Studies on Genetic Variation of Soluble Solids, Acidity and Carotenoid Contents in Tomato Fruits from Germplasm (국내외에서 수집된 토마토에서 당도, 산도, 카로티노이드 색소의 유전변이에 관한 연구)

  • Tomato (Lycopersicon esculentum M.) is one of the most important crops to the fresh vegetable market and the food processing industry. To evaluate genetic variation in tomato fruits, major characteristics such as soluble solids, acidity and carotenoid contents were analyzed for 771 genetic resource lines. Lines in red color was about 85% which is the largest one followed by peach color, yellow, green, orange, and black. The sweetness of juice ranged from 2.2 to 11.5% (in brix), the average being 5.6%. The acidity ranged from 0.124% to 1.665%, and the average was 0.881%. The lycopine contents was up to 80.4 ${\mu}g/g$, and 43.4 ${\mu}g/g$ in average. ${\beta}$-carotine ranged 1.8 to 48.8 ${\mu}g/g$ and it average was 10.8 ${\mu}g/g$. Statistical analysis indicates that there is coefficient of correlation between acidity and sweetness, acidity and pH, pH and lycopine, lycopine and ${\beta}$-carotine. It is expected that the result of this study can be used for breeding more competitive species with respect to contents in sugar or functional chemicals from the selected characteristic species.

Effects of Modified Atmosphere Packaging and Gamma Irradiation on NO-Mb Contents and Color of Sausage (가스치환 포장 및 감마선 조사된 소시지의 NO-Mb 함량과 색 특성)

  • Combined effect of modified atmosphere packaging and gamma irradiation on color characteristics of emulsion-type cooked pork sausage were investigated. Nitrosomyoglobin (NO-Mb) contents and CIE color values of sausage with aerobic, $CO_2, \;N_2$, and $CO_2/N_2$ packagings were observed during storage for 4 weeks. Irradiation reduced NO-Mb content in sausage, inducing denitrosylation of nitrite-cured meats. $CO_2$ and $CO_2/N_2$ packagings were effective for maintaining NO-Mb content. In CIE color values, $a^*$ was significantly reduced by irradiation at 5 kGy and above. The $a^*$ values of sausages with $CO_2$ and $CO_2/N_2$ packagings were higher than those of aerobic or $N_2$ packaging. Results indicated that modified atmosphere packaging, such as $CO_2$ or $CO_2/N_2$ packaging, was effective for minimizing the loss of red color in sausage caused by irradiation.

Conservation and Analysis of Wall Painting Fragments of Goguryeo Possessed by National Museum of Korea (국립중앙박물관 소장 고구려 벽화 편의 보존과 분석)

  • Conservation and analysis of wall painting Goguryeo was performed to classify the unknown fragments. The conservation naked eye observation, optical microscopy, and infrared examination were carried out in order to figure out the structure, quality of constituting materials, and damages such as cracks, and discolored fragments of colored areas. Based on such investigation, conservation was proceeded. and it was completed with strengthening the weakened pigment layer of wall blocks. In addition tombs where the wall painting fragments were excavated were investigated by making comparison with gelatin dry plates and copies possessed by National Museum of Korea. According to the result, they were Kaemachong, Gosan-ri Tomb No.1 Gamsinchong, and Wonbong-ri Tomb. The components of colors with which Goguryeo wall painting fragments were painted and the mineral pigments of the wall layer were analyzed. Portable µ-XRF spectrometer and X-ray diffractometer were employed. It showed that lime (CaCO3) used for the wall layer, and the brown color is hematite(Fe2O3) and cerusite (PbCO3) and lead oxide(PbO) were identified. In the red color, cinnabar (HgS) were detected.

Characteristics of Seed Germination and Seedling Growth of Native Hydrangea serrata for. acuminata (자생 산수국의 종자 발아와 유묘 생육 특성)

  • This work aims to obtain basic information for seed propagation of Hydrangea serrata for. acuminata. The germination percentage of the seeds taken on 15 November, 30 December, and 23 January was $90.0{\pm}4.16%$, $84.4{\pm}5.52%$, and $88.9{\pm}2.40%$, respectively. This suggest that seeds of Hydrangea serrata for. acuminata are non-dormant seeds. The optimum temperature for germination was $25^{\circ}C$ and light was necessary. Most of the growth parameters (shoot and leaf length, stem diameter, root length, no. of roots, T/R ratio, and fresh and dry wts.) were significantly greater at $25/20^{\circ}C$ and $25^{\circ}C$ than at the other temperatures. Low T/R ratio at relatively cool temperatures (15 and $20^{\circ}C$) was caused by suppressed top growth. In light quality treatment, red light (RL) significantly enhanced stem elongation. The greatest photosynthetic pigments (total chl, chl a/b, and carotenoid) were observed in seedlings grown in blue light (BL), followed by seedlings grown in RL+BL. When blue light was added, higher pigment contents were found. Effect of plug cell size (50, 72, 128, 162 and 200 cells) on the growth of seedlings was investigated. The highest top growth was observed in seedlings grown in 50 cell trays, followed by seedlings grown in 72, 128, 162, and 200 cell trays. However, there was no significant differences between 162 and 200 cell trays. Especially, smaller size leaves were observed in seedlings grown in smaller cell trays (lower volume and high plant density).

