Monoterpenoids were recently found as main biologically active compounds which is responsible for various physiological effect in goat's beard (Aruncus dioicus). Ethyl acetate extract of A. dioicus (ADE) was treated to B16F10 melanoma cells for the examination of whitening activity. MTT assay was performed to evaluate cell toxicity and the result showed that slight cell toxicity (> 10%) by over $500{\mu}g{\cdot}mL^{-1}$. Thus, 0, 5, 10, or $50{\mu}g{\cdot}mL^{-1}$ ADE was used for further experiments. We found that tyrosinase activity was decreased according to ADE concentration, and the total melanin content was also dramatically reduced. Especially with $50{\mu}g{\cdot}mL^{-1}$ ADE treatment tyrosinase activity was reduced to 35.6%, and 58.8% of melanin content was lowered. In addition, whitening related proteins including tyrosinase, tyrosinase related protein 1 (TRP1), TRP2, microphthalmia associated transcription factor (MITF) and cAMP and protein kinase A (PKA) were reduced by ADE treatment. It caused decreased phosphorylation of cAMP response binding protein (CREB) but increased phosphorylation of extracellular signal related kinase (ERK). Therefore, in this paper we would like to suggest the potent usage of A. dioicus natively grown in Ulleungdo, Korea as materials of functional cosmetics by confirming whitening activity related with melanin content.

Acquired pigmentary skin diseases such as abnormal melanogenesis, vitiligo, chloasma and inflammatory pigmentation are related to regulate the melanin production, In this study, an ethanol extract of Hovenia dulcis Thunb.(EHD) makedly inhibited melanin biosynthesis and suppressed, the protein expression of tyrosinase, tyrosinase-related protein 1(TRP-1), and tyrosinase-related protein 2(TRP-2) in B16F10 cells. On the other hand, EHD did not inhibit mushroom tyrosinase activity. These results indicate that EHD may contribute to the inhibition of melanin biosynthesis through regulating tyrosinase activity and expression, and serve as a new candidate in the design of new skin-whitening or therapeutic agents.
Objectives : The purpose of this study was to investigate melanogenesis inhibition of ethanol extract of Polyporus (EP) by using B16F10 melanoma cells. Methods : We measured antioxidant effect of EP by using 1,1-Diphenyl-1-picrylhydrazyl (DPPH) assay and we confirmed melanin contents and tyrosinase activity of EP in cells. Additionally, the expression of tyrosinase-related protein-1 (TRP-1) and TRP-2 was observed by Western blot. Results : EP showed significantly high radical scavenging activity and inhibition of melanogenesis in dose-dependent manner by decreasing cellular tyrosinase activity and melanin content with or without α-melanin stimulating hormone. TRP-1 and TRP-2 expressions were also suppressed by EP in B16F10 cells. Conclusions : These results suggest that EP inhibits the melanogenesis and it could be a new organic ingredient for hyper-pigmentation.

Melanin synthesis is regulated by melanocyte specific enzymes and related transcription factors. $\beta$-carboline alkaloids including harmaline and harmalol are widely distributed in the environment including several plant families and alcoholic beverages. Presently, melanin content and tyrosinase activity were increased in melanoma cells by harmaline and harmalol in concentration- and time-dependent manners. Increased protein levels of tyrosinase, tyrosinase-related protein-1 (TRP-1), and TRP-2 were also evident. In addition, immunofluorescence and Western blot analyses revealed harmaline and harmalol increased cAMP response element binding protein phosphorylation and microphthalmia-associated transcription factor expression. In addition to studying the signaling that leads to melanogenesis, roles of the p38 MAPK pathways by the harmaline and harmalol were investigated. Harmaline and harmalol induced time-dependent phosphorylation of p38 MAPK. Harmaline and harmalol stimulated melanin synthesis and tyrosinase activity, as well as expression of tyrosinase and TRP-1 and TRP-2 indicating that these harmaline and harmalol induce melanogenesis through p38 MAPK signaling.

The anti-melanogenic substance was isolated from the root of Angelica gigas Nakai by silica gel column chromatography, preparative HPLC and TLC. As a result of the structure analysis by mass, $^1H$-NMR, and $^{13}C$-NMR spectrometry, the compound was identified as demethylsuberosin. Demethylsuberosin reduced melanin contents of B16F1 melanoma cells in a dose-dependent manner and decreased to about 74% at a concentration $5{\mu}g/ml$. Demethylsuberosin inhibited the expression in microphthalmia associated transcription factor (MITF), tyrosinase, tyrosinase related protein-1 (TRP-1), and tyrosinase related protein-2 (TRP-2) in melanocytes. These results suggest that the whitening activity of demethylsuberosin may be due to the inhibition of the melanin synthesis by down-regulation of MITF, tyrosinase, TRP-1 and TRP-2 expression. Thus, our results provide evidence that demethylsuberosin might be useful as a potential skin-whitening agent.

