Screening Test of Wild Mushroom Extracts for Fibrinolytic Activity

  • To investigate the fibrinolytic activities of the Korean basidiomycetes, the Extracts of 50 wild mushrooms were tested for their fibrinolytic activites. Extract from Tricholoma sejunctum showed 175% increased activity to that of plasmin 1.0 U/ml. Marasmius siccus showed 54% of activity, and Laetiporus sulphureus var. miniatus and Macrolepiota procera, 43% and 26% activities, respectively to that of plasmin. But, Cystoderma amianthinum, Lepiota sp., Coprinus sp., Lycoperdon sp. were less than 10% of plasmin activity.

혈전용해능을 갖는 버섯류의 탐색 (Screening of Mushrooms Having Fibrinolytic Activity)

  • 혈전용해능을 가진 균류 (버섯류) 5종 Daedaleopsis styracina, Trichaptum abietium, Coriolus versicolor, Pisolithus tinctorius 그리고 Tricholomopsis decora 등을 선발하여 혈전용해효소 활성을 측정하였고, 기질 특이성을 조사하였다. 버섯 추출액을 조제하여 혈전 용해도를 측정한 결과 plasmin 1.0 unit 보다 $3{\sim}4$배의 높은 활성을 보였으며, 그 중 Pisolithus tinctorius이 가장 높은 활성(4.71 plasmin unit)을 나타내었고, Tricholomopsis decora가 Plasmin에 대한 기질 N-p-Tosyl-Gly-Pro-Lys p-nitroanilide에 대해 가장 높은 특이성(1.32 plasmin unit)을 보였다. 버섯의 균사체로부터 추출한 효소를 SDS-fibrin zymography 활성확인법에 의해 분석한 결과 분자량이 54와 61 kDa의 공통된 활성을 가지는 단백질을 확인하였으며, Trichaptum abietium는 100 kDa의 그리고 Tricholomopsis decora는 84 kDa의 강한 활성을 가지는 혈전용해효소를 확인하였다.

Plasminogen Activator Inhibitor-1 Antisense Oligodeoxynucleotides Abrogate Mesangial Fibronectin Accumulation

  • Excessive extracellular matrix (ECM) accumulation is the main feature of chronic renal disease including diabetic nephropathy. Plasminogen activator inhibitor (PAI)-1 is known to play an important role in renal ECM accumulation in part through suppression of plasmin generation and matrix metalloproteinase (MMP) activation. The present study examined the effect of PAI-1 antisense oligodeoxynucleotide (ODN) on fibronectin upregulation and plasmin/MMP suppression in primary mesangial cells cultured under high glucose (HG) or transforming growth factor (TGF)-${\beta}1$, major mediators of diabetic renal ECM accumulation. Growth arrested and synchronized rat primary mesangial cells were transfected with $1\;{\mu}M$ phosphorothioate-modified antisense or control mis-match ODN for 24 hours with cationic liposome and then stimulated with 30 mM D-glucose or 2 ng/ml TGF-${\beta}1$. PAl-1 or fibronectin protein was measured by Western blot analysis. Plasmin activity was determined using a synthetic fluorometric plasmin substrate and MMP-2 activity analyzed using zymography. HG and TGF-${\beta}1$ significantly increased PAI-1 and fibronectin protein expression as well as decreased plasmin and MMP-2 activity. Transient transfection of mesangial cells with PAI-1 antisense ODN, but not mis-match ODN, effectively reversed basal as well as HG- and TGF-${\beta}1$-induced suppression of plasmin and MMP-2 activity. Both basal and upregulated fibronectin secretion were also inhibited by PAI-1 antisense ODN. These data confirm that PAI-1 plays an important role in ECM accumulation in diabetic mesangium through suppression of protease activity and suggest that PAI-1 antisense ODN would be an effective therapeutic strategy for prevention of renal fibrosis including diabetic nephropathy.

