Bactericidal Efficacy of a Disinfectant Composed of Povidone-iodine Against Clostridium Perfringens and Mycobacterium Fortuitum

  • Clostridium perfringens (C. perfringens) and Mycobacterium fortuitum (M. fortuitum) are associated with considerable diseases in animals and human. In this study, the disinfection efficacy of a commercial disinfectant composed to povidone-iodine (PVI) was evaluated against C. perfringens and M. fortuitum. A bactericidal efficacy test by broth dilution method was used to determine the lowest effective dilution of the disinfectant following exposure to C. perfringens and M. fortuitum for 30 min at $4^{\circ}C$. The disinfectant and test bacteria were diluted with hard water (HW) or organic matter suspension (OM) according to treatment condition. On HW condition, the bactericidal activity of the disinfectant against C. perfringens and M. fortuitum was 50 and 80 fold dilutions, respectively. On OM condition, the bactericidal activity of the disinfectant against both C. perfringens and M. fortuitum was 15 fold dilutions. As the disinfectant composed to PVI possesses bactericidal efficacy against C. perfringens and M. fortuitum, the disinfectant solution can be used to control the spread of bacterial diseases.

  • Disinfectants/sanitizers, showing the bactericidal activity of $5log_{10}$ reduction against E. coli ATCC 10536 and Staphylococcus aureus ATCC 6538 used far assessing disinfectants/sanitizers efficacies, were examined whether showing similar efficacies against Salmonella spp. and Listeria monocytogenes, the major food poisoning bacteria, isolated from foods in current Korean market. The bactericidal efficacies on sodium hypochlorite and benzalkonium chloride were assessed by quantitative suspension tests in both 'clean' and 'dirty' conditions, respectively. Test organisms were consisted of E. coli ATCC 10536, Staphylococcus aureus ATCC 6538, Salmonella typhimurium ATCC 13311, Salmonella Enteritidis ATCC 13076, Listeria monocytogenes ATCC 19111, 10 kinds of Salmonella spp. from foods, and 11 kinds of Listeria monocytogenes from foods. More than $5log_{10}$ reduction in viable count for all strains was only achieved with benzalkonium chloride at the tolerance exemption concentration under dirty conditions. However, all strains were achieved more than $5log_{10}$ reduction under clean conditions.

Influence of water temperature, soaking period, and chemical dosage on Bakanae disease of rice (Gibberella fujikuroi) in seed disinfection (벼 종자소독시 수온 처리시간 및 약량이 벼 키다리병 발병에 미치는 영향)

  • In order to develop effective control methods for Bakanae disease caused by Gibberella fujikuroi (Saito) Ito during rearing of rice seedlings, we investigated the disease resistance of 15 rice varieties to G. fujikuroi and control effect of six seed disinfectants, and tried to improve the using methods of the seed disinfectants. Disease resistance was tested by investigating the disease incidence on each rice cultivar grown in rice seedling box infested with or not infested with G. fujikuroi at 30 days after sowing seeds and 20 days after heading date. The results showed that Hwayongbyeo, Dongjinbyeo, Hwoanbyeo, Nonghobyeo, Nampyeongbyeo, and Hwojinbyeo were resistant G. fujikuroi, meanwhile Keulubyeo, Sobibyeo, Odaebyeo, Junambyeo, Samchonebyeo, Sangjubyeo, and Hwabongbyeo were susceptible. Three seed disinfectants, prochloraz, fludioxonil, and carproamid + thiram + fludioxonil controlled Bakanae disease of rice very well, while bonomyl + thiram, thiophanate-methyl + thiram and thiophanate-methyl + triflumizole did not suppress the disease enough. Water temperature was turned to be an important factor for controlling the disease by treating seed disinfectants. Prochloraz showed 61% control value on the disease at $10^{\circ}C$, but it showed above 95% control value at the range of $30-35^{\circ}C$. It was confirmed that the control effect of seed disinfectants increased with increasing water temperature. Meanwhile soaking period of rice seeds in the suspension of seed disinfectants and chemical dosage had no high relation to control the disease. This results suggest that rice varieties, water temperature, and optimal selection of suitable seed disinfectants are very important to control Bakanae disease effectively.

