HCl/에탄올로 유발된 급성 위염에서 Artemisia frigida Willd의 위장 보호 효과 (Gastroprotective Effect of Artemisia frigida Willd in HCl/Ethanol-induced Acute Gastritis)

  • Artemisia frigida Willd (AW, Fringed sagewort), which is widespread in Mongolia, is a well-known medicinal plant as a member of the Compositae family. This study aims to explore the gastroprotective effect of water extract of AW on 150 mM HCl/60% ethanol-induced acute gastritis in 5 week old male ICR mice. Total polyphenols, total flavonoid contents, and anti-oxidant activity in vitro in AW were evaluated. First, the gross area of gastric mucosal damage was measured. Then western blot analysis was conducted to determine the possible mechanisms of action underlying the effects of AW. AW administration decreased gastric mucosal damage. Moreover, the group with AW treatment effectively inhibited nicotinamide adenine dinucleotide phosphate (NADPH) oxidase expression associated with oxidative stress. AW treatment enhanced an anti-oxidant effect through the increase of anti-oxidant proteins. Besides, the increased expressions of inflammatory cytokines induced by nuclear factor-kappa B (NF-κB) activation are alleviated through AW treatment. Taken together, AW exerted a gastroprotective effect against gastric mucosal damage. These results indicate that AW could have the potential used as a natural therapeutic drug for the treatment of acute gastritis.

  • It has been reported that various anti-oxidant substances stimulate non-specific immune responses in fishes. In this study it was examined whether N-acetylcysteine (NAC), a precusor for anti-oxidant glutathione (GSH) synthesis, can modulate non-specific immune function in Far Eastern catfish Silurus asotus. Immune functions were assessed using the respiratory burst activity monitored by chemiluminescence (CL) responses in isolated leucocyte. NAC stimulated CL responses with doses of 10 or 100 mg/kg, but not with 1 mg/kg after 48 hr injection. It was observed with 10 mg/kg NAC that CL activity continued to elevate from 24 hr through 96 hr post-dosing, and returned to the near preinjection level by 10 days. To understand whether NAC can also activate CL activity in vitro, NAC was directly added to isolated catfish leucocytes. It was observed, however, that NAC can not stimulate CL at reasonable concentration ranges in vitro. As NAC is a precursor for the strong anti-oxidant glutathione (GSH), a putative immune stimulator, it was assessed whether GSH can also stimulate CL responses. Observed results show that GSH activated CL both in vivo and in vitro. The data obtained collectively support the proposition that NAC indirectly stimulates non-specific immune functions in catfish by enhancing GSH biosynthesis, but not by direct action of NAC. Such effects may have beneficial significance in aquaculture for practical utilization.

목과(木瓜)추출물이 급성 간손상 흰쥐에 미치는 효과 (The Effect of Chaenomelis Fructus Extract on Acute Hepatic Injury in Rats)

  • Objective: This study was conducted to investigate the effect of Chaenomelis Fructus (CF) water extract on thioacetamide (TAA)-treated rats. Methods: Rats were divided into five groups: one normal group (n=8) and four with TAA-induced hepatic injury. These treatment groups were administered distilled water (n=8); silymarin 100 mg/kg (n=8); CF 100 mg/kg (n=8); and CF 200 mg/kg (n=8). In the TAA groups, the acute liver injury was induced via IP injection (200 mg/kg), and the silymarin and CF extract were then orally administered for three days. Subsequently, serum levels of GOT, GPT, and ammonia were confirmed as well as protein expressions using liver tissue. Results: In the liver injury-induced rats, CF administration reduced tissue damage and serum levels of GOT, GPT, and ammonia. In addition, CF increased the anti-oxidant proteins Nrf2, Keap1, HO-1, and catalase and significantly regulated matrix metalloproteinases (MMP-2 and MMP-9) and their tissue inhibitors (TIMP-1 and TIMP-2). Conclusions: In this animal model of liver injury induced by TAA, CF extract is determined to have a hepatoprotective effect by increasing anti-oxidant proteins that relieve damage and by regulating the expression of matrix metalloproteinases.

Enhancement of performance and anti-oxidant variables in broiler chicken fed diets containing sub-optimal methionine level with graded concentrations of sulphur and folic acid

