  • Milk and dairy products are the high value foods for the elderly population. In particular, fermented milk is the best source of calcium for people in the specific age group of over 79 years. It provides a good source of protein. Regular exercise and active lifestyle are recommended to slow down the muscle loss. However, exercising without proper nutrient intake is simply not sufficient at this age. Milk and dairy products provide the iron and protein content required for effective exercise-assisted growth. Milk nutrients have the advantage of being produced in various food forms, such as liquid, semi-solid, and powder types. Fat-soluble vitamins such as retinol and vitamin K can be encapsulated using various technologies for milk and dairy products. Using the encapsulation method, spray drying and fluidized-bed coating have been used for adding the micro-nutrients to the food. Microencapsulation technology is being applied in case of the fermented dairy products too. In particular, various wall materials are being developed to enhance the viability of probiotics. In the near future, advanced high-efficiency technologies that can effectively nourish the dairy products with nutrients will be developed to produce targeted high-nutrition value food for the elderly.

The Effect of Solvents on Sold Dispersion of Ipriflavone with Polyvinylpyrrolidone In Vivo

  • ABSTRACT -Ipriflavone is a synthetic flavonoid derivate that improves osteoblast cell activity inhibiting bone resorption. In order to improve the bioavailability, solid dispersions of ipriflavone with PVP (poly-N-vinylpyrrolidone, MW=40,000 g/mole) were prepared by a spray-drying method. During the manufacturing of solid dispersion, various solvents [ethanol (EtOH), acetonitrile, methylene chloride and cosolvent-EtOH:acetone=1:1] were used to dissolve the ipriflavone and PVP. Scanning electron microscopy (SEM) and differential scanning calorimetry (DSC) were used to evaluate the physicochemical interaction between ipriflavone and PVP. Particle size, crystallinity and the area of the endotherm $({\Delta}H)$ of solid dispersed ipriflavone using the acetonitrile as solvent were much smaller than those of the other preparation types. Bioavailability of ipriflavone in vivo was changed by solvents. When considering the result of in vivo test, solid dispersion of ipriflavone using the acetonitrile as solvent showed the best choice.

Qualitative Analysis of the Major Constituents in Traditional Oriental Prescription Bang-poong-tong-sung-san by Liquid Chromatography/Ultraviolet Detector/Ion-Trap Time-of-Flight Mass Spectrometry

  • An advanced and reliable high performance liquid chromatography (HPLC)/ultraviolet detector (UV)/ion-trap time-of-flight (IT-TOF) mass spectrometry was developed for the simultaneous quantification of 19 marker compounds in Bang-poong-tong-sung-san (BPTS), a traditional oriental prescription. Various parameters affecting HPLC separation and IT-TOF detection were investigated, and optimized conditions were identified. The separation was achieved on a Capcell PAK C18 column ($1.5mm{\times}250mm$, $5{\mu}m$ particle size) using a gradient elution of acetonitrile and water containing 0.1% formic acid at a flow rate of 0.1 mL/min. The column temperature was maintained at $40^{\circ}C$ and the injection volume was $2{\mu}L$. IT-TOF system was equipped with an electrospray ion source (ESI) operating in positive or negative ion mode. The optimized electrospray ionization parameters were as follows: ion spray voltage, +4.5 kV (positive ion mode), or -3.5 kV (negative ion mode); drying gas ($N_2$), 1.5 L/min; heat block temperature, $200^{\circ}C$. Automatic $MS^n$ (n = 1~3) analyses were carried out to obtain structural information of analytes. Elemental compositions and their mass errors were calculated based on their accurate masses obtained from a formula predictor software. The marker compounds in BPTS were identified by comparisons between $MS^n$ spectra from standards and those from extracts. Moreover, the libraries of $MS^2$ and $MS^3$ spectra and accurate masses of parent and fragment ions for marker compounds were constructed. The developed method was successfully applied to the BPTS extracts and identified 17 out of 19 marker compounds in the BPTS extracts.

Control of Algal Blooms in Eutrophic Water Using Porous Dolomite Granules

  • The use of aluminum-based coagulants in water pretreatment is being carefully considered because aluminum exposure is a risk factor for the onset of Alzheimer's disease. Lightly burned-dolomite kiln dust (LB-DKD) was evaluated as an alternative coagulant because it contains high levels of the healthful minerals calcium and magnesium. An organic pore forming agent (OPFA) was incorporated to prepare porous granules after OPFA removal through a thermal decomposition process. A spray drying method was used to produce uniform and reproducible spherical granules with low density, since fine dolomite particles have irregular agglomeration behavior in the hydration reaction. The use of fine dolomite powder and different porosity granules led to a visible color change in raw algae (RA) containing water, from dark green to transparent colorlessness. Also, dolomite powders and granules exhibited a mean removal efficiency of 48.3% in total nitrogen (T-N), a gradual increase in the removal efficiency of total phosphorus (T-P) as granule porosity increased. We demonstrate that porous dolomite granules can improve the settling time and water quality in summer seasons for the emergent treatment of excessive algal blooms in eutrophic water.