수종 식물의 Plasmin 저해 활성 검색 (Plasmin Inhibitory Activity of Medicinal Plants)

  • Binding of urokinase-type plasminogen activator to its cellular receptor accelerate production of plasmin from plasminogen on the cell surface. Plasmin can digest extracellular matrix components and basement membranes through activating certain proMMPs, which is related to the invasiveness to the cells. Plasmin also acts the regulation of blood coagulation and relates closely to cardiovascular diseases such as stroke and coronary occlusion. Therefore, its inhibitors may be useful as antimetastatic agents and to the treatment of cardiovascular diseases. To search for plasmin inhibitors from plant resources, we screened plasmin inhibitory activities with 76 methanol extract of plant species. Among them, three plant samples showed strong inhibitory activities (>70%) and thirteen plant samples showed more than 50% inhibitory activities of plasmin. Their inhibitory activities were not directly related with uPA inhibitory activites and cell viability.

Screening Test of Wild Mushroom Methanol Extracts for Fibrinolytic and $\alpha-Glucosidase$ Inhibitory Activity

  • We investigated the fibrinolytic and ${\alpha}-glucosidase$ inhibitory activity of 55 wild mushroom methanol extracts. Among them, 14 mushrooms showed fibrinolytic activity. In particular, Amanita virgineoides showed the greatest enzyme activity (3.9 plasmin units/ml) by a fibrin plate assay. The fibrinolytic activities of Suillus pictus and Polypolellus varius were 3.8 plasmin units, and the activity of Gomphus fujisanensis was 2.8 plasmin units. Leccinum extremiorientale and Xerocomus nigromaculatus had the same activity with 2.3 plasmin units. In a ${\alpha}-glucosidase$ inhibitory activity test, Lactarius sp. showed the greatest inhibitory activity at 97.3%. The ${\alpha}-glucosidase$ inhibitory activities of Clitocybe odora, Xerocomus nigromaculatus, Melanoleuca melaleuca, Suillus pictus, and Gyroporus castaneus were 84.3%, 77.9%, 74.6%, 68.7%, and 65.4%, respectively. According to the results, because Suillus pictus and Xerocomus nigromaculatus have strong fibrinolytic and ${\alpha}-glucosidase$ inhibitory activities, the two mushrooms will be used as materials for the development of new biofunctional food.

야생버섯 메탄올추출물의 혈전용해 활성과 α-Glucosidase 저해 활성 (Fibrinolytic and α-Glucosidase Inhibitory Activities of Wild Mushroom Methanol Extracts)

  • 오대산에 자생하는 야생버섯 60종의 메탄올추출물을 이용하여 혈관계 질환과 당뇨병의 치료에 효과 있는 버섯을 확인하기위해, 혈전용해 활성과 ${\alpha}-glucosidase$ 저해 활성을 확인한 결과 11종의 버섯이 혈전용해 활성을 나타냈고, 6종의 버섯이 50% 이상의 ${\alpha}-glucosidase$ 저해 활성을 나타냈다. 뽕나무버섯류가 4.2 plasmin units의 가장 큰 혈전용해 활성을 나타냈으며, 독우산광대버섯, 미치광이 버섯류, 민자주방망이버섯이 2.3 plasmin units, 애괄대버섯은 2.1 plasmin units, 흰보라끈적버섯은 2.0 plasmin units 의 높은 혈전용해 활성을 나타냈다. ${\alpha}-Glucosidase$ 저해 활성 측정결과 노란다발버섯이 98.5%의 가장 큰 저해 활성을 나타냈고 비단그물버섯과 점마개버섯도 60% 이상의 높은 저해 활성을 나타냈다. 본 연구는 뽕나무벗섯류와 노란다발버섯은 혈관계질환 치료용 의약이나 기능성 식품으로 개발될 수 있음을 보여준다.

