산초(Zanthoxylum Schinifolium) 뿌리, 줄기 및 잎 추출물의 항산화, DDPH Radical 소거 작용 및 항혈전 효과 (Effects of Antioxidative, DPPH Radical Scavenging Activity and Antithrombogenic by the Extract of Sancho (Zanthoxylum Schinilolium))

  • Effects of root, stem and leaf extract of sancho (Zanthoxylum schinifolium) on the inhibition of lipid peroxidation in the hepatic microsome of rat, DPPH radical scavenging activity and activated partial thromboplastin times (APTT) were examined in vitro. The highest inhibition of hepatic microsomal lipid peroxidation was observed by ethyl acetate fraction than that of methylene chloride fraction of the root and stem extracts. The high inhibition of lipid peroxidation was determined in the leaf, the root and the stem in order. The DPPH radical scavenging activity of ethyl acetate fraction was higher than that of n-butanol fraction and it was similar to the root and the steam extract. It was similar to the inhibition of hepatic microsomal lipid peroxidation. The DPPH radical scavenging activity was the highest in 2.500mg/mL of ethyl acetate fraction and it was 4.4 fold higher than that of $\alpha-tocopherol$, as an antioxidant standard. The DPPH radical scavenging activity was dependent on the extract concentration in the range of 0.125-5.000 mg/mL. The throm-boplastin times were higher than that of n-butanol fraction and it was similar to the root and the steam extracts. The leaf extract showed the highest antithrombogenic effect followed by the stem and then the root extract. The activated partial thromboplastin times were ependent on the extract concentration in the range of 0.100-2.000 mg/mL. Consequently, the effects of antioxidative, DPPH radical scavenging activity and antithrombogenic of Z. schinifolium was observed due to the inhibition of lipid peroxidation and the DPPH radical scavenging activity by methylene chloride, n-butanol and ethyl acetate fraction of the leaf extract. 천연약용식물의 활성산조종에 의한 산화적 손상의 보호 효과 (Protective Effect of Natural Medicinal Plants against Oxidative Damage Induced by Reactive Oxygen Species)

  • In order to evaluate anti -oxidant activities and protective effect against oxidatve damage, DPPH radical scavenging activity and lipid peroxidation inhibitory activity were measured among methanol extracts prepared from natural medicinal plants. Fourteen natural medicinal plants which were reported to have anti -oxidative or anti-inflammatory effects were selected based on our previous report. In addition to the total methanol extracts, n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions were prepared from each total extract. DPPH radical scavenging assay was performed against 14 total extracts and all samples showed dose-dependent activities in various extent. Among those, 6 samples, methanol extracts of Euryale ferox, paeonia suffruticosa, Areca catechu var. dulcissima, Cinnamomun cassia, Alpinia katsumadai and Betula platyphlla var. japonica showed IC$\sub$50/ value lower than 6.0 $\mu\textrm{g}$/ml. The highest DPPH radical scavenging activity was found in ethylacetate fraction of paeonia suffruticosa with IC$\sub$50/ value of 1.1 $\mu\textrm{g}$/ml. Analysis of lipid peroxidation inhibitory activity on hydrogen peroxide-induced oxidative damage in Chinese hamster lung fibroblast (V79-4) cells revealed that the highest inhibitory effect was observed in methanol extract of Betula platyhpylla var. japonica. Lipid peroxidation inhibitory activity was observed as a dose-dependent manner in all samples used in this study. Among fraction samples, ethylacetate fraction of Alpinia katsumadai had the strongest inhibitory activity with IC$\sub$50/ value of 0.9 $\mu\textrm{g}$/ml.

해당화의 항산화 효과 (Antioxidant Activity of Rosa rugosa)

  • 본 연구에서는 염생식물의 하나인 해당화의 항산화효과를 탐색하기 위해 해당화의 dichloromethane과 methanol의 조추출물을 합하여 n-hexane, 15% aq. MeOH, n-BuOH, $H_2O$로써 순차적으로 용매 분획하여 DPPH 라디칼, peroxynitrite의 소거 활성과 지질 과산화 저해 활성을 측정하였다. 그 결과 각각의 실험에서 해당화의 유의적인 항산화 효과가 확인되었으며, 특히, 4개의 분획물들 중 n-BuOH 획분이 가장 뛰어난 항산화 효과가 있는 것으로 나타났다. 따라서 해당화의 항산화 효과는 비교적 극성이 큰 2차 대사산물에 의한 것임을 추측할 수 있으며, 해당화의 많은 생물학적인 활성들 중 peroxynitrite 소거 효과와 마우스 간 균질물을 이용한 지질 과산화물 생성 억제 활성에 관한 보고는 이것이 처음이다.

