The Structural Characterization of Recombinant Bovine Somatotropin Expressed in Escherichia coli (재조합 소성장호르몬의 구조적 특성)

  • In this paper we have described the structural characterization of recombinant bovine somatotropin produced in Escherichia coli. Recombinant bovine somatotropin consists of 191 amino acid residues with a calculated molecular weight of 21,802 Da. For fragmentation of recombinant bovine somatotropin, we have used trypsin, Staphylococcus aureus V8 pretease, CNBr, and mild acid hydrolysis method. Digestion and cleavage with these proteases and chemicals yielded peptides of various size for amino acid sequence determination. The N-terminal sequence analysis was carried out up to thirty residues. Because the design of the recombinant bovine somatotropin gene for expression was such that the coding sequence begins with an initiation codon, AUG, before Ala, the first amino acid of bovine somatotropin, we could expect the initial amino acid as N-formyl Met. But the first amino acid of this protein, expressed in E. coli cells as inclusion bodies, was Ala. And the amino acid composition of RP-HPLC purified recombinant bovine somatotropin was determined and no essencial difference was observed. The amino acid sequence of the recombinant bovine somatotropin was identical to that predicted from its recombinant gene. There was no processing or replacement of amino acid residues in recombinant bovine somatotropin expressed in E. coli. The hydropathy plot of recombinant bovine somatotropin revealed a hydrophobic region at the NH2-terminus and hydrophilic region at the COOH-terminus. The E. coli expression system is thought to be valuable for the expression of recombinant bovine somatotropin because protein was processed to remove the N-terminal Met residue by methionyl-aminopeptidase autonomously.

Characterization and N-Terminal Amino Acid Sequence Analysis of Catechol 2,3-dioxygenase Isolated from the Aniline Degrading Bacterium, Delftia sp. JK-2 (Aniline 분해세균 Delftia sp. JK-2에서 분리된 catechol 2,3-dioxygenase의 특성 및 N-말단 아미노산 서열분석)

  • The aim of this work was to investigate the characterization and sequence of catechol 2,3-dioxygenase isolated from Delfia sp. JK-2, which could utilize aniline as sole carbon, nitrogen and energy source. In initial experiments, several characteristics of C2,3O separated with ammonium sulfate precipitation, DEAE-sepharose were investigated. Specific activity of C2,3O was approximately 4.72 unit/mg. C2,3O demonstrated its enzyme activity to other substrates, catechol and 4-methylcatechol. The optimum temperature of C2,3O was $$Cu^{2+}$^{\circ}C$, and the optimal pH was approximately 8. Metal ions such as $Ag^{+}$, $Hg^{+}$, and $Cu^{2+}$ showed inhibitory effect on the activity of C2,3O. Molecular weight of the enzyme was determined to approximately 35 kDa by SDS-PAGE. N-terminal amino acid sequence of C2,3O was analyzed as $^{1}MGVMRIG-HASLKVMDMDA- AVRHYENV^{26}$, and exhibited high sequence homology with that of C2,30 from Pseudomonas sp. AW-2, Comamonas sp. JS765, Comamonas testosteroni and Burkholderia sp. RPO07. PCR product was amplified with the primers derived from N-terminal amino acid sequence. In this work, we found that the amino acid sequence of Delftia sp. JK-2 showed high sequence homology of C2,3O from Pseudomonas sp. AW-2 (100%) and Comamonas sp. JS765 (97%).

  • We consider bilateral operator weighted shift T on $L^2$(K) with weight sequence ${[A_{n}]_{n=-{\infty}}}^{\infty}$ of positive invertible diagonal operators on K. We give a characterization for T to be hypercyclic, and show that the conditions are far simplified in case T is invertible.

  • We provide quantitative two weight estimates for the commutator of the Hilbert transform under certain conditions on a pair of weights (u, v) and b in $Carl_{u,v}$. In [10] and [11], Bloom's inequality is shown in a modern setting, and the boundedness of the commutators is provided by assuming both weights u, v are $A_2$ and $b{\in}BMO_{\rho}$. In the present paper we show that the condition on b can be replaced by $Carl_{u,v}$ by using the joint $A^d_2$ condition.

  • In this paper we first characterize the hereditarily hypercyclicity of the unilateral (or bilateral) weighted shifts on the spaces $L^2(\mathbb{N},\mathcal{K})$ (or $L^2(\mathbb{Z},\mathcal{K})$) with weight sequence {$A_n$} of positive invertible diagonal operators on a separable complex Hilbert space $\mathcal{K}$. Then we give the necessary and sufficient conditions for the supercyclicity of those weighted shifts, which extends some previous results of H. Salas. At last, we give some conditions for the supercyclicity of three different weighted shifts.

  • Let $W_{\alpha}$ be a weighted shift with positive weight sequence ${\alpha}=\{\alpha_i\}_{i=0}^{\infty}$. The semi-cubical hyponormality of $W_{\alpha}$ is introduced and some flatness properties of $W_{\alpha}$ are discussed in this note. In particular, it is proved that if ${\alpha}_n={\alpha}_{n+1}$ for some $n{\geq}1$, ${{\alpha}_{n+k}}={\alpha}_n$ for all $k{\geq}1$.

  • Let ${\alpha}:1,(1,{\sqrt{x}},{\sqrt{y}})^{\wedge}$ be a weight sequence with Stampfli's subnormal completion and let $W_{\alpha}$ be its associated weighted shift. In this paper we discuss some properties of the region ${\mathcal{U}}:=\{(x,y):W_{\alpha}$ is semi-cubically hyponormal} and describe the shape of the boundary of ${\mathcal{U}}$. In particular, we improve the results of [19, Theorem 4.2].

  • Injection Molding is the most effective process for mass production of plastic parts. The injection molding process is composed with several steps such as Filling, Packing, Holding, Cooling, Ejecting. Among them, filling and packing process should be considered carefully to improve accuracy of dimension, surface quality of plastic parts. Usually the quality above-mentioned is managed with weight of part after molding on the field. In this paper, a series of experiment for molding automotive front bumper was conducted with cavitity temperature sensor to optimize switch-over time(V-P switching), hot runner vale gate sequence time during filling and packing step for the purpose of uniform quality, weight at every molding. As a result, it was found that it is effective method to use temperature sensor in injection molding for quality control of plastic molding.

Antioxidative Activity of Low Molecular Weight Biocompounds Purified from Anchovy (Engraulis japonicus) Sauce (멸치액젓 유래 저분자 물질의 항산화활성)

  • Antioxidative activities of low molecular weight biocompounds purified from anchovy (Engraulis japonicus) sauce fermented at $15\pm3^{\circ}C$ for 5 years were investigated. The fermented anchovy sauce showed 5 peaks on gel chromatography pattern. Antioxidant activity of peak 2 was $82.7\%$ followed by $42.6\%$ of peak 1. Main antioxidant compounds of peak 1 were glutamic acid and lysine, but those of peak 2 were not confirmed by amino acid sequence analysis.

  • This study presents the decision of priority for optimal design guide to improve disassembly process. Disassembly process is divided into recognition, transfer and disassembly of assembly point and recognition, transfer and remove of grasp point. Significant influential factors are derived from analyzing the above process. And those factors are used for making the check list to evaluate the properties of parts in each process. Furthermore, the weight with considering disassembly process is also used to determine weight of each process. On the base of the above sequence, qualitative score of disassemblability of each process that is enabled to compare different disassembly processes can be acquired. Ultimately the score helps to decide the priority of design guide for disassembly process.