된장으로부터 혈전용해능을 가진 Bacillus sp.의 분리 및 동정 (lsolation of Bacillus Strains Secreting Fibrinolytic Enzymes from Doen-Jang)

  • 우리 나라의 전통 대두발효식품인 된장으로부터 혈전용해능을 가진 미생물을 분리하였다. 그 중 5종류의 균주를 Bergey's manual of systematic bacteriology에 의거하여 동정한 결과 Bacillus sp. 균주로 밝혀졌다. 분리된 Bacillus 속 미생물을 효소 유도배지에서 배양한 결과 B. amyloliquefaciens는 2.46 plasmin unit/ml, B pantothenticus는 3.82 plasmin unit/ml 의 혈전용해능을 가지고 있었고, B. subtilis는 4.94 plasmin unti/ml의 높은 혈전용해효소 생산능을 보여주었다. 분리 균주에 의하여 생산된 세포외 단백질을 SDS-PAGE와 reverse fibrin zymogram 활성측정법에 의해 확인한 결과 각 균주별로 서로 다른 혈전용해효소가 생산되었음을 확인하였다.

Spinosin Attenuates Alzheimer's Disease-Associated Synaptic Dysfunction via Regulation of Plasmin Activity

  • Hippocampal synaptic dysfunction is a hallmark of Alzheimer's disease (AD). Many agents regulating hippocampal synaptic plasticity show an ameliorative effect on AD pathology, making them potential candidates for AD therapy. In the present study, we investigated spinosin as a regulating agent of synaptic plasticity in AD. Spinosin attenuated amyloid β (Aβ)-induced long-term potentiation (LTP) impairment, and improved plasmin activity and protein level in the hippocampi of 5XFAD mice, a transgenic AD mouse model. Moreover, the effect of spinosin on hippocampal LTP in 5XFAD mice was prevented by 6-aminocaproic acid, a plasmin inhibitor. These results suggest that spinosin improves synaptic function in the AD hippocampus by regulating plasmin activity.

치악산 버섯추출물로부터 Fibrin 분해활성의 검색 (The Screening of Fibrinolytic Activities of Extracts from Mushrooms in Mt. Chiak)

  • 치악산에 자생하는 야생버섯 65종에서 fibrin 분해활성을 검색하였으며, 그 결과 9종의 버섯이 활성을 나타냈다. Agrocybe sp.와 Stropharia rugosoannulata는 작은 활성률을 보였으며, Lepiota sp.와 Coprinus comatus는 plasmin 1.5 unit의 $56%{\sim}58%$의 fibrin 분해활성을 보인 반면 Collybia maculata는 plasmin 1.5 unit와 거의 같은 활성을 보였고 Armillariella mellea와 Calocybe sp.와 Lepista nuda와 Trichaptum abietinum은 plasmin 1.5 unit의 약 2배의 fibrin 분해활성을 나타냈다.

Effect of Homogenization Pressure on Plasmin Activity and Mechanical Stress-Induced Fat Aggregation of Commercially Sterilized Ultra High Temperature Milk during Storage

  • Commercially sterilized ultra high temperature (UHT) milk was manufactured at different homogenization pressures (20, 25, and 30 MPa), and changes in fat particle size, mechanical stress-induced fat aggregation, plasmin activity, and lipid oxidation were monitored during ambient storage of the UHT milk for up to 16 wk. The particle sizes of milk fat globules were significantly decreased as homogenization pressure increased from 20 to 30 MPa (p<0.05). The presence of mechanical stress-induced fat aggregates in milk produced at 20 MPa was significantly higher than for UHT milk produced at either 25 or 30 MPa. This difference was maintained all throughout the storage. There were no significant differences in plasmin activity, trichloroacetic acid (12%, w/v) soluble peptides, and the extent of lipid oxidation. Based on these results, an increase of homogenization pressure from 20 (the typical homogenization pressure employed in the Korea dairy industry) to 25-30 MPa significantly decreased mechanical stress-induced fat aggregation without affecting susceptibility to lipid oxidation during storage.
