A Study on Hydraulic Control Characteristics of a Swashplate Type Axial Piston Pump-Regulator System by Linearization Analysis (사판식 액셜피스톤 펌프-레귤레이터계의 선형화해석에 의한 유압제어특성 고찰)

  • The regulator system has been modeled and combined to a swashplate type axial piston pump. Linear approximation has been performed for nonlinear coefficient terms of an axial piston pump-regulator model without significantly affecting accuracy. Based on the mathematical model of an axial piston pump-regulator system, a couple of characteristic curves of negative flow control and horsepower control are drawn, which show a good correlation with those of experimental results. So the simplified axial piston pump-regulator model in this paper is expected to be utilized not only for the design and analysis of hydraulic circuit of excavator but also for prevention of engine overload.

Subcloning and DNA Sequencing of the Phenol Regulatory Genes in Ralstonia eutropha JMP134 (Ralstonia eutropha JMP134에서 페놀분해에 관여하는 조절유전자의 Subcloning 및 염기서열 분석)

  • In this study, chromosomal DNA fragment related to the regulation of phenol metabolism in Ralstonia eutropha JMP 134 was cloned and sequenced. The result has shown that two open reading frames (ORF1 and ORF2) exist on this regulatory region. ORF1, which initiates from 454 bp downstream of the stop codon of the phenol hydroxylase genes, was found to be composed of 501 amino acids. ORF2, whose start codon is overlapped with the stop codon of ORFl, was found to contain 232 amino acids. The comparison of amino acid sequences with other proteins has revealed that ORF1 belongs to the family of NtrC transcriptional activator, whereas ORF2 shares high homology with the family of GntR protein, which is known to be a negative regulator. ORF1 and ORF2 were designated as a putative positive regulator, phlR2 and a negative regulator phlA, respectively. Possible regulatory mechanisms of phenol metabolism in this strain was discussed.

  • This paper proposes the design of STATCOM(static synchronous compensator) stabilizer for improving power system stability using fuzzy logic controller(FLC). The STATCOM DC voltage regulator contributes negative damping to the power system as the installation of STATCOM DC voltage regulator. STATCOM stabiliser is superimposed on the AC voltage regulator to compensate the negative damping effect. To evaluate usefulness of the proposed method, we perform the nonlinear simulation on a single-machine infinite bus system. As results of the simulations, the proposed method shows better control performance than PI controller in terms of damping effects.

  • Geraniin, a hydrolysable tannin, used in traditional medicine in Southeast Asia, is known to exhibit various biological activities. As an antioxidant it is known to up-regulate phase II enzyme Heme oxygenase-1 (HO-1). However its mechanism is not clearly understood. Nuclear factor erythroid-derived 2 related factor 2 (Nrf-2) is transcriptionally up-regulated by Extracellular signal-regulated kinase (ERK) 1/2 and retained in nucleus due to inactivated Glycogen synthase kinase 3 beta ($GSK-3{\beta}$). Geraniin additionally down-regulates expression of microRNA 217 and 377 (miR-217 and miR-377) which target HO-1 mRNA. Expression of BTB and CNC homolog 1 (BACH-1), another regulator of HO-1, is also down-regulated by up-regulating microRNA 98 (miR-98), a negative regulator of BACH-1. Thus, geraniin up-regulates HO-1 expression both through activating its positive regulator Nrf-2 and by down-regulating its negative regulator BACH-1. Up-regulation of HO-1 also confers protection to HepG2 cells from tertiary butyl hydroperoxide (TBH) induced cytotoxicity.

Protein Kinase A Functions as a Negative Regulator of c-Jun N-terminal Kinase but not of p38 Mitogen-activated Protein Kinase in PC12 Cells

  • Cyclic-AMP-dependent protein kinase (PKA) seems to function as a negative regulator of the c-Jun $NH_2-terminal$ kinase (JNK) signaling pathway. We demonstrate here that the activity of the PKA catalytic subunit (PKAc) is reduced in apoptotic PC12 pheochromocytoma cells. Apoptotic progress was inhibited by dibutyryl cyclic AMP (dbcAMP), an analog of cAMP. The rescue by dbcAMP was attributable to inhibition of the JNK but not of the p38 signaling pathway, due to the induction of PKA activity. JNK was present in immunocomplexes of PKAc, and PKAc phosphorylated JNK in vitro. Presence of p38 kinase, however, was not prominent in immunocomplexes of PKAc. Our data suggest that JNK is a target point of negative regulation by PKAc in the JNK signaling pathway.

