Study on Anti-oxidant and Anti-inflammatory Activity of Eggplant-cheongyeolsodokum (가지-청열소독음(淸熱消毒飮)의 항산화 및 항염 효능에 관한 연구)

  • The purpose of this study is verification of the anti-oxidant effect and anti-inflammatory effect of Eggplant-cheongyeolsodokum composed of 8 herbs (Solanum melongena L., Lonicera japonica Thunb., Glycyrrhiza uralensis Fisch., Ligusticum chuanxiong Hort., Angelica gigas Nakai., Coptis deltoidea C. Y. Cheng et Hsiao., Gardenia jasminoides J. Ellis., Forsythia suspensa Vahl) to confirm the possibilities as useful cosmetic material. We used the modified prescription of 'cheongyeolsodokum' contained in Korean traditional medical book 'Donguibogam' as composition of Eggplant-cheongyeolsodokum and their proportions. Eggplant-cheongyeolsodokum were extracted with hot water, 70% ethanol and then powdered. To confirm anti-oxidant effect, we investigated radical scavenging ability (DPPH, $ABTS^+$, superoxide), superoxide dismutase (SOD)-like activity, total polyphenolic contents. Also to confirm anti-inflammatory effect, we investigated inhibition effect of nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages, and Inhibition effect of the expression of inflammatory-related proteins (iNOS, COX-2) by western blot analysis. As a result, Eggplant-cheongyeolsodokum showed good anti-oxidant and anti-inflammation effects, we suggest that it can be used as an active ingredient for cosmetics.

Study on Anti-oxidant Effect of Extracts from Pyrus ussuriensis Leaves (산돌배나무(Pyrus ussuriensis) 잎 추출물의 항산화활성에 관한 연구)

  • The aim of the study was to assess the cosmeceutical activity of Pyrus ussuriensis leaves and it is possible that can be used as a cosmetic ingredient for application of cosmetic industries. P. ussurensis leaf was extracted with various solvents including water, 70% ethanol and 60% acetone. In the result of DPPH (1,1-diphenyl-2-picryl-hydrazyl) scavenging radical activity, water, ethanol and acetone extract of P. ussuriensis leaf were higher than 70%, 80% and 85% at 50 ppm concentration, respectively. Xanthine oxidase inhibition activity and superoxide dismutase (SOD)-like activity by P. ussuriensis extract were higher than 30%. In addition, SODlike activity of all extracts showed tendency of the significant increase with the increase of concentration. In the anti-inflammatory test, P. ussuriensis leaf extract inhibited generation of nitric oxide (NO) stimulated by LPS in the macrophage cell line (raw 264.7) after 12 to 24 hours. As above results, P. ussuriensis has a great potential as a cosmeceutical raw material as well as anti-oxidant and anti-inflammatory ability.

Effects of Spatholobi Caulis MeOH Extract on the Production of NO and Pro-inflammatory Cytokines in LPS-activated Raw264.7 Cells (계혈등 메탄올추출물이 LPS로 활성화된 Raw264.7 Cell에서 Nitric Oxide 및 Pro-inflammatory Cytokines 생성에 미치는 영향)

  • Objectives: Spatholobi Caulis is the stem of Spatholobus suberectus Dunn., which has been used for treating menstrual disorders, anemia, and rheumatic arthralgia in traditional Oriental medicine, This study was conducted to evaluate the inhibitory effects of Spatholobi Caulis extract (SCE) on the production of nitric oxide (NO) and pro-inflammatory cytokines in LPS-activated Raw264,7 cells. Methods : Cell viability was determined by MIT assay. Relative levels of NO were measured with Griess reagent and pro-inflammatory cytokines were detected by ELISA Expression of iNOS and COX-2 proteins were determined by western blotting. Results : NO production and iNOS expression were increased by incubation with LPS for 24 h. However, the augmented NO was reduced by SCE in a dose-dependent manner. Expressions of iNOS and COX-2 were inhibited by the treatment with SCE, SCE also suppressed the production of pro-inflammatory cytokines, such as IL-1beta and IL-6. Conclusions : These results indicate that the MeOH extract of Spatholobi Caulis warrant further development as an anti-inflammatory agent for the treatment of Gram-negative bacterial infections.

