The purpose of this study is to analyze the writing activity materials presented in the thirteen middle school textbooks and determine if the writing activity materials are appropriate for the goals and contents of the 7th national curriculum. The writing activity materials are analyzed based on three criteria. First, the writing activity materials are analyzed based on Rivers'(1981) five writing guidances, which are specified with detailed writing activity materials. The first criterion was suggested by Young-Ran Lee(2004). Second, the writing activity materials in the textbooks are analyzed according to the criterion prescribed by the 7th national curriculum, which are specified with writing activities presented by Rivers(1981), Raimes(1983), Hedge(1988), and Beverly and Carol(1988). Third, the writing activity materials related to other skills, such as listening, speaking, and reading, in the textbooks are selected and analyzed according to the proportion of the integration of skills. The results are as follows: First, writing activity materials in the textbooks belong to the type of 'reproduction', ie. the second stage of Rivers' writing guidance, 'recombination, which is the third stage of Rivers' writing guidance, and the type of 'guided writing', noted on the fourth stage of the guidance. Second, the writing activity materials satisfy the criterion in the 7th national curriculum, as for the proportions, but they are not balanced. Third, most of the writing activity materials contain writing activities that involve non-writing skills such as listening, speaking and reading, but they are not balanced. Particularly, writing activity materials related to reading activities are common.
Bioassay-directed chromatographic fractionation of a methylene chloride extract of Ailanthus altisshima indicated the presence of 20(S), 24(R), epoxy-25-hydroxydammarane-3-one (compound 1, ocotillone) which was isolated from this plant, for the first time. Antimicrobial activity of compound 1 was measured by inhibition of bacterial and fungal cells growth and by a hemolytic assay with human erythrocytes, respectively. The results revealed that compound 1 had potent antibacterial activity against Cram-negative bacteria, P. aeruginosa and S. typhimurium, that were without hemolytic activity, whereas it had weak antimicrobial activity against Gram-positive bacteria and fungi. These results demonstrated that the compound 1 has more antibacterial activity against 6ram-negative bacteria, which have no hemolytic activity, than Gram-positive bacteria and fungi. This is the first report on the biological activities of the compound 1.

Mulberry leaves containing 1-deoxynojirimycin (DNJ) have been recognized as a potentially important source for prevent or treat hyperglycemia. However, DNJ content of natural mulberry leaf are as low as 0.1%. Thus, the most effective method for increasing ${\alpha}$-glucosidase inhibitory activity with the DNJ high-production is needed. In this study, we investigated the influence of ${\alpha}$-glucosidase inhibitory activity according to different pH values (6-9) and inoculation amounts (0.1-0.5%) when Bacillus subtilis cultured on mulberry leaf powder media. We confirmed that ${\alpha}$-glucosidase inhibitory activity was difference according to culture conditions of different pH values, inoculation amounts, and fermentation times. The results of mulberry leaf fermentation according to pH values and inoculation amounts were shown that the optimal conditions for ${\alpha}$-glucosidase inhibitory activity were defined as pH 7 and 9, inoculation amount 0.4%, and incubation until 2 to 4 days. These results can be provided a basic data for the optimal culture conditions increasing ${\alpha}$-glucosidase inhibitory activity from mulberry leaf fermentation.
This Study was tarried out to investigate the relationship ACE inhibitory activity and degradations of sulfur containing materials in Dolsan leaf mustard juice (DLMJ). The changes of sulfur containing materials which were treated with autolysis, myrosinase, ascorbate and papain were studied, as well as the changes of ACE inhibitory activity in DLMJ. At $37^{\circ}C$, sulfur contain-ing materials by autolysis decreased most rapidly from $0.43\%$ to $0.13\%$ in the second day. Conversely. ACE inhibitory activity increased most from $66\%$ to $87\%$. in the second day at $37^{\circ}C$. As myrosinase concentrations increased more, sulfur containing materials in DLMJ decreased more. The ACE inhibitory activities at 0, 0.5, 1, 2, and 4 Units of myrosinase for 240 min later were 70, 74, 75, 82, and $85\%$, respectively. At 1 mM ascorbate. concentrations of Sulfur containing materials in DLMJ decreased more significantly on the second day than on the other days. At 1 mM ascorbate for 6 days, ACE Inhibitory activity reached a maximum of about $92\%$. And, an increase of papain concentration was noted in accordance with a decreased sulfur containing materials. The maximum rate of AEC inhibitory activity at control, 3, 6, and 12 Units of papains treatments was shown as 70, 70, 75, and $78\%$ at 60 min, respectively. These results suggested that the degradation of sulfur containing materials led to the increase of ACE inhibitory activity. Consequently, it was suggested that ACE inhibiting was significantly related to the degradatives of sulfur containing materials.

