왕모시풀의 페놀성 성분 및 항당뇨 활성 (Phenolic Constituents of Boehmeria pannosa and α-Glucosidase Inhibitory Activity)

  • Boehmeria pannosa (Urticaceae) is a perennial herb widely distributed in Korea. In this study, investigation on the constituents of B. pannosa was conducted by chromatographic techniques and spectroscopic analysis. As a result, nine compounds including eight phenolic compounds, 3-hydroxy-1-(4-hydroxyphenyl)propan-1-one (1), β-hydroxypropiovanillone (2), C-veratroylglycol (3), 3-hydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl)-propan-1-one (4) 2,3-dihydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl) -1-propanone (5), p-coumaric acid (6), 4-hydroxybenzoic acid (7), vanillic acid (8) and one lignan, (-)-(7R,8S)-dihydrodehydrodiconiferyl alcohol (9) were isolated from the EtOAc-soluble fraction of B. pannosa. Among them, compounds 5, 6 and 9 inhibited α-glucosidase inhibitory activity more than 50% at the concentration of 100 μM.

음나무 수피의 화학적 성분 (Chemical Components from the Stem Barks of Kalopanax septemlobus)

  • The stem barks of Kalopanax septemlobus were extracted with MeOH, and successively partitioned with $CH_2Cl_2$, EtOAc, BuOH and water. Repeated column chromatographic separation of the $CH_2Cl_2$ fraction resulted in the isolation of four compounds. Their structures were identified as ${\beta}-sitosterol$ (1), oleanolic acid (2), 3,3'-bis(3,4-dihydro-4-hydroxy-6-methoxy-2H-1-benzopyran) (3) and (-)-balanophonin (4). This is the first report on the isolation of 3,3'-bis(3,4-dihydro-4-hydroxy-6-methoxy-2H-1-benzopyran) (3) and (-)-balanophonin (4) from Kalopanax spp.

The Extract of Limonium tetragonum Protected Liver against Acute Alcohol Toxicity by Enhancing Ethanol Metabolism and Antioxidant Enzyme Activities

  • The protective effect of EtOAc fraction of Limonium tetragonum extract (EALT) against alcohol-induced hepatotoxicity was assessed following acute ethanol intoxication in Spraque-Dawley rats. EALT (200 mg/kg p.o.) was administrated once before alcohol intake (8 g/kg, p.o.). Blood ethanol concentration, and the activities of alcohol metabolic enzymes, alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) in the liver were measured. Also, the formation of malondialdehyde (MDA) and the activities of antioxidant enzymes, superoxide dismutase (SOD), glutathione peroxidase (GSH-px), catalase were determined after acute alcohol exposure. Pretreatment of rats received ethanol with EALT significantly decreased blood ethanol concentration and elevated the activities of ADH and ALDH in liver. The increased MDA level was decreased, and the reduced activities of SOD, GSH-px and catalase were markedly preserved by the treatment with EALT. This study suggests that EALT prevent hepatic injury induced by acute alcohol which is likely related to its modulation on the alcohol metabolism and antioxidant enzymes activities.

  • In the present study, we investigated the antioxidant activity of methanol extract and its organic fractions (n-hexane, $CH_2Cl_2$, EtOAc, n-BuOH and $H_2O$) of Capsiccum annuum leaves against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. As results, methanol extract, and its ethyl acetate and n-butanol fractions showed good scavenging effect at 100 ${\mu}g/ml$. Four compounds were isolated from the ethyl acetate fraction through silica gel and Sephadex LH-20 column chromatographies. Their chemical structures were elucidated as apigenin (1), vanillic acid (2), uracil (3) and apigenin $7-O-{\beta}-D-apiofuranosyl(1{\rightarrow}2)-{\beta}-D-glucopyranoside$ (4) by comparison of spectral data with those in reference. $IC_{50}$ value of compound 2 was 14.7 ${\mu}g/ml$, while compounds 1, 3 and 4 had no effect on the DPPH radical.

  • Repeated $SiO_2$ and octadecyl silica gel (ODS) column chromatographies of the EtOAc fraction from Magnolia obovata fruits, 10 neolignans, named magnolol (1), honokiol (2), isoobovatol (3), isomagnolol (4), obovatol (5), obovatal (6), 9-methoxyobovatol (7), magnobovatol (8), obovaaldehyde (9), and 2-hydroxyobovaaldehyde (10) were isolated and identified. All isolated compounds were evaluated for in vitro cytotoxicity against seven human cancer cell lines.