  • Objective: An experiment was conducted to determine the effects of supplementing graded concentrations of inorganic sulphur (S) without and with folic acid (FA) in maize-soybean meal diets on performance, slaughter and anti-oxidant variables, immune responses and serum protein fractions in broiler chicken. Methods: Inorganic S was supplemented at 0.05%, 0.10%, 0.15%, and 0.20% alone or in combination with FA (4 mg/kg) in basal diet (BD) containing no supplemental methionine (Met) and FA. A control group was fed with the recommended concentration of Met. Each diet was offered to 10 pens of 5 male broiler chicks (Cobb 400) and fed ad libitum from day 1 to 42. Results: The broilers fed the BD had lower body weight gain (BWG), feed efficiency (FE), higher lipid peroxidation (LP), lower activity of glutathione peroxidase (GSHPx), lower lymphocyte proliferation ratio (LPR), and reduced concentrations of total protein, albumin, and globulin in serum. Supplementation of FA and S to the BD improved the BWG (all concentrations of S) and FE (0.20% S) similar to the control group. Similarly, the combination of S and FA significantly improved the concentrations of total protein, albumin, and globulin in serum, reduced the LP and increased the activity of GSHPx and LPR. However, responses in the above parameters were related to the concentration of S in the diet. The slaughter variables and antibody titres against the Newcastle disease were not affected with the treatments. Conclusion: Based on the results, it is concluded that the combination of S (0.2%) and FA (4 mg/kg) improved the BWG and FE, similarly supplementation of these nutrients improved the concentration of protein fractions and reduced the stress (reduced LP and improved GSHPx) variables in serum and improved the cell mediated immune response (LPR) in broilers fed sub-optimal concentrations of Met in diet.

Effect of methyl donors supplementation on performance, immune responses and anti-oxidant variables in broiler chicken fed diet without supplemental methionine

  • Objective: Methionine (Met) is involved in methyl group transfer besides protein synthesis. As the availability is limited and cost is high for synthetic Met, reductions in its inclusion in broiler diet may be possible by supplementing the low Met diets with methyl donors (MD) like betaine (Bet), folic acid (FA), vitamin B12 (B12), and biotin (Bio). An experiment was conducted to study the effects of supplementing the MD on performance (average daily gain [ADG], daily feed intake, feed efficiency [FE]), anti-oxidant variables, immune responses and serum protein concentration in broilers fed sub-optimal concentrations of dietary Met. Methods: Maize-soybean meal diet was used as control (CD). Different MD like Bet (0.2%), B12 (0.1 mg), FA (4 mg), or Bio (1.5 mg/kg) were supplemented to basal diet (BD) having no supplemental Met. The BD without MD was kept for comparison. Each diet was fed ad libitum to 10 replicates of 25 chicks in each from 1 to 42 d of age. Results: At the end of experiment, the ADG in MD group was higher than BD and lower than CD. The FE improved with FA or Bet compared to the BD. Breast meat weight was higher in Bet compared to the BD, while it was intermediate between BD and CD in other groups. The lipid peroxidation reduced with Bio, B12, or Bet, while the glutathione peroxidase activity improved with Bio or B12 compared to the BD. Lymphocyte proliferation improved with Bet compared to the BD. The serum protein concentrations increased with FA, Bio, or Bet compared to those fed BD. Conclusion: It can be concluded that the ADG can be improved partially with supplementation of MD while the FE improved with FA or Bet. Some MD also reduced the stress indices and improved immune responses compared to the BD fed broilers.

Effect of Rhodiola Extract Supplementation on Blood Lipid Concentrations and Anti-Oxidant Status in Rats Fed Highly Oxidized Linoleic Acid Diets

  • The influence of Rhodiola extract on tissue antioxidant status, plasma lipid levels, cholesterol contents of liver and fores were investigated in rats find oxidized linoleic acid. Groups of five-week old male Sprague-Dawley rats fed ad libitum with a diet containing 20% oxidized linoleic acid with or without 300 mg/kg body weight freeze-dried Rhodiola water extract. The antioxidant effect of dietary Rhodiola extract supplementation on the peroxidation potential of rats was investigated. The microsomal thiobarbiruric acid reactive substance (TBARS) contents were changed significantly by Rhodiola extract supplementation. Hepatic Catalase activities were increased in Rhodiola supplemented rats, whereas hepatic Manganese Superoxide Dismutase (MnSOD) or Copper Zinc Superoxide Dismutase (CuZnSOD) were not elevated. In addition, plasma cholesterol lowering effect was observed along with the stimulated excretion of cholesterol through the feces were observed with Rhodiola feeding. Supplementation with Rhodiola extract did not alter high density lipoprotein (HDL) cholesterol. These results support that Rhodiola extract may be effective in protection against oxidative stress, and prevention and treatment of blood dyslipidemia. It demonstntes that Rhodiola extract has a potential to exert anti-atherogenic properties antioxidative capacities .

영지 에탄올 추출물의 마우스 유래 뇌 해마세포 보호효과 (Neuroprotective Effects of Ethanol Extract of Ganoderma lucidum L. on murine hippocampal cells)

  • Ganoderma lucidum L. (GL) is a traditional oriental medicine that has been widely used as anti-inflammatory, antitumor and anti-oxidant in Korea and other Asian countries. In this study, we investigated the ethanol extract of GL has neuroprotective effects in murine hippocampal HT22 cells. GL ethanol extract has the potent neuroprotective effects on glutamate-intoxicated cells by inducing the expression of heme oxygenase (HO)-1 in HT22 cells. GL ethanol extract increased JNK phosphorylation. Obviously, When we treated the GL extract with c-Jun N-terminal kinase (JNK) inhibitor (SP600125), HO-1 expression was reduced. Moreover, we found that GL treatment caused the nuclear accumulation of Nrf2. In conclusion, the ethanol extract of GL significantly protects glutamate-induced oxidative damage by induction of HO-1 via Nrf2, JNK pathway in mouse hippocampal HT22. These results suggest that GL ethanol extract would be a good source for taking active compounds and may be a potential pharmaceutical products for brain disorder induced by neuronal damage and oxidative stress.