Chitosan, a cationic polysaccharide, has been widely used as a dietary supplement and in a variety of pharmacological and biomedical applications. The antifungal activity and mechanism of action of low molecular weight water-soluble chitosan (LMWS-chitosan) were studied in fungal cells and vesicles containing various compositions of fungal lipids. LMWS-chitosan showed strong antifungal activity against various pathogenic yeasts and hyphae-forming fungi but no hemolytic activity or cytotoxicity against mammalian cells. The degree of calcein leakage was assessed on the basis of lipid composition (PC/CH; 10:1, w/w). Our result showing that LMWS-chitosan interacts with liposomes demonstrated that chitosan induces leakage from zwitterionic lipid vesicles. Confocal microscopy revealed that LMWS-chitosan was located in the plasma membrane. Finally, scanning electron microscopy revealed that LMWS-chitosan causes significant morphological changes on fungal surfaces. Its potent antibiotic activity suggests that LMWS-chitosan is an excellent candidate as a lead compound for the development of novel anti-infective agents.
This study was carried out to investigate the antibacterial activity against pathogens of acne and the anti-inflammatory effect of 75% ethanol extract and its fractions from the leaves of Prunus sargentii. In the antibacterial activity by the disc diffusion assay, the extract showed the highest effect against Propionibacterium acnes, Staphylococcus aureus and Staphylococcus epidermidis in 5 mg/disc. However, the ethyl acetate fraction showed the highest antibacterial activity in 1 mg/disc. On the other hand, the hexane and chloroform fraction showed strong nitric oxide (NO) production inhibitory effect in lipopolysaccharide (LPS)-stimulated Raw 264.7 cell. In the cell viability of Raw 264.7 by MTT assay, the extract and all fractions were exhibited normal viabilities as nontoxic result. Consequently, the extract from the leaves of P. sargentii and its ethyl acetate fraction could be applicable to functional materials for antibacterial activity related fields. Moreover, the hexane and chloroform fraction could be applicable to candidate materials as anti-inflammatory agent.
Silybin has a potent antibacterial activity, more potent than silymarin II, against gram-positive bacteria without hemolytic activity, whereas it has no antimicrobial activity against gram-negative bacteria or fungi. The mode of action of silybin against the gram-positive bacterial cell was examined by investigating the change in plasma membrane dynamics of bacterial cells using 1 ,6-diphenyl-1,3,5-hextriene (DPH) as a membrane probe and by assessing the inhibition of macromolecular synthesis using radiolabeled incorporation assay. The results showed that silybin inhibited RNA and protein synthesis on gram-positive bacteria.
Four flavonoids (1-4), a phenylethyl glycoside (5), and a phenylpropanoid glycoside (6) were isolated from the flowers of Buddleia officinalis (Loganiaceae). Their structures were determined by chemical and spectral analysis. Among the isolated compounds, luteolin (1) and acteoside (6) exhibited the most potent antioxidative activity on the NBT superoxide scavenging assay. In addition, compounds 1-6 revealed weak antifungal activity, and no hemolytic activity.
Eukaryotic replication protein A (RPA) is a single-stranded(ss) DNA binding protein with multiple functions in DNA replication, repair, and genetic recombination. The 70-kDa subunit of eukaryotic RPA contains a conserved four cysteine-type zinc-finger motif that has been implicated in the regulation of DNA replication and repair. Recently, we described a novel function for the zinc-finger motif in the regulation of human RPA's ssDNA binding activity through reduction-oxidation (redox). Here, we show that yeast RPA's ssDNA binding activity is regulated by redox potential through its RPA32 and/or RPA14 subunits. Yeast RPA requires a reducing agent, such as dithiothreitol (DTT), for its ssDNA binding activity. Also, under non-reducing conditions, its DNA binding activity decreases 20 fold. In contrast, the RPA 70 subunit does not require DTT for its DNA binding activity and is not affected by the redox condition. These results suggest that all three subunits are required for the regulation of RPA's DNA binding activity through redox potential.

The antioxidant activity of silkworm powder treated by proteolytic enzyme was investigated. Total protein content of silkworm power was assayed using BCA, Bradford assays and SDS-polyacrylamide gel electrophoresis (PAGE) with alkaline protease treatment conditions including temperature and pH. The optimum condition of alkaline protease treatment for silkworm powder was found to be $60^{\circ}C$ and pH 7. The alkaline protease treatment resulted in increased contents of free amino acids, total polyphenol and total flavonoid compared to control group. The silkworm hydrolysates showed excellent antioxidant activities in various in vitro models such as 2,2 diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, 2,2 - azino-bis(3-ethylbenzthiazoline-6)-sulfonic acid (ABTS) radical scavenging activity. These results provide useful information for using silkworm powder as an ingredient in functional foods and for exploiting alkaline protease treatment to improve the extractability and bioactivity of a raw material.