  • This study deals with a dynamic behavior analysis of pump control regulator varied the swash plate tilting angle with the positive and negative direction. To accomplish it's purpose, modeling and displacement response analysis about principal parts (spool, servo piston, feedback lever, sleeve) of pump control regulator was performed. We have been able to verifying the propriety of servo mechanism and design parameters of pump control regulator by research results. So, it respect to utilized with useful research data at variable displacement control valve development of variable displacement piston pump.

  • The regulator problem can be considered as some impulsive disturbance rejection one. In this point of view, the rate of decay is one of important factors for regulation and depends on how negative the real parts of the eigenvalues of closed-loop system. The algorithm that the closed-loop system has eigenvalues lying within a vertical. strip is useful for rapid disturbance rejection. This paper presents a design method for a linear quadratic regulator of two-time scale system with eigenvalues in a vertical strip by use of time-scale separation property.

HDAC3 acts as a negative regulator of angiogenesis

  • Histone deacetylase-3 (HDAC3) is involved in cellular proliferation, apoptosis and transcriptional repression. However, the role of HDAC3 in angiogenesis remains unknown. HDAC3 negatively regulated the expression of angiogenic factors, such as VEGF and plasminogen activator inhibitor-1 (PAI-1). HDAC3 showed binding to promoter sequences of PAI-1. HDAC3 activity was necessary for the expression regulation of PAI-1 by HDAC3. VEGF decreased the expression of HDAC3, and the down-regulation of HDAC3 enhanced endothelial cell tube formation. HDAC3 negatively regulated tumor-induced angiogenic potential. We show the novel role of HDAC3 as a negative regulator of angiogenesis.

LAMMER Kinase Lkh1 Is an Upstream Regulator of Prk1-Mediated Non-Sexual Flocculation in Fission Yeast

  • The cation-dependent galactose-specific flocculation activity of the Schizosaccharomyces pombe null mutant of $lkh1^+$, the gene encoding LAMMER kinase homolog, has previously been reported by our group. Here, we show that disruption of $prk1^+$, another flocculation associated regulatory kinase encoding gene, also resulted in cation-dependent galactosespecific flocculation. Deletion of prk1 increased the flocculation phenotype of the $lkh1^+$ null mutant and its overexpression reversed the flocculation of cells caused by lkh1 deletion. Transcript levels of $prk1^+$ were also decreased by $lkh1^+$ deletion. Cumulatively, these results indicate that Lkh1 is one of the negative regulators acting upstream of Prk1, regulating non-sexual flocculation in fission yeast.

Pleckstrin homology domain of phospholipase D2 is a negative regulator of focal adhesion kinase

  • Phospholipase D2 (PLD2) has been implicated in the tyrosine kinase-mediated signaling pathways, but the regulation events are yet to be identified. Herein, we demonstrate that pleckstrin homology (PH) domain of PLD2 (PLD2-PH) exerts an antitumorigenic effect via the suppression of PLD2 and focal adhesion kinase (FAK). The kinase domain of FAK interacts with PLD2-PH and induces tyrosine phosphorylation and activation of PLD2. Furthermore, PLD2 increased tyrosine phosphorylation of FAK. However, ectopic expression of the PLD2-PH competes for binding to FAK and reduces the interaction between PLD2 and FAK, thereby suppressing FAK-induced PLD activation and tyrosine phosphorylation of FAK. The PLD2-PH suppressed the migration and invasion of glioblastoma cells, as well as tumor formation in a xenograft mouse model. This study uncovers a novel role of PLD2-PH as a negative regulator of PLD2 and FAK.
