본 연구는 돼지 골수에서 존재하는 조혈 줄기 세포 및 전구 세포를 이용해 이종 동물 모델인 태아 마우스 복강 생체 이식을 통하여 돼지 조혈 세포의 이종 조혈 조직에서의 증식과 분화 특성을 규명하였다. 선천성 면역 부전 마우스인 NOD/SCID 마우스 태아 조혈 환경에 돼지 골수 유래 조혈 줄기 세포 및 전구 세포를 이식하고, 이식 후 5주령에 마우스 조혈기관에서의 돼지 조혈 세포의 증식과 분화 특성을 돼지 특이적 항체 면역 염색으로 유세포 분석을 실시한 결과, 마우스 조혈 조직인 골수, 흉선, 간장, 비장 및 림파절에서 돼지 조혈 세포의 분화 및 증식이 관찰되었다. 특히 돼지의 T 면역세포가 골수계 세포에 비해서 높은 chimerism이 관찰되어 태생 초기의 NOD/SCID 조혈 환경에 의한 특이적 T 면역세포의 증식에 적합한 조혈 환경을 제공하고 있다는 사실이 밝혀졌다. 본 마우스 신생 NOSD/SCID 복강 이식 동물 모델을 이용해 돼지 T 면역세포의 분화 발달 연구 및 이종 장기 이식 기전 연구에 좋은 모델로서 활용이 기대된다.

조혈 줄기 세포에의 효과적인 유전자 전달은 유전자 치료의 새로운 가능성을 제시할 수 있다. 레트로바이러스를 이용한 유전자 전달 기술은 많은 기초 연구와 임상 시도가 이루어진 대표적인 바이러스이다. 그러나 현재 사용되고 있는 in vitro에서의 조혈 줄기 세포에의 유전자 도입은 조혈 줄기 세포의 분화 유도, 자기 복제 능력과homing 능력의 저하 등 많은 문제점이 있다. 본 연구는 이러한 문제점을 극복하기 위한 방법으로서 마우스의 대퇴골에 직접 레트로바이러스를 이식하는 IBM (Intra-Bone Marrow) 방법을 이용하여 조혈 줄기 세포에의 효과적인 유전자 도입을 시도하였다. IBM 이식 2주 후 마우스의 각 조직을 분석한 결과, 골수뿐 아니라 림파절, 비장, 간장 세포 등에서 유전자가 안정적으로 발현하는 것을 관찰하였다. 또한, $6.4{\pm}2.7%$의 골수조직 존재 조혈줄기/전구세포에서 도입된 유전자가 안정적으로 발현하고 있는 사실을 확인하였다. 본 연구의 결과를 바탕으로 IBM 이식 방법을 이용한 생체 조직 내 레트로바이러스의 유전자 도입은 조혈 줄기 세포를 이용한 유전자 치료에 매우 효과적인 방법이라는 사실을 시사해주고 있다.

Pronuclear DNA microinjection has been the most universal method in transgenic animal production but its success rate of transgenesis in mammals are extremely low. To address this long-standing problem, we used retrovirus- and lentivirus-based vectors carrying the enhanced green fluorescent protein (EGFP) gene under the control of ubiquitously active cytomegalovirus (CMV) promoter to deliver transgenes to bovine embryos. The rate of transgenesis was evaluated by counting EGFP positive blastocysts after injection of concentrated virus stock into the perivitelline space of the bovine oocytes in metaphase II. Among two different types of lentivirus vectors derived from FIV (feline immunodeficiency virus) and HIV (human immunodeficiency virus), the former scored the higher gene transfer efficiency; almost 100% of the blastocysts developed from the oocytes infected with FIV-based vector were EGFP positive. As for the vectors derived Com HIV lentivirus, the transgenesis rate of the blastocysts was reduced to 39%.

Bone marrow mesenchymal stem cells (BMMSCs) have the capacity for self-renewal and differentiation into a variety of cell types. They represent an attractive source of cells for gene and cell therapy. The purpose of this study is to direct the specific expression of the DsRed reporter gene in $Sca-1^+$ BMMSCs differentiated into a cardiomyogenic lineage. We constructed the prMLC-2v-DsRed vector expressing DsRed under the control of the 309 tp fragment of the rat MLC-2v 5'-flanking region. The specific expression of the DsRed reporter gene under the transcriptional control of the 309 bp fragment of the rat MLC-2v promoter was tested in 5-azacytidine healed-$Sca-1^+$ BMMSCs over 2 weeks after the prMLC-2v-DsRed transfection. The prMLC-2v-DsRed was specifically expressed in the $Sca-1^+$ BMMSCs with cardiomyogenic lineage differentiation and it demonstrates that the 309 bp sequences of the rat MLC-2v 5'-flanking region is sufficient to confer cardiac specific expression on a DsRed reporter gene. The cardiac-specific promoter-driven reporter vector provides an important tool for the study of stem cell differentiation and cell replacement therapy in ischemic cardiomyopathy.

