고추에서 분리한 오이모자이크바이러스(CMV-GTN)의 특성과 고추 품종의 저항성 평가 (Characteristics of Cucumber mosaic virus-GTN and Resistance Evaluation of Chilli Pepper Cultivars to Two Cucumber mosaic virus Isolates)

  • 오이모자이크바이러스(CMV)는 고추에 심한 피해를 주는 바이러스이다. 2013년 고추 품종 청양의 선단부위에 괴저증상이 나타난 식물체로부터 CMV를 순수 분리하였고 이 분리주를 CMV-GTN으로 명명하였다. CMV-GTN과 기존에 특성이 잘 알려진 CMV-Ca-P1을 대상으로 외피단백질의 아미노산 서열과 여러 기주식물에서 생물적 반응을 비교하였다. CMV-Ca-P1의 외피단백질 아미노산은 217개로 구성되어 있었으나, CMV-GTN은 아미노산 서열 57번째에 발린이 추가된 218개로 구성되어 있었다. CMV-GTN과 GeneBank에 등록된 다양한 CMV 분리주들의 외피단백질유전자 유사성은 96-99%였다. 생물 검정에서 토마토와 고추에서 병원성이 강하게 발현되는 CMV-GTN을 고추 품종에 대한 저항성 스크린을 위하여 선발하였다. 고추 시판 품종 135종에 대하여 CMV-GTN으로 검정하였고 CMV-Ca-P1과 저항성 반응을 비교하였다. 그 결과 CMV-GTN에 대하여 저항성 반응을 표현한 품종은 프레미엄, 중도 저항성 품종은 핫스타, 카이저, 굿쵸이스였다. 한편 CMV-Ca-P1에 대한 저항성 품종은 베로따와 카이저였다.

  • 식물 바이러스는 작물 생산량 손실을 일으키는 주요 병원체 중 하나로, 돌연변이 발생이 빈번하고 치료 약제가 개발되어 있지 않아 방제가 매우 어렵다. 이러한 바이러스병을 방제하기 위한 가장 효과적인 방법은 저항성 품종을 재배하는 것이며, 바이러스 저항성 품종을 개발하기 위해서는 바이러스와 기주 식물 간의 다양한 유전자적 상호작용에 대한 정확한 이해가 필요하다. 열성 저항성은 병원체가 살아가는데 필요한 식물 유전자가 결핍되었을 때 획득되는데, 저항성 유전자(R gene)에 의해 유도되는 우성 저항성에 비해 넓은 범위의 저항성을 발현하고 돌연변이 출현에 쉽게 저항성이 깨지지 않는 특성을 보인다. 현재까지 알려진 바이러스병에 대한 열성 저항성 유전자는 대부분 순행유전학(forward genetics)를 통해 밝혀졌으나, 최근 CRISPR/Cas9 등을 이용한 유전자 교정 기술의 급속한 발전에 힘입어 역유전학(reverse genetics)을 통한 열성 저항성 작물개발의 가능성이 열리고 있다. 이러한 역유전학적 접근을 통한 열성 저항성 작물 개발은 먼저 바이러스 단백질과 상호작용하는 기주 인자를 밝히고 이들간의 상호작용을 억제하도록 하는 기주 인자에 대한 유전자 교정을 통해 이루어 질 수 있다. 본 논문에서는 열성 저항성에 대한 소개와 새로운 열성 저항성 후보 유전 소재 발굴을 위한 기주 인자 연구의 중요성 및 방법을 소개하고, 열성 저항성 작물 개발에 적용할 수 있는 유전자 교정기술의 최신 동향에 관해 정리하였다.