우엉 에탄올 추출물의 항산화활성과 항돌연변이 효과 (Antioxidative and Antimutagenic Effects of $Arctium$ $lappa$ Ethanol Extract)

  • The antioxidant activities of the ethanol extract of Arctium lappa were assessed by measuring the 1,1-diphenyl-2-picrylhydrazyl( DPPH) radical scavenging effect, inhibition of $Fe^{2+}$-induced lipid peroxidation, inhibition of malondialdehyde(MDA)-bovine serum albumin(BSA) conjugation reaction and antimutagenic capacities using the Ames test. The DPPH radical scavenging activity and inhibition of $Fe^{2+}$-induced lipid peroxidation of the $Arctium$ $lappa$ ethanol extract significantly increased in a dose-dependent manner. In the radical scavenging assay using DPPH, the $IC_{50}$ of the Arctium lappa extract was 296 ${\mu}g$/assay(1.29 mg of dry sample). In addition, the $IC_{50}$ in the inhibition of $Fe^{2+}$-induced lipid peroxidation was 1,759 ${\mu}g$/assay(7.65 mg of dry sample). This extract also significantly inhibited the MDA-BSA conjugation reaction with an $IC_{50}$ of 57.58 mg/assay(250 mg of dry sample). However, no inhibitory effects against the direct and indirect mutagenicities in $Salmonella$ Typhimurium TA98 and TA100 were observed. Based on these results, the ethanol extract of $Arctium$ $lappa$ was shown to display considerable antioxidative activities.

  • The effects of aqueous extracts from Korean commercial teas on excessive free radicals were examined utilizing spin trapping, 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical and lipid peroxidation. A potent scavenging effect of green tea and oriental senna tea was dound using sipin trapping. The most effective teasagainst the DPPH radical was green tea, followed inorder by pine leak tea, Chinese gutta percha tea and orietnal senna tea. Similar to the effects of DPPH radical , green tea, pine leaf tea, Chinese gutta percha tea and oriential senna tea had an inhibitory effect on lipid peroxidation. These findings predict that Korean tea is a promising material for scavenging free radicals, and for curing diseases related to free-radical reactions.

Schisandra Chinensis Inhibits Oxidative DNA Damage and Lipid Peroxidation Via Antioxidant Activity

  • Schisandra chinensis have been traditionally used in Asia for the treatment of dyspnea, cough, mouth dryness, spontaneous diaphoresis, nocturnal diaphoresis, nocturnal emission, dysentery, insomnia and amnesia. The purpose of this study is to evaluate the protective effects of Schisandra chinensis on oxidative DNA damage and lipid peroxidation induced by ROS in non cellular and cellular system. DPPH radical, hydroxyl radical and hydrogen peroxide scavenging assay were used to measure the antioxidant activities. Phi X-174RF I plasmid DNA cleavage assay and intracellular DNA migration assay were used to evaluate the protective effect on oxidative DNA damage. MTT assay and lipid peroxidation assay were used for evaluating the protective effect on oxidative cell damage. It was found to scavenge DPPH radical, hydrogen peroxide and hydroxyl radical and it inhibited oxidative DNA damage, lipid peroxidation and cell death induced by hydroxyl radical. These data indicate that Schisandra chinensis possesses a spectrum of antioxidant and DNA-protective properties

Antioxidant Effect of Enzymatic Hydrolyzate from a Kelp, Ecklonia cava

  • The potential antioxidative activity of water-soluble enzymatic hydrolyzates from a kelp, Ecklonia cava was evaluated by free radical scavenging and lipid peroxidation assays. To prepare water-soluble hydrolyzates from E. cava the seaweed was enzymatically hydrolyzed by five carbohydrases (Viscozyme, Celluclast, AMG, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase). Among all the hydrolyzates, Celluclast hydrolyzate effectively scavenged free radicals released from DPPH (1,1-diphenyl-2- pricrylhydrazyl) and recorded around 73% scavenging activity at the concentration of 4 mg ${\cdot}ml^{-1}$. This hydrolyzate was thermally stable and DPPH radical scavenging activity remained 80% or higher at heating temperatures of 40 and 60$^{\circ}C$ up to 12 h and around 80% at 100$^{\circ}C$ up to 8 h. AMG and Ultraflo hydrolyzate inhibited the lipid peroxidation of fish oil as that of $\alpha$-tocopherol. These results suggested that an enzymatic extraction will be an effective way for the production of a potential antioxidant from seaweeds.

