Saprolegniasis is one of the most devastating oomycete diseases in freshwater fish which is caused by species in the genus Saprolegnia including Saprolegnia parasitica. In this study, we isolated the strain of S. parasitica from diseased rainbow trout in Korea. Morphological and molecular based identification confirmed that isolated oomycete belongs to the member of S. parasitica, supported by its typical features including cotton-like mycelium, zoospores and phylogenetic analysis with internal transcribed spacer region. Pathogenicity of isolated S. parasitica was developed in embryo, juvenile, and adult zebrafish as a disease model. Host-pathogen interaction in adult zebrafish was investigated at transcriptional level. Upon infection with S. parasitica, pathogen/antigen recognition and signaling (TLR2, TLR4b, TLR5b, NOD1, and major histocompatibility complex class I), pro/anti-inflammatory cytokines (interleukin $[IL]-1{\beta}$, tumor necrosis factor ${\alpha}$, IL-6, IL-8, interferon ${\gamma}$, IL-12, and IL-10), matrix metalloproteinase (MMP9 and MMP13), cell surface molecules ($CD8^+$ and $CD4^+$) and antioxidant enzymes (superoxide dismutase, catalase) related genes were differentially modulated at 3- and 12-hr post infection. As an anti-Saprolegnia agent, plant based lawsone was applied to investigate on the susceptibility of S. parasitica showing the minimum inhibitory concentration and percentage inhibition of radial growth as $200{\mu}g/mL$ and 31.8%, respectively. Moreover, natural lawsone changed the membrane permeability of S. parasitica mycelium and caused irreversible damage and disintegration to the cellular membranes of S. parasitica. Transcriptional responses of the genes of S. parasitica mycelium exposed to lawsone were altered, indicating that lawsone could be a potential anti-S. parasitica agent for controlling S. parasitica infection.
Previously, agar obtained from Gelidum sp. has a small molecular weight and has the disadvantage of inherent viscosity properties and poor functionality as a dietary fiber. In order to improve aforementioned disadvantages, agar having a fluidity that can be added to food at a higher concentration that a powder agar having a gelling property at low concertation was manufactured. In addition, the anti-inflammatory activity of agar hydrolysates was evaluated to confirm their potential as a functional material. As a result, agar hydrolysates significantly reduced NO levels secreted by LPS-activated macrophages and inhibited the expression of iNOS and COX-2, which are inflammatory mediators that regulates NO secretion in macrophages. Furthermore, in in vivo zebrafish embryos model results demonstrated significant reduction of LPS induced NO production after the treatment of agar hydrolysate hydrolyzed for 360 min. In addition, ROS production and cell death by stresses were also reduced in LPS-exposed embryos after the treatment of agar hydrolysis product hydrolyzed for 360 min. Taken together, agar hydrolysate hydrolyzed for 360 min can be easily added into food due to their fluidity and used as a food ingredient that inhibits inflammation due to their anti-inflammatory property.
We investigated the protective effects of 3-bromo-4,5-dihydroxybenzaldehyde (BDB) from Polysiphonia morrowii Harvey against hydrogen peroxide ($H_2O_2$)-induced apoptosis in Vero cells. BDB exhibited scavenging activity for DPPH, hydroxyl, and alkyl radicals. BDB also inhibited $H_2O_2$-induced lipid peroxidation, cell death, and apoptosis in Vero cells by inhibiting the production of ROS. To evaluate the molecular mechanisms of apoptosis inhibition, the expression of Bax/Bcl-xL and $NF-{\kappa}B$ was assessed by western blot assay. BDB significantly suppressed the cleavage of caspase-9 and PARP and reduced Bax levels in $H_2O_2$-induced Vero cells. Besides, BDB suppressed the phosphorylation of $NF-{\kappa}$B and the translocation of p65 in $H_2O_2$-induced cells. Furthermore, we evaluated the effect of BDB on ROS production, cell death, and lipid peroxidation in an $H_2O_2$-stimulated zebrafish embryo model. Taken together, these results indicated that ROS generation and cell death were significantly inhibited by BDB in zebrafish embryos, thereby proving that BDB exerts excellent antioxidant activity in vitro and in vivo.