In this study, the whitening activity of Nymphoides indica extract in B16F10 cells were measured. Inhibition rate of tyrosinase from mushroom was 42% at $1,000{\mu}g/mL$. And inhibition of tyrosinase and melanin biosynthesis in B16F10 cells were 26 and 25% at $5{\mu}g/mL$, respectively. The expression levels of cAMP and protein kinase A (PKA), which are higher levels of melanin-related factors, were found to be decreased in a dose-dependent manner. In addition, the expression rate of protein and mRNA of tyrosinase, tyrosinase related protein 1 (TRP1), tyrosinase related protein 2 (TRP2) and microphthalmia associated transcription factor (MITF). In this study, it was confirmed that the N. indica extract effectively inhibited the activity of tyrosinase, TRP1, TRP2 and MITF as well as the activity of PKA by effectively inhibiting cAMP. Therefore, it was confirmed that the N. indica extract has high value as a functional material.

This study was peformed to investigate the mechanism of ethanol extract of Eclipta Prostrata (L.) L. (EEP) induced melanogenesis. EEP enhanced tyrosinase activity and melanin contents of B16F10 cells. Moreover, EEP increased the protein expression of tyrosinase and tyrosinase-related protein 1 (TRP-1). But EEP did not increase the protein expression of tyrosinase-related protein 2 (TRP-2). These results suggest that melanogenesis-promoting effect of EEP was involved in regulation of tyrosinaase and TRP-1 protein, and EEP may be a potent pigmentation darkening agent in hypopigmentation condition.
For the purpose of the development of skin-whitening or therapeutic agents against hyperpigmentation, aqueous extract from Nardostachys chinensis (AENC) was evaluated for melanogenesis inhibitory activity in B16F10 melanoma cell. The treatment with AENC at the 0.2, 0.5 and 1.0 mg/ml level significantly inhibits the biosynthesis of melanin compared with untreated control. The tyrosinase activity also significantly decreased in AENC-treated cells at the 0.2 and 0.5 mg/ml level and inhibitory effects were more efficient than commercial arbutin at 0.1 mg/ml. The Western analyses confirmed the significantly decreased expression of tyrosinase and tyrosinase-related protein-2 by AENC treatment. These results indicate that AENC may contribute to the inhibition of melanin biosynthesis through regulating the expression as well as activity of tyrosinase and AENC may be useful as a new candidate in the design of new skinwhitening or therapeutic agents.

The purpose of this study was to investigate anti-melanogenesis effects of mulberry seed extracts (MSE). MSE inhibited melanogenesis in melan-a cells at $10{\mu}g/mL$ without cytotoxicity. Also, MSE decreased tyrosinase, tyrosinase-related protein-1 (TRP-1) protein expression in the melan-a cells. To identify the signaling pathway of MSE, the ability of MSE to influence extracellular signal-regulated protein kinase (ERK) activation was investigated. MSE induced ERK protein expression in a dose-dependent manner. In addition, MSE presented inhibition of the body pigmentation in vivo zebrafish model. These results suggest that MSE may be an effective anti-melanogenesis agent regulating the expression of ERK protein and melanogenic enzymes.

Poria cocas has been reported to be effective in skin whitening. However, the direct effect of P. cocas ethanol extracts (PCEE) on melanin synthesis has not been scientifically studied. To elucidate the direct effect of PCEE on melanogenesis, a 3,4-dihydroxyindole-2-carboxylic acid (DOPA) synthesis assay, tyrosinase activity assay, and Western blotting for melanogenic proteins, including tyrosinase, tyrosinase-related protein (TRP)-1, and TRP-2 were performed in mouse B16F1 cells. The results revealed that PGEE inhibited melanin production in a dose-dependent manner by blocking the synthesis of DOPA. Although the activation of tyrosinase was not affected, the expression levels of TRP-1 and TRP-2 were controlled. These results suggest that PCEE has a whitening effect, indicating that it may be a useful agent in the development of whitening cosmetics.