토마토황화잎말림바이러스병에 대한 저항성 품종과 항바이러스 활성 물질 3종의 효과 검증 (Efficacy of Three Antiviral Agents and Resistant Cultivars on Tomato Yellow Leaf Curl Virus in Tomato)

  • 최근에는 작물의 유도 저항성을 이용한 항바이러스제 개발에 관한 많은 연구가 수행되고 있으나 실제 농업현장에 널리 보급되지 못하고 있는 실정이다. 본 실험은 시설토마토 재배현장에 외생 살리실산, 키토산, 유제놀 처리에 따른 토마토황화잎말림바이러스(Tomato yellow leaf curl virus, TYLCV) 감염억제효과를 검증하고자 수행되었다. 실내검정에서 TYLCV에 감수성 품종인 '슈퍼도태랑'은 항바이러스제 처리 후 TYLCV에 감염된지 12일 후 VP (virus infected control plants)에서 바이러스 증상이 나타나기 시작했다. 접종 32일 후 TYLCV 발병도는 VP에서 98.8%였고, SAT (2 mM salicylic acid)+VP, EGT (200 ㎍/ml eugenol)+VP에서는 각각 98.8%, 98.7%로 발병도가 높았으나, CHT (0.1% chitosan)+VP는 85.7%로 다른 처리들과 통계적으로 유의한 차이를 보였다. 그러나 TYLCV 농도는 CHT+VP에서 OD값이 0.3으로 오히려 가장 높게 나타났으며, 토마토의 초장, 지상부 및 지하부 생체중에서도 뚜렷한 효과를 보이지 않았다. 여름작형 시설재배지에서 도태랑 계열의 토마토품종 '도태랑솔라'를 사용하여 항바이러스 3종의 효과를 조사한 결과, 수확기에 모든 처리구에서 100.0%에 가까운 TYLCV 감염률은 나타냈으며, 수확량에도 처리간의 통계적 유의차가 인정되지는 않았다. 이와 대조적으로 Ty-1과 Ty-3a 유전자를 보유한 TYLCV에 내병성 품종인 'TY자이언츠'는 저항성 유묘검정의 전 조사기간 동안 바이러스 증상이 전혀 관찰되지 않았고, 식물체내 바이러스 농도도 무접종 수준이었다. 본 실험 결과 'TY자이언츠' 품종은 TYLCV 발생이 만연한 지역 및 재배시기에 감수성 품종을 대체할 수 있을 것으로 생각된다. 반면, 저항성 유도물질인 살리실산, 유제놀, 키토산의 항바이러스 효과는 입증되지 않았기 때문에, 아직 시설토마토 재배현장에 적용하기는 어려울 것으로 판단된다.

Susceptibility of Koi, Koi×Red Common Carp, and Red Common Carp×Koi to Koi Herpesvirus (KHV)

  • The disease-causing koi herpes virus (KHV), also known as cyprinid herpesvirus-3 (CyHV3), causes mass mortality of koi and carp. Koi (Cyprinus carpio) is a host for KHV, one of 12 virus species in the Alloherpesviridae family. We examined the effects of KHV disease koi (KK), and on koi×red common carp (KR) and red common carp×koi (RK) cross, using a virus challenge test. The infected fish had clinical signs that included gill necrosis and skin lesions. The RK and KR were highly more resistant (cumulative mortality: RK; 6% and KR; 8%) to KHV infection than KK fish (cumulative mortality: 28%). KHV DNA was confirmed in the tissues of all dead fish in groups by use of polymerase chain reaction (PCR), and the presence of the KHV protein in kidney was confirmed by immunohistochemistry. Histological analysis showed severe gill lesions and fusion of the lamellae in KK fish, but less severe damage in RK fish. In immunohistochemistry analysis, the KHV protein localized in the cytoplasm of infected kidney cells of KK, but the cross groups had lower levels of KHV antigen. Our data indicate that the cross groups had increased resistance to KHV disease.

