Effect of pH values and inoculation amounts for α-glucosidase inhibitory activity in mulberry leaf fermentation

  • Mulberry leaves containing 1-deoxynojirimycin (DNJ) have been recognized as a potentially important source for prevent or treat hyperglycemia. However, DNJ content of natural mulberry leaf are as low as 0.1%. Thus, the most effective method for increasing ${\alpha}$-glucosidase inhibitory activity with the DNJ high-production is needed. In this study, we investigated the influence of ${\alpha}$-glucosidase inhibitory activity according to different pH values (6-9) and inoculation amounts (0.1-0.5%) when Bacillus subtilis cultured on mulberry leaf powder media. We confirmed that ${\alpha}$-glucosidase inhibitory activity was difference according to culture conditions of different pH values, inoculation amounts, and fermentation times. The results of mulberry leaf fermentation according to pH values and inoculation amounts were shown that the optimal conditions for ${\alpha}$-glucosidase inhibitory activity were defined as pH 7 and 9, inoculation amount 0.4%, and incubation until 2 to 4 days. These results can be provided a basic data for the optimal culture conditions increasing ${\alpha}$-glucosidase inhibitory activity from mulberry leaf fermentation.

Screening Test of Wild Mushroom Methanol Extracts for Fibrinolytic and $\alpha-Glucosidase$ Inhibitory Activity

  • We investigated the fibrinolytic and ${\alpha}-glucosidase$ inhibitory activity of 55 wild mushroom methanol extracts. Among them, 14 mushrooms showed fibrinolytic activity. In particular, Amanita virgineoides showed the greatest enzyme activity (3.9 plasmin units/ml) by a fibrin plate assay. The fibrinolytic activities of Suillus pictus and Polypolellus varius were 3.8 plasmin units, and the activity of Gomphus fujisanensis was 2.8 plasmin units. Leccinum extremiorientale and Xerocomus nigromaculatus had the same activity with 2.3 plasmin units. In a ${\alpha}-glucosidase$ inhibitory activity test, Lactarius sp. showed the greatest inhibitory activity at 97.3%. The ${\alpha}-glucosidase$ inhibitory activities of Clitocybe odora, Xerocomus nigromaculatus, Melanoleuca melaleuca, Suillus pictus, and Gyroporus castaneus were 84.3%, 77.9%, 74.6%, 68.7%, and 65.4%, respectively. According to the results, because Suillus pictus and Xerocomus nigromaculatus have strong fibrinolytic and ${\alpha}-glucosidase$ inhibitory activities, the two mushrooms will be used as materials for the development of new biofunctional food.

Inhibitory Effect of Buthus martensi Karsch Extracts on ${\alpha}$-Glucosidase Enzyme

  • While searching for ${\alpha}$-glucosidase inhibitors, the active compound was found in a methanol extract of Burthus martensi Kirsch. The separation of the active compound was performed using various chromatography methods and the physico-chemical properties of the purified compound were characterized. The compound showed very potent inhibitory activity against ${\alpha}-glucosidase$ with an $IC_{50}$ value of $5.3\;{\mu}g/ml$. Lineweaver-Burk plot indicated that its inhibition of ${\alpha}-glucosidase$ was competitive.

Inhibitory Effects of Quinoline Isolated from Ruta chalepensis and Its Structurally Related Derivatives against α-Amylase or α-Glucosidase

  • This study was to isolate an active component of the chloroform fraction from the methanol extract of Ruta chalepensis leaves and to measure inhibitory effects against ${\alpha}$-glucosidase or ${\alpha}$-amylase. The inhibitory compound of R. chalepensis leaves was isolated using chromatographic methods and identified as quinoline. Quinoline and its structurally related derivatives were tested for their inhibitory activities by evaluating the $IC_{50}$ values against ${\alpha}$-amylase or ${\alpha}$-glucosidase and were compared with that of acarbose. Based on the $IC_{50}$ values, quinazoline exhibited the greatest inhibitory activity ($20.5{\mu}g/mL$), followed by acarbose ($66.5{\mu}g/mL$), and quinoline ($80.3{\mu}g/mL$) against ${\alpha}$-glucosidase. In case of ${\alpha}$-amylase, quinazoline had potent inhibitory activity, followed by quinoline ($179.5{\mu}g/mL$) and acarbose ($180.6{\mu}g/mL$). These results indicate that R. chalepensis extract, quinoline, and quinazoline could be useful for inhibiting ${\alpha}$-glucosidase or ${\alpha}$-amylase.

