Plant parasitic nematodes are causing significant damage in crop production. There is a need to develop eco-friendly nematicide that reduces the damage of nematode and has little effect on the environment and human. In this study, we have isolated a substance having nematicidal activity from Ginkgo biloba L. outer seedcoat. Studies of G. biloba L. outer seedcoat are insufficient compared to the seed and leaves due to their odor and toxicity. The dried G. biloba L. outer seedcoat was extracted with dichloromethane:methanol (1:1) and fractionated into hexane, ethyl acetate and H2O. Four steps TLC were performed from EtOAc fraction to purely isolate GB4-3 with nematicidal activity. To compare nematicidal activity, G. biloba L. seedcoat methanol extract and purified GB4-3 were investigated in terms of treatment concentration and time. As a result, the nematicidal activity increased with concentration and time. In the place treated with 20 ㎍/mL of crude G. biloba L. seedcoat MeOH extract, strong activity appeared after 12 hours, and 46% nematicidal activity shown after 18 hours. About 69% of nematicidal activity was confirmed in the place where GB4-3 purified from outer seedcoat was treated with 20 ㎍/mL, and the possibility of development as nematicide was very high. This study could be used as a basic data for the development of a nematode preparation from G. biloba L. outer seedcoat.
The nematicidal and egg haching inhibitory effects of extracts from 30 herbal plants (total 32 samples) against Meloidogyne hapla J2 juveniles and eggs was tested using the dipping method. At 1,000 ppm, extracts of Daphne genkwa flower buds, Eugenia caryophyllata flowers, Quisqualis indica fruits, and Zingiber officinale rhizomes produced > 80% mortality in J2 juveniles. At 125 ppm, extracts of D. genkwa and Q. indica produced 91 and 99% mortality, respectively. The toxicity of 5 selected plant extracts to M. hapla differed depending on the solvent used (i.e. hexane, methanol, hot water, or cold water). Hot water extracts of Z. officinale and Q. indica produced nematicidal efficacies of 99 and 99%, compared to 36 and 98%, respectively, with cold water extraction. Q. indica extract was highly active against M. hapla regardless of extraction method. The inhibitory effects of Areca catechu, D. genkwa, Desmodium caudatum, Pharbitis nil, Q. indica, and Z. officinale extracts on egg hatching of M. hapla was evaluated. At 1,000 ppm, D. genkwa, P. nil, and Q. indica extracts significantly reduced hatching at 7, 14 and 21 days after treatment. Numbers of juveniles in soil treated with the methanol extract D. genkwa (1,000 ppm) were significantly lower than in untreated soil in trials in pots and in a ginseng (Phanax ginseng) field. These results indicate that Q. indica extracts could be used as an environmental friendly control agent of M. hapla.

Southern root-knot nematode (SRKN, Meloidogyne incognita) is known to be responsible for annual economic losses of 38.2 million USD in cucumber cultivation in Korea. Nematicides are frequently used to manage SRKN in plastic greenhouses. A field experiment was conducted to assess the effect of a soil fumigant (dimethyl disulfide, DMDS) and a non-fumigant (fluopyram, FL) against SRKN during the winter fallow season in a greenhouse from October to December. Nematicidal efficacy was assessed at 43 days after treatment, and the root gall index was measured 4 months after transplanting cucumber seedlings. DMDS effectively reduced second-stage SRKN juvenile density with 91% control efficacy, while FL showed no control efficacy against SRKN. Root gall index values were significantly different(p=0.020) in DMDS treatment and controls at 1.0±0.00 and 4.3±0.58, respectively. This study showed that DMDS could be considered an effective nematicide for controlling SRKN in the fallow period in winter.