  • Acyclovir (ACV) is one of the most effective and selective agents against viruses of the herpes group. Because of low solubility, bioavailability of ACV has shown below 30% with oral dosage form. In our previous study, we reported that the fabrication of solid dispersion of ACV was possible and the solid dispersion of ACV and PVP was the most useful in all samples. In this study, we examined the effect of mixture ratio of polymers (PEG and PVP) to ACV. Solubility of ACV was dramatically increased up to 25 mg/ml in $80^{\circ}C$ distilled water. So water was used as a solvent to eliminate problem of residual solvent. Spray drying method was used for the solid dispersion of ACV as solvent extraction. Different scanning calorimeter was used to check degradation of drug. Polymer carriers were PEG 6,000 and PVP. In summary, ACV-PVP (1:3) showed the best solubility in distilled water.

친수성 고분자가 BCNU 함유 PLGA 웨이퍼로부터 BCNU의 방출에 미치는 효과 (Effect of Hydrophilic Polymers on the Release of BCNU from BCNU-loaded PLGA Wafer)

  • 1, 3-bis[2-chloroethyl]-1-nitrosourea (BCNU, carmustine)는 악성 뇌종양 치료를 위하여 화학요법적 임상에서 널리 사용되는 약물이다. 또한, poly(D,L-lactide-co-glycolide) (PLGA, 분자량: 20000 g/mole, 락타이드와 글리콜라이드 몰비 75 : 25)는 약물전달시스템을 위한 약물 전달체로써 사용되어지는 잘 알려진 생분해성 초분자이다. 본 연구에서 폴리비닐피롤리돈 (PVP) 또는 폴리에틸렌옥사이드 (PEO)를 함유하고 있는 BCNU 함유 PLGA 웨이퍼들의 BCNU 방출거동과 웨이퍼에 포접된 친수성 고분자의 효과를 조사하였다. 친수성 고분자의 첨가 또는 첨가 없이 BCNU 함유 PLGA 미분말은 분사건조법에 의해 제조하였으며, 제조된 BCNU 함유 PLGA 미분말은 압축성 형법에 의해 웨이퍼형태고 제조하였다. 친수성 고분자가 첨가된 BCNU 함유 PLGA 미분말의 포접율은 85∼97%였고, PLGA에 포접된 BCNU의 결정성은 현저히 감소하였다. 약물 방출 경향과 분해 거동에서 친수성 고분자의 함량이 증가할수록 BCNU의 초기방출량과 방출속도는 증가됨을 확인하였다. 방출시험 기간동안 웨이퍼의 형태변화와 무게변화를 측정함으로써 친수성 고분자의 함량이 증가할수록 PLGA의 수차와 분해가 촉진됨을 관찰하였다.

  • 골 재흡수 치료를 목적으로 파미드로네이트를 지속적으로 방출하는 제형으로 제조하기 위하여 락타이드-글리콜라이드 공중합체 (PLGA, 락타이드 : 글리콜라이드 몰비 = 75 : 25, 분자량 20000 g/mole 및 90000 g/mole)를 이용하여 직접압축 성형방법으로 생분해성 웨이퍼를 제조하였다. 약물과 고분자의 함량비 웨이퍼의 두께, PLGA 분자량 등을 조절하여 PLGA 웨이퍼를 제조하였고 이들의 형태학적 특성과 방출거동 및 분해거동을 살펴보았다. 웨이퍼의 제조는 혼합된 분말을 웨이퍼 제작용 몰드에 넣은 후 프레스를 이용하여 일정 압력으로 일정시간 동안 상온에서 가압하여 제조하였다. 제조된 웨이퍼는 약물의 초기함량이 증가할수록 방출속도가 빠르게 나타났으며, 제형의 두께가 두꺼워질수록 시간이 경과함에 따라 약물의 방출속도가 증가하였다. 또한 고분자의 분자량이 큰 것이 작은 것에 비해 상대적으로 초기 약물 방출량이 적고 방출되는 속도 또한 느려져. 저분자보다 오랫동안 약물이 방출되었다. 이러한 약물전달 시스템은 압축성형방법에 의해 제조하므로 제조가 간단하고 약물방출속도를 정확하게 제어할 수 있으므로 이식을 위한 제형으로 제조시 유용하게 쓰일 것으로 예상되었다.