RT-RPA Assay Combined with a Lateral Flow Strip to Detect Soybean Mosaic Virus

  • Soybean (Glycine max L.) is one of the most widely planted and used legumes in the world, being used for food, animal feed products, and industrial production. The soybean mosaic virus (SMV) is the most prevalent virus infecting soybean plants. This study developed a diagnostic method for the rapid and sensitive detection of SMV using a reverse transcription-recombinase polymerase amplification (RT-RPA) technique combined with a lateral flow strip (LFS). The RT-RPA and RT-RPA-LFS conditions to detect the SMV were optimized using the selected primer set that amplified part of the VPg protein gene. The optimized reaction temperature for the RT-RPA primer and RT-RPA-LFS primer used in this study was 38℃ for both, and the minimum reaction time was 10 min and 5 min, respectively. The RT-RPA-LFS was as sensitive as RT-PCR to detect SMV with 10 pg/µl of total RNA. The reliability of the developed RT-RPA-LFS assay was evaluated using leaves collected from soybean fields. The RT-RPA-LFS diagnostic method developed in this study will be useful as a diagnostic method that can quickly and precisely detect SMV in the epidemiological investigation of SMV, in the selection process of SMV-resistant varieties, on local farms with limited resources.

Antiviral Activity of a Type 1 Ribosome-inactivating Protein from Chenopodium album L.

  • The antiviral activity of CAP30 from Chenopodium album, a type1 ribosome-inactivating protein (RIP), was examined against 5 different plant viral pathogens, and its activity against Tobacco mosaic virus was compared to those of well known antiviral proteins such as Pokeweed Antiviral protein from leaves and seeds. When the inoculating concentration of Tobacco mosaic virus was varied from 0.4 to $400{\mu}g/ml$, it was observed that CAP30 at the concentration of $1{\mu}g/ml$ suppressed the viral infection of C. amaranthicolor and C. quinoa almost completely up to $40{\mu}g/ml$ Tobacco mosaic virus. Results from the assays for the inhibitions of in vitro translation of rabbit reticulocyte lysate and the suppression of Tobacco mosaic virus infection ($10{\mu}g/ml$) to C. quinoa indicated that CAP30 is a strong inhibitor of protein synthesis and virus infection. The infection of several viruses other than Tobacco mosaic virus to host plants were also inhibited by $5{\mu}g/ml$ CAP30, suggesting that a gene encoding CAP30 can be used to develop transgenic virus-resistant plants.

Identification of Glycine max Genes Expressed in Response to Soybean mosaic virus Infection

  • Identification of host genes involved in disease progresses and/or defense responses is one of the most critical steps leading to the elucidation of disease resistance mechanisms in plants. Soybean mosaic virus (SMV) is one of the most prevalent pathogen of soybean (Glycine max). Although the soybeans are placed one of many important crops, relatively little is known about defense mechanism. In order to obtain host genes involved in SMV disease progress and host defense especially for virus resistance, two different cloning strategies (DD RT-PCR and Subtractive hybridization) were employed to identify pathogenesis- and defenserelated genes (PRs and DRs) from susceptible (Geumjeong 1) and resistant (Geumjeong 2) cultivars against SMV strain G7H. Using these approaches, we obtained 570 genes that expressed differentially during SMV infection processes. Based upon sequence analyses, differentially expressed host genes were classified into five groups, i.e. metabolism, genetic information processing, environmental information processing, cellular processes and unclassified group. A total of 11 differentially expressed genes including protein kinase, transcription factor, other potential signaling components and resistant-like gene involved in host defense response were selected to further characterize and determine expression profiles of each selected gene. Functional characterization of these genes will likely facilitate the elucidation of defense signal transduction and biological function in SMV-infected soybean plants.

