  • Human lymphotoxin (LT) was produced in E. coli as a soluble protein. The level of recombinant human LT production was about 4% of the total souble proteins of E. coli extracts. Recominant human LT was purified to apparant homogeneity by a simple procedure utilizing FPLC on Mono Q and Mono S columns. The specific activity of the purified LT was $10\times10^7\;units/mg$.

  • A side scan sonar system uses the towfish installed sonars, It is an equipment that search images of the bottom surface of the sea in real time. It is a typical equipment that is related to a sea investigation such as a geological survey, seabed communication cable and power line cable placing repair investigation, fish breeding ground investigation, sea purification, relic and mineral investigation, and mine and submarine search. It used to fined objects and investigate on the seabed surface. But, recently, it is used to sea purification and geological survey that require information of the correct surface of the seabed. So, it needs various filtering technique and image processing techniques development to acquire high resolution image. therefore, this research develops a side scan sonar using multi-beam sensors that supply various information with the fast scan speed and correct high resolution that is not a simple underwater investigation equipment.

  • Glycopeptide antibiotics, teicoplanin was purified from a mutant strain of Actinoplanes teichomyceticus ATCC31121, A. teichomyceticus MSL2211. We developed a simple procedure to separate and purify the teicoplanin from the fermentation broth. Teicoplanin was purified by two-step purification system, hydrophobic adsorption and sugar affinity chromatography in combination with HPLC analysis based on the properties of hydrophobic acyl chain and sugar moiety in teicoplanin. Teicoplanin was separated from the culture broth by Diaion HP-20 and further purified by concanavalin A affinity column chromatography. As an adsorbent resin, Diaion HP-20 in broth eliminated toxic effects on growth, reduced feedback repression of teicoplanin production, and assisted In rapid recovery of teicoplanin. The teicoplanin displayed the final yield of 80% and 95% of purity.

  • A simple sequence of membrane concentration and DEAE-Cellulose chromatography has been optimized to give a purified dextransucrase from Leuconostoc mesenteroides B-512FMCM with the highest specific activity (248.8 IU/mg protein) ever reported in high yield (overall 88.7%) for dextransucrase. When there was no sucrose in the dextransucrase and the dextran reaction digest, the dextransucrase hydrolyzed glucose from dextran. The glucose was transferred to the other glucoses from dextran and formed isomaltose and isomaltodextrin. The transglycosylation efficiency of glucose from dextran was much higher with acceptors. The dextransucrase can be used for the production of various kinds (or structures) of oligosaccharides using dextran and various acceptors with almost 100% theoretical yield.

  • The study was carried out on the biochemical characters of Cd-BP(I) after isolation and purification of the protein from the liver of rat injected intraperitoneally with Cd. A c ontinued study has been doing whether Cd-BP(I) could be induced by Cd or by other metals such as Zn and Cu. Antisera were made against the Cd-BP(I) from NewZealand white rabbits. Carried out were ${\gamma}$-globulin purification, then Ouchterlony test adn gel immunodiffusion test. Cd-BP(I) was also found in normal tissues of rat. It was induced up to a considerable level by Cd, whose induced level was higher than that of Cu or Zn treatment. The level of induction by Cu or Zn pretreatment plus Cd treatment was lower than that by simple treatment of Cu or Zn. Such a result was presumably related to the Cd toxicity.

  • A simple procedure for the extraction of the lipolytic enzyme from rice bran has been developed. High activity of lipolytic enzyme was obtained by first defatting the rice bran to remove lipid components with various extraction conditions. Then, after five cycles of aqueous extraction, rice bran lipolytic enzyme was purified using micro- and ultrafiltration apparatus. Lipolytic enzyme activity was estimated by its hydrolytic action of tributyrin. The result indicated that the standard activity curve of butyric acid showed that the potential rice bran enzyme is a hydrolytic lipase enzyme. In addition, it showed higher lipolytic activity and specific enzyme activity with further purification by micro- and ultrafiltration.

A Simple and Efficient Method to Determine Rivaroxaban in Rat Plasma Using Liquid-Liquid Extraction and LC-MRM

  • Rivaroxaban (RRN) is the first available active direct factor Xa inhibitor (anticoagulant) with oral administration. Due to its success in market, there have been efforts to develop various RRN formulations, and the development of good analytical methods for its in vivo evaluation is an essential prerequisite. Thus, here, a simple and efficient method to determine RRN in rat plasma using liquid-liquid extraction (LLE) and liquid chromatography and multiple reaction monitoring (LC-MRM) was presented. The use of ethyl acetate as the LLE solvent results appropriate extraction and purification of RRN and it also helps the significant reduction of rat plasma volume required for RRN quantitation. The developed method showed good analytical performance including specificity, linearity ($r^2{\geq}0.999$ within 0.5 - 500 ng/mL), sensitivity (the lower limit of quantitation at 0.5 ng/mL), accuracy (89.3 - 107.0%), precision (${\geq}12.7%$), and recovery (89.2 - 105.7%). Additionally, RRN in sample extracts showed good stability. Finally, the applicability of the validated method to the PK evaluation of RRN was confirmed after its oral administration to normal rats. The present method is the first analytical method employing LLE for the simple and efficient extraction and purification of RRN in rat plasma. Therefore, the present method can contribute to the development of new RRN formulations as well as to the monitoring of RRN in special clinical situations through its efficient determination in various samples with or without minor modification.

Optimization of Adsorbent Treatment Process for the Purification of Paclitaxel from Plant Cell Cultures of Taxus chinensis (주목 식물세포(Taxus chinensis)배양으로부터 파클리탁셀 정제를 위한 흡착제 처리 공정 최적화)

  • Biomass-derived tar and waxy compounds have a highly negative effect on the separation and purification of paclitaxel and should be removed prior to final purification. Adsorbent treatment is a simple, efficient method for removal of tar and waxy compounds from plant cell cultures. In this study, we optimized the important process parameters (adsorption temperature, time, solvent type and adsorbent amount) of adsorbent treatment with Sylopute to remove the tar and waxy compounds in a pre-purification step. The optimal adsorption temperature, adsorption time, solvent type, and crude extract/Sylopute ratio were $30^{\circ}C$, 15 min, methylene chloride, and 1:1(w/w), respectively. This result could be confirmed by HPLC analysis of the absorbent after treatment and TGA of the organic substances that were bound to the adsorbent. In adsorbent treatment step, the purity seemed to show a small improvement but this treatment had a significant effect on convenience and feasibility of following steps by the removal of tar and waxy compounds.