  • In ornamental crops, the color and shape of flowers are one of the important traits. Generally, flower colors are determined by accumulating pigments such as carotenoids, flavonoids, and betalains. Among them, flavonoids are responsible for broad ranges of colors. Chrysanthemums are one of the most popular ornamental crops in the world, and there have been many efforts to change their flower color. In chrysanthemum flowers, cyanidin-based anthocyanin confers pink or red color, whereas terpenoid-based carotenoids are mainly responsible for yellow and green colors. However, blue colored chrysanthemums do not occur in nature. To date, there have been attempts to obtain blue or violet-colored chrysanthemum flowers through the introduction of a novel gene for accumulating delphinidin-based anthocyanins, while other studies have reported changing endogenous metabolites through the reconstruction of flavonoid biosynthesis. Since various transcription factors are involved in the regulation of flavonoid biosynthesis, it is important to understand not only the structural genes, but also the transcription factors required for the modification of flavonoid-based flower color. Therefore, in this paper, we describe the flavonoid biosynthetic pathway and its regulation, and review previous studies on the change in flower color through modification of flavonoid biosynthesis. This effort could be an important milestone in successfully achieving the modification of chrysanthemum flower color by means of plant biotechnology.

Production of Carotenoids by Bacteria; Carotenoid Productivity and Availability (박테리아에 의한 카로티노이드 생산; 카로티노이드 생산성 및 활용 가능성)

  • Carotenoids are red, orange, and yellow fat-soluble pigments that exist in nature, and are known as physiologically active substances with various functions, such as provitamin A, antioxidant, anti-inflammatory, and anticancer. Because of their physiological activity and color availability, carotenoids are widely used in the food, cosmetics, and aquaculture industries. Currently, most carotenoids used industrially use chemical synthesis because of their low production cost, but natural carotenoids are in the spotlight because of their safety and physiologically active effects. However, the production of carotenoids in plants and animals is limited for economic reasons. Carotenoids produced by bacteria have a good advantage in replacing carotenoids produced by chemical synthesis. Since carotenoids produced from bacteria have limited industrial applications due to low productivity, studies are continuously being conducted to increase the production of carotenoids by bacteria. Studies conducted to increase carotenoid production from bacteria include the activity of enzymes in the bacterial carotenoid biosynthesis pathway, the development of mutant strains using physical and chemical mutagens, increasing carotenoid productivity in strain construction through genetic engineering, carotenoid accumulation through stress induction, fermentation medium composition, culture conditions, co-culture with other strains, etc. The aim of this article was to review studies conducted to increase the productivity of carotenoids from bacteria.

Heat Shield Property of Nanostructural-regulated Fe2O3/TiO2 Composites Filled with Polyacrylate Paint (나노구조 변화에 의한 Fe2O3/TiO2 복합재료를 충전한 Poly Acrylate 도료의 열차단 특성)

  • Fe2O3 nanoparticles with the mixed structure of cubic and nanorod were synthesized by precipitation, hydrothermal, sol-gel method, etching process and heat treatment. Fe2O3/TiO2 core-shell (CS) of type Fe2O3@TiO2 composite was fabricated on a 20 nm nanolayer of TiO2 coated on the surface of Fe2O3 nanoparticles. Fe2O3/TiO2 yolk-shell (YS) composite was prepared by chemical etching and heat treatment of Fe2O3/TiO2 CS nanoparticles. Physical properties of Fe2O3, Fe2O3@TiO2 CS and Fe2O3@TiO2 YS nanoparticles were characterized by FE-SEM, HR-TEM and X-ray diffraction. The solar reflectance, commission internationale de l'Elcairage (CIE) color coordinate and heat shield temperatures of Fe2O3, CS and YS type Fe2O3@TiO2 pigments filled with poly acrylate (PA) paints were investigated by UV-Vis-NIR spectrometer and homemade heat shield temperature measuring device. The Fe2O3@TiO2 YS red pigment filled PA composite exhibited excellent near infrared light reflecting performance and also reduced the heat shield temperature of 13 ℃ than that of Fe2O3 filled counterparts.

Changes in Carotenoid and Anthocyanin Contents, as well as Antioxidant Activity during Storage of Lettuce (저장에 따른 상추의 카로티노이드와 안토시아닌 함량 및 항산화능 변화)

