  • 강황(Curcuma longa)으로부터 bisdemethoxycurcumin (BDMC), demethoxycurcumin (DMC) 및 curcumin의 생물 활성을 offline-ABTS 측정법과 on-line screening high-performance liquid chromatography (HPLC)-ABTS 측정법을 적용한 빠른 스크리닝을 통해 정량 및 성분 분리를 하였다. 이때, off-line-ABTS와 on-line screening HPLC-ABTS 비교는 미미한 오차를 보여주었다.

  • On-line screening high performance liquid chromatography (HPLC)-$ABTS^+$ assay를 이용한 청호로부터 chlorogenic acid, caffeic acid, vitexin, queretin, aremisinin의 항산화 활성을 온라인스크리닝 하였다. 이때 침적방법을 적용한 추출시간과 추출용매 조성으로부터 추출효율을 확인하였다. 이 결과 100% 물 추출물에서 수율이 가장 좋왔고 chloroenic acid의 항산화 활성이 가장 높았다. 또한 on-line screening HPLC-$ABTS^+$ assay 분석은 천연물에서 항산화 활성을 신속하게 탐색하는데 효율적이다.

  • Acer tegmentosum was a traditional Korean herbal medicine showing various pharmacological activities. In this work, A. tegmentosum were extracted with boiling water and then successively partitioned with dichloromethane, ethyl acetate, n-butyl alcohol (n-BuOH), and water. Salidoside, the target compound, was purified in n-BuOH phase using a chromatography method. For the analysis of salidoside, TLC and LC-MS were used as well as on-line screening $HPLC-ABTS^+$ assay with three different wavelength of 254, 280, and 320 nm. An amount of 1.34 g of salidoside were obtained in n-BuOH phase fromAcer tegmentosum was a traditional Korean herbal medicine showing various pharmacological activities.   • To determination of antioxidant substance homoorientin, from Phyllostachys bambusoides leaves, the ultrasonic extraction and HPLC on-line $ABTS^+$ screening method were empolyed. Also, the various experimental variables such as the frequency and time of ultrasonic system were investigated and homoorientin was extracted efficiently at the low frequency 35 kHz and the extraction time 60 min. The values were positive peak 1574.71 (relative area, 23.67%) and negative peak 6924.34 (relative area, 1.23%), respectively. This HPLC on-line $ABTS^+$ screening method was rapid and efficient to search for antioxidants from natural products. These results will provide a database for investigating the constituents of natural products and the resources of pharmaceutical and cosmetic products.

  • 한국산 녹차로부터 HPLC On-line $ABTS^{+}$ screening기법을 사용하여 catechin compounds의 특성중의 하나인 항산화활성을 빠르게 분석하였으며 녹차로부터 catechin compound의 추출을 다양한 온도와 시간의 추출방법을 적용하였다. 전 처리한 추출액에 포함된 catechin compounds을 분석하고 최적의 추출조건을 실험적으로 모색하였다. 실험결과에 의하면 추출온도 $60^{\circ}C$, 추출시간 3분으로 추출한 시료가 항산화활성이 가장 우수하였다.

  • 땅콩새싹은 많은 아시아 국가에서 오랜 시간 동안 식품으로 사용되어왔다. 그중에서 resveratrol은 항산화 및 항암 등의 생리 활성 효과가 있다. 본 연구는 resveratrol은 침적방법(추출시간 3h, $25^{\circ}C$)을 이용하여 추출하였다. 그리고 on-line screening HPLC-ABTS을 사용하여 추출 효율 및 함량을 확인 하였다. 추출액은 상업용 C18 역상 고성능액체 크로마토그래피에 의한 이동상으로 물과 아세토나이트릴을 사용하여 선형기울기용매법으로 분리 분석을 하였다. 유속은 1.0mL/min, 주입부피는 $10{\mu}L$, 컬럼오븐 온도 $40^{\circ}C$, 파장 320nm에서 수행되었다. 실험결과, 50% 에탄올에서 resveratrol 함량이($0.310-0.346{\mu}g/mL$) 100% 물과 에탄올 조성보다 높았다. 또한 100% 물 추출물의 수율은 2.040-2.145%로 낮았다. 위의 결과 on-line HPLC-ABTS 분석은 식물추출물의 생물활성을 빠르고 특화된 강력한 기술이 될 수 있음을 보여주었다. 이것은 제약 및 기능성 소재로서의 응용 가능성을 나타내었다.

  • GCSB-5 preparation is a purified extract from a mixture of 6 medicinal plants(Acanthopanacis Cortex, Achyranthis Radix, Saposhnikoviae Radix, Cibotii Rhizoma, Glycine Semen Nigra, Eucommiae Cortex) that have been widely used for the treatment of various bone disorders. The aim of this study was to investigate HPLC analysis method and screening of standard compound on Saposhnikoviae Radix for quality standardization of a medicinal crude drug GCSB-5. Standard compound of Saposhnikoviae Radix was decided with cimifugin by isolation and instrumental analysis such as NMR. HPLC analysis method for the simultaneous determination of cimifugin was established for the quality control of the medicinal plants of Saposhnikoviae Radix species, GCSB-5 raw material and preparation. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy following ICH guideline.

  • Sulfonamides, a therapeutically important group of antimicrobial drugs, are widely used to treat and prevent the acute systemic and skin infections in dairy cattle. They also pose an economic hazard through inhibition of growth of dairy starter cultures. This study was carried out to compare four screening methods for detection of sulfamethazine in milk and determine the positive milk sample by HPLC method. Sulfamethazine was used to spike at five levels of sulfamethazine. The Lac-Tek test and CharmII test were also consistent better than TTCII test and Delvo SP test in sulfamethazine detection. Analysis probabilities of obtaining a positive response with TTCII test and Delvo SP test assay at 50 ppb sulfamethazine level in milk samples were only 14%, 42% each. Whereas using the Lac-Tek test and CharmII test would have resulted in 100% identification of the five levels. Determination of sufamethazine using the HPLC method in the spiked milk were 10.64, 19.30, 30.76,38.83 and 50.23 ppb, respectively.

Simple Screening Method for Double-strand DNA Binders Using Hairpin DNA-modified Magnetic Beads

  • We designed an effective screening method for double strand DNA (dsDNA) binders using DNA-modified magnetic particles. Hairpin DNA was immobilized on the surface of magnetic particle for a simple screening of dsDNA binding materials in a solution containing various compounds. Through several magnetic separation and incubation processes, four DNA-binding materials, DAPI, 9AA, AQ2A, and DNR, were successfully screened from among five candidates. Efficiency of screening was demonstrated by HPLC analysis using a C2/18 reverse-phase column. In addition, their relative binding strengths were verified by measuring the melting temperature ($T_m$). If hairpin DNA sequence is modified for other uses, this magnetic bead-based approach can be applied as a high-throughput screening method for various functional materials such as anti-cancer drugs.
