Grapefruit 종자추출물을 이용한 Aspergillus parasiticus의 생육 및 Aflatoxin 생성억제 효과 (Inhibitory Effects of Grapefuit Seed Extract on Growth and Aflatoxin Production of Aspergillus parasiticus)

  • 농산물 및 그 가공식품을 수확·저장·가공하는 기간중에 Aspergillus parasiticus가 오염되어 aflatoxin을 생성하는 것을 방지할 목적으로 생육배지에 graefruit 종자추출물(GFSE)을 처리하여 곰팡이의 생육 및 aflatoxin 생합성을 저해하는 뚜렷한 효과를 관찰할 수 있었다. 4000 ppm의 GFSE농도 처리로 Aspergillus parasiticus의 생육을 92% 저해하였으며, 5000ppm 농도에서는 곰팡이 생육을 완전히 저해하였고 3000ppm 이상의 농도에서 aflatoxin 의생성을 100% 억제하였다. 한편, GFSE 처리에 의하여 aflatoxin 생합성 경로증, averufin, versiconal acetate 등의 중간대사산물이 aflatoxin으로 주입되는 것이 완전히 저해된 반면, versicolorin A, versicolorin A hemiacetal, sterigmatocystin 등은 aflatoxin 으로 전환되는 결과를 보여주어 GFSE는 versiconal acetate를 포함한 그 이전의 중간 대사물이 반응기질이 되는 효소 반응계를 저해하는 것으로 나타났다. 전자현미경을 이용한 곰팡이균체 및 포자의 형태변화는 GFSE처리에 의하여 세포막기능에 파괴되어 세포구성 내용물이 유실되고 곰팡이가 사멸하고 aflatoxin 생합성 기작이 중단되는 결과를 초래하였다.

파종시기 및 파종량이 헤어리 벳치의 수량 및 사료가치에 미치는 영향 (Effect of Seeding Dates and Rates on the Productivity and Nutritive value of Hairy Vetch(Vicia villosa Roth))

  • This experiment was conducted to investigate the effect of seeding dates and rates on the productivity and nutritive value of hairy vetch(Vicia villosa Roth) at experimental field of Grassland and Forage Crops Division, National Livestock Research Institute, RDA, Suwon from 1999 to 2000. The experimental design was consisted of split-plot design with three replications. The main plot was seeding dates(25 Aug., 15 Sep. and 5 Oct.) and the subplot was seeding rates(20, 30 and 40 kg/ha). Late seeding dates increased the plant height and delayed the flowering stage as 1 or 2 days. The dry matter(DM) content was decreased with low seeding rates, and the trend was same in the all plots. Average crude protein(CP) content was $24.1\%$ and increased with delayed seeding date and lower seeding rate. The content of acid detergent fiber(ADF) and neutral detergent fiber(NDF) was increased with higher seeding rate, but seeding date did not affect. In vitro dry matter digestibility(IVDMD) of hairy vetch tended to decrease with high seeding rate, and total digestible nutrient(TND) did not show significant difference among seeding rates. Average relative feed value(RFV) of hairy vetch showed 140 which means very high quality. Dry matter and CP yield was decreased with delayed seeding date. The results of this experiments indicated that seeding in middle-August with 40 kg/ha would be recommended to produce the highest yield and quality f3r the cultivation of hairy vetch in middle part of Korea

Effects of Trichostatin A on In Vitro Development of Porcine Parthenogenetic and Nuclear Transfer Embryos

  • Developmental potential of cloned embryos is related closely to epigenetic modification of somatic cell genome. The present study was to investigate the effects of applying histone deacetylation inhibitor, trichostatin A (TSA) to activated porcine embryos on subsequent development of porcine parthenogenetic and nuclear transfer embryos. Electrically activated oocytes were treated with 5 nM TSA for different exposure times (0, 1, 2 and 4 hr) and then the activated embryos were cultured for 7 days. The reconstructed embryos were treated with different concentrations of 0, 5, 10 and 25 nM TSA for 1 hr. Also 5 nM TSA was tested with different exposure times of 0, 0.5, 1, 2 and 4 hr. And fetal fibroblast cells were treated with 50 nM TSA for 1, 2 or 4 hr and with 5 nM TSA for 1 hr. Cumulus-free oocytes were enucleated and reconstructed by TSA-treated donor cells and electrically fused and cultured for 6 days. In parthenogenetic activation experiments, 5 nM TSA treatment for 1 hr significantly improved the percentage of blastocyst developmental rates than the other groups. Total cell number of blastocysts in 1 hr group was significantly higher than other groups or control. Similarly, blastocyst developmental rates of porcine NT embryos following 5 nM TSA treatment for 1 hr were highest. And the reconstructed embryos from donor cells treated by 50 nM TSA for 1 hr improved the percentage of blastocyst developmental rates than the control group. In conclusion, TSA treatment could improve the subsequent blastocyst development of porcine parthenogenetic and nuclear transfer embryos.