  • To select the operating condition of 0.5 MWth chemical looping combustion system, minimum fluidization velocity, transition velocity to fast fluidization and solid circulation rate were measured using mass produced new oxygen carrier (N016-R4) which produced by spray drying method for 0.5 MWth chemical looping combustion system. A minimum fluidization velocity decreased as the pressure increased. The measured transition velocity to fast fluidization was 2.0 m/s at ambient temperature and pressure. The measured solid circulation rate increased as the solid control valve opening increased. We could control the solid circulation rate from 26 to $93kg/m^2s$. Based on the measured minimum fluidization velocity and transition velocity to fast fluidization, we choose appropriate operating conditions and demonstrated continuous solid circulation at high pressure condition (5 bar-abs) up to 24 hours.

  • 본 연구에서는 마이크로 크기의 실리콘 입자와 탄소나노튜브를 활용하여 고용량을 갖는 실리콘/탄소나노튜브/탄소 복합입자를 제조하여 리튬이차전지용 음극활물질로서의 적용가능성을 확인하고자 하였다. 실리콘/탄소나노튜브/탄소 복합입자 제조를 위해 분무건조 방식을 이용하여 실리콘입자가 탄소나노튜브에 의해 균일하게 분산되어 비정질탄소로 결합된 구조를 갖는 구형의 복합입자를 제조하였다. 제조한 복합입자는 실리콘 입자 주변에 탄소나노튜브의 네트워크 구조를 형성하며 비정질 탄소에 의해 실리콘 입자와 탄소나노튜브의 입자들이 결합한 상태를 유지하는 구조로 이루어진다. 이러한 복합입자의 구조적인 특성으로 인해 계속적인 충방전 과정에서 실리콘의 부피팽창이 효과적으로 완충되고 이에 따라 전기적 접촉 손실 및 SEI 막 형성에 따른 비가역 반응이 제어되어 우수한 수명 특성 및 충전출력 특성을 갖는 것으로 나타난다.

LED배양조건에서 미세조류 Nannochloropsis oculata의 생산 효율성을 높이는 공정 최적화 (Optimized Processing Condition of Production of Nannochloropsis oculata under Light-emitting Diode (LED) Condition)

  • 100 l scale 미세조류 배양 시스템은 LED 광을 기반으로 자체 제작하였으며 Nannochloropsis oculata를 f/2 medium에서 광도($100{\mu}mol/m^2/s$), 배양온도($20^{\circ}C{\pm}1^{\circ}C$), LD cycle (12 hr)으로 배양하였다. 그 결과 LED blue (475 nm)에서 $100{\mu}mol/m^2/s$의 광도, 24 mg/l의 nitrate 농도에서 100 l 대량배양 결과 최대 biomass인 1.07 g/l를 배양하였다. 이렇게 배양된 미세조류에서 유용물질을 추출의 위한 추출방법을 확보하고 나아가 산물의 추출, 농축 및 건조공정 기술을 최적화하고자 하였다. 추출은 물리적인 방법인 sonicator, homogenizer를 이용한 파쇄법과 화학적인 방법인 0.5M HCl 파쇄법을 이용한 결과 chlorophyll a, chlorophyll b, carotenoid 함량은 물리적인 파쇄법인 sonicator를 이용 시 1.6, 0.5, 0.3 mg/g cell, homogenizer 이용 시 1.0, 0.6, 0.2 mg/g cell로 측정되었다. 0.5M HCl의 화학적 파쇄법 이용 시 chlorophyll a, chlorophyll b 함량은 0.9, 0.8 mg/g cell으로 측정되었다. 파쇄시간당 추출되는 영양물질이 가장 높은 시간은 15 min 파쇄 시 3.6 mg/g cell로 측정되었으며 acetone, methanol, ethanol의 3가지 용매 중 acetone이 3.6 mg/g cell로 효과적이 용매로 측정되었다. 농축은 2종류의 연속원심분리장비(tubular type, disk type), 마이크로필터, 필터프레스 4종 장비를 이용하여 시험한 결과 마이크로필터, disk type 연속원심분리기, Tubular type 연속원심분리기의 경우 16.0, 1.1, 0.5 g/l의 순으로 시간당 수율을 확인하였다. 건조공정 최적화를 위하여 열풍건조기, 진공건조기, 분무건조기, 동결건조기 등의 4종 시생산설비를 이용해 미세조류의 시간당 회수율 분무건조기는 Dextrin 0.5 kg이 첨가되면 회수율이 80%이지만 순도가 떨어졌고 동결건조기 회수율은 60%로 측정되었다.
