Acute Oral Toxicity Study of Dictamnus dasycarpus Turcz in SD Rats (백선피(Dictamnus dasycarpus Turcz) 추출물의 급성 경구투여 독성 연구)

  • Objectives : This study was carried out to evaluate the acute oral toxicity of Dictamnus dasycarpus Turcz in Sprague-Dawley(SD) rats. Methods : Male and female rats were administered orally with Dictamnus dasycarpus Turcz water extract of 1,000 mg/kg(low dosage group), 2,000 mg/kg(middle dosage group) and 4,000 mg/kg(high dosage group). We daily observed number of deaths, clinical signs and gross findings for 7 days. After 7 days, we measured body and organs weight. Also we analyzed hematological changes. Results : No dead SD rats and no clinical signs were found during the experiment period. Also other specific changes were not found between control and treated groups in hematology. But we found out subtle changes in body weight and individual organ weight of the female group. In addition specific changes were observed in serum biochemical value of female group. Conclusions : These results suggest that water soluble extract of Dictamnus dasycarpus Turcz has not acute oral toxicity and oral $LD_{50}$ value was over 4,000 mg/kg in SD rats. Also Dictamnus dasycarpus Turcz is expected to be sensitive with respect to the female.

A literature study on dermatological efficacy and drug induced liver injury of Dictamnus dasycarpus Turcz (백선피(白鮮皮)의 피부과적 효능과 약인성 간손상에 대한 문헌 연구)

  • Objectives : The root bark of Dictamnus dasycarpus has been frequently used to treat patients with skin diseases in Korea. Recently, wild root of D. dasycarpus are reported to induce liver injury. Methods : Traditional uses of D. dasycarpus for skin diseases were analysed bibliographically. In addition, reported cases were collected and analysed using pubmed and national digital library. Results : In taiwan, D. dasycarpus revealed to be one of major herbs for skin diseases and many researchers in worldwide had reported its dermatological efficacies. Reported cases related in liver injury described that hepatocellular or cholestatic liver injury were seen in patients eating wild root of D. dasycarpus. In addition, 6 cases in worldwide and 1 case in Korea showed that patients with drug induced liver injury (DILI) ingested not root bark of D. dasycarpus but prescriptions containing root bark of D. dasycarpus. These mean that wild root of D. dasycarpus (Bongsam or Bongwhangsam) was estimated to be closely related in DILI. Whereas, it was difficult to confirm direct correlation between root bark of D. dasycarpus used as herbal medicine by doctor of Korean medicine and DILI. Conclusions : these results imply that wild root of D. dasycarpus is closely related in DILI and strong recommendation not to take it without consultation by experts is needed. In addition, although there are no evidences of direct correlation between root bark of D. dasycarpus and DILI, doctor of Korean medicines should pay attention to use root bark of D. dasycarpus in their clinics.

Screening for Antifungal Medicinal Plants against Brown Patch and Large Patch Diseases of Turfgrass (잔디 Brown patch와 Large patch병 방제를 위한 향균성 약용식물 탐색)

  • Of crude extracts from 30 species belonging to the 21 families of medical plants, the crude extracts from Cinnamomum cassia, Paeonia moutan, Dictamnus dasycarpus showed highly antifungal activities against Rhizoctonia solani AG1-1 and R. solani AG2-2. The antifungal activity of C. cassia and P. moutan against R. solani AG1-1 were the highest through 100% methanol extraction, but the one of D. dasycarpus were the highest through 80% methanol extraction. On the other hand, against R. solani AG2-2 the antifungal activity of them were the highest through 100% methanol. C. cassia P. mouton and D. dasycarpus showed over 60% inhibition of mycelial growth against R. solani AG1-1 and R. solani AG2-2 on up to 500 fold dilution. The crude extract of P. moutan showed broader antifungal spectrum compared with those of C. cassia and D. dasycarpus. The crude extract of C. cassia was not phytotoxic at over 250 fold dilution and the crude extract of P. moutan was not phytotoxic at over 100 fold dilution. The crude extract of D. dasycarpus was not phytotoxic even at full concentration. The control values of P. moutan and D. dasycarpus were increased 75.0% and 79.1% respectively 30 days after treatment.

Simultaneous Determination of Dictamine, Obacunone and Fraxinellone in Dictamnus dasycarpus cortex by HPLC-DAD (HPLC-DAD를 이용한 백선피의 Dictamine, Obacunone, Fraxinellone의 동시분석법 확립)

  • A high performance liquid chromatographic (HPLC) method for the simultaneous determination of dictamine, obacunone and fraxinellone was established for the quality control of traditional herb Dictamnus dasycarpus cortex. Separation and quantification were successfully achieved with a Shiseido C18 column ($5\;{\mu}m$, 4.6 mm I.D. ${\times}$ 250 mm) by gradient elution of a mixture of methanol and water at a flow rate of 1.0 ml/min. The diode-array UV/Vis detector (DAD) was used for the detection and the wavelength for quantification was set at 236 nm. The presence of dictamine, obacunone and fraxinellone in this extract was ascertained by retention time, spiking with each authentic standard and UV spectrum. All three compounds showed good linearity ($r^2$ > 0.999) in relatively wide concentration ranges. The R.S.D. recovery of each compound was 101.0~103.7% with R.S.D. values less than 1.0%. This method was successfully applied to the determination of contents of dictamine, obacunone and fraxitnellone in three commercial products of D. dasycarpus cortex. These results suggest that the developed HPLC method is simple, effective and could be readily utilized as a quality control method for commercial products.