본 연구는 체세포 복제란 이식우의 분만에 있어서 혈중 스테로이드호르몬, TGF-${\beta}_1$ 농도와 분만지연의 상관 관계를 살펴보고자 실시하였다. 대조군으로는 인공수정(AI)을 통하여 임신한 암소(cow)들을 사용하였다(AI-R). 모든 AI-R들은 자연분만(n=5, 임신 $284{\pm}0.71$일)을 하였다. 분만징후를 보이지 않는 체세포 복제란 이식우(n=5, SCNT-R)들은 분만 예정일보다 10일 정도 지난 임신 292일째에 제왕절개(Caesarean section, C-sec)를 실시하여 분만하였다. 혈액 및 태반 샘플을 분만 전.후에 채취하여 형태 및 중량 등을 측정하였다. 혈장호르몬인 Progesterone(P4)와 Estradiol-$17{\beta}$(E2) 농도는 방사선동위원소 면역 분석 시험(RIA) 방법을 이용하여 측정하였다. 혈장 및 태반분엽의 TGF-${\beta}_1$ 농도는 ELISA 방법으로 측정하였다. SCNT-R에서 회수한 태반의 무게는 AI-R의 것과 비교하여 유의적으로 무거웠다(p<0.05). 분만 직전 SCNT-R들의 혈장 내 P4 농도는 AI-R들의 그것과 비교하여 유의하게 높았다(p<0.01). 하지만 SCNT-R들의 혈장 내 E2 농도는 AI-R과 비교하여 상대적으로 낮게 나타났다(p<0.01). 한편, 분만 전.후 SCNT-R들에서 혈장 또는 태반분엽의 TGF-${\beta}_1$ 단백질 발현 수준은 AI-R들과 비교하여 각각 유의적으로 높은 수준을 유지하였다(p<0.01). 이상의 결과를 종합하여 보면, 분만 시 P4 및 E2의 이상 발현과 높은 수준의 혈장 및 태반 내 TGF-${\beta}_1$ 단백질은 체세포 복제태아의 분만지연을 야기하는 중요한 요인들 중의 하나일 것이라 사료된다.

DNA 메틸화는 조직특이적인 유전자 조절에 관여하고, 정상적인 배 발달에 필수적이다. POU5F1은 octamer-binding transcription factor 4 (Oct-4)를 encode하며, 초기 분화에 중요한 전사인자이다. 본 실험에서 소의 Oct-4가 조직특이적이고 발달의존적인 epigenetic 표지 인지를 검토하고자, 착상 전 수정란에서 Oct-4 전사산물과 상류 promoter 영역의 CpGs의 메틸화를 조사하였다. Oct-4 전사산물은 정자 그리고 2-cell에서 8-cell 수정란까지 낮은 수준으로 존재하지만, 상실배와 배반포에서 높게 검출되었다. 이러한 결과는 배 발달 과정의 상실배 단계에서 Oct-4의 de novo 발현이 시작됨을 의미한다. Oct-4 상류 promoter 영역에는 메틸화 가변 영역 (tissue-dependent differentially methylated region, T-DMR)이 존재한다. Oct-4 메틸화 가변 영역의 메틸화 상태는 정자, 성체 체조직과 난자에서 서로 다르고, 수정란으로부터 배반포 단계까지 변화하였는데, 이는 착상 전 초기 배 발달 과정에 active 메틸화와 탈메틸화가 일어남을 의미한다. 이상의 결과, Oct-4 유전자 상류 promoter 영역은 DNA 메틸화의 타깃이고, 그 메틸화 상태는 소 수정란 발달 동안에 다양하게 변화한다.

Most studies on transgenic bioreactors have focused on expression levels of interest genes. In this study we examined whether transgenic bioreactors would inherit expression level of the Oansgene to long-term generations independently of transgene sources. We employed three transgenic mice, which were separately reported, carrying different transgenes and copy numbers, 27 kb of hLF and 22 kb of hIL-10 genomic sequences, and 1.3 kb of hTPO cDNA, respectively. Three females of the transgenic lineages crossbred with a wild-type male up to 20 generations to test transgenic frequencies of their progenies and to determine expression levels of the transgenes. Ultimately, transmission rates of kLF, hIL-10, and hTPO were $64.3{\pm}7.0$, $59.3{\pm}9.8$, and $56.1{\pm}9.7$, respectively, appeared following Mendelian pattern of inheritance. Notably, we found that levels of expressions of hLF, hIL-10, and hTPO in milk were sustained to high numbers of generations. No transgene silencing of expression was observed in every generations of all transgenic mice. In conclusion, we suggest that once established animal bioreactors could consistently transmit the transgene to continual generations, without loss of expressional activity, independently of transgene sources.