안개초의 잎 절편체를 이용한 기내재분화 (In Vitro Regeneration Using Leaf Segment in Gypsophila paniculata L. 'Bristol Fairy')

  • 안개초 잎조직의 재분화는 TDZ가 BA와 kinetin에 비하여 월등히 효과적이었고 TDZ $3.0mg{\cdot}L^{-1}$에서 가장 높은 재분화율을 보였다. 한편 BA처리에서는 투명묘 발생율이 높았고 kinetin은 재분화에 영향을 주지 못하였다. NAA와 IAA의 비교에서 IAA $0.3mg{\cdot}L^{-1}$를 첨가한 배지에서 재분화율과 잎당 부정아수가 많았다. Agar 농도에 따른 잎조직의 재분화는 1.0, 1.2 및 1.6%에서 야기되었다. 배양초기 10일간의 암처리는 재분화율을 75%까지 증가시켰다. 생장점배양 후 3회 계대배양한 묘에서 채취한 잎 절편에서 재분화율이 84%로 가장 높았고 잎당 부정아수도 6.8개로 가장 많았다.

Evaluation of Herbicidal Potential of Essential Oils and their Components under In vitro and Greenhouse Experiments

  • The present study aimed to evaluate the phytotoxic potential of essential oils. For this purpose, 18 essential oil samples extracted from Korean plants and 64 commercial essential oils were screened for their phytotoxic potential against the seedling growth of Brassica napus L. (rapeseed). Among the 82 samples, 11 commercial oils (cinnamon, citronella, clove, cumin seed, geranium, jasmine, lemongrass, palmarosa, pimento, rose otto and spearmint) strongly inhibited the seedling growth with $GR_{50}$ value < $150{\mu}g\;mL^{-1}$. Major components from these effective essential oils were identified by solid phase microextraction/gas chromatography-mass spectrometry (SPME/GC-MS). GC-MS analyses revealed that the effective samples mainly consist of benzyl benzoate, carvone, citral, citronellol, eugenol, geraniol, D-limonene and terpinene. Subsequently, bioactivity of these individual components was evaluated against the seedling growth of B. napus, Echinochloa crus-galli and Aeschynomene indica. The components from different chemical groups exhibited different potency in inhibiting the seedling growth with varied $GR_{50}$ values ranged from $29{\mu}g\;mL^{-1}$ to > $1000{\mu}g\;mL^{-1}$. In the greenhouse experiment, citral and geraniol completely suppressed the growth of all the tested 10 plants at $100kg\;ha^{-1}$. In conclusion, the individual essential oil components geraniol and citral could be used as natural herbicides for weed management.

새로운 항궤양제(IY-80843과 IY-80845)의 기니픽 위점막세포 $H_2$-수용체에 대한 결합반응연구 (Study on the Binding of New Anti-ulcer Agent(IY-80843 and IY-80845) to Histamine $H_2$-Receptor in Isolated Guinea Pig Gastric Glands)

  • Receptor binding study was carried out as an in vitro assay to test the anti-ulcer effect for newly synthesized test compounds(IY-80843 and IY-80845) which were reported to have a strong anti-secretory effect in Shay-ligated rats. The histamine H$_{2}$-receptor fraction was prepared from the membranes of the isolated gastric glands in guinea pigs and $^{3}$H-cimetidine was used as a radioligand. The binding of $^{3}$H-cimetidine to the membranes of the isolated gastric glands was found to be time dependent, saturable and confined to a single population of binding sites with $K_{D}$ value of 0.13$\pm$0.03 $\mu{M}$ and B$_{max}$ value of 52.5$\pm$1.5 pmol/mg. From the competition experiments, both IY-80843 and IY-80845 were shown to have a strong blocking effect against binding of $^{3}$H-cimetidine to the histamine H$_{2}$-receptor. The IC$_{50}$, Ki, and Hill coefficient(nH) values for IY-80843 were 0.18$\pm$0.02 $\mu{M}$, 0.16$\pm$0.02 $\mu{M}$, and 0.97$\pm$0.15, respectively and those values for IY-80845 were 0.27$\pm$0.02 $\mu{M}$, 0.24$\pm$0.02 $\mu{M}$, and 0.82$\pm$0.13, respectively. The results demonstrated that the blocking effects of IY-80843 and IY-80845 were 7 and 5 times stronger than that of cimetidine, respectively. Therefore, the newly synthesized compounds, IY-80843 and IY-80845, appeared to be the highly potent competitive inhibitors of histamine on the H$_{2}$-receptor.