Neuroprotective Effect of the Water-insoluble fraction of Root Barks of Dictamnus dasycarpus 70% Ethanolic Extract on Glutamate-Induced Oxidative Damage in Mouse Hippocampal HT22 Cells (백선피 70% 에탄올 추출물의 비수용성 분획물의 뇌세포 보호 효과)

  • Oxidative stress or accumulation of reactive oxygen species (ROS) leads neuronal cellular death and dysfunction, and it contributes to neuronal degenerative disease such as Alzheimer's disease, Parkinson's disease and stroke. Glutamate is one of the major excitatory neurotransmitter in the central nervous system (CNS). Glutamate contributes to fast synaptic transmission, neuronal plasticity, outgrowth and survival, behavior, learning and memory. In spite of these physiological functions, high concentration of glutamate causes neuronal cell damage, acute insults and chronic neuronal neurodegenerative diseases. Heme oxygenase-1 (HO-1) enzyme plays an important role of cellular antioxidant system against oxidant injury. NNMBS020, the water-insoluble fraction of the 70% EtOH extract of root barks of Dictamnus dasycarpus, showed dominant neuroprotective effects on glutamate-induced neurotoxicity in mouse hippocampal HT22 cells by induced the expression of HO-1 and increased HO activity. In mouse hippocampal HT22 cells, NNMBS020 makes the nuclear accumulation of Nrf2 and stimulates extracellular signal-regulated kinase (ERK) pathway. The ERK MAPK pathway inhibitor significantly reduced NNMBS020-induced HO-1 expression, whereas the JNK and p38 inhibitors did not. In conclusion, the water-insoluble fraction of the 70% EtOH extract of root barks of D. dasycarpus (NNMBS020) significantly protect glutamate-induced oxidative damage by induction of HO-1 via Nrf2 and ERK pathway in mouse hippocampal HT22 cells.

The effects of water extract from Dictamnus dasycarpus Turcz on Hepatocellular Damage in vitro (백선 추출물의 간세포 손상에 대한 연구)

  • Objectives : This study was carried out to evaluate whether the water extract from cause the cellular damage in HepG2 cell line. It was reported that Dictamnus dasycarpus Turcz(DDT) intake induce poisoning symptoms in human population. These symptoms was closely related to liver toxicity, however, mechanisms for liver toxicity caused by DDT have not been elucidated exactly. Here, hepatotoxicity caused by DDT was evaluated using HepG2 cell line. Methods : Water extract of DDT was treated into HepG2 cell with various doses such as 0, 0.1, 0.5, 1.0 and $5.0mg/m{\ell}$. In order to cell viability, both MTT and LDH assay were carried out. Also, apoptosis array kit was used to identify whether cell death caused by DDT is due to apoptosis or not. In addition, reactive oxygen species (ROS) was measured after treatment of water extract. Results : We found out significant changes in the apoptosis related factors of hepatocyte. The cell viability of HepG2 treated with DDT water extract was decreased in dose-dependent. Also most of the apoptosis related factors were significantly increased. We found out that Caspase 3, Cytochrome C and ROS had increased in dose-dependent. In addition, other apoptosis related factors Bcl 2 and Bax, which were also constant changes. However, there was no significance. Conclusions : These results suggest that water soluble extract of DDT is expected to have oral toxicity, including hepatocellular damage Therefore, it is suggested that DDT could cause various side effects and toxicity of clinical conditions.

Anti-Toxoplasmosis effects of herbal extracts against Toxoplasma Gondii (항톡소포자충 효과를 가진 천연물 유래 추출물에 관한 연구)

  • Toxoplasmosis is a parasitic disease caused by Toxoplasma gondii, with very few therapeutic treatment options. The choices for treatment are pyrimethamine and sulfadiazine, however their utility is limited because of drug toxicity and serious side effects. In this study, ethanol extracts of 13 traditional medicines used to treat Toxoplasma gondii were tested in vitro for their anti-Toxoplasma gondii cytotoxicity. The median effective concentration ($EC_{50}$) values for the herbal extracts ranged from 173 mg/mL to 1995.35 mg/mL. In HeLa cell, the selectivity of Alpinia oxyphylla (2.75), Mucunae Caulis (2.96), Dictamnus dasycarpus (7.52) which was higher than sulfadiazine (2.08). This indicates that Alpinia oxyphylla, Mucunae Caulis, Dictamnus dasycarpus extracts may be sources of new anti-Toxoplasma gondii compounds.

Analyses of Morphological Characteristics, Antibiotic Effect, and Molecular Cytogenetics in Baegseon (Dictamnus dasycarpus Turcz) (백선의 형태적 특성, 항균 효과 및 분자 세포학적 분석)

  • The importance of wild plant resources along with the development of high biotecyhnology is highlighted for exploitation of new materials which can make the added value. The goal of this study is providing fundamental data bases for developing new materials through the analyses of morphological characteristics, antibiotics and molecular cytogenetics in Baegseon (Dictamnus dasycarpus Turcz.). Baegseon has several characteristics; there are two types of flower color, pink and white, the seed germination starts about February 20th, the maximum flowering season is around May 17th in southern Korea, the growth period is about 60 days. The number of chromosomes are 2n=2x=36, the size of chromosomes in metaphase is $4.2{\sim}8.1{\mu}m$. The amount of 2C nuclear DNA is 1.93pg, and there is no variation of genome size amoung varieties. The extraction juice of baegseon young roots has the excellent antibiotic activity against the mold (Botrytis cinerea).