Research on ANIOS Disinfection Efficiency to Prevent Infection in Radiography Room (방사선촬영실 감염 방지를 위한 에니오설프의 소독효율 연구)

  • In this study, microbes were collected before and after disinfection using ANIOS(ANIOSURF Premium NPC) and compared the areas where the radiological technologist and the patient frequently contacted the chest X-ray. From September 1st to September 7th, 2020. in P Hospital in Deagu, 4 region were collected in a 10×10 size using a sterile cotton swab of the transport medium, and before and after disinfection results were obtained through the colorimetric method. As a result, n the X-ray tube handle Proteus mirabilis, Staphylococcus epidermidis, Bacillus spp., Candida spp., and in the Chin region Proteus mirabilis, Enterococcu faecium, Pseudomonas aeruginosa, NTM, and in the Chest region Proteus mirabilis, Enterococcu faecium, Pseudomonas aeruginosa, and in the Palm region NTM, Candida spp. were detected, and 103 CFU(Colony Forming Unit) or more were measured. After disinfection, only X-ray tube handle was detect Bacillus spp. and more than 102 CFU was measured. Microorganisms found prior to disinfection can cause opportunistic infections, Experimental results showed that Aniosulf(0.25%) is more economical and disinfectant than ethanol(70-90%) and isopropyl alcohol(70-90%). However, further research is needed on the detection of Bacillus spp. resultingly this research is useful basic data of infection control in Radiography room and prevention secondary infections.

Evaluation of Cryptosporidiurn Disinfection by Ozone and Ultraviolet Irradiation Using Viability and Infectivity Assays (크립토스포리디움의 활성/감염성 판별법을 이용한 오존 및 자외선 소독능 평가)

  • In the ozone disinfection unit process of a piston type batch reactor with continuous ozone analysis using a flow injection analysis (FIA) system, the CT values for 1 log inactivation of Cryptosporidium parvum by viability assays of DAPI/PI and excystation were $1.8{\sim}2.2\;mg/L{\cdot}min$ at $25^{\circ}C$ and $9.1mg/L{\cdot}min$ at $5^{\circ}C$, respectively. At the low temperature, ozone requirement rises $4{\sim}5$ times higher in order to achieve the same level of disinfection at room temperature. In a 40 L scale pilot plant with continuous flow and constant 5 minutes retention time, disinfection effects were evaluated using excystation, DAPI/PI, and cell infection method at the same time. About 0.2 log inactivation of Cryptosporidium by DAPI/PI and excystation assay, and 1.2 log inactivation by cell infectivity assay were estimated, respectively, at the CT value of about $8mg/L{\cdot}min$. The difference between DAPI/PI and excystation assay was not significant in evaluating CT values of Cryptosporidium by ozone in both experiment of the piston and the pilot reactors. However, there was significant difference between viability assay based on the intact cell wall structure and function and infectivity assay based on the developing oocysts to sporozoites and merozoites in the pilot study. The stage of development should be more sensitive to ozone oxidation than cell wall intactness of oocysts. The difference of CT values estimated by viability assay between two studies may partly come from underestimation of the residual ozone concentration due to the manual monitoring in the pilot study, or the difference of the reactor scale (50 mL vs 40 L) and types (batch vs continuous). Adequate If value to disinfect 1 and 2 log scale of Cryptosporidium in UV irradiation process was 25 $mWs/cm^2$ and 50 $mWs/cm^2$, respectively, at $25^{\circ}C$ by DAPI/PI. At $5^{\circ}C$, 40 $mWs/cm^2$ was required for disinfecting 1 log Cryptosporidium, and 80 $mWs/cm^2$ for disinfecting 2 log Cryptosporidium. It was thought that about 60% increase of If value requirement to compensate for the $20^{\circ}C$ decrease in temperature was due to the low voltage low output lamp letting weaker UV rays occur at lower temperatures.