지구상에서 가장 오래된 해양 미세조류로 알려진 스피룰리나는 인체에 필요한 영양성분을 대부분 함유하고 있는 것으로 알려져 있다. 그 구성성분으로는 phycocyanin, chlorophyll, β-carotene 과 같은 다양한 물질을 다량 함유한다고 보고되고 있으며, 노화 및 미백효과가 있다고 알려져 있다. 본 연구에서는 스피룰리나 정제수 추출물의 UVB로 유도된 활성산소종 ROS (reactive oxygen species) 소거능과 항산화능을 확인하였다. 스피룰리나 정제수 추출물 0.05, 0.10, 0.50 1.0 mg/mL의 DPPH 라디칼 소거능, FRAP 환원능 및 ABTS+ 라디칼 소거능을 측정하여 항산화효과를 확인하였다. 대체실험동물모델인 Zebrafish를 이용하여 스피룰리나 정제수 추출물을 0.05, 0.10, 0.50 mg/mL 농도로 처리하여 응고율, 부화율, 심장독성을 측정하였으며, 스피룰라나 추출물을 0.05, 0.10, 0.50 mg/mL 농도로 처리한 후, DCFH-DA로 염색하여 UVB로 유도된 ROS 저해효과를 확인하였다. 항산화효과 측정 결과 양성대조군인 ascorbic acid와 비교시 DPPH, Frpa, ABTS 모두 농도 의존적으로 항산화 효과가 있음을 확인하였다. 대체 실험동물인 Zebrafish를 이용하여 응고율과 부화율, 심박수를 측정한 결과 0.5 mg/mL을 제외한 0.05, 0.10 mg/mL에서는 대조군과 비교 시 독성이 없음을 확인 하였다. UVB로 유도된 Zebrafish의 ROS 소거능은 양성대조군에 비해 높은 ROS 감소 효과를 나타내었다. 본 연구의 결과는 스피룰라나 정제수 추출물이 자외선 및 피부 보호 화장품 소재로 사용가치가 있음을 시사한다.
본 연구는 오디씨 에탄올 추출물(MSE)의 멜라닌 합성 저해 효과를 밝히는 것이다. 먼저 MSE의 melan-a 세포를 이용한 멜라닌 합성 저해 실험결과, 독성을 보이지 않는 $10{\mu}g/mL$의 농도까지 멜라닌 합성 저해 효과를 나타내었다. 또한 tyrosinase, tyrosinase-related protein-1 (TRP-1) 단백질의 발현이 저해되었으며, extracelluar signal-regulated protein kinase (ERK)의 발현을 농도 의존적으로 증가시키는 MSE의 기전을 확인하였다. 뿐만 아니라, 제브라피쉬를 이용한 in vivo 모델의 실험에서도 색소 발생이 저해됨을 관찰하였다. 따라서 오디씨로 부터 획득한 에탄올 추출물이 ERK 단백질의 발현으로 인해 멜라닌 생합성을 억제할 수 있음을 밝혔다.
Background: Ginsenoside Rg1 is a class of steroid glycoside and triterpene saponin in Panax ginseng. Many studies suggest that Rg1 suppresses adipocyte differentiation in 3T3-L1. However, the detail molecular mechanism of Rg1 on adipogenesis in 3T3-L1 is still not fully understood. Methods: 3T3-L1 preadipocyte was used to evaluate the effect of Rg1 on adipocyte development in the differentiation in a stage-dependent manner in vitro. Oil Red O staining and Nile red staining were conducted to measure intracellular lipid accumulation and superoxide production, respectively. We analyzed the protein expression using Western blot in vitro. The zebrafish model was used to investigate whether Rg1 suppresses the early stage of fat accumulation in vivo. Results: Rg1 decreased lipid accumulation in early-stage differentiation of 3T3-L1 compared with intermediate and later stages of adipocyte differentiation. Rg1 dramatically increased CAAT/enhancer binding protein (C/EBP) homologous protein-10 (CHOP10) and subsequently reduced the $C/EBP{\beta}$ transcriptional activity that prohibited the initiation of adipogenic marker expression as well as triglyceride synthase. Rg1 decreased the expression of extracellular signal-regulated kinase 1/2 and glycogen synthase kinase $3{\beta}$, which are also essential for stimulating the expression of $CEBP{\beta}$. Rg1 also reduced reactive oxygen species production because of the downregulated protein level of nicotinamide adenine dinucleotide phosphate hydrogen (NADPH) oxidase 4 (NOX4). While Rg1 increased the endogenous antioxidant enzymes, it also dramatically decreased the accumulation of lipid and triglyceride in high fat diet-induced obese zebrafish. Conclusion: We demonstrated that Rg1 suppresses early-stage differentiation via the activation of CHOP10 and attenuates fat accumulation in vivo. These results indicate that Rg1 might have the potential to reduce body fat accumulation in the early stage of obesity.
후각기관은 주변 환경의 다양한 화학물질을 감지하는 기관으로 생존, 종족번식에서 감정에 이르기까지 다양하고 중요한 역할을 하고 있다. 유전적, 환경적 등 다양한 요소에 의해 후각장애가 발생할 수 있으며, 일시적인 경우에는 약물치료 등으로 회복될 수 있지만, 신경세포에 문제가 생긴 영구적인 손상의 경우는 치료가 어렵다. 따라서, 신경세포의 사멸을 억제하거나 재생을 유도하는 치료제의 개발이 필요하다. 본 연구에서는 후각신경세포 특이적으로 GFP 형광단백질을 발현하는 형질전환동물을 제작하여 생체 내 후각신경세포를 관찰하고자 하였다. 또한, 다양한 화학물질을 처리하여 후각신경세포 손상을 인위적으로 유도할 수 있는 방법을 고안하였고, 후각신경세포의 손상 및 재생 과정을 실시간으로 모니터링하였다. 본 연구를 통해 확립된 후각신경세포의 손상 및 재생 모니터링 시스템은 향후 후각신경세포 재생 메커니즘 연구 및 치료제 개발에 유용하게 사용될 것으로 기대된다.