Antioxidant effects and tyrosinase and elastase inhibitory activities of mountain ginseng adventitious roots extracts at different ethanol concentrations

  • To use mountain ginseng adventitious roots (MGAR), which is superior in functionality compared to ginseng, and to mass produce raw materials as a functional cosmetic material, the antioxidant and cosmeceutical activities of the extracts of MGAR at different ethanol concentrations (0, 20, 40, 60 and 80%) were compared and analyzed. At first, different ethanol concentrations were compared to determine the best solvent for the extraction of phenolic compounds from MGAR. An 80% ethanol extract with 10.07 mg of gallic acid equivalents/g sample and 0.94 mg of quercetin equivalents/g sample was the best solvent in the extraction of phenolic compounds. However, the 60% ethanol extract had the highest antioxidant activity, evident by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, Trolox equivalent antioxidant capacity (35.01 mM Trolox/g sample) and ferric reducing antioxidant power (460.94 mM $FeSO_4/g$ sample). However, the inhibitory activities of the 80% ethanol extract from the MGAR against tyrosinase and elastase were higher than those of the other extracts. The results of this study show that the extract of MGAR can be used as an easily accessible source of natural antioxidant and as a possible cosmetic ingredient in the cosmeceutical industry. However, the components responsible for the antioxidant and cosmeceutical activities of the MGAR extracts are currently unclear. Therefore, it is suggested that further studies should be carried out to isolate and identify the antioxidant and cosmeceutical components in MGAR.

Anti-Inflammatory Effect of Carex scabrifolia Steud. Extract in RAW264.7 Cells

  • This research was designed to evaluate the possible anti-inflammatory effects of Carex scabrifolia Steud. extract using RAW264.7 cells. The assessments of these effects were based on cell viability assay, mRNA expression levels of interleukin-1 alpha (IL-1α), interleukin-1 beta (IL-1β), IL-6, tumor necrosis factor alpha (TNFα), and levels of nitric oxide (NO)/prostaglandin E2 (PGE2) production. Quantitative real-time polymerase chain reaction showed that treatment with C. scabrifolia Steud. extract decreased the mRNA levels of iNOS, COX2, IL-1α, IL-1β, IL-6, and TNFα. Furthermore, from the production levels of PGE2/NO, it can be inferred that C. scabrifolia Steud. extract exhibited anti-inflammatory properties. These results suggest that C. scabrifolia Steud. extract contains anti-inflammatory compound(s), and consequently, that it may have applications as a potent cosmeceutical material.

Study on Cosmeceutical Activities and Anti-inflammatory Activities of Magnolia biondii Extracts (신이화(辛夷花, Magnoliae Flos)추출물의 화장품약리활성 및 항염증효과에 관한 연구)

  • Existing pharmaceutical studies show that Magnolia biondii is effective in treating rhinitis and in reducing cholesterol, given its endogenous, volatile ingredients. The study herein seeks to assess the cosmeceutical activities and anti-inflammatory activities of Magnolia biondii extracts for possible application as cosmetic ingredients. The cosmeceutical and anti-inflammatory activities were investigated using hydroxyl radical scavenging, superoxide dismutase (SOD)-like activity, xanthine oxidase (XO) inhibition, cell viability, nitric oxide (NO) inhibition, and inducible nitric oxide synthase (iNOS) expression by Western blotting. Magnolia biondii extracts were identified to have antioxidant activities in hydroxyl free radical scavenging, SOD-like activity, and XO inhibition. In testing the anti-inflammatory activities of the extracts, NO production was inhibited in a dose-dependent manner. Additionally, in a dose-dependent manner, the Magnolia biondii extracts were able to suppress iNOS expression in LPS-stimulated RAW 264.7 macrophage cells. From these results, Magnolia biondii showed adequate potential for application in cosmetic production and related industries as well as a functional material.

Effect of Red Ginseng Extract on the Inflammatory Response of LPS-Induced RAW 264.7 cell (홍삼추출물이 LPS로 유도된 RAW 264.7 cell의 염증반응에 미치는 효과)

  • We conducted this study to investigate anti-inflammatory possibilities of applying cosmetic material about extracts from red ginseng. For this we carried out biological active evaluation about anti-inflammatory by using extracts of red ginseng. In order to evaluate the anti-inflammatory effects of the samples in macrophages (RAW 264.7 cells), MTT assay was used to evaluate the toxicity of red ginseng extracts and the inhibitory activity of nitric oxide production and the expression levels of inflammation-related proteins and genes. The inhibitory activity of nitric oxide in the LPS-induced RAW 264.7 cells was 71.2% at 25 ㎍/ml concentration and western blot analysis showed that the expression of iNOS and COX-2 protein decreased in a concentration-dependent manner. These results suggest that extracts from red ginseng may have value as the potential cosmetic materials.