Genotypes of Hepatitis C Virus in Relapsed and Non-respondent Patients and their Response to Anti-Viral Therapy in District Mardan, Khyber Pakhtunkhawa, Pakistan

  • Hepatitis C is a blood-borne infectious disease of liver, caused by a small enveloped, positive-single stranded RNA virus, called the hepatitis C virus (HCV). HCV belongs to the Flaviviridae family and has 6 genotypes and more than 100 subtypes. It is estimated that 185 million people are infected with HCV worldwide and 5% of these are in Pakistan. The study was designed to evaluate different genotypes of HCV circulating in District Mardan and to know about the behavior of these genotypes to different anti-viral regimes. In this study 3,800 patients were exposed to interferon alfa-2a plus Ribavirin treatment for 6-months and subjected to real-time PCR to check the viral response. Among these 3,677 (97%) patients showed no detectable HCV RNA while 123 (3%) patients (non-responders) remained positive for HCV RNA. Genotypes of their analyzed showed that most of them belonged to the 3a genotype. Non-responders (123) and relapsed (5) patients were subjected to PEG-interferon and Ribavirin therapy for next 6 months, which resulted into elimination of HCV RNA from 110 patients. The genotypes of the persisting resistant samples to anti-viral treatment were 3b, 2a, 1a and 1b. Furthermore, viral RNA from 6 patients remained un-typed while 4 patients showed mixed infections. HCV was found more resistant to antiviral therapy in females as compared to mals. The age group 36-45 in both females and males was found most affected by infection. In general 3a is the most prevalent genotype circulating in district Mardan and the best anti-viral therapy is PEG-interferon plus Ribavirin but it is common practice that due to the high cost patients receive interferon alfa-2a plus Ribavirin with consequent resistance in 3% patients given this treatment regime.

Tobamovirus Coat Protein CPCg Induces an HR-like Response in Sensitive Tobacco Plants

  • When inoculated into sensitive tobacco Xanthi-nn plants, the crucifer and garlic-infecting Tobacco mosaic virus (TMV-Cg) induces local necrotic lesions that resemble those seen in the hypersensitive response (HR) of resistant tobacco plants. However, unlike these, tobacco Xanthi-nn plants do not become resistant to infection and the virus spreads systemically causing a severe disease characterized by necrotic lesions throughout the plant. To identify the viral protein that elicits this necrotic response, we used a set of hybrid viruses constructed by combination of TMV-Cg and the tobacco mosaic virus strain U1 (TMV-U1). In this study we present evidence that the coat protein of TMV-Cg (CPCg) is the elicitor of the necrotic response in tobacco Xanthi-nn plants. Local and systemic necrotic lesions induced by TMV-Cg and by the hybrid U1-CPCg -that carries CPCg in a TMV-U1 context- are characterized by cell death and by the presence of autoflorescent phenolic compounds and $H_2O_2$, just like the HR lesions. In addition, defense-related genes and detoxifying genes are induced in tobacco Xanthi-nn plants after TMV-Cg and U1-CPCg inoculation. We postulate that in our system, CPCg is recognized by sensitive tobacco plants that mount an incomplete defense response. We call this an HR-like since it is not enough to induce plant resistance.

배나무잎검은점병 무독수의 선발 및 품종 저항성 (Selection of Pear Trees Free from Pear Black Leaf Spot Disease and Resistant Test for Pear Tree Cultivars)

  • 배나무잎검은점병에 대한 종합방제 대책을 수립하고자 시험을 수행하였다. 배 생산단지로부터 무독모수를 선발하기 위해서 본 병에 대한 감염 여부를 검정한 결과, 배나무잎검은점병이 발생하지 않는 6주를 확인하였다. 본 바이러스는 $40/32^{\circ}C$에서 3주간, $37/30^{\circ}C$에서 4주간 변온처리에 의해서 무독화가 가능하였으나 신초의 활착률이 매우 낮았다. 본 바이러스를 접종하여 병징이 발현하는 품종은 신고 등 7품종이었고, 발현하지 않는 품종은 수황배등 11종이었다. 품질이 우수한 저항성 품종인 수황배를 배나무잎검은점병이 발생하고 있는 신고 배나무에 고접한 결과 배나무잎검은점병이 전혀 발생하지 않았다.