Clover cyst nematode (Heterodera trifolii, CCN) is one of the important plantparasitic nematodes in the Republic of Korea, causing serious damage to Kimchi-cabbage in Gangwon province since 2017. Soil fumigants are the preferred choice for managing CCN. However, cabbage fields in Gangwon province have a slope, making it challenging to apply soil fumigants and use plastic mulch. Consequently, alternative materials should be developed for managing CCN. Nematicidal cover crops and resistant cultivars are among the alternatives. We conducted resistance screening of 82 white mustard(Sinapis alba) resources from a genebank at RDA against CCN for use as nematicidal cover crops or breeding materials. In the first assay (1st, 2nd, and 3rd pot experiments), 15 white mustard resources were selected, while others were susceptible to CCN. To confirm the reproducibility of resistance to CCN for 13 selected resources, the second assay was performed. As a result, five white mustard resource (IT297309, IT297312, IT302951, IT302953, IT302954) demonstrated resistance to CCN. These findings indicate the potential use of these five white mustard resources as nematicidal cover crops or for breeding cultivars of Kimchi-cabbage in Republic of Korea.

The mixture (MX) of acetic acid (AA) and lactic acid (LA) was examined for its effectiveness in the control of the pine wood nematode Bursaphelenchus xylophilus contaminated in cymbidium culture medium. Nematode mortality in vitro was nearly 100% in AA and MX at the concentrations of 5.0-1.0% (pH 2.6 - 4.2) and in LA only at 5.0% (pH 3.5), but lowered at concentrations of 0.5-0.1% (pH 5.1-6.9) more significantly in LA than AA and MX. MX of most concentrations caused higher nematode mortality than the average response to AA and LA. All treatments of MX (0.5% and 0.25%), fosthiazate (standard and double concentrations) and culture dilution of Paenibacillus polymyxa GBR-1 ($10^7$colony-forming units/ml) reduced significantly the nematode populations in the cymbidium culture medium, compared to non-treatment control, with no significant difference among the treatments. No phytotoxicity occurred in all treatments. pH of the medium with the time after treatment and growths of 2-year-old cymbidium were not significantly different among treatments. Considering the safety and price of the organic acids, use of MX in the processes for culturing cymbidium may be a practically reliable and eco-friendly way in the control of the quarantine nematode in cymbidium.
To evaluate the efficacy of natural nematicides for the control of root-knot nematode in strawberry greenhouses, commercial essential oils were examined by 24-well culture plate bioassay for their nematicidal activities against second-stage juveniles and eggs of Meloidogyne hapla. Based on the mortality of M. hapla juveniles at a concentration of $125{\mu}g/mL$, the most active essential oil was Alpinia galanga (100%), followed by Carum carbi (22.3%), Eugenia caryophyllata (9.4%), Cinnamonum zeylanicum (7.2%), Mentha pulegium (2.4%), and Foeniculum vulgare (2.1%). Moreover, A. galanga significantly reduced hatching at 7, 14, and 21 days after treatment. The volatile constituents identified in the A. galangal oil were methyl cinnamate (87.4%), 1,8-cineole (4.4%), ${\beta}$-pinene (2.5%), ${\alpha}$-pinene (2.2%), and p-cymene (1.1%), as major constituents. Results of this study show that A. galangal essential oil and its major constituents may serve as an environmental friendly agent of a promising natural nematicide to control Meloidogyne spp.