  • Lettuce (Lactuca sativa) is an important dietary leafy vegetable that is primarily consumed as a fresh or salad material. It has a number of cultural varieties with green and/or red color. Carotenoids and anthocyanins are known to be responsible for these two colors, respectively. In this study, carotenoid and anthocyanin contents were determined to evaluate the stability of these functional pigments during storage at home. Analyses were carried out at the beginning, 3, 6, 9, and 12 days after harvest. In the course of storage at room temperature, total carotenoid levels rapidly decreased, and the decrease was found to be greatest during the first 3 days. Meanwhile, carotenoid level slightly changed within the first 9 days at $4^{\circ}C$ after harvest. This result suggests that carotenoids in green lettuce are more stable when refrigerated than at room temperature. Meanwhile, total anthocyanin content in red lettuce did not significantly decrease during storage at room temperature and $4^{\circ}C$, which indicates that anthocyanins have higher stability during storage compared with carotenoids in green lettuce. Anthocyanin extract exhibited higher antioxidant activity than carotenoid extract based on 2,2'-azino-bis(3-ethylbenzothazoline-6-sulfonic acid) (ABTS) radical scavenging assay. Antioxidant activity of anthocyanin extract may also be estimated directly by the presence of another potent hydrophilic antioxidant compound, which is ascorbic acid in this extract. In addition, anthocyanin extract showed about a 5-fold higher amount of anthocyanins than carotenoids in the carotenoid extract. The high correlation between carotenoid content with ABTS radical scavenging activity indicates that ABTS assay is more suitable than 1,1-diphenyl-2-picrylhydrazyl radical scavenging assay for detecting antioxidant capacity of carotenoid extract from lettuce.

The Effects of Anoxic Treatments on Color and Mechanical Property in Fabrics, Natural Dyed Fabrics, Papers, Natural Dyed Papers and Paints (저산소 농도 살충처리가 직물, 염색 직물, 종이, 염색지 및 채색편의 색상 및 기계적 성질에 미치는 영향)

  • Fabrics, natural dyed fabrics, papers, natural dyed papers and paints were examined effects of colors and mechanical properties for materials of museum collections under anoxic treatment. Anoxic conditions using nitrogen and argon were oxygen concentration 0.01%, temperature($20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$), 50% RH and exposure time 30 days. Examined fabrics were raw silk fabric, UV irradiated raw silk fabric, degummed silk fabric, UV irradiated degummed silk fabric, cotton fabric, and UV irradiated cotton fabric. Natural dyed silk and cotton fabrics were dyed with fresh indigo, indigo, safflower, gromwell, madder sappanwood, amur cork tree, turmeric, gardenia, barberry root, pagoda tree flower, cochineal, lac, alnus japonica, gallnut, chestnut shell, and combination(indigo and safflower, indigo and amur cork tree, indigo and pagoda tree flower, indigo and sappanwood). Papers were Korean papers(mulberry paper, mulberry(70%) and rice straw(30%) mixed paper), Japanese paper(gampi paper), cotton paper, refined linen paper, cotton, linen & manila mixed fibre furnish, copy paper, news print, and alum sized mulberry paper. Natural dyed papers were dyed with indigo, sappanwood, madder, safflower, gardenia, amur cork tree, and pagoda tree flower. Paints were painted on alum-sized papers and silk fabrics using glue and pigments(azurite, malachite, cinnabar, vermilion, orpiment, gamboge, red lead, haematite, iron oxide red, indigo(lake), lac, cochineal, safflower, madder root lake, celadonite, smalt, ultramarine blue, lapis lazuli, prussian blue, kaolin, lead white, oyster-shell white, and clam-shell white). The color differences(${\Delta}E^*$) of all examined materials were below 1.5 or lowered than control samples after anoxic treatment. The variations of tenacity of yarns of fabrics and natural dyed fabrics after anoxic treatment were within that of standard silk and cotton fabrics. Gases(nitrogen and argon) and temperatures of anoxic treatment did not also affected color differences and variations of tenacity of materials.

Effect of Halophilic Bacterium, Haloarcula vallismortis, Extract on UV-induced Skin Change (호염 미생물(Haloarcula vallismortis) 용해물의 자외선유발 피부변화에 대한 효과)

  • Skin carrys out protective role against harmful outer environment assaults including ultraviolet radiation, heavy metals and oxides. Especially, ultraviolet-B (UVB) light causes inflammatory reactions in skin such as sun burn and erythma and stimulates melanin pigmentation. Furthermore, the influx of UVB into skin cells causes DNA damage in keratinocytes and dermal fibroblasts, inhibition of extracellular matrix (ECM) synthesis which leads to a decrease in elasticity of skin and wrinkle formation. It also damages dermal connective tissue and disrupts the skin barrier function. Prolonged exposure of human skin to UVB light is well known to trigger severe skin lesions such as cell death and carcinogenesis. Haloarcula vallismortis is a halophilic microorganism isolated from the Dead Sea, Its growth characteristics have not been studied in detail yet. It generally grows at salinity more than 10%, but the actual growth salinity usually ranges between 20 to 25%. Because H. vallismortis is found mainly in saltern or salt lakes, there could exist defense mechanisms against strong sunlight. One of them is generation of additional ATP using halorhodopsin which absorbs photons and produces energy by potential difference formed by opening the chloride ion channel. It often shows a color of pink or red because of their high content of carotenoid pigments and it is considered to act as a defense mechanism against intense UV irradiation. In this study, the anti-inflammatory effect of the halophilic microorganism, H. vallismortis, extract was investigated. It was found that H. vallismortis extract had protective effect on DNA damage induced by UV irradiation. These results suggest that the extract of halophilic bacterium, H. vallismortis could be used as a bio-sunscreen or natural sunscreen which ameliorate the harmful effects of UV light with its anti-inflammatory and DNA protective properties.