Bactericidal Efficacy of Fumagari OPP®, Fumigant Against Escherichia coli and Salmonella typhimurium (훈증소독제, Fumagari OPP®의 Escherichia coli와 Salmonella typhimurium에 대한 살균효과)

  • This test was performed to evaluate the bactericidal efficacy of Fumagari OPP$^{(R)}$, fumigation disinfectant, containing 20% ortho-phenylphenol against Escherichia coli (E. coli) and Salmonella typhimurium (S. typhimurium). In preliminary tests, both E. coli and S. typhimurium working culture suspension number (N value) was $4.0{\times}10^8$ CFU/mL. And all of the colony numbers on the carriers exposed the fumigant (n1, n2, n3) were higher than 0.5N1 (the number of bacterial test suspentions by pour plate method), 0.5N2 (the number of bacterial test suspentions by filter membrane method) and 0.5N1, respectively. In addition, the mean number of bacteria recovered on the control-carriers (T value) was $3.4{\times}10^6$ CFU/mL. In the bactericidal effect of the fumigant, the reduction number of S. typhimurium and E. coli (d value) was 5.26 and 5.64 logCFU/mL, respectively. According to the French standard for the fumigant, the d value for the effective bactericidal fumigant should be over than 5 logCFU/mL. With the results of this study, Fumagari OPP$^{(R)}$ has an effective bactericidal activity, then the fumigant can be applied to disinfect food materials and kitchen appliances contaminated with pathogenic bacteria.

Efficacy of Electrolyzed Water and Aqueous Chlorine Dioxide for Reducing Pathogenic Microorganism on Chinese Cabbage (전기분해수 및 이산화염소수 처리에 따른 배추의 미생물 제어 효과)

  • This study evaluated the efficacy of strong acidic electrolyzed water (SAcEW), low alkaline electrolyzed water (LAlEW) and aqueous chlorine dioxide (ACD) for reducing pathogenic bacteria($Escherichia$ $coli$, $Bacillus$ $cereus$, $Salmonella$ Typhimurium, $Stapylococcus$ $aureus$) on Chinese cabbage. Artificially inoculated Chinese cabbage was immersed for 1, 5 and 10 min with TW, NaClO, EW and ACD. Generally, leaves showed more effective reduction than stems. Regarding the inhibitory effect, ACD treatment showed the highest effects rather than other treatments. When Chinese cabbage was immersed for 3 min in sterilized water, it was reduced to a minimum of 1.33 log CFU/g at LAlEW and a maximum of 4.70 log CFU/g at ACD. Compared to NaClO, ACD and LAlEW which showed a reduction of 3.2 log CFU/g ($Sal.$ Typhimurium) and 2.7 log CFU/g($B.$ $cereus$), respectively. Furthermore, the others had similar inhibitory effects compared to NaClO.

Evaluation of the Efficacy of Sanitizers on Food Contact Surfaces Using a Surface Test Method (표면시험법을 이용한 식품접촉표면 재질에 따른 살균소독제의 유효성 평가)

  • The study was undertaken to provide information on the efficacy of sanitizers against bacteria with and without organic road dried on to food contact surfaces using the surface test method which EU and USA are currently implementing as one of their official test methods. Escherochia coli ATCC 10536 or Staphylococcus aureus ATCC 6538 was inoculated on to food contact surfaces, such as stainless steel, polypropylene, and silicon, which was then treated with benzalkonium chloride, sodium hypochlorite, or ethanol as a sanitizer for 5minutes at $20^{\circ}C$. Results indicated that the type of surface had little affected the efficacy of various sanitizers. In addition, 200 ppm of benzalkonium chloride or 200 ppm of sodium hypochlorite showed no definite reduction of bacterial populations in the present of organic load, while 40% ethanol showed reduction to $4\;cfu\;\log_{10}$/carrier or more in viable count in the organic load.

  • To investigate the comparative susceptibility of E. coli and S. aureus strains isolated from Korean foods to sanitizer/disinfectants, the bactericidal efficacies of sodium hypochlorite and benzalkonium chloride were assessed by quantitative suspension tests in both 'clean' and 'dirty' conditions, respectively. The sanitizers/disinfectants achieved over 5-log reduction in viable counts of all strains at the tolerance exemption concentration, except in the case of S. aureus exposure to sodium hypochlorite under dirty condition. There were no significant differences in the susceptibility of antibiotic-resistrant and antibiotic-sensitive isolated strains to the sanitizers/disinfectants (p>0.05).