Carbohydrate, Lipid Inhibitory Activity and Antioxidant Activity of Extracts from Several Economic Resource Plants in Vitro

  • The objective of this study was determined to evaluate ${\alpha}$-amylase, ${\alpha}$-glucosidase, pancreatic lipase inhibition in vitro and DPPH radical scavenging activity of the several Korean resources plants. The ${\alpha}$-amylase inhibitory activity of Salicornia herbacea, Erythronium japonicum (flower) and Phragmites communis (root) in water extract showed relatively high 62.8%, 66.5% and 69.3%, respectively. The ${\alpha}$-amylase inhibitory activity of Citrus junos (pericarp) and Cornus officinalis in methanol extract was found to have an effect with 32.8% in Citrus junos (pericarp) and 60.9% in Cornus officinalis. Corylopsis coreana in both water and methanol extract had the highest ${\alpha}$-glucosidase inhibitory activity of 81.7% and 89.5%, while the extract of Portulaca oleracea, Ficus carica and Citrus junos was not measured ${\alpha}$-glucosidase inhibitory activity at given experiment concentration. Depending on the extraction solvent and the plant species, it was observed that there was a significant difference in ${\alpha}$-glucosidase inhibitory activity. The pancreatic lipase inhibitory activity showed relatively higher in the methanol extract than water extract except pericarp of Citrus junos. The DPPH radical scavenging activity of selected plants was much difference between measured plant species, and showed that the increase was proportional to the concentration. These results suggested that selected plants had the potent biological activity on carbohydrate, lipid Inhibitory activity and antioxidant activity, therefore these plant resources could be a good materials to develop medicinal preparations, nutraceuticals or health functional foods for diabetes or obesity.

인삼종자오일의 α-Glucosidase 및 α-Amylase 저해작용 (Inhibitory Effects of Ginseng Seed Oil on α-Glucosidase and α-Amylase Activity)

  • This study was to evaluate the effect of ginseng (Panax ginseng) seed oil on the ${\alpha}$-glucosidase and ${\alpha}$-amylase. Each ginseng seed oils (HE, SE, EE) exhibited a significant inhibitory effect (p<0.001) at all concentrations (10 and 20 mg/ml) on ${\alpha}$-glucosidase activity. HE is the highest inhibitory activity (86.92%) at a concentration of 20 mg/ml, SE and EE showed an inhibitory effect of 77.13% and 65.83%, respectively. And also, Each ginseng seed oils (HE, SE, EE) exhibited a significant inhibitory effect (p<0.001) at all concentrations (1 and 2 mg/ml) on ${\alpha}$-amylase activity. HE is the highest inhibitory activity (89.68%) at a concentration of 2 mg/ml, SE and EE showed an inhibitory effect of 76.99% and 65.70%, respectively.

야생버섯 메탄올추출물의 혈전용해 활성과 α-Glucosidase 저해 활성 (Fibrinolytic and α-Glucosidase Inhibitory Activities of Wild Mushroom Methanol Extracts)

  • 오대산에 자생하는 야생버섯 60종의 메탄올추출물을 이용하여 혈관계 질환과 당뇨병의 치료에 효과 있는 버섯을 확인하기위해, 혈전용해 활성과 ${\alpha}-glucosidase$ 저해 활성을 확인한 결과 11종의 버섯이 혈전용해 활성을 나타냈고, 6종의 버섯이 50% 이상의 ${\alpha}-glucosidase$ 저해 활성을 나타냈다. 뽕나무버섯류가 4.2 plasmin units의 가장 큰 혈전용해 활성을 나타냈으며, 독우산광대버섯, 미치광이 버섯류, 민자주방망이버섯이 2.3 plasmin units, 애괄대버섯은 2.1 plasmin units, 흰보라끈적버섯은 2.0 plasmin units 의 높은 혈전용해 활성을 나타냈다. ${\alpha}-Glucosidase$ 저해 활성 측정결과 노란다발버섯이 98.5%의 가장 큰 저해 활성을 나타냈고 비단그물버섯과 점마개버섯도 60% 이상의 높은 저해 활성을 나타냈다. 본 연구는 뽕나무벗섯류와 노란다발버섯은 혈관계질환 치료용 의약이나 기능성 식품으로 개발될 수 있음을 보여준다.