본 연구는 근생성 및 근위축 완화효능을 가진 유효소재 발굴의 필요성에 의해 polygalacin D가 근원세포 분화 및 미토콘드리아에 미치는 영향과 항암제 유도 근위축에 대한 완화효과를 각각 세포 및 동물실험을 통해 확인하고자 하였다. 그 결과, polygalacin D는 다핵을 지닌 근관세포의 수와 분화 종결인자인 MHC isoforms의 발현량을 증가시켰고 근육 내 단백질 분해 인자인 MuRF1, Smad2/3의 발현량은 유의적으로 감소시켰다. 또한 미토콘드리아 생합성 조절인자인 Pgc1α의 발현은 증가시키고 미토콘드리아 분열인자인 Drp1과 Fis1의 발현은 감소시켰다. 한편 zebrafish 동물모델을 통해 항암제 유도 근위축에 대한 개선효과를 확인한 결과, polygalacin D는 항암제에 의해 유도된 근위축과 미토콘드리아 손상을 완화시켰다. 이상의 결과들은 polygalacin D가 미토콘드리아 기능 증진을 매개로 근원세포 분화 촉진 및 근육 단백질 분해 저하 효과를 지닐 뿐만 아니라, 미토콘드리아 손상을 개선하여 항암제로 유도된 근위축에 대한 완화 효과를 나타냄을 시사한다. 따라서 본 연구를 통해 polygalacin D가 근생성 및 근위축 예방과 치료를 위한 잠재적인 유효소재로서의 가능성을 제시하였다.
가습기 살균제 성분(PHMG, PGH, CMIT/MIT)의 노출에 의한 다양한 장기에 대한 독성들에 대해서 피해사례는 계속 증가하고 있으나, 세포모델과 동물모델에서의 연구와 보고는 아직 부족한 실정이다. 심혈관 독성, 간 독성, 배아 독성에 대해서는 최근 알려져 있으나 뇌신경 독성과 피부 독성에 대해서는 상대적으로 적게 알려져 있다. 본 연구에서는 이들 세 가지 성분들의 피부 독성과 뇌신경 독성을 사람 피부섬유세포와 제브라피쉬 동물모델을 대상으로 각각 평가하였다. 사람피부섬유세포에 세 가지의 성분들을 0, 2, 4, 6, 8, 16 mg L-1 (최종농도)로 처리하였을 때, 세포 생존율은 PHMG가 33%로 가장 낮았고, PGH가 49%, CMIT/MIT가 40%의 생존율을 보였다. 세포배양액 내의 산화물을 정량해본 결과, PHMG 처리된 세포가 28 nmol MDA로 가장 높았고, PGH가 13 nmol MDA, CMIT가 21 nmol MDA를 보였다. 제브라피쉬 사육수조에 PHMG, PGH, CMIT를 40 mg L-1의 최종농도가 되도록 희석한 후, 제브라피쉬를 30분간 노출시킨 후 중뇌의 광시개영역(optic tectum)을 횡면 미세절단하여 산화물의 생성정도를 비교해본 결과, CMIT/MIT를 처리한 그룹에서 대조군 대비 17배 많은 산화물의 생성이 있었고, PGH를 처리한 그룹에서는 15배, PHMG를 처리한 그룹에서는 11배 많은 산화물이 관찰되어 심각한 뇌신경계 독성을 보여주었다. 결론적으로 세 가지 종류의 가습기 살균제 성분들에서 모두 심각한 피부세포 독성과 뇌신경계 독성이 나타났는데, 피부 독성은 특히 PHMG가, 뇌신경계 독성은 특히 CMIT/MIT가 가장 심각하였다. 이들 결과들은 가습기 살균제에 노출된 어린이들이 뇌신경계 독성을 통하여 언어장애, 운동장애, 발달장애 등을 겪게 될 수도 있음을 실험적으로 제시한다.
Ginsenosides, which are the active materials of ginseng, have biological functions that include anti-osteoporotic effects. Aqueous ginseng extract inhibits osteoclast differentiation induced by receptor activator of NF-${\kappa}B$ ligand (RANKL). Aqueous ginseng extract produces chromatography peaks characteristic of ginsenosides. Among these peaks, ginsenoside Re is a major component. However, the preventive effects of ginsenoside Re against osteoclast differentiation are not known. We studied the effect of ginsenoside Re on osteoclast differentiation, RANKL-induced tartrate-resistant acid phosphatase (TRAP) activity, and formation of multinucleated osteoclasts in vitro. Ginsenoside Re hampered osteoclast differentiation in a dose-dependent manner. In an in vivo zebrafish model, aqueous ginseng extract and ginsenoside Re had anti-osteoclastogenesis effects. These findings suggest that both aqueous ginseng extract and ginsenoside Re prevent bone resorption by inhibiting osteoclast differentiation. Ginsenoside Re could be important for promoting bone health.