Anti-inflammatory Effect of Ishige foliacea in RAW 264.7 Cells (넓패추출물에 의한 RAW 264.7 세포에서의 항염효과)

  • This study was carried out to identify the anti-inflammatory effects of Ishige foliacea (I. foliacea) extract on skin using RAW 264.7 cells. The anti-inflammatory effects of I. foliacea extract on RAW 264.7 cells were assessed by cell viability assay, mRNA expressions, and nitric oxide (NO)/prostaglandin E2 (PGE2) productions. The anti-inflammatory effects of I. foliacea extract were elucidated by analysis of IL-1α/IL-1β/IL-6/TNFα gene expressions and PGE2/NO production. Quantitative real-time polymerase chain reaction showed that I. foliacea extract decreased the gene expression levels of iNOS/COX2/IL-1α/IL-1β and IL-6. Furthermore, PGE2/NO production also revealed that I. foliacea extract exhibited anti-inflammatory properties. These results suggest that I. foliacea extract is an anti-inflammatory compound. It could be a potent cosmeceutical material for anti-inflammatory effects. Further studies on the anti-inflammatory mechanisms of broadleaf extracts are expected to help identify pharmacological mechanisms related to inflammation in addition to cosmeceuticals.

Anti-Oxidant and Antiinflammatory Effects of Rosa multiflora Root (찔레나무뿌리(Rosa multiflora root)의 항산화 및 항염증효과)

  • Rosa multiflora thunberg belonging to Rosaceae is widely distributed in East Asia including Korea and Japan, and has been reported to have tormentic acid and rosamultin. To develop a new natural anti-inflammatory agent for cosmetics, we investigated the inhibitory effects of inflammation in Rosa multiflora root (R. multiflora root). The biological activity and anti-inflammatory effects were investigated by water, ethanol, methanol and acetone extracts of R. multiflora root. The measurements of polyphenol content from R. multiflora root were highest in water and acetone extracts, at 57.48 ${\pm}$ 0.88 mg/g and 67.05 ${\pm}$ 0.56 mg/g, respectively. The result of DPPH, ABTS and superoxide anion radical scavenging effects showed over 50% efficacy at 50 ${\mu}g/ml$ in ethanol, methanol and acetone extracts. Hyaluronidase inhibition effect showed over 60% efficacy at 500 ${\mu}g/ml$ in ethanol, methanol, and acetone extracts. Nitric oxide radical inhibition effect of R. multiflora root ethanol extracts showed over 30% efficacy at 500 ${\mu}g/ml$. We investigated the effect of R. multiflora root extracts on nitric oxide (NO) production of inducible nitric oxide synthase (iNOS) in LPS-induced RAW 264.7 macrophage cells. The result showed that R. multiflora root extracts have an inhibitory effect on NO production and iNOS expression and also can be used as an anti-inflammatory agent. These antioxidant and anti-inflammatory effects of R. multiflora root show applicant potential application as a functional cosmetic material.

The Study of Cosmeceutical Activities from Lentinula edodes extracts and Application a Natural Cosmetic Material (표고버섯 추출물의 화장품약리활성 검증과 천연화장품 소재로써의 활용에 관한 연구)

  • This study is for checking the possibility of Lentinula edodes as cosmetic materials. For this we carried out biological active evaluation about anti-oxidant and anti-inflammatory effects by Lentinula edodes extracts. We extracted Lentinula edodes with water and 70% ethanol and then in order to evaluate anti-oxidant activity we treated samples by concentrations (100, 500, 1000) ${\mu}g/ml$ and carried out 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, the activity of 2,2'-azino-bis ( 3-ethylbenzothiazoline-6-sulphonic acid )-diammonium salt (ABTS) cation radical scavenging and superoxide dismutase(SOD) like activity. Also, in order to evaluate effect of anti-inflammatory the samples in macrophages(RAW 264.7 cells), we carried out evaluation of cell viability, nitric oxide inhibitory activity western blot. The results of DPPH, $ABTS^+$ radical scavenging activity and SOD-like activity of the Lentinula edodes extracts increased in dose-dependent manner. The cytotoxic of samples by MTT assay showed no toxicity at the concentrations of 10, 25 and $50{\mu}g/ml$ of Lentinula edodes extract. Nitric oxide inhibition activity results showed that the extracts reduced NO productions in a concentration-dependent manner. Expression of inflammatory cytokines as $TNF-{\alpha}$, $PGE_2$ and $IL-1{\beta}$ decreased in a concentration-dependent manner and iNOS and COX-2 proteins expression rates were decreased significantly in western blot analysis. From the results of the experiment, it was comfirmed that the Lentinula edodes extracts had excellent anti-oxidant and anti-inflammatory effect and could be used as a safe natural cosmetic material in the future.
