It has been believed that supplementation of human diets with soybean products markedly reduces human cancer mortality rates. Specifically the black colored soybean has been considered as having a medicinal effect since ancient times in Korea. Six genotypes of black colored soybean and a normal variety were investigated for their biological effect. While the isoflavone content of black colored soybean was higher than the normal soybean, saponin content of the seeds was not different among varieties. In the case of antioxidant activity, black colored soybean showed over 3 times increase in the active effect than the control variety. When feeding soybean extract, a general change of alcohol resolving effect, such as alcohol dehydrogenase activity, was observed in the blood and liver of rats.
This study was carried out to establish the extraction method of red ginseng extract without saponin decomposition. Red ginseng was extracted with impulse vacuum system and multi-stage extraction method. Crude saponin content of red ginseng extract (RGE) from impulse vacuum system was 5.4-5.9%, while that of RGE from multi-stage extraction method was 8.2-8.3%. However, HPLC Patterns indicated that saponins of RGE from impulse vacuum system were hardly decomposed, while those of RGE from multi-stage extraction method were decomposed, especially in ginsenoside -Rgl and -Re saponin. Also, the yields of red ginseng by impulse vacuum system were 15 to 20 times higher than that of multi-stage extraction method.

Microwave extraction system equipped with closed vessels was applied to confirm its capability of extracting ginseng solubles including saponin, which was monitored to optimize extraction con ditions by response surface methodology. Total yield increased with the decrease in particles size and more extraction steps. Soluble components were completely extracted by operating the system within 6 min per one step, which should be repeated by 3 or 4 times. Optimized conditions for maxi mum extraction of response variables(total yield, crude saponin) were 40~50% of ethanol concentration, about 140oC of extraction temperature, and within 6 min of the extraction time. The extraction effi ciency of total yield was dependent on the decrease of ethanol concentration, while crude saponin content was favored on the higher degree of ethanol concentration. According to the temperature guideline, below 90oC of extraction temperature, the optimal ranges of extraction conditions were predicted as 30~50% of ethanol concentration and 2.5~6.5 min of extraction time. Estimated values of total yield and crude saponin were in good agreement with experimental values.
Background: To obtain useful cosmetic resources, this study aimed to determine the non-saponin fatty acid and inhibitory activities of collagenase and elastase by treatment of Saccharomyces cerevisiae in supercritical fluid extracted oil of the adventitious root culture of wild mountain ginseng. Methods and Results: We performed supercritical fluid extraction at various conditions such as pressure, temperature, time, and use of co-solvents, unlike the n-hexane extraction for the adventitious roots culture of wild mountain ginseng. The non-saponin-fatty acid obtained from the oil of the adventitious roots culture was incresed by treatment with S. cerevisiae. The supercritical fluid extraction was conducted using gas chromatography. Non-saponin-fatty acid content, in the oil of adventitious roots culture of wild mountain ginseng treated with S. cerevisiae for 2 days were three times higher than that in the control. In addition, the oil of the adventitious roots culture treated with S. cerevisiae was investigated for the anti-wrinkle effect by using collagenase and elastase. The oil of adventitious roots culture treated with S. cerevisiae exhibited higher collagenase and elastase inhibitory activities than those in the control. Conclusions: Supercritical fluid extracted oil of the adventitious roots culture of wild mountain ginseng treated with S. cerevisiae was found to have decreased ratio of saturated fatty acids and incresed ratio and content of unsaturated fatty acids increased. Furthermore, it showed anti-wrinkle effects in vitro.

