Improvement of Fibrinolytic Activity of Bacillus subtilis 168 by Integration of a Fibrinolytic Gene into the Chromosome

  • Fibrinolytic enzyme genes (aprE2, aprE176, and aprE179) were introduced into the Bacillus subtilis 168 chromosome without any antibiotic resistance gene. An integration vector, pDG1662, was used to deliver the genes into the amyE site of B. subtilis 168. Integrants, SJ3-5nc, SJ176nc, and SJ179nc, were obtained after two successive homologous recombinations. The integration of each fibrinolytic gene into the middle of the amyE site was confirmed by phenotypes (Amy-, SpecS) and colony PCR results for these strains. The fibrinolytic activities of the integrants were higher than that of B. subtilis 168 by at least 3.2-fold when grown in LB broth. Cheonggukjang was prepared by inoculating each of B. subtilis 168, SJ3-5nc, SJ176nc, and SJ179nc, and the fibrinolytic activity of cheonggukjang was 4.6 ± 0.7, 10.8 ± 0.9, 7.0 ± 0.6, and 8.0 ± 0.2 (U/g of cheonggukjang), respectively at 72 h. These results showed that construction of B. subtilis strains with enhanced fibrinolytic activities is possible by integration of a strong fibrinolytic gene via a marker-free manner.

  • KARI precision attitude determination system has been developed for high accurate geo-coding of KOMPSAT-2 image. Sensor data from two star trackers and a IRU are used as measurement and dynamic data. Sensor data from star tracker are composed of QUEST and unit vector filter. Filter algorithms consists of extended Kalman filter, unscented Kalman filter, and least square batch filter. The type of sensor data and filter algorithm can be chosen by user options. Estimated parameters are Euler angle from 12000 frame to optical bench frame, gyro drift rate bias, gyro scale factor, misalignment angle of star tracker coordinate frame with respect to optical bench frame, and misalignment angle of gyro coordinate frame with respect to optical bench frame. In particular, ground control point data can be applied for estimating misalignment angle of star tracker coordinate frame. Through the simulation, KPADS is able to satisfy the KOMPSAT-2 mission requirement in which geo-location accuracy of image is 80 m (CE90) without ground control point.

Comparison of Methods for Stable Simultaneous Expression of Various Heterologous Genes in Saccharomyces cerevisiae (출아효모에서 다양한 이종 유전자의 안정적 동시발현을 위한 방법의 비교)

  • We compared two integration systems for stable expression of heterologous genes in Saccharomyces cerevisiae. A Candida glabrata-derived gene was used as the selective marker for the Cre/loxP system, and XYLP, XYLB, GRE3, and XYL2 genes were used as model heterologous genes and ligated into the universal pRS-CMT vector. The resulting pRS-XylP, pRS-XylB, pRS-Gre3, and pRS-Xyl2 plasmids were sequentially integrated into yeast chromosome VII by four integration processes (marker rescue and gene integration). The four introduced genes were successfully expressed. Further, the pRS-PBG2 plasmid harboring expression cassettes for the four genes was constructed for one-step integration. The four genes that were introduced were stably maintained as a gene cluster and were simultaneously expressed. The one-step integration was more effective for the simultaneous integration and expression of the four genes related to xylan/xylose metabolism. This method will enable the generation of a useful biosystem through appropriate use of gene integration methods.

  • Due to the broad host suitability of viral vectors and their high gene delivery capacity, many researchers are focusing on viral vector-mediated gene therapy. Among the retroviruses, foamy viruses have been considered potential gene therapy vectors because of their non-pathogenicity. To date, the prototype foamy virus is the only retrovirus that has a high-resolution structure of intasomes, nucleoprotein complexes formed by integrase, and viral DNA. The integration of viral DNA into the host chromosome is an essential step for viral vector development. This process is mediated by virally encoded integrase, which catalyzes unique chemical reactions. Additionally, recent studies on foamy virus integrase elucidated the catalytic functions of its three distinct domains and their effect on viral pathogenicity. This review focuses on recent advancements in biochemical, structural, and functional studies of foamy virus integrase for gene therapy vector research.

Highly accurate family of time integration method

  • In this study, the acceleration vector in each time step is assumed to be a mth order time polynomial. By using the initial conditions, satisfying the equation of motion at both ends of the time step and minimizing the square of the residual vector, the m+3 unknown coefficients are determined. The order of accuracy for this approach is m+1, and it has a very low dispersion error. Moreover, the period error of the new technique is almost zero, and it is considerably smaller than the members of the Newmark method. The proposed scheme has an appropriate domain of stability, which is greater than that of the central difference and linear acceleration techniques. The numerical tests highlight the improved performance of the new algorithm over the fourth-order Runge-Kutta, central difference, linear and average acceleration methods.

  • Mushroom transformation requires a series of experimental steps, including generation of host strains with a desirable selective marker, design of vector DNA, removal of host cell wall, introduction of foreign DNA across the cell membrane, and integration into host genomic DNA or maintenance of an autonomous vector DNA inside the host cell. This review introduces limitations and obstacles related to transformation technologies along with possible solutions. Current methods for cell wall removal and cell membrane permeabilization are summarized together with details of two popular technologies, Agrobacterium tumefaciens-mediated transformation and restriction enzyme-mediated integration.

  • In the automobile industry, the design of optimal blank shape becomes a significant part of the stamping. It provides many evident advantages, sush as enhancement of formability, reduction of material cost and product development period. However, the nesting process, required for the optimal usage of materials in the blanking becomes more complicated as the blank shape becomes complicated, like most optimal blank shape. In this study, stamping process optimization system for the optimal usage of material has been developed through the integration of optimal blank design and optimal nesting. For optimal blank design, a radius vector method, the modified version of the initial nodal velocity method, the past work of the present author, have been proposed. Both the optimal blank design and optimal nesting programs have been developed under the GUI environment for the convenience of engineers. The efficiency of the optimization system has been verified with some chosen problems.

  • Flow visualization is one of visualization techniques and it means a visual expression of vector data using 2D or 3D graphics. It aims for human to easily understand a special feature of the vector data. Flow visualization can be classified into various criterions such as visualization technique, data dimension, type of the flow, and so on. Visualization technique can be categorized into direct method, integration method and derived data based method. Data dimension can be divided into 2D, 2.5D and 3D. Type of flow data may be classified into steady and unsteady. In this paper, various LIC based flow visualization methods will be introduced which is one of representative integration based techniques. Those methods will be categorized with more detailed criterions such as dimension and type of flows.

GPS/INS Integration using Vector Delay Lock Loop Processing Technique

  • Conventional DLLs estimate the delay times of satellite signals individually and feed back these measurements to the VCO independently. But VDLL estimates delay times and user position directly and then estimate the feedback term for VCO using the estimated position changes. In this process, input measurements are treated as vectors and these vectors are used for navigation. First advantage of VDLL is that noise is reduced in all of the tracking channels making them less likely to enter the nonlinear region and fall below threshold. Second is that VDLL can operate successfully when the conventional independent parallel DLL approach fails completely. It means that VDLL receiver can get enough total signal power to track successfully to obtain accurate position estimates under the same conditions where the signal strength from each individual satellite is so low or week that none of the individual scalar DLL can remain in lock when operating independently. To operate VDLL successfully, it needs to know the initial user dynamics and position and prevents total system from the divergence. The suggested integration method is to use the inertial navigation system to provide initial dynamics for VDLL and to maintain total system stable. We designed the GPS/INS integrated navigation system. This new type of integrated system contained the vector pseudorange format generation block, VDLL signal processing block, position estimation block and the conversion block from position change to delay time feedback term aided by INS.