  • Nowadays, implantable hearing aids have been developed to solve the problems of conventional hearing aids. In case of fully implantable hearing aids, an implantable microphone is necessary to receive sound signal beneath the skin. Normally, an implantable microphone has poor frequency response characteristics in high frequency bands of acoustic signal due to the high frequency attenuation effect of skin after implantation to human body. In this paper, the implantable microphone is designed to reduce the high frequency attenuation effect of a skin by putting its resonance frequency at the attenuated range through a finite element analysis (FEA) simulation. The designed implantable microphone through the simulated results has been fabricated by manufacturing process using bio-compatible materials. By the several in-vitro experiments with pig skin, it has been verified that the designed implantable microphone has a resonance frequency around the starting part of the attenuated range and reduces the attenuation effect.

자생 식물로 부터 Cyclin-dependent Kinase 4/Cyclin D1 저해물질의 탐색 (Searching of Cyclin-Dependent Kinase 4/Cyclin D1 Enzyme Inhibition Materials from the Native Plants)

  • 인체 세포의 세포주기를 조절하는 Cyclin-dependent Kinase(CDK) 중 CDK4/Cyclin D1 효소 저해 물질을 찾기 위하여 국내에 자생하는 37과 87종의 식물 추출물을 탐색한 결과, 50%$(5\;{\mu}g/ml)$ 이상의 CDK4/Cyclin D1 호소 저해활성을 보인 것은 단삼(Salvia miltiorrhiza), 노박덩굴(Celastrus orbiculatus), 삼백초(Saurus chinensis), 목단(Paeonia suffruticosa) 및 오이풀(Sanguisorba officinalis)이었다. 이들 중에서 목단과 오이풀 추출물이 가장 높은 56%의 저해율을 각각 나타내었다. 또한 추출물에서 활성물질을 추출 정제하여 2차원 NMR 등을 포함한 각종 기기분석을 통하여 cryptotanshinone으로 동정하였다.

  • 히말라야산의 동토에서 발견된 Preussia sp.로부터 신규 벤조산 유도체 benzyl 2,4-di(benzyloxy)benzoate (1)와 6개의 알려진 화합물들(2-7)이 분리되었다. 신규물질 및 다른 화합물들의 구조는 1D-와 2D NMR실험 및 문헌값과의 비교에 의하여 결정되었다. 화합물 2-7은 동 속 및 Sporormiaceae과에서 처음으로 얻어졌다. 분리된 물질들은 쥐의 대식세포 RAW 264.7 cells에서 lipopolysaccharide (LPS) 처리에 의하여 유도된 nitric oxide (NO)의 생성량 억제를 측정하였다. 화합물 1번과 2번은 100 mg/ml의 농도로 처리했을 때 LPS에 의하여 유도된 NO 생성량을 각각 50.7% 및 88.5%까지 억제하는 것을 보였다.

한지형 잔디의 종자발아에 미치는 Ethephon,ABA,BA,NAA 침지처리 효과 (The Effect of Ethephon, ABA, BA and NAA Soaking Treatment of Seed Germination of Cool Season Turfgrass -es)

  • The study is aimed to understand effects of ethephon(lmg/1), ABA(0.lrng/l), BA(0.lmg/l). and NAA (0.lmg /1) soaking treatment on the seed germination of perennial ryegrass 'Dandy', creeping bentgrass 'Penlinks', tall fescue 'Arid' and kentucky bluegrass 'Nasaw'. These experiments are carried in vitro and their results are summarized as the followings ; In case of ethephon (lmg /1) soaking treatment of seed, the early germination of tall fescue 'Arid' and perennial ryegrass 'Dandy' was good without the soaking times. creeping bentgrass 'Penlinks' and kentucky bluegrass 'Nasaw' obtained the best results at 12 hr. soking treatment, and the longer the soaking time, the higher the germination percentage. The early germination percentage of tall fescue 'Arid' and perennial ryegrass 'Dandy' increased more highly at ABA(0. tmg /1) soaking treatment than at the control. The germination percent-age of creeping loentgrass 'Penlink' and kentucky bluegrass 'Nasaw' had respectively the best results at 6hr. and l2hr. soaking treatment. The early germination percentage of tall fescue 'Arid' and kentucky bluegrass 'Nasaw'increased more highly at BA (0.lmg /1) soaking treatment than at the control, and especially had the best results in 6hr. soaking treatment. perennial ryegrass 'Dandy' had the best results at l0min. soaking treatment, and the longer the soaking time, the lesser the germination percentage. The early germination of perennial ryegrass 'Dandy'. creeping bentgrass 'Penlinks' and tall fes- cue 'Arid' increased more highly at NAA (0.lmg /1) soaking treatment than at the control, and especially had the best results at l2hr. soaking treatment. However, the germination of Kentucky bluegrass 'Nasaw' seeds inhibited at NAA (0.lmg /1) soaking treatment. Key words: Ethephon, ABA, BA, NAA, Soaking treatment, Seed germination.