This study investigated quality characteristics of 4 year-old ginseng by enzymatic hydrolysis conditions to increase utilization. Ginseng was ground after steaming and was each treated with hydrolase A, B, C and D. When quality characteristics by the enzymes were examined, no significant difference was observed with pH of 5.5~5.6 and the sugar content of 4.0~4.33. The crude saponin content was the highest in ginseng treated with D, followed by B, C and A. The crude saponin, the reducing sugar and the total sugar contents increased until 0.3% (w/w) concentration in enzyme D with no significant difference by its concentration. Although active ingredients increased with time passage of hydrolysis, no significant change was found after three hours and the crude saponin content was the highest when ginseng was treated at $60^{\circ}C$. From these results, optimum conditions for 4 year-old ginseng were $60^{\circ}C$ for 3 hours with 0.3% (w/w) enzyme D, and under these conditions the reducing sugar, the total sugar and the crude saponin contents recorded 18.11, 36.21 and 4.23 mg/g, respectively. Therefore, enzymatic hydrolysis was found to be effective in increasing active ingredients of 4 year-old ginseng with various usages expected.
This study was conducted to compare the root weight, yield, quality of fresh and red ginseng roots and crude saponin content in roots between fixing light transmittance(Control) and changing light transmittance(C.L.T.) during the ginseng growing seasons. The root weight in C.L.T. was higher than control by $35{\%}$ in early growth stage, $28{\%}$ in middle growth stage and $26{\%}$ in late growth stage in 6 years old ginseng plant. Root yield per 10a in C.L.T. was increased about $40{\%}$ as compared with that of control, also 1st and 2nd grade of fresh ginseng roots in C.L.T. was higher $(50.3{\%})$ compared with that $(12.9{\%})$ of control. The specific gravity of ginseng roots grown under the C.L.T. was exhibited the significant difference than control during the growing season in 4 and 6 years old ginseng plants. Red ginseng quality in C.L.T. was not only improved remarkably due to the increasement of heaven and earth grade red ginseng but also increased in crude saponin content than control. Therefore it needs to change the light transmittance(increasing light during low temperature periods and decreasing light during high temperature period) during the growing season for high yield and good qualities of ginseng roots.
Panax ginseng leaf tea was developed for the functional benefit of health, preference and convenience. The leaves of 4-year-old ginseng were selected in July and August. The ginseng leaf was treated by three methods : heat processed tea(HPT), aged tea(AGT) and hot-air dried tea(DRT). The contents and compositions of their crude saponin of ginseng leaves were measured. 1. The content of crude saponin of HPT was the higher than other treatments. The content of HPT was 18.72∼18.82%, ACT 18.24∼18.29% and DRT 17.02∼17.17%. 2. The harvest time and treatment methods were not affect the composition of ginsenoside in ginseng leaf tea. The ginsenoside-Re was shown the highest value as 1.97∼2.15. And ginsenoside-Rd was 1.48∼1.79, -Rg$_1$ 1.33∼1.58 and -Rb, -Rb$_2$, -Rc in the order. 3. The content of protopanaxadiol(PD) and protopanaxatriol(PT) was shown that DRT was 1.11∼1.13, HPT 1.09~l.12 and AGT 0.92∼1.02. The content of PD and PT were shown similar result at any harvest time. 4. The contents of crude saponin extracted by hot-water at 5 min was the higher ratios in HPT and harvested in July than other treatments. The content of crude saponin of ginseng leaf harvested in July was 15.88% and HPT was 16.88%. The order of contents of ginsenoside were -Re, -Rd, -Rg$_1$, -Rb$_1$, -Rb$_2$, and - Rc. The extraction ratio of crude saponin extracted by the circulated extraction method in 8 hours and 5 min extraction were 81.74∼84.38%. And HPT of ginseng leaf harvested in July was the highest value 84.3% but the extraction ratio of ginsenoside was 78.00~88.13%. But the extraction ratio of ginsenoside was similar trend in all treatments.
The difference of ginsenosides content according to placement of ginseng planting (line) under shading net in 5-year-old ginseng roots were examined. The total saponin ($Rb_1$, $Rb_2$, Rc, Rd, Re, and $Rg_1$) contents were 15.01 mg/g and 21.79 mg/g in the main roots, 35.93 mg/g and 43.32 mg/g in the lateral roots, 87.85 mg/g and 105.51 mg/g in the fine roots for the front $1st{\sim}2nd$ lines in Yunpoong and Landrace variety (purple-stem variant), respectively. In the middle $3rd{\sim}5th$ lines the total saponin contents were 18.73 mg/g and 23.19 mg/g in the main roots, 44.92 mg/g and 43.50 mg/g in the lateral roots, 92.97 mg/g and 110.70 mg/g in the fine roots in Yunpoong and Landrace variety, respectively. In the rear $6th{\sim}7th$ lines the total saponin contents were 21.88 mg/g and 26.68 mg/g in the main roots, 38.41 mg/g and 44.89 mg/g in the lateral roots, 101.03 mg/g and 107.06 mg/g in the fine roots in Yunpoong and Landrace variety, respectively. The differences in total and individual ginsenosides content in the main, lateral and fine roots among the lines were not significant but total ginsenosides contents in the main roots were different in case of Yunpoong variety. The ratios of protopanaxadiol (PD) type saponin to protopanaxatriol (PT) type saponin in roots were lower in the front lines compared to the middle and rear lines and the ratios were significantly different among the parts of roots.

This study investigated the changes of saponin and $\beta$-glucan content on the cultured ginseng with mushroom mycelia of Phellinus linteus (PL), Ganoderma lucidum (GL), and Hericium erinaceum (HE). Cultured ginsengs with mushroom mycelia were extracted with 80% ethanol, fractionated with n-buthanol, and analysed for ginsenosides by high performance liquid chromatography (HPLC). Crude saponin content of raw ginseng was 4.11% (d.b) but cultured ginseng with mushroom mycelia of PL, GL, and HE were increased to 6.74, 6.77 and 6.23% (d.b), respectively. Ginsenoside-Rd, among the 12 ginsenosides which were available for analysis, was remarkably increased to 13.61, 24.26, and 32.69 mg/g, respectively (raw ginseng: 0.80 mg/g). The $\beta$-glucan content of cultured ginseng with mushroom mycelia of PL, GL, and HE were decreased to 8.85, 5.51 and 5.46% rather than mushroom mycelia of 29.14, 19.44, and 23.39% (d.b), respectively.
This study was conducted to provide basic information about the drying methods (daylight, hot-air, and freeze drying) used for Platycodon grandiflorum radix. We investigated the mineral, free sugar, and saponin contents of dried P. grandiflorum. The potassium and calcium contents of hot-air-dried samples were the highest (22.6 and 9.2 mg%, respectively), when compared to those of daylight- or freeze-dried samples. Glucose and sucrose contents were the highest in freeze-dried samples (1,552 and 145.0 mg%, respectively), while fructose content was the highest in hot-air-dried samples (611.9 mg%). Platycodin D content was the highest in hot-air-dried samples (622.0 mg%); however platycodin D3, polygalacin D, and deapioplatycodin D contents were the highest in daylight-dried plant (113.5, 756.6, and 109.2 mg%, respectively). Glucose content was highly negatively correlated (p<0.01) with platycodin D, platycodin D3, and deapioplatycodin D (-0.924, -0.957, -0.861, p<0.01, respectively). These results suggest that the drying method affects the saponin content of P. grandiflorum and daylight and hot-air drying methods are more suitable and beneficial than freeze-drying.