Antidiabetic Activity of an Ayurvedic Formulation Chaturmukha Rasa: A Mechanism Based Study

  • Objectives: The objective of this study was to evaluate antidiabetic activity of Chaturmukha rasa based on streptozotocin induced diabetes model, alpha amylase inhibitory activity, alpha Glucosidase inhibitory activity and inhibition of sucrase. Methods: Chaturmukha rasa was prepared as per Ayurvedic formulary. Antidiabetic activity was measured in experimentally streptozotocin induced rats. The dose was taken as 45 mg/kg, i.p. The antidiabetic activity of Chaturmukha rasa was compared Triphala Kwatha, a marketed formulation. Further In vitro $\acute{\alpha}$- Amylase Inhibitory Assay, In vitro salivary amylase Inhibitory Assay, In vitro ${\alpha}-Glucosidase$ Inhibitory Assay and In vitro Sucrase Inhibitory Assay was performed with respect to Chaturmukha rasa. The IC50 value was calculated for all the above activity. Results: Streptozotocin with Acarbose showed significant decrease in blood glucose level whereas streptozotocin with Triphala kwatha showed more decrease in blood glucose level than Streptozotocin with Acarbose. The combination of Streptozotocin + Triphala kwatha + Chaturmukha rasa showed a significant decrease in blood glucose level on 21st day. In vitro $\acute{\alpha}$- Amylase Inhibitory Assay the Chaturmukha rasa showed IC50 value $495.94{\mu}l$ when compared with Acarbose $427.33{\mu}l$, respectively. In the ${\alpha}-Glucosidase$ Inhibitory Assay Chaturmukha rasa showed IC50 value $70.93{\mu}l$ when compared with Acarbose $102.28{\mu}l$, respectively. In vitro Sucrase Inhibitory Assay Chaturmukha rasa showed IC50 value $415.4{\mu}l$ when compared with Acarbose $371.43{\mu}l$, respectively. Conclusion: This study supports that Chaturmukha rasa may inhibit diabetes by inhibition of salivary amylase or alpha Glucosidase or sucrase. This may be the mechanism by which Chaturmukha rasa inhibits diabetes. Further this study supports the usage of Chaturmukha rasa for the management of diabetes.

2-Hydroxyquinoline and Its Structural Analogs Show Antidiabetic Effects against α-Amylase and α-Glucosidase

  • This study investigated the inhibitory activities of 2-hydroxyquinoline and its analogs against ${\alpha}$-glucosidase and ${\alpha}$-amylase. Based on the $IC_{50}$ values of 2-hydroxyquinoline analogs tested against ${\alpha}$-glucosidase and ${\alpha}$-amylase, 2-hydroxyquinoline had potent inhibitory activity (64.4 and $130.5{\mu}g/mL$, respectively), while 2-methyl-8-hydroxyquinoline showed weakly inhibitory activity (90.7 and $215.4{\mu}g/mL$, respectively). 2-Methylquinoline demonstrated no activity against ${\alpha}$-glucosidase and ${\alpha}$-amylase. In conclusion, 2-hydroxyquinoline analogs, with the existence of a methyl group and hydroxyl on quinoline, can be useful as a new diabetes treatment.

In vitro $\alpha$-Glucosidase Inhibitory Potential and Antioxidant Activity of Selected Lamiaceae Species Inhabited in Korean Penninsula

  • In the current study, inhibitory activity of 8 selected Korean edible plants of Lamiaceae family against $\alpha$-glucosidases, prepared from rat small intestine acetone powder was evaluated. Total flavonoids and oxygen radical absorbance capacity (ORAC) were also investigated. Methyl alcohol extracts of Scutellaria indica (SI) had the highest $\alpha$-glucosidase inhibitory activity relevant for potentially managing hyperglycemia, followed by Clinopodium gracile (CG) and Thymus quinquecostatus (TQ). These 3 species also showed significant antioxidant activity in ORAC system. The $\alpha$-glucosidase inhibitory activity of the extracts was compared to selected phenolics. Among the standard phenolics tested quercetin which was major flavonoid in the extracts had the highest $\alpha$-glucosidase inhibitory activity. CG, TQ, and SI which had high quercetin content and ORAC values also exhibited significant sucrase inhibitory activity. Results suggested that selected 3 Korean Lamiaceae species have the potential development of effective dietary strategy for postprandial hyperglycemia and oxidative stress-linked diabetes complications.