We have previously produced transgenic (TG) mice expressing the human lactoferrin (hLF), interleukin-10 (hIL-10), and thrombopoietin (hTPO) proteins in the milk. In this study, we examined whether simple crossbreeding between two kids of a single transgenic mouse can produce double transgenics co-expressing two human proteins.. The hLF male, and the hIL-10 male were crossbred with the hIL-10 and hTPO females, and the hTPO female, respectively. PCR analysis for genotyping showed 32%, 23% and 24% double transgenic rates for hLF/hIL-10, hLF/hTPO, and hIL-10/hTPO transgenes, respectively. We analyzed the expression levels of the human proteins from double transgenic mice and compared those with their single transgenic siblings. All double transgenic co-expressed two human proteins at comparable levels to singles', unless hTPO was not co-expressed: for hLF, 1.1 mg/ml in hLF/hIL-10, whereas 0.5 mg/ml in hLF/hTPO; for hIL-10, 4.1 mg/ml in hIL-10/hLF, whereas 1.4 mg/ml in hIL-10/hTPO. Ihe downregulation of hTPO to half level of singles' was observed in double transgenic mice. The possible reason why hTPO co-expressed might lead to down-regulation of another human protein was discussed. These results suggested that double transgenic generated by crossbreeding between two singles' could be useful system for bioreactor.

본 연구는 체세포 복제 한우 송아지의 생산에서 생시체중과 생존성과의 관계에 대하여 살펴보고자 실시하였다. 293두의 한우 대리모에 580개의 복제란을 이식하였다. 복제란의 임신율은 이식 후 50일까지 72.3%로 높았으나, 이후 급격하게 감소하였다. 평균 임신 기간은 복제 송아지에서 287일($279{\sim}295$일)이었으며, 인공수정 송아지도 287일($255{\sim}293$일)로 각각 나타났다. 복제 송아지의 생시체중(30.3kg)은 인공수정 송아지(23.7kg)에 비하여 유의하게 높은 것으로 나타났다(p<0.05). 자연분만(n=17, 29.9kg)과 제왕절개(n=14, 32.3kg)로 태어난 복제 송아지의 생시체중은 차이가 없었다. 하지만, 생후 175일 이전에 사망한 복제 송아지(n=18, 32.8kg)의 생시체중이 175일 이상 생존한 복제 송아지(n=11, 28.3kg)보다 유의적으로 높게 나타났다(p<0.05). 흥미로운 점은 생시체중이 15kg 이하(n=5) 또는 35kg 이상(n=9)인 복제 송아지들은 모두 생후 175일 이전에 폐사하였다. 생후 175일 이전에 폐사한 복제 송아지들(n=20)의 사망 원인은 미성숙 개체 2두(10.0%), 폐와 간 이상 2두(10.0%), 폐의 원인 4두(20.0%), 기형 4두(20.0%), 출생 후 급사(sudden death syndrome) 4두(20.0%) 및 기타 원인불명이 4두(20.0%) 등으로 분류되었다. 이상의 결과를 종합하여보면, 복제 송아지의 정상 생시체중이 6개월 이상을 생존하는데 가장 중요한 요소들 중의 한 가지임을 확인하였다.

본 연구는 정액의 보존 기간 동안 정액의 질적 변화를 알아보고자 시행하였다. 돼지 정액을 Beltsville Thawing Solution (BTS)에 희석한 후 $17^{\circ}C$에서 5일 동안 보존하였다. 보존 기간 동안 정자의 운동성(%)과 linearity는 3일째부터 유의하게 감소하였으나, 다른 운동 역학 변수에서는 유의적 변화를 나타내지 않았다. 또한, 5일 동안 정액을 보존할 경우 첨체의 온전성에도 변화가 없었다. 그러나 제 4일째부터 첨체 변화가 야기된 정자는 유의적으로 증가하였으나, 수정능 획득이 일어난 정자는 유의적으로 감소하였다. 정액의 보존 기간 동안 첨체의 온전성의 유의적 변화가 없었다. 즉, 보존 기간 3일동안 정자의 질적 운동성 및 첨체 온전성에는 유의적인 변화가 없었으므로 상업용 돼지 액상정액은 $17^{\circ}C$에서 적어도 3일간 수정능력을 만족스럽게 유지함을 보여준다.

Members of the Vps (Vacuolar protein sorting) protein family involved in the formation of the retromer complex have been discovered in a variety of species such as yeast, mouse, and human. A mammalian retromer complex is composed of Vps26, Vps29, and Vps35 proteins and plays and important role in cation-independent mannose-6-phosphate receptor retrieval from the endosome to the trans-Golgi network. In this study, we have identified the full-length sequences of the retromer components of Vps26, Vps29, and Vps35 in micro pigs. The cDNA sequences of these retromer components have been determined and the result showed there is 99% homology among the component counterparts from mouse, micro pigs, and humans. In addition, the retromer complexes formed with hetero-components were found in the brain of micro pigs. Based on above results, we suggest mammalian Vps components are well conserved in micro pigs.
