GGEx18 modulates fatty acid β-oxidizing enzymes (In vitro 동물세포에서 GGEx18의 ethyl acetate 분획물에 의한 지방산 β-산화효소 유전자 발현의 조절)

  • Objectives : This study was undertaken to investigate the effects of the GGEx18 ethyl acetate fraction (EF) on lipid accumulation and gene expression of fatty acid-oxidizing enzymes using 3T3-L1 adipocytes, C2C12 skeletal muscle cells, and NMu2Li liver cells. Methods : PPAR${\alpha}$, AMPK and UCPs transactivation was examined in NMu2Li hepatocytes, C2C12 myocytes, and 3T3-L1 preadipocytes using transient transfection assays. Results : 1. Compared with control, EF significantly increased the mRNA expression of VLCAD in 3T3-L1 adipocytes. 2. Compared with control, EF (0.1 ${\mu}g/ml$) significantly inhibited lipid accumulation in 3T3-L1 adipocytes. 3. EF significantly increased the mRNA expression of AMPK${\alpha}$1, AMPK${\alpha}$2 and PPAR${\alpha}$ in C2C12 skeletal muscle cells compared with control. 4. EF significantly increased the mRNA expression of genes involved in fatty acid ${\beta}$-oxidation, such as thiolase, MCAD, and CPT-1 in C2C12 skeletal muscle cells compared with control. 5. EF significantly increased the mRNA expression of UCP2 involved in energy expenditure in C2C12 skeletal muscle cells compared with control. 6. Compared with control, EF (10 ${\mu}g/ml$) significantly inhibited lipid accumulation in C2C12 skeletal muscle cells. 7. EF (10 ${\mu}g/ml$) significantly increased the mRNA expression of ACOX, HD, VLCAD and MCAD in NMu2Li liver cells compared with control. Conclusions : These results suggest that EF may prevent obesity by increasing the mRNA expression of mitochondrial fatty acid ${\beta}$-oxidizing enzymes in 3T3-L1 adipocytes, by not only regulating the fatty acid oxidation through activation of AMPK and PPAR${\alpha}$, but also increasing the UCP2 mRNA expression in C2C12 skeletal muscle cells, and by stimulating the mRNA expression of fatty acid-oxidizing enzymes in NMu2Li liver cells.

Effect of β-glucan on immune parameters in the Manila clam Ruditapes philippinarum (β-glucan이 바지락의 면역력에 미치는 영향)

  • ${\beta}$-Glucan is a polysaccharide that is widely used as an adductive in fish feed to facilitate immune stimulation. This study aimed to investigate the effect of ${\beta}$-glucan on immune responses in the Manila clam Ruditapes philippinarum. For this purpose, three groups of R. philippinarum were exposed to 0%, 0.1%, or 1% ${\beta}$-glucan in sea water for 1 hr/day for 2 weeks using an immersion method. Thereafter, two immune parameters-phagocytic rate and antibacterial activity-were measured. R. philippinarum exposed to 1% ${\beta}$-glucan showed an approximate 30% significant increase in phagocytic rate. In addition, ${\beta}$-glucan significantly limited the growth of the pathogenic bacteria Vibrio tapetis, V. parahaemolyticus, and V. ordalii. Moreover, the mortality rates of ${\beta}$-glucan-treated clams decreased during a 17-day experiment. Our study suggests that treatment with ${\beta}$-glucan significantly increases the immune responses in R. philippinarum, and that immersion is a simple and effective method for immune stimulation in this species.

Study of Innate Immunity Suppression of Yeonsan Ogye listed on Dong-eui-bo-gam (동의보감에 수재된 오계(烏鷄)의 선천면역반응 억제에 관한 연구)

  • The aim of the study is to evaluate immune-enhancing effects of Yeonsan Ogye. Various extract of Yeonsan Ogye (200 and 400 mg/kg/daily) was treated orally to Balb/c mice for 1 week, before acute inflammation was induced by LPS. After cytokine (IFN-γ, TNF-α, IL-6, and IL-1β) and immune cells (white blood cell, neutrophil, lymphocyte, and monocyte) level by serum and blood were counted. As a result, Oral treatment of Yeonsan Ogye extract to the Balb/c mice were significantly decreased cytokine level in serum, in comparison with control group. in addition, production of white blood cell and monocyte in blood was decreased and granulocyte was increased respectively, in comparison with control. Our results demonstrated that Yeonsan Ogye extracts seem to have significant immune-enhancing. Thus, Yeonsan ogye may be developed as a raw material for new health food and medicine to ease the symptoms related with inflammatory and immune.
