FA/Mel@ZnO nanoparticles as drug self-delivery systems for RPE protection against oxidative stress

  • Drug self-delivery systems can easily realize combination drug therapy and avoid carrier-induced toxicity and immunogenicity because they do not need non-therapeutic carrier materials. So, designing appropriate drug self-delivery systems for specific diseases can settle most of the problems existing in traditional drug delivery systems. Retinal pigment epithelium is very important for the homeostasis of retina. However, it is vulnerable to oxidative damage and difficult to repair. Worse still, the antioxidants can hardly reach the retina by non-invasive administration routes due to the ocular barriers. Herein, the targeted group (folic acid) and antioxidant (melatonin) have been grafted on the surface of ZnO quantum dots to fabricate a new kind of drug self-delivery systems as a protectant via eyedrops. In this study, the negative nanoparticles with size ranging in 4~6 nm were successfully synthesized. They could easily and precisely deliver drugs to retinal pigment epithelium via eyedrops. And they realized acid degradation to controlled release of melatonin and zinc in retinal pigment epithelium cells. Consequently, the structure of retinal pigment epithelium cells were stabilized according to the expression of ZO-1 and β-catenin. Moreover, the antioxidant capacity of retinal pigment epithelium were enhanced both in health mice and photic injury mice. Therefore, such new drug self-delivery systems have great potential both in prevention and treatment of oxidative damage induced retinal diseases.

Nano-TiO2를 이용한 자외선차단 마이카 복합체 제조 (Preparation of blocking ultraviolet mica composites using Nano-TiO2)

  • 자외선차단 화장품은 기능성 화장품 중의 하나로서, 유 무기 자외선차단물질이 함유되어 있다. 무기계 자외선차단제는 주로 산화아연, 이산화티탄 등이 있다. 무기계 자외선차단제는 입자의 지름이 60 ~ 100 nm로 자외선 A, B의 차단능이 좋은 것으로 알려져 있다. 또한자외선을 포함한 태양광선에 대해 비활성이 크고 안전성이 우수하다. 그리고 유기계 자외선차단제처럼 피부에 흡수 또는 축적되지 않으므로 피부자극이나 알레르기를 유발하지 않는다. 본 연구에서는 판상 무기안료인 마이카, 자외선차단 효과를 갖는 이산화티탄 나노입자, 소수성 실리카를 각각 계면활성제로 표면처리 하였고, 각 물질의 전하 차이에 따른 비화학적인 상호 인력 작용에 의해 마이카에 이산화티탄 나노입자, 실리카를 물리적으로 흡착시켰다. 이후, 소수성 표면처리제인 실란을 표면처리 하여 소수성을 갖는 자외선 차단 판상 마이카 복합체를 제조하였다. 자외선 차단 판상 마이카 복합체는 일반적인 나노입자 이산화티탄의 응집성을 개선하고 균일한 분산에 따른 자외선차단 효과가 증대되었으며, 소수성으로 표면처리를 하여 화장품 제형에서의 분산안정성을 크게 개선할 수 있었다. 안료의 표면전하는 제타전위로 평가하였으며, 제조된 자외선차단 마이카 복합체의 특성 평가는 FE-SEM, XRD, FT-IR, UV-VIS 등으로 확인하였다.

현미경, Flow Cytometer, HPLC 색소자료 및 원격탐사를 이용한 이어도 관측기지 주변수의 식물플랑크톤 연구 (Phytoplankton in the Waters of the Ieodo Ocean Research Station Determined by Microscopy, Flow Cytometry, HPLC Pigment Data and Remote Sensing)

  • Phytoplankton community structure and distribution pattern in the surface water around the Ieodo Ocean Research Station were investigated during seven cruises carried out from July, 2003 to October, 2004. Samples were analyzed using various tools including a microscope, flow cytometer, and HPLC. Satellite images were used to analyze spatio-temporal phytoplankton biomass distribution. SeaWiFS chlorophyll a (chl a) images showed that spring blooms occurred in April-May near the Ieodo Station, and these waters were under the influence of Changjiang Dilute Water during July-October. Also, during the July-October period, HPLC pigments data showed increasing zeaxanthin concentrations, a marker pigment of cyanobacteria whereas increasing concentrations of various other pigments such as fucoxanthin, peridinin, prasinoxanthia alloxanthin, 19'-hexanoyloxyfucoxanthin and chlorophyll b were noted during spring blooms. Such pigment marker data were consistent with picoplankton data analyzed by flow cytometer and nano-microplankton analyzed by microscope. The pigment-CHEMTAX method was used to drive the phytoplankton group apportioned chi a. Diatoms, chlorophytes, dinoflagellates, and cryptophytes comprised 25.8, 20.7, 15.9, and 14.1%, respectively, of the total chl a in May. Average cyanobacteria concentrations in July-October contributed 25.4% of the total concentration. This was the highest percent contribution and was followed by chlorophytes, diatoms, and prymnesiophytes. This study discusses results from various methods, similarities and differences in the results among those methods, and the application range of the results from different analytical methods. Also, the study reveals a detailed phytolpankton community structure in the waters around the Ieodo Station, and suggests future monitoring considerations in relation to cell morphology, ecology and diversity factors according to taxonomic groups.