  • Campsis grandiflora K. Schum. flower was extracted with 80% aqueous MeOH, and concentrated extract was successively partitioned with EtOAc, n-BuOH, and $H_2O$. From n-BuOH fraction, two cyclohexylethanoids were isolated through repeated silica gel and Sephadex LH-20 column chromatographies. Based on physico-chemical data obtained from NMR, MS, and IR, chemical structures of compounds were determined as 1,4-dihydroxy-3,4-(epoxyethano)-5-cyclohexene (1) and cornoside (2). These compounds were isolated for the first time from C. grandiflora K. Schum flower.

  • A total of 20 extracts derived from different plant family commonly used in Indonesian traditional inflammation medicine were screened for their inhibitory effect on soybean lipoxygenase (SBL) and hyaluronidase (HAse) activity. Three methanol extracts, the bark of Cinnamomum burmanni (CB), the leaves of Piper betel (PB), and fruit of Barringtonia acutangula (BA) were found to have high inhibitory effects, whereas the methanol extract of the leaves of Mimusops elengi (ME) have medium inhibitory effect. The IC50 of CB, PB, BA and ME were found to be 21.7, 16.9, 39.1 and 62.8 g/$m\ell$, respectively. Among the tested extracts, only CB inhibited HAse (IC50 = 27g/$m\ell$). CB was successively fractionated with n-hexane, ethyl acetate, butanol and water. The EtOAc fraction having the strongest activity was fractionated and some compounds were isolated and purified by a preparative HPLC(Develosil ODS-HG-5 column). Coumarin and 2-hydroxy cinnamaldehyde. were identified through the analyses of UV-Vis absorption 1H-NMR, 13C-NMR and FAB+-MS spectra.

Anti-rheumatoidal Effect of Sulfuretin Isolated from the Heartwood of Rhus veniciflua in Rats and Mice

  • The present study was undertaken to evaluate the anti-rheumatoidal arthritis effect of the R. verniciflua heartwood extract, its EtOAc fraction, and its primary flavonoids, sulfuretin and fustin. All test samples showed variably significant inhibitory effects on hind paw edema and trypsin inhibitor activity induced by Freund's complete adjuvant reagent (FCA reagent), and on vascular permeability caused by acetic acid. Treatment with 10 mg/kg (i.p.) sulfuretin for seven days inhibited edema formation by 54.2$\pm$3.0%. Test samples, especially sulfuretin, shifted the values of biochemical parameters such as serum-cholesterol, serum-triglyceride and serum-total protein toward the normal and restored the numbers of leucocytes and platelets. These results suggest that the heartwood of R. verniciflua reduces immunological injuries caused by FCA reagent provides evidence that suluretin is an active anti-rheumatoid arthritis agent.

  • Eight compounds were isolated from the EtOAc soluble fraction of Euphorbia supina MeOH extract as the radical scavengers for antioxidant activity. Their structures were identified as kaempferol (1), quercetin (2), juglanin (3), avicularin (4), astragalin (5), isoquercitrin (6), hyperin (7), and nicotiflorin (8) by spectroscopic analysis. The antioxidant activity was evaluated by the ORAC (oxygen radical absorbance capacity) assay, which measures scavenging activity against peroxy radicals induced by 2,2'-azobis (2-methylpropionamidine) dihydrochloride, and the ORAC value is expressed as relative trolox equivalent. Compounds 4, 6, and 7 exhibited potent antioxidant activity, whereas the other compounds showed weaker activity than trolox.

Anticoagulant 1,2,3,4,6-Pentagalloyl-$\beta$-D-Glucopyranose Isolated from Geranium (Pelargonium inquinans Ait)

  • Geranium (Pelargonium inquinans Ait) leaves were extracted with $80\%$ MeOH, and partitioned into n-hexane, ethyl acetate, BuOH and $H_2O$ to isolate the anticoagulant principles. The EtOAc fraction was found to be the most active, and was further purified using silica and octadecylisilane column chromatography employing a bioassay-guided fractionation method. The active compound was isolated and identified as $1,2,3,4,6-pentagalloyl-\beta-D-glucopyranose$(PGG) (compound I). The isolated anticoagulant significantly prolonged the activated partial thrombin time (APTT) and thrombin time (TT) using normal human plasma. One microgram of $1,2,3,4,6-pentagalloyl-\beta-D-glucopyranose$ showed 0.063 heparin units in the APTT and 2.73 heparin units in the TT for anti-thrombosis. This is the first report of the isolation of PGG from geranium plants.