Antioxidant Effect of Flavonoids Isolated from the Root of Clematis trichotoma Nakai

  • The antioxidant effect of methanol extract (ME) and water extract (WE) from Clematis trichotoma was evaluated as primary study to scavenge stable 1,1-diphenyl-2-picrylhydrazyl radicals (DPPH), inhibited iron-induce lipid peroxidation in linoleic acid emulsion, peroxidation of liposome induced by $Fe^{3+}/H_2O_2/ascorbie$ acid, and on $Fe^{2+}/H_2O_2$ induced the mitochondrial lipid peroxidation. In secondary study, five flavonoids as luteolin (1), quercetin (2), apigenin (3), hirsutrin (4), kaempferol-3-O-glucoside were isolated (5). Among them, compounds 1 and 2 showed good activities in all the model systems. Compound 3 exhibited moderate antioxidant activities in both radical scavenging and these lipid peroxidation systems tested. Compound 4 showed significant inhibitions in liposome peroxidation and compound 5 displayed weak inhibition in all four tested systems. All the results presented herein indicate that products of C. trichotoma maybe useful in inhibiting membrane lipid peroxidation and preventing free radical-linked diseases.

쇠비름(Portulace oleracea) 추출물의 DPPH radical 소거능과 in vitro 지질과산화 억제 효과와 그 활성성분 (DPPH Radical Scavenging Effect and in vitro Lipid Peroxidation Inhibition by Portulaca oleracea)

  • 쇠비름 (Portulaca oleracea)의 항산화 효과를 탐색하기 위해 MeOH과 CH$_2$Cl$_2$를 이용해서 추출하여 crude extract를 조제하였다. 그리고 이 조추출물 (crude extract)에 대해 n-hexane, 15% aq. MeOH, EtOAc, n-BuOH, $H_2O$로 순차적으로 용매분획하였다. 쇠비름의 조추출물과 각 분획물들에 대해 DPPH radical 소거능과 마우스 간 균질물 (homogenate)을 이용한 과산화지질 억제능을 측정하였다. 그 결과 쇠비름 조추출물이 DPPH 라디칼에 대한 강한 소거작용과 in vitro 과산화지질 억제효과가 있음이 확인되었다. 조추출물의 DPPH radical 소거효과에 대한 $IC_{50}$/은 53.33 $\mu\textrm{g}$/$m\ell$로 천연 항산화제인 L-ascorbic acid (1.25 $\mu\textrm{g}$/$m\ell$)와 a-tocopherol (6.99 $\mu\textrm{g}$/$m\ell$)보다는 약하지만, 합성 항산화제로 널리 사용되고 있는 BHT(87.84 $\mu\textrm{g}$/$m\ell$)보다는 월등히 뛰어난 라디칼 소거능이 관찰되었다. 또한 각 분획물 중 EtOAc와 n-BuOH 분획물의 DPPH라디칼 소거효과가 가장 우수하였다. 마우스 간 균질물(mouse liver homogenate)을 이용한 과산화지질 저해효과에 대해서도 쇠비름 조추출물의 농도의존적인 (0.1 mg/$m\ell$~5 mg/$m\ell$) 상관성이 관찰되었으며, 특히 n-BuOH 분획물은 L-ascorbic acid의 효과와 거의 대등한 것을 나타났다. 이것으로 쇠비름 조추출물이 항산화효과 (in vitro)가 있음이 증명되었으며, 이 항산화활성은 극성이 비교적 큰 화합물들에 의한 것임을 추정할 수 있다. 현재 쇠비름 추출물로부터 항산화활성성분을 분리하기 위한 연구가 진행 중이다.

  • An antioxidative activity of Portulaca oleracea was tested by in vitro experimental models. The antioxidative activities were determined by evaluation the DPPH radical scavenging activity and by measuring lipid peroxide using 2-thiobarbituric acid (TBA). The crude extract was sequentially partitioned with n-hexane, 15% aq. MeOH, EtOAc, n-BuOH, $H_2O$. Among them, a remarkable antioxidative effect was observed in the fractions of EtOAc and n-BuOH. The DPPH radical scavenging effect $(IC_{50}=17.90{\mu}g/ml)$ of the n-BuOH soluble fraction was comparable with that of natural antioxidant, ${\alpha}-tocopherol(IC_{50}=\;6.99{\mu}g/ml)$ and the inhibitory effect of lipid peroxidation in mouse liver homogenate was similar to that of natural antioxidant, L-ascorbic acid at a concentration of 0.1mg/ml to 5mg/ml. From the BuOH soluble fraction yielded two biophenolic glycosides, 3-hydroxy-1-(2-hydroxyethyl)phenyl-4-O-${\beta}$-D-glucopyranoside(1) and 2-(3,4-dihydroxyphenyl)ethyl-O-${\beta}$-D-glucopyranoside(2) using column chromatography and revered-phase HPLC. In particular, the DPPH radical scavenging activity of 2 was comparable to that tocopherol$(IC_{50}=6.59{\mu}g/ml)$.