Control of nematode has become difficult owing to the restricted use of effective soil fumigant, methyl bromide, and other non-fumigant nematicides. Therefore, it is urgently necessary to develop microbial nematicide to replace chemical nematicides. In this study, the 50% aqueous methanol extraction solution of fermentation broths of 2,700 actinomycete strains were tested for their nematicidal activity against second stage of juveniles (J2s) of Meloidogyne incognita. As the results, only the 50% aqueous methanol extraction solution of AN110065, at 20% equivalent to 10% fermentation broth, showed strong nematicidal activity with 78.9% of mortality 24 h after treatment and 94.1% of mortality at 72 h. The 16S rRNA gene sequencing showed that the strain sequence was 99.78% identical to Streptomyces netropsis. The extract of S. netropsis AN110065 fermentation broth was successively partitioned with ethyl acetate and butanol and then the ethyl acetate, butanol and water layers were investigated for their nematicidal activity against the M. incognita. At $1000{\mu}g/ml$, ethyl acetate layer showed the strongest activity of 83.5% of juvenile mortality 72 h after treatment. The pot experiment using the fermentation broth of AN110065 on tomato plant against M. incognita displayed that it evidently suppressed gall formation at a 10-fold diluent treatment. The tomato plants treated with the fermentation broth of S. netropsis AN110065 did not show any phytotoxicity. The results suggest that S. netropsis AN110065 has a potential to serve as microbial nematicide in organic agriculture.
This study was investigated to compare the suppression of Meloidogyne arenaria by different treatments of Pasteuria penetrans which is known for biological control agent against Meloidogyne spp.. In order to select proper number of P. penetrans showing good suppression effect, P. penetrans were mixed with M, arenaria for attachment using three different concentration such as $3{\times}10^4$, $3{\times}10^5$ and $3{\times}10^6$ endospores/5 g medium, followed by treating them onto the roots of tomato. After 14 weeks incubation, P. penetrans at $3{\times}10^6$ endospores showed highest activity against the formation of gall caused by M, arenaria. At a dose of $3{\times}10^5$ endospores/5 g medium, P. penetrans was treated into soil either mixing with soil or spray onto soil surface for comparing of suppressive efficacy. When the antagonistic bacterium was treated by the former method, it suppressed more effectively Using P. penetrans at $3{\times}10^6$ endospores and mixing with soil method, suppression was compared among P. penetrans, $PASTORIA^{(R)}$(Japan) and $Fosthiazate^{(R)}$(Korea). P. penetrans was more potent than $PASTORIA^{(R)}$(Japan) and as similar as $Fosthiazate^{(R)}$(Korea). Therefore, these results suggested that P. penetrans can be used for controling of M. arenaria as biological control agent. Furthermore, thess results can be provided to develop environmentally-friendly nematicide.

An nematophagous fungi Arthrobotrys thaumasia Nema-1 and Pseudomonas putida C-5, which degrade the collagen and gelatin, was isolated from controlled horticultural soils in Seonnam-myun, Sungju-gun, Kyungpook and Woosung-myun, Gongju-shi, Chungnam to develop biological nematode pesticide. When $5,000mL\;L^{-1}$ of A. thaumasia Nema-1 culture was treated to Meloidogyne incognita, the nematicidal activity resulted in 55% at 72 hours after treatment. While the nematicidal activity increased to 65% by treating the culture mixture of $5,000mL\;L^{-1}$ Nema-1 and P. putida C-5 after 72 hours. Furthermore, the nematicidal activity of the mixture containing cinnamon extract $50mg\;L^{-1}$, each $5,000mL\;L^{-1}$ of Nema-1 and C-5 culture was elevated to 89% at 72 hours after treatment, comparing to the result showed 17% and 57% of the nematicidal activity, respectively by the treatment of chemical nemato pesticide Fosthiazate $50mg\;L^{-1}$ and neem oil $2,000mL\;L^{-1}$. These results suggested that the mixture of microorganisms and plant extract were more effective biological nematicide than the case of only microorganism or plant extract for nematode control.
Plant disease caused by root-knot nematode is a major problem in crop production. Using of chemical pesticides, one of the most efficient methods to control nematodes, have raised issues in toxicity to humans and animals and environmental pollution. In this study, to select actinomycete strains that have potential to serve as a microbial agent for control of nematodes, we investigated nematicidal activity of culture broth from 670 Streptomyces isolates. A culture filtrate of KRA15-528 isolate that was identified as S. flavogriseus on the basis of 16S rRNA sequence analysis, showed strong nematicidal activity against second stage of juveniles of Meloidogyne incognita and inhibited egg hatching; exposure to 10% of culture filtrate resulted in 71% juvenile mortality at 48 hours afters treatment and suppressed egg hatching by 54% at 9 days after treatment. When the KRA15-528 culture filtrate was partitioned with ethyl acetate and butanol, ethyl acetate layer exclusively showed strong activity; 91%, 53%, 30% of mortality at 1,000, 500, $250{\mu}g/ml$, respectively. Additionally, the culture filtrate suppressed gall formation on cucumber plant by M. incognita with no phytotoxicity. These results suggest that S. flavogriseus KRA15-528 has potential to serve as a microbial nematicide for the control of root-knot nematode disease.