신나무 유래 Acertannin의 인체 모유두 세포 Apoptosis 조절 효능 (Modulative Effect of Human Hair Dermal Papilla Cell Apoptosis by Acertannin from the Barks and Xylems of Acer ginnala Maxim)

  • We isolated gallotannin, 2,6-digalloyl-1,5-anhydroglucitol, known as acertannin (1), from the barks and xylems of Acer ginnala Maxim. It is a genus of Acer species of shrubs in the family Aceraceae. A. ginnala grows in Korea, Japan and Mongolia. We accomplished the structure elucidation by confirming that the result of $^1H$,$^{13}C-NMR$,MS spectrum data was similar to previous references. We measured DPPH and ABTS radical scavenging activity in vitro to evaluate anti-oxidative activities on acertannin isolated from A. ginnala. Acertannin from A. ginnala exhibited potent DPPH and ABTS radical scavenging activities. We examined the antioxidant and apoptosis modulative effects. This examination shows that A. ginnala has not only 1,1-diphenyl-2-picryhydrazyl(DPPH) radical scavenging activity and ABTS radical scavenging activity, but also human hair dermal papilla cell protection effects. These results indicate that the barks and xylems of A. ginnala might be developed as a potent anti-oxidant, hair growth agent, and ingredient for related new functional cosmetic materials.

에탄올 유발 위점막 손상에 대한 황련 물 추출물의 방어효과 (Protective Effects of Coptidis Rhizoma on Ethanol-induced Gastric Ulcer in Mice)

  • The exposure of gastric mucosa to ethanol produces acute ulcers mediated by inflammatory processes, hemorrhagic erosions and increase of reactive oxygen species. The purpose of this study was to assess the effects of Coptidis Rhizoma(CR) aqueous extracts on hydrochloride (HCl)/ethanol induced gastric ulcer in mice as compared with rebamipide (30 mg/kg) and ranitidine (100 mg/kg). Stomach ulcers were induced by oral ingestion of HCl/ethanol. CR extracts (125, 250 and 500 mg/kg) were orally administered, once a day for 7 continuous days, and 1 hr after last 7th treatment of CR extracts stomach ulcers were induced. Effects of CR extracts on HCl/ethanol-induced gastric ulcer were evaluated based on gross and microscopic observations with anti-oxidant activities. All three different dosages of CR extract significantly decreased HCl/ethanol-induced gastric ulcer compared with the HCl/ethanol control mice. CR extracts also strengthened the antioxidative defense systems - decreased the level of lipid peroxidation but increased the level of catalase, superoxide dismutase and nitrate/nitrite compared with the HCl/ethanol control. The effects of CR extract 500 mg/kg were similar to that of 30 mg/kg rebamipide, and CR extract 250 mg/kg showed similar anti-ulcer effects as compared with ranitidine 100 mg/kg. These results suggest that the gastroprotective effects of CR extracts on mice ulcer models can be attributed to its ameliorating effect on oxidative damages.

박달나무로부터 분리된 페놀성 화합물의 항산화 및 Tyrosinase 저해 활성 연구 (Identification of Anti-Oxidant and Anti-Tyrosinase Activity of Phenolic Components Isolated from Betula schmidtii)

  • The aim of study to investigate the phytochemicals and biological activities the bark of Betula schmidtii. The studies consisted of the solvent extraction, followed by the isolation of phenolic components 1~3 from ethyl acetate-soluble fraction of Betula schmidtii Bark. Their chemical structures were identified as arbutin (1), ρ-coumaric acid (2) and ferulic acid (3) using Ultraviolet-Visible (UV-Vis) Spectrophotometer, Electrospray Ionization Mass Spectrometry (ESI-MS) (negative ion mode), 1H-Nuclear Magnetic Resonance (NMR), 13C-NMR, 1H-1H Correlation Spectroscopy (COSY) and 1H-13C Hetero Nuclear Multiple Quantum Correlation (HMQC) spectral data. Compounds 1~3 shows the anti-oxidant effect with IC50 values of 29.74±1.52, 21.32±1.07 and 34.41±1.24 in 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity, respectively. Also, compounds 1~3 exhibited mushroom tyrosinase inhibitory activity with IC50 values of 31.14±1.07, 42.54±1.46 and 69.22±1.43 µM, respectively.