안료배합용 실리카 입자사이즈에 따른 근적외선 반사율을 향상시킨 고기능성 도료 제조 (The development of highly functional paints improving NIR reflectance by investigating silica particles size for pigment mixing)

  • In order to overcome the urban heat island effect, highly functional paint is attracting attention as a promising means by shielding heat on the structure (building) surface. When a paint was prepared containing nano-sized silica particles, the heat-insulating performance was relatively higher than that of paints with other sizes. In addition, developed paints showed enhanced properties such as chemical resistance and abrasion resistance test because of the presence of nano-sized silica included in functional paint.

  • The black/white electronic ink containing high mobility white nano particles and the organic black pigment particles dispersed in dielectric fluid were prepared. A charge control agent affects the electrophoretic zeta potentials of white particle, which show the maximum value in zeta potential. The electronic ink panel fabricated with the charged white particles and the black particles exhibits more than 15:1 contrast ratio at 10V.

Biocidal Activity of Metal Nanoparticles Synthesized by Fusarium solani against Multidrug-Resistant Bacteria and Mycotoxigenic Fungi

  • Antibiotic resistance by pathogenic bacteria and fungi is one of the most serious global public health problems in the 21st century, directly affecting human health and lifestyle. Pseudomonas aeruginosa and Staphylococcus aureus with strong resistance to the common antibiotics have been isolated from Intensive Care Unit patients at Zagazig Hospital. Thus, in this study we assessed the biocidal activity of nanoparticles of silver, copper and zinc synthesized by Fusarium solani KJ 623702 against these multidrug resistant-bacteria. The synthesized Metal Nano-particles (MNPs) were characterized by UV-Vis spectroscopy, transmission electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction, and Zeta potential. The Fourier transform infrared spectroscopy (FTIR) result showed the presence of different functional groups such as carboxyl, amino and thiol, ester and peptide bonds in addition to glycosidic bonds that might stabilize the dispersity of MNPs from aggregation. The antimicrobial potential of MNPs by F. solani against the multidrug-resistant (MDR) P. aeruginosa and S. aureus in addition to the mycotoxigenic Aspergillus awamori, A. fumigatus and F. oxysporum was investigated, based on the visual growth by diameter of inhibition zone. Among the synthesized MNPs, the spherical AgNPs (13.70 nm) displayed significant effect against P. aeruginosa (Zone of Inhibition 22.4 mm and Minimum Inhibitory Concentration 21.33 ㎍/ml), while ZINC oxide Nano-Particles were the most effective against F. oxysporum (ZOI, 18.5 mm and MIC 24.7 ㎍/ml). Transmission Electron Microscope micrographs of AgNP-treated P. aeruginosa showed cracks and pits in the cell wall, with internalization of NPs. Production of pyocyanin pigment was significantly inhibited by AgNPs in a concentration-dependent manner, and at 5-20 ㎍ of AgNPs/ml, the pigment production was reduced by about 15-100%, respectively.

  • A number of research groups worldwide are studying electronic implants that can be mounted on retinal optic nerve/visual cortex to restore vision of patients suffering from retinal degeneration. The implants consist of a neural interface made of biocompatible materials, one or more integrated circuits for stimuli generation, a camera, an image processor, and a telemetric channel. The realization of these classes of neural prosthetic devices is largely due to the explosive development of micro- and nano-electronics technologies in the late $20^{th}$ century and biotechnologies more recently. Animal experiments showed promise and some human experiments are in progress to indicate that recognition of images can be obtained and improved over time. We, at NBS-ERC of SNU, have started our own retinal implant project in 2000. We have selected polyimide as the biomaterial for an epi-retinal stimulator. In-vitro and in-vivo biocompatibility studies have been performed on the electrode arrays. We have obtained good affinity to retinal pigment epithelial cells and no harmful effect. The implant also showed very good stability and safety in rabbit eye for 12 weeks. We have also demonstrated that through proper stimulation of inner retina, meaning vision can be obtained.

HPLC를 이용한 광합성색소 분석과 CHEMTAX 프로그램을 이용한 식물플랑크톤 군집조성 연구 (Application of Photosynthetic Pigment Analysis Using a HPLC and CHEMTAX Program to Studies of Phytoplankton Community Composition)

  • High-Performance Liquid Chromatograph(HPLC)를 이용하여 식물플랑크톤의 광합성색소를 분석한 결과를 토대로 CHEMTAX 프로그램을 이용하여 식물플랑크톤 군집조성을 추정하는 방법이 많이 이용되고 있다. CHEMTAX 프로그램은 식물플랑크톤 군집별(class level)로 체내에 존재하는 주요색소의 chlorophyll a에 대한 상대적인 비 값을 기초로 분석을 한다 본 연구는 국내 해역에서 분리한 식물플랑크톤 군집별 주요 종들이 가지는 광합성색소의 상대적인 비 값을 파악하고 이 자료를 기초로 남해안에 위치한 여자만과 가막만에서 측정한 광합성색소 분석결과에 대해서 CHEMTAX 프로그램을 이용하여 식물플랑크톤의 군집조성을 파악하였다. 국내해역에 존재하는 같은 군집종들 사이에서도 주요색소의 상대적인 비 값은 차이를 보였으며, 국외의 다른 해역에서 제시된 상대적인 비 값과도 상당한 차이를 보였다. 본 연구에서 얻은 비 값을 기초로 식물플랑크톤 군집조성을 분석한 결과, 다른 해역에서 제시된 비 값을 이용하여 분석한 결과와 유의한 차이를 보였다. 우점하는 군집(bacillariophyceae와 dinophytes)에 대해서는 5% 이내에서 차이를 보였으며, 비록 chlorophyll a에 대해서 낮은 기여도를 보이지만 pico와 nano 크기의 군집들(cyanophytes와 prasinophytes)에서는 상당한 차이(2배 이상)를 보였다. 비록 HPLC-CHEMTAX 분석방법은 식물플랑크톤 군집조성 연구에 유용하게 이용되고 있지만, 보다 정확한 결과와 식물플랑크톤의 생리, 생태를 이해하기 위해서는 연구해역에 존재하는 식물플랑크톤 종들에 대한 주요색소의 상대적인 비 값에 대한 연구가 계속적으로 진행되어야 할 것으로 판단된다.

어트리션밀을 이용한 CoAl2O4 나노 무기 안료의 제조 및 특성 평가 (Preparation and characterization of CoAl2O4 blue ceramic nano pigments by attrition milling)

  • $CoAl_2O_4$는 열적 및 화학적으로 매우 안정한 물질이며 독특한 광학적 특성으로 인해 도자기 제품, 유리, 페인트, 플라스틱을 장식하는 세라믹 착색제로 사용되어져 왔다. 본 연구에서는 착체중합법(Polymerized complex method)과 고상합성법(Solid state reaction)을 이용하여 단상의 $CoAl_2O_4$ 청색 무기 안료를 합성하였고, 잉크젯 프린팅용 잉크로의 응용 시 노즐 막힘 및 분산안정성과 같은 문제점을 방지하기 위해 고에너지 밀링 공정 중 하나인 어트리션밀을 이용하여 입자를 미세화하였다. 밀링 공정은 직경 1 mm의 지르코니아 볼을 이용하여 BPR(Ball to powder weight ratio) 100 : 1, 800 rpm의 조건에서 3시간 동안 진행되었다. 합성된 안료는 XRD, FE-SEM, TEM, PSA, 그리고 CIE $L^*a^*b^*$ 측정을 통하여 어트리션밀링 전후의 특성을 분석하였다. XRD 측정 결과 단상의 $CoAl_2O_4$ 청색 무기 안료가 합성되었고, 고에너지 밀링 공정을 통하여 100~200 nm의 입자 크기를 가지는 청색 나노 무기 안료를 얻었다.

Isolation of Lactobacillus plantarum subsp. plantarum Producing C30 Carotenoid 4,4'-Diaponeurosporene and the Assessment of Its Antioxidant Activity

  • Carotenoids are organic pigments with antioxidant properties and are widespread in nature. Here, we isolated five microbes, each forming yellow-colored colonies and harboring C30 carotenoid biosynthetic genes (crtM and crtN). Thereafter, Lactobacillus plantarum subsp. plantarum KCCP11226, which showed the highest carotenoid production, was finally selected and the produced pigment was identified as C30 carotenoid 4,4'-diaponeurosporene. This strain exhibited the highest survival rate under oxidative stress and its carotenoid production was also enhanced after exposure to 7 mM H2O2. Moreover, it showed the highest ability to scavenge DPPH free radical. Our results suggested that L. plantarum subsp. plantarum KCCP11226, which produces 4,4'-diaponeurosporene as a natural antioxidant, may be a functional probiotic.
